다양한 PCR용 DNA 추출법에 의한 패류 내 Megalocytivirus의 검출 (Detection of Megalocytivirus in shellfish using PCR with various DNA extraction methods)

  • 패류 내에 오염되어 있는 바이러스의 검출에 있어서 정성 정량적 분석이 기능하며 신속하고 간편한 방법의 개발을 이루고자 하였다. 5g의 굴 중장선 조직을 Glycine buffer 및 PEG 8000 용액을 사용하여 제조한 농축 시료 (T5g-D)와 50mg의 굴 중장선 조직으로부터 직접적으로 제조한 시료(sT5Omg-D)를 대상으로 2-step PCR을 실시하여 검출감도를 비교하였다. 동일한 1 ${\mu}l$의 DNA template T5g-D와 sT50mg-D를 사용하였을 때, 35cycles의 1-step PCR에서 양성의 결과를 얻을 수 있었다. 인위적으로 sT50mg-D 혼합 시료를 사용하여 2-step PCR (35cycles)을 실시하였을 때 T5g-D를 사용한 것과 비교하여 뚜렷한 차이를 나타내지 않았다. 또한 megalocytivirus에 오염된 양성시료 0.5~50mg을 사용하여 조제한 각각 $50{\mu}l$의 template DNA 1 ${\mu}l$를 사용하여 qPCR을 하였을 때 6.14E+00~1.2E+02/${\mu}l$의 농도를 함유하고 있는 것으로 나타났다. 조제한 template DNA에 6.14E+00 copies/${\mu}l$ 이하의 농도가 있을 때는 qPCR에서 positive의 결과를 얻을 수 없었다. 결론적으로 패류 내 mega1ocytivirus의 오염 유무 판단을 위한 2-step PCR 및 qPCR에서 50mg의 중장선을 사용하는 것은 i) 굴의 조직으로부터 오염된 megalocytivirus의 정성 및 정량을 위한 최소 검출 한계에 벼하여 충분한 양이었으며 ii) 개체별 분석이 가능하다는 장점이 있었으며 iii) 이를 토대로 국내에서 서식하고 있는 굴에서 megalocytivirus의 오염을 확인할 수 있었다.

산초나무 잎녹병균의 중간기주 및 형태학적 특징 (Morphological Features of Coleosporium xanthoxyli and Its Alternate Host in Korea)

  • 경상남도 진주와 하동지역의 산초나무 재배단지에서 잎녹병에 의한 피해가 매년 지속적으로 발생하고 있다. 산초나무 잎에 형성된 여름포자세대의 녹포자세대 기주를 밝히고 녹병균을 동정하기 위하여 본 병해의 발생지 주변 곰솔에서 채집된 녹포자를 산초나무에 인공접종하였다. 그 결과 산초나무에서 성공적으로 여름포자가 형성되어 이 녹병균은 녹포자세대를 곰솔에서, 여름포자 및 겨울포자세대를 산초나무에서 형성하는 장세대형 생활사를 갖는 C. xanthoxyli로 동정하였다. 대부분의 형태적 형질은 기존 기록과 일치하였으나 녹포자와 여름포자의 표면돌기 수에는 차이가 있었다. 곰솔과 산초나무에서 채집된 C. xanthoxyli의 건조표본을 이용하여 녹포자세대 및 여름포자세대의 형태학적 특징을 기술하였다. 경남 진주와 하동지역에서 조사된 산초나무에서의 최초 병발생시기는 6월 하순${\sim}$7월 초순이었으며 8월 중의 이병엽율은 17.8%${\sim}$58.7%이었다.

장래교통수요예측을 고려한 도로 유지관리 방안 (Road Maintenance Planning with Traffic Demand Forecasting)

  • PURPOSES : This study aims to examine the differences between the existing traffic demand forecasting method and the traffic demand forecasting method considering future regional development plans and new road construction and expansion plans using a four-step traffic demand forecast for a more objective and sophisticated national highway maintenance. This study ultimately aims to present future pavement deterioration and budget forecasting planning based on the examination. METHODS : This study used the latest data offered by the Korea Transport Data Base (KTDB) as the basic data for demand forecast. The analysis scope was set using the Daejeon Metropolitan City's O/D and network data. This study used a traffic demand program called TransCad, and performed a traffic assignment by vehicle type through the application of a user equilibrium-based multi-class assignment technique. This study forecasted future traffic demand by verifying whether or not a realistic traffic pattern was expressed similarly by undertaking a calibration process. This study performed a life cycle cost analysis based on traffic using the forecasted future demand or existing past pattern, or by assuming the constant traffic demand. The maintenance criteria were decided according to equivalent single axle loads (ESAL). The maintenance period in the concerned section was calculated in this study. This study also computed the maintenance costs using a construction method by applying the maintenance criteria considering the ESAL. The road user costs were calculated by using the user cost calculation logic applied to the Korean Pavement Management System, which is the existing study outcome. RESULTS : This study ascertained that the increase and decrease of traffic occurred in the concerned section according to the future development plans. Furthermore, there were differences from demand forecasting that did not consider the development plans. Realistic and accurate demand forecasting supported an optimized decision making that efficiently assigns maintenance costs, and can be used as very important basic information for maintenance decision making. CONCLUSIONS : Therefore, decision making for a more efficient and sophisticated road management than the method assuming future traffic can be expected to be the same as the existing pattern or steady traffic demand. The reflection of a reliable forecasting of the future traffic demand to life cycle cost analysis (LCCA) can be a very vital factor because many studies are generally performed without considering the future traffic demand or with an analysis through setting a scenario upon LCCA within a pavement management system.

편향하중 조건 보행시 인체의 적응 작용에 대한 분석 (Joint moments and muscle forces during walking with sided load as one of activities of daily living)

  • The trunk is inclined to the loaded side when carrying an object as one of activities of daily living. As the reaction to this behavior the human body may be inclined to his/her trunk to unloaded side. The present study investigated the biomechanical effects of weight variation for sided load carriage during walking upon joint moments and muscle torques, through the tracker agent and joint driving dynamic analysis. To perform the experiment one male was selected as subject for the study. Gait analysis was performed by using a 3D motion analysis system. Thirty nine 14mm reflective markers, according to the plug-in marker set, were attached to the subject. We used BRG.LifeMOD(Biomechanics Research Group, Inc., USA), for skeletal modeling and inverse and joint driving dynamic simulation during one gait cycle. In walking with a sided load carriage, the subject modeled held the carriage with the right hand, which weighed 0, 5, 10, 15kg, 20kg respectively. The result of this simulation showed that knee and hip in the coronal plane were inclined to the loaded side and loaded side had larger moments as the sided load carriage was increased. On the other hand thoracic and lumbar in the coronal plane had larger negative values as the sided loaded carriage was increased. The thoracic and lumbar in the transverse plane also had larger values as the sided load was increased. And the several muscles of loaded side were increased as increasing sided load. It could be concluded that human body is adopted to side loaded circumstances by showing more biologic force. These results could be very useful in analysis for delivery motion of daily life.

Root Bark of Morus alba L. and Its Bioactive Ingredient, Ursolic Acid, Suppress the Proliferation of Multiple Myeloma Cells by Inhibiting Wnt/β-Catenin Pathway

  • The root bark of Morus alba L. has cytotoxic activity against several types of cancer cells. However, little is known about its chemopreventive mechanisms and bioactive metabolites. In this study, we showed that M. alba L. root bark extracts (MRBE) suppressed β-catenin response transcription (CRT), which is aberrantly activated in various cancers, by promoting the degradation of β-catenin. In addition, MRBE repressed the expression of the β-catenin/T-cell factor (TCF)-dependent genes, c-myc and cyclin D1, thus inhibiting the proliferation of RPMI-8226 multiple myeloma (MM) cells. MRBE induced apoptosis in MM cells, as evidenced by the increase in the population of annexin VFITC-positive cells and caspase-3/7 activity. We identified ursolic acid in MRBE through LC/mass spectrum (MS) and observed that it also decreased intracellular β-catenin, c-myc, and cyclin D1 levels. Furthermore, it suppressed the proliferation of RPMI-8226 cells by stimulating cell cycle arrest and apoptosis. These findings suggest that MRBE and its active ingredient, ursolic acid, exert antiproliferative activity by promoting the degradation of β-catenin and may have significant chemopreventive potential against MM.

  • 자나방과(Geometridae), 가지나방아과(Ennominae)에 속하는 줄마디가지나방(Chiasmia cinerearia)은 중부지방의 회화나무 가로수 및 조경수를 가해하는 대표적인 수목 해충이다. 본 연구의 결과 줄마디가지나방은 주로 잎 위에 250~300개의 알을 무더기로 산란하였으며, 유충은 총 5령기를 거쳐 용화되었다. 성충은 5월 초순과 7월 말경에 2회 우화하였으며, 1세대 경과일수는 약 48일로 조사되었다. 유충의 몸길이는 25~30 mm 정도이며 연녹색을 띠고 복부의 옆선은 황색이었다. 성충의 날개편 길이는 30~35 mm 정도이며 회갈색을 띠며 앞날개 외횡선을 중심으로 4각형의 무늬가 무리지어 있는 것이 특징이다.

  • 미군의 아키텍처프레임워크는 3원화관점에 의해서 산출물을 정의하였으나, 관점간의 전환과정이 불명확하고, 개발절차가 미흡하여, 아키텍처개발자가 아키텍처를 개발하기 어렵다. 그리고, 개발하더라도 독립적인 방법으로 산출물을 작성하여, 개발된 산출물의 수준이 상이하고 일관성이 없는 경우가 많다. 따라서, 본 논문에서는 우리 군 실정에 맞추어 산출물간의 연계성과 일관성을 유지시키기 위한 신규 산출물을 추가로 정의하였고, 아키텍처 개발절차를 보다 상세히 제시하였으며, 정보체계 수명주기별로 아키텍처 활용 용도를 제공하는 국방아키텍처프레임워크(MND AF)를 개발하였다. 이를 통하여 아키텍처산출물의 일관성과 추적성을 제공할 수 있게 되고, 체계통합 및 상호운용성 기반을 구축하게 되었으며, 아키텍처 개발자가 좀 더 쉽게 아키텍처를 개발할 수 있도록 하였다.

  • The BESS(Battery Energy Storage System) is an useful device for load leveling, but the high cost, installation space and safety issues are the main barriers for supplying it widely. The important factor in supplying BESS to customers successfully is the payback period. As most of the H/W cost factors are uncontrollable, the optimization of storage volume can be useful factor in improving payback period. In order to obtain optimized BESS volume, the load factor, demand ratio, peak shaving ratio, electric rates and benefits from DR participation of customer should be analyzed. In this paper, we could verify the peak cutting capability and cost effectiveness under the some proposed conditions and changing value of PCS and battery based on the customers data after volume optimization process was applied, and we can identified the saturation point of load factor and shortening of customer's payback period.

  • 미 국방성은 플랫폼중심전의 상향식소요제기에서 네트워크중심전의 하향식소요제기로 변화하고 능력기반전력구조개발에 힘쓰고 있으며, 한국군도 다변화하는 안보위협과 전시작전권전환을 앞두고 능력기반전력구조로의 전환을 시도하고 있으나 통합능력의 개념과 개발방법이 부족한 상태이다. 따라서 이 연구는 능력기반전력구조 개발을 위해 상 하위 조직 전체의 능력과 현재의 전력운용관점과 미래의 전력개발관점을 동시에 포함하는 통합능력프레임워크를 제시한다.

  • Plastics that we use and simply throw away have a life span of about 500 years and barely decompose. The practice of producing and using common plastics needs to be challenged. Until now, they have been useful in the industrial structure of mass production, but it can be said that there is a lack of research into new materials to introduce and apply in terms of material recycling. As a result of this, we have come to the uncomfortable realization of the fact that we cannot incinerate or reuse these precious resources indiscriminately. No matter how well-designed a product is, it has a competitive advantage if production and consumption activities, waste, collection, sorting and treatment are considered in terms of a continuous cycle, and in this respect, Extended Producer Responsibility (EPR) can help. We are implementing the EPR system, and active industrialization in the field of recycling is required, which is also a challenge for producers to participate actively in recycling and seek to save and recycle resources in design and manufacturing. Against this backdrop, We would like to examine the possibilities, through various studies and developments on product design of recyclable materials, which is being conducted mainly in Europe. In particular, we would like to examine the methods, and value of solving environmental problems and the active efforts to achieve this in the design world, and in particular the case of product design using recycled plastics.
