Transcriptional Regulation of the Methuselah Gene by Dorsal Protein in Drosophila melanogaster

  • The Drosophila methuselah (mth) mutant has an approximately 35 percent increase in average lifespan, and enhanced resistance to various forms of stress, including starvation, high temperature, and dietary paraquat. To examine the transcriptional regulation of mth, we used luciferase assays employing Drosophila S2 cells. Two positive control elements were found at -542 ~ -272 (PE1) and +28 ~ +217 (PE2), where putative binding sites for transcription factors including Dorsal (Dl) were identified. Cotransfection of a Dl expression plasmid with a mth-luciferase reporter plasmid resulted in decreased reporter activity. PE1 and PE2, the minimal elements for strong promoter activity, were required for maximal repression by Dl protein. The N-terminal Rel homology domain (RHD) of Dl was not sufficient for repression of mth. We demonstrated by chromatin affinity precipitation (ChAP) assays in S2 cells that Dl bound to the putative PE1 binding site. Unexpectedly, semi-quantitative RT-PCR analysis revealed that the level of mth transcripts was reduced in dl flies. However, the in vivo result support the view that mth expression is regulated by dl, since it is well known that Dl functions as both a transcriptional activator and repressor depending on what other transcription factors are present. These findings suggest that both innate immunity and resistance to stress are controlled by Dl protein.

Isolation and Characterization of an Eosinophilic GH 16 β-Agarase (AgaDL6) from an Agar-Degrading Marine Bacterium Flammeovirga sp. HQM9

  • A special eosinophilic agarase exo-type ${\beta}$-agarase gene, AgaDL6, was cloned from a marine agar-degrading bacterium, Flammeovirga sp. HQM9. The gene comprised 1,383-bp nucleotides encoding a putative agarase AgaDL6 of 461 amino acids with a calculated molecular mass of 52.8 kDa. Sequence analysis revealed a ${\beta}$-agarase domain that belongs to the glycoside hydrolase family (GH) 16 and a carbohydrate-binding module (CBM_4_9) unique to agarases. AgaDL6 was heterologously expressed in Escherichia coli BL21 (DE3). Enzyme activity analysis of the purified protein showed that the optimal temperature and pH of AgaDL6 were $50^{\circ}C$ and 3.0, respectively. AgaDL6 showed thermal stability by retaining more than 98% of activity after incubation for 2 h at $50^{\circ}C$, a feature quite different from other agarases. AgaDL6 also exhibited outstanding acid stability, retaining 100% of activity after incubation for 24 h at pH 2.0 to 5.0, a property distinct from other agarases. This is the first agarase characterized to have such high acid stability. In addition, we observed no obvious stimulation or inhibition of AgaDL6 in the presence of various metal ions and denaturants. AgaDL6 is an exo-type ${\beta}$-1,4 agarase that cleaved agarose into neoagarotetraose and neoagarohexaose as the final products. These characteristics make AgaDL6 a potentially valuable enzyme in the cosmetic, food, and pharmaceutical industries.

The Study of Associations among Serum Lipids, Anthropometric Measurements, Food Intake Frequency and Nutrient Intake in Healthy Adults (중.노년층이 섭취한 식품 종류와 영양소섭취상태, 신체지수 및 혈중지질과의 관계)

  • The purpose of this study was to investigate the association among food intake frequency, nutrient intake, anthropometric measurements, and serum lipids in 100 women and 85 men aged 40-74yrs residing in Jeonju City. Food intake frequency and nutrient intake were obtained from one-day dietary recalls. The results were as follows : The mean height, body weight and BMI were 161 cm, 62.5 kg, and 24 kg/$m^2$respectively. The mean of SBP and DBP were 118 mmHg, 77 mmHg respectively. The mean of TC and TG were 185.5 mg/dl, 153 mg/dl respectively. The mean daily energy and carbohydrate intake was 85. 7%, 91% of RDA for Koreans respectively and that protein intake was 118.5% of RDA. The TC levels of subjects less than 75%, 75-125%, more than 125% of RDA energy were 176 mg/dl, 192 mg/dl, 185 mg/dl respectively(p < 0.01). The TC levels less than 75%, 75-125%, more than 125% of RDA protein were 180 mg/dl, 190 mg/dl, 175 mg/dl respectively(p < 0.05). TC levels less than 75%, 75-125%, more than 125% of RDA fat were 181 mg/dl, 193 mg/dl, 194 mg/dl respectively(p < 0.05). Of the subjects, 18% ate mushrooms. The BMI of the mushroom eaten was 24 and that of non-mushroom eaten was 25(p <0.05). Of the subjects, 28% consumed milk and milk produce. The mean level of TC and TG in milk and milk products eaters were 181 mg/dl, 131 mg/dl and that of non milk and milk products eaters were 197 mg/dl, 159 mg/dl(p < 0.001, p < 0.05). Of the subjects, 58% ate fish. The HDL-C of the fish eaten was 50 mg/dl and that of non-fishes eaten was 45 mg/dl(p < 0.05).

Composition and Yield of Korean Breast Milk and Maternal Intakes of Foods and Nutrients (수유부의 식품 및 영양소 섭취와 유즙 생성량 및 조성)

  • This study was conducted to investigate the relationship between lactational capacity and intakes of energy and energy-yielding nutrients. Food consumption, intakes of carbohydrate, protein, fat and energy and quantity and proximate composition of milk of 11 lactating Korean mothers were determined at 1, 2 and 3 months postpartum longitudinally. Food consumption was estimated using a 24-hour recall method ; intakes of energy and energy-yielding nutrients were calculated according to the Food Composition Table. Daily milk production was estimated using a 72-hour test-weighing method ; protein, fat, lactose and energy concentrations were analyzed. Average intakes of energy and protein were 1974$\pm$386㎉/day and 67.0$\pm$12.3㎉/day, these were 73% and 74% of the Korean Recommended Allowances, respectively. Average milk yield was 720.1$\pm$123.3ml/day containing energy 59.6$\pm$9.5㎉/dl, protein 1.1$\pm$0.1g/dl, fat 2.7$\pm$0.8g/dl, and lactose 6.3$\pm$0.3g/dl. No relationship existed between the intakes of carbohydrate, protein, fat and energy and the quantity and proximate composition of the milk. However, the intakes of energy, carbohydrate and vegetable protein were inversely related to the concentrations of energy and lactose in the milk. This result indicates that lactational capacity may be affected by the other factors excluding intakes of energy and energy-yielding nutrients and a high intake of energy may not guarantee optimal lactational capacity.

Microparticle-based Immunonassay for the Detection of C-reactive Protein in Serum (미립자 응집반응을 이용한 C-reactive Protein의 면역 측정 법에 관한 연구)

  • The C-reactive protein(CRP) from ascitic and pleural fluid was purified using calcium dependent affinity chromatography of CNBr activated Sepharose-4B covalently coupled to p-diazonium phenylphosphorylcholine(DPPC) and hydroxylapitite chromatography. Polyclonal antibody was prepared from rabbit by immunizing the purified CRP. Specific immunoglobulin G was isolated using affinity chromatography and coupled to microparticles. A sensitive microparticle-based immunoassay was developed to measure CRP within 3 mins. The detection range was between 0.5mg/dl) and 20mg/dl in serum, showing strong response in the range of 0.7~2.9 mg/dl, weak response in 5.0~13.2 mg/dl and zone phenomenon over 28mg/dl. The average value of CRP in 74 samples was 3.8mg/dl and most of the values were lower than 10mg/dl .The CRP values of serum samples were determined by our microparticle-based immunoassay, and were compared with those obtained using the other commercial products(B Co., France and I Co., Japan). Good correlations were shown between the values obtained by our developed microparticle-based immunoassay system and those by other commercial products. All performance characteristics evaluated make our developed microparticles-based immunoassay suitable for a simple, rapid, and reliable screening of CRP in serum.

Anaphylactoid Reaction Induced by Administration Dirofilaria immitis Extracts (개 심장사상충 추출액 투여에 의한 Anaphylactoid Reaction)

  • This study was performed to clarify the induction possibility of anaphylactoid reactions by the administration with the heartworm extracts, and, if any, to elucidate different virulences in terms of the protein concentrdtions and sexes of Dirofilaria immitis. Twenty three clinically healthy D. immitis-free adult dogs were used in the present study. The experimental animals were divided into 5 groups. Group A (5 heads) was administered with an female heartworm extract containing 0.1 g/dl protein concentration. Group B (4 heads) was administered with an male heartworm extract containing 0.1g/dl protein concentration. Group C (5 heads) was administered with an female heartworm extract containing 0.2 g/dl protein concentration. Group D (4 heads) was administered with an male heartworm extract containing 0.2 g/dl protein concentration. Group E (5 heads) was administered with an female heartworm extract containing 0.4 g/dl protein concentration. The changes of clinical symptoms and vital signs (body temperature, heart rate and respiration rate) were examined before and 5 minutes, 15 minutes, 30 minutes, 1 hour, 2 hours and 24 hours after injection with the extracts, respectively. In addition, the changes of hematological values (RBC, PCV and total leukocytes counts), serum chemical values (ALP and CK) were determined. It was considered that heartworm extract could induce anaphylactoid reaction and adult female heartworm extract was more affective than those of adult male heartworm extract in the changes of clinical symptoms, vital signs, hematological values and serum chemical values.

Relation among Calcium Intake , Bone Metabolism Parameters , Serum Protein and Lipids of Female College Students in Chungnam (충남지역 일부 여대생의 칼슘 섭취상태와 혈청 골대사지표 , 단백질 및 지질 수준과의 관계에 대한 연구)

  • The purpose of this study was to investigate the relationship among calcium intake, blood parameters related with bone metabolism, and serum lipids in healthy adults on self-selected diet. Subjects were consisted of 40 female college students residing in Chungnam. Anthropometric measurements, dietary intake measurements and blood collection were conducted. Serum concentrations of total protein, albumin, alkaline phosphates, leucine amino peptidase, BUN, calcium, inorganic phosphorus, and lipids were measured by biochemical analyzer and ICP spectrometer. The results were as follows. The mean age of subjects was 22.34 years and weight, height and BMI were 52.89kg, 161.29cm and 20.34, respectively. The daily mean energy and calcium intakes were 81.75% and 64.38% of RDA. The mean animal 1:2. The mean serum concentrations were 6.54g/dl(total protein), 4.12g/dl(albumin), 123.24U/(alkaline phosphates), 36.59U/l(leucine amino peptidase), 8.26mg/dl(calcium), 3.29mg/dl(inorganic phosphorus), 60.73mg/dl(triglyceride), 138.49mg/dl(total cholesterol), 65.95mg/dl(HDL-cholesterol), and 60.39mg/dl(LDL-cholesterol). There were no significant differences among calcium intake, bone metabolism parameters, and serum lipids when analyzed by Pearson's correlation coefficient. More systematic studies are required to investigate the roles of calcium in healthy persons on self-selected diets containing different levels of calcium.

Studies on the Blood Chemistry of Korean Native Cattle and Pigs (한우 및 돼지의 혈액화학치에 관한 연구)

  • In order to know the range in blood chemical values and enzyme activities of korean native cattle and pigs being raised in kyongbuk province, the mean values of serum protein compositions, enzyme activities(GOT, GPT, ALP), cholesterol, glucose, bilirubin, creatinine, urea nitrogen and uric acid were determined and the results obtained were as follows ; 1. Mean contents of total protein, albumin, globulin and A / G ratio for the korean native male cattle were $7.28{\pm}0.86$ g / dl, 43.39{\pm}0.65$ g / dl, 43.83{\pm}0.92$ g / dl & 40.95{\pm}0.34$;for the male pigs were 47.39{\pm}0.84$ g / dl, 43.63{\pm}0.43$ g / dl, 43.77{\pm}0.88$ g / dl, 40.97{\pm}0.26$;for the female pigs were 47.60{\pm}0.68$ g/ dl, 43.54{\pm}0.58$ g / dl, 43.90{\pm}0.74$ g / dl, 41.02{\pm}0.34. 2.$ Mean activities of GOT, GPT and ALP for the korean native male cattle were 448.3{\pm}11.6$ U, 429.6{\pm}4.6$ U, 413.7{\pm}2.8$ U; for the male pigs were 458.6{\pm}10.6$ U, 438.6{\pm}5.3$ U, 424.9{\pm}9.2$ U; for the female pigs were 443.9{\pm}10.8$ U, 440.5{\pm}4.6$ U, 413.9{\pm}3.4$ U. 3. Mean contents of cholesterol, glucose and bilirubin for the korean native male cattle were 4108.7{\pm}31.8$ mg / dl, 488.2{\pm}26.4$ mg / dl, $0.69{\pm}0.52$ mg / dl; for the male pigs were 4105.9{\pm}18.3$ mg / dl, 4102.9{\pm}22.3$ mg / dl, 40.70{\pm}0.18$ mg / dl: for the female pigs were $113.1{\pm}20.9$ mg / dl , 4103.2{\pm}23.9$ mg / dl, 40.49{\pm}0.21$ mg / dl. 4. Mean contents of creatinine, urea nitrogen and uric acid for the korean native male cattle were 42.86{\pm}0.69$ mg / dl, $18.1{\pm}5.0$ mg / dl, 41.38{\pm}0.66$ mg /dl; for the male pigs were 42.20{\pm}0.51$ mg / dl, $18.2{\pm}4.3$ mg / dl, 40.29{\pm}0.10$ mg /dl: for the female pigs were 42.36{\pm}0.70$ mg / dl, $20.5{\pm}5.4$ mg / dl, 40.69{\pm}0.27$ mg / dl.

Effect of Levels of Sodium DL-malate Supplementation on Ruminal Fermentation Efficiency of Concentrates Containing High Levels of Cassava Chip in Dairy Steers

  • Four rumen-fistulated dairy steers were randomly assigned according to a $4{\times}4$ Latin square design to investigate effects of supplementation levels of sodium dl-malate in concentrates on rumen ecology, ruminal fermentation, nitrogen balance, feed intake and digestibility of nutrients and ruminal microbial protein synthesis. The dietary treatments were cassava concentrate-based, containing sodium dl-malate supplementation at 0, 9, 18 and 27 g/hd/d with urea-treated rice straw (UTS) fed ad libitum. The experiment was conducted for four periods, each period lasting 21 days. Ruminal pH increased with incremental addition of malate (p<0.05). Additionally, molar proportions of propionate were higher in supplemented groups and was highest at 18 g/hd/d of malate supplement (p<0.05). Microbial protein synthesis tended to be higher in dairy steers receiving sodium dl-malate supplements and also was the highest at 18 g/hd/d. Variable bacterial populations, such as amylolytic, proteolytic and cellulolytic species were increased (p<0.05). Furthermore, protozoal populations were decreased significantly (p<0.05), while fungal zoospores were dramatically increased in dairy steers receiving sodium dl-malate supplement (p<0.05). These results suggested that supplementation of concentrate containing a high level of cassava chip at 18 g/hd/d with UTS in dairy steers could improve rumen fermentation efficiency and rumen microbial protein synthesis.

Composition of Serum Protein in Korean Fragile X Syndrome Patients (한국인 Fragile X 환자들의 혈청단백질 구성)

  • The karyotype and the concentration of serum protein were investigated in Korean unclassified mental retardees. The results were as follows. Fragile X chromosomes were identified in three patients, and the frequencies of fragile X chromosome were 4~15%. The concentration of serum protein was 5.73$\pm$0.89(g/dl), and the A/G ratio was 0.86$\pm$0.14 in fragile X syndrome patients. The concentration of serum protein was 6.83 $\pm$0.72(g/dl), and the A/G ratio was 0.87$\pm$0.17 unclassified mental retardees. The results revealed that the level of globulin concentration and A/G ratio in fragile X syndrome patients and unclassified mental retardees were lower than in normal group and Down's patients.
