편백나무 추출액의 항균 및 항산화 활성 (Antibacterial and Antioxidant Activity of Chamaecyparis obtusa Extracts)

  • 본 연구에서는 편백나무(Chamaecyparis obtusa) 추출액의 항산화 활성 및 항균활성을 검토하였다. 편백나무 추출액의 항산화 활성을 평가하기 위하여 DPPH radical 소거능, ABTS radical 소거능 및 SOD 유사 활성을 분석하였다. 그 결과, 편백나무 추출액의 DPPH radical 및 ABTS radical 소거 활성은 농도 의존적으로 증가하였으며, $50{\mu}l/ml$ 농도에서 78% 및 62%의 최대 활성을 나타내었다. 또한, 편백나무 추출액은 높은 SOD 유사 활성을 보였으며, $50{\mu}l/ml$ 농도에서 92.85 %의 최대 활성을 나타내었다. 한편, 편백나무 추출액의 항균 활성을 측정하기 위해 Paper disc agar diffusion 법을 이용하여 식중독 및 질병을 일으키는 6종의 균주에 대하여 검토하였다. 편백나무 추출액은 B. cereus, E. coli, L. monocytogenes, S. aureus, S. typhi, V. parahaemolyticus에 대한 항균 활성이 나타났고, 이중 B. cereus에 대하여 가장 강한 항 박테리아 활성을 보였다. 이상과 같이 항균 활성이 나타난 6종의 균주에서 편백나무 추출액의 MIC 및 MBC는 $30{\sim}40{\mu}l/ml$로 확인되었다. 이러한 결과는 편백나무 추출액을 이용하여 식중독과 같은 병원균의 성장을 억제하는 항균 소재로 개발한다면 산업적 가치가 높을 것으로 생각된다.

어성초(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성 (Antimicrobial Activity of Fractional Extracts from Houttuynia cordata Root)

  • 본 연구는 어성초(Houttunynica cordata) 뿌리의 에탄올추출물과 클로로포름, 에틸아세테이트, 부탄올 및 수용성 분획물을 이용하여 disc diffusion의 방법에 의해 항균활성에 대한 연구를 수행하였다. 건조된 어성초 뿌리로부터 획득한 에탄올 추출물의 수율은 전체 건조 중량의 15.7%로 나타났으며, 용매의 극성을 이용한 분획물의 수율은 수용성 분획물(5.9%)>클로로포름 분획물(0.5%)>에틸아세테이트 분획물(0.1%) 순으로 나타났다. 어성초뿌리의 에탄올 추출물은Gram 양성균에 대하여 항균활성이 강하게 나타났으며, 특히, B. cereus, C. xerosis 및 S. aureus에 대하여 10.7∼11.8mm의 inhibition zone을 보였다. 또한 에틸아세테이트 분획물은 1.25mg/disc의 낮은 농도에서 강한 항균활성을 보였으며, 5.00mg/disc에서는 5종의 Gram 양성균과 9종의 Gram 음성균에서 강한 항균활성을 나타내었다. 이에 반해서 부탄올 분획물은 에틸아세테이트 분획물보다 2배정도 높은 농도에서 비슷한 항균활성을 나타내었다. 그러나, 클로로포름과 수용성 분획물은 20.00 mg/disc의 높은 농도에서도 각각 3종 및 5종의 검정균에 대해서만 약한 항균활성을 보였다. 어성초 뿌리의 에탄을 추출물과 각 순차분획물의 총폴리페놀 함량은 에틸아세테이트 분획물>부탄올 분획물>에탄올 추출물>클로로포름 분획물>수용성 분획물의 순으로 나타났으며, 페놀성 물질이 항균 활성을 나타낸다는 기존의 보고와 동일하게 에탄올 추출물과 각 순차분획물에 함유된 페놀성 물질의 함량과 항균 활성이 정의 비례 관계를 나타내는 것을 확인할 수 있었다.

  • 항균력이 있는 것으로 예측되는 몇몇 천연물로부터의 추출물과 유산균인 L. mesenteroides, L. citreum, P. pentosaceus의 대사산물인 박테리오신의 항균특성을 알아보기 위하여 최소억제농도 (MIC) 측정법, disc diffusion assay와 challenge법으로 항균력을 조사하였다. L. mesenteroides, L. citreum, P. pentosaceus 유산균에 의해 생산된 박테리오신은 P. aeruginosa, S. aureus, E. coli, B. subtillis 균에 대하여 뛰어난 항균특성을 보인 반면 C. albicans에 대해서는 상대적으로 약한 항균력을 보였다. 반면에 phenonip과 sandal wood 추출물은 C. albicans에 대해서는 강한 항균력을 나타냈으나 다른 대표균들에 대해서는 매우약하거나 거의 항균력을 나타내지 않았다.

식품 부패미생물의 증식을 억제하는 천연 항균성물질의 검색 (Screening of Natural Antimicrobial Plant Extract on Food Spoilage Microorganisms)

  • 31종의 식물에서 필요 부위를 선정하여 75% 에탄올이나 물로 추출하여 얻은 추출물로서, 일반적으로 식품의 부패 혹은 변질에 관여한다고 알려진 Bacillus subtilis, Lactobacillus plantarum, Leuconostoc mesenteroides, Pseudomonas fluorescens, Bacillus cereus 등 5종의 세균과 Saccharomyces cerevisiae 등 1종의 효모를 대상으로 항균성을 시험하였다. 실험대상 6종의 모든 미생물에 대하여 항균성을 나타낸 것은 황백의 에탄올 추출물이었으며 산초, 참깨묵, 지치, 졸참나무는 효모를 제외한 5종에 대하여 댓잎, 오동나무, 리기다는 B. subtilis 및 S. cerevisiae 혹은 L. mesenteroides 및 S. cerevisiae 또는 L. mesenteroides 및 P. fluorescens, B. cereus를 각각 제외한 4종에 대하여 항균성을 나타냈고, 균종에 따라 약간의 차이는 있었지만 황백이 가장 높은 항균성을 보였다. B subtilis, L. plantarum 및 L. mesenteroides에는 황백이, P. fluorescens에는 황백과 오배자가 disc diffusion method에서 가장 높은 항균성을 보였다. 에탄올 추출물과 물 추출물을 비교한 결과 거의 대부분 에탄올 추출물의 항균성이 높았으나, 균종에 따라서는 일부 물 추출물에서 항균성을 따로 보이거나 더 높은 경향이 있기도 하였다. 이상과 같이 천연물 중에도 상당한 항균성물질이 존재한다는 것이 확인되었다.

Lactobacillus casei subsp. 및 Streptococcus faecium이 생산한 항균성물질의 성상 (Characteristics of the antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium)

  • Antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium were examined for its antibacterial effects against some pathogenic bacteria. They were partially purified with ammonium sulfate precipitation, methanol-acetone extraction, G-50 gel filtration and examined its characteristics. When L casei subsp. and Str faecium were cultivated in MRS broth, stationary phase of L casei is until 24 hours and Str faecium is 20 hours. pH change of the cultured medium was both decreased after 12 hours and then constant at pH 4.5~4.6 after 28 hours. MRS broth culture fluids of L casei subsp. and Str faecium appeared the antibacterial effects by the spot-on-the-lawn method against ETEC, Sal pullorum and Sta aureus. Culture filtrates of L casei subsp. and Str faecium also appeared the antibacterial effects by the disc diffusion method. Culture filtrates of L casei sub. rhamnosus 7469 produced 0.032M of lactic acid and 0.01M of acetic acid. Str faecium 27273 also produced 0.027M of lactic acid and 0.01M of acetic acid. Protein concentrations of culture filtrates produced by L casei sub rhamnosus 7469 and Str faecium 27273 was $495{\mu}g/m{\ell}$ and $594{\mu}g/m{\ell}$, respectively. Antibacterial substances which are partially purified by ammonum sulfate precipitation, methanol-acetone extraction and G-50 gel filtration inhibit the growth of ETEC, Sal pullorum and Sta aureus. Characteristics of purified antibacterial substances was examined. Its molecular weight was about 31Kd, stabilized at $100^{\circ}C/20min.$ and some of proteolytic enzyme treatment.

병원성 미생물에 대한 다빈도 51종 한약처방의 항균 활성 연구 (Antimicrobial Activities of 51 Herbal Formulae on Pathogenic Microorganisms)

  • Objectives: The purpose of this study was to establish experimental evidence for the antimicrobial effects of 51 herbal formulae commonly used in traditional Korean medical institutions. Methods: The antimicrobial activities of herbal formulae were screened using the disc diffusion method against 10 pathogenic microorganisms (Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, Escherichia coli DH5α, E. coli O157, Salmonella enteritidis, Yersinia enterocolitica, Shigella flexneri, and Helicobacter pylori). Of the 51 herbal formulae, 13 herbal formulae with antimicrobial activity were selected and their dose-dependency were confirmed. Results: Nine herbal formulae, including Gyeji-tang, Dangguisu-san, Saengmaek-san, Samul-tang, Ssanghwa-tang, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, and Paljung-san, presented antibacterial activity against B. cereus. The effects of Saengmaek-san and Paljung-san was sustained for 48 hr. On L. monocytogenes, Dangguisu-san and Hyangsapyeongwi-san showed antimicrobial activity, but only Hyangsapyeongwi-san maintained the activity for 48 hr. Thirteen herbal formulae such as Galgeun-tang, Gyeji-tang, Dangguisu-san, Mahwang-tang, Banhasasim-tang, Saengmaek-san, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, Cheonwangbosim-dan, Palmijihwang-tang, Paljung-san, and Hwanglyeonhaedok-tang showed antimicrobial activity against V. parahaemolyticus, and the activity was maintained for 48 hr. The 51 herbal formulae did not show any antimicrobial activity against seven strains such as E. coli DH5α, E. coli O157, S. aureus, S. enteritidis, Y. enterocolitica, S. flexneri, and H. pylori. Conclusions: Nine, two, and thirteen herbal formulae showed antimicrobial activities against B. cereus, L. monocytogenes, and V. parahaemolyticus in a dose-dependent manner, respectively. The results of antimicrobial activity of 51 herbal formulae against 10 microorganisms might be used as the basis for new application of herbal formulae.

천연 약용자원 추출물의 인수공통 감염 세균에 대한 in vivo 및 in vitro에서의 항균 효과 (In vivo and In vitro Antimicrobial Effects of Natural Antibiotics Present in Crude Extracts of Various Medicinal Plants)

  • Bacteria are among the most common causes of severe diseases in both plants and animals. Salmonella spp. has deleterious effects and is the cause of various transmittable diseases. Because of strains resistivity, side effects and high prices of synthetic antibiotics, it has become essential to explore safe and economical natural sources of antibiotics. In this study, growth inhibitory effects of natural antibiotics present in crude extracts of Galla rhois, Thujae semen, Paeonia japonica, and Armeniacae semen were investigated both in vivo and iv vitro. Ethanol extracts of the above-mentioned plants were prepared and tested against seven serovars of Salmonella and Escherichia coli by disc diffusion method. In addition, the antibacterial effects of the plant extracts were determined in vivo using ducks as model animals. Reverse transcription-polymerase chain reaction was performed using blood and fecal samples of control, infected, and treated groups of the ducks to determine the gene expression levels of the bacteria. Our results confirmed that the Galla rhois ethanol extract had the highest antibacterial activity among the plant extracts when they were used individually. However, the Galla rhois, Thujae semen, and P. japonica ethanol extracts showed stronger antibacterial effects against all the bacterial species used when the extracts were combined at a ratio of 3:3:2, respectively.

In vitro Study and Clinical Trial of Natural Essential Oils and Extract Against Malassezia Species

  • Background: Malassezia, a lipophilic yeast, is a causative agent for dandruff and seborrheic dermatitis. Many biological agents have been studied for anti-Malassezia effect but further studies are needed for their clinical application. Objective: The study was conducted to evaluate the inhibitory effect of different natural essential oils and a fruit extract on Malassezia species in an in vitro study and a clinical trial. Methods: The antifungal effects of natural essential oils and a fruit extract on Malassezia species (M. furfur and M. sympodialis) were evaluated by measuring the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and using the disc diffusion method. Natural essential oils of citron seed, lavender, and rosemary and citrus junos fruit extract were used for the in vitro study. The clinical trial was conducted with a shampoo containing four ingredients. A total of 22 subjects used the shampoo every day for 4 weeks and were evaluated using clinical photography, trichoscopy, and sebumeter at baseline, 2 weeks, and 4 weeks after treatment. Results: Antifungal activity of agents was relatively lower in lavender and rosemary essential oils at MIC and MFC. Disc diffusion method revealed same results. In the clinical trial, the amount of sebum decreased statistically significantly and erythema, dandruff, and lesion extent also improved. Conclusion: The natural essential oils and fruit extract are effective for suppressing Malassezia activity, therefore these might be used as an alternative for treatment of dandruff and seborrheic dermatitis.

아토피 피부염 외치치료제 처방 개발을 위한 실험적 연구 (An experimental study for the development of prescription on atopic dermatitis)

  • Objectives : Atotang was composed of 10 kinds of traditional medicinal herb. This research was performed to examine biological effects of Atotang for the development of prescription on atopic dermatitis. Methods : Atotang was extracted with 80% EtOH. Free radical scavenging assay has tested for anti-oxidative activity as well as the contents of total polyphenol. We observed the production of ROS, nitric oxide(NO) and the inflammatory cytokines such as interleukin-1beta(IL-${\beta}$), IL-6, tumor necrosis factor-alpha(TNF-${\alpha}$), Prostaglandin E2($PGE_2$) in Raw 264.7 cells stimulated by LPS. We used Disc diffusion method to investigate antibacterial activity on Candida albicans, Staphylococcus aureus and Staphylococcus epidermis. Result : Content of total phenolic compound of Atotang was 36.3 mg/g ext. DPPH and ABTS scavenging activities were 77% and 46% at 200 ug/ml respectively, showing dose-dependent increase. The amounts of ROS and NO in RAW 264.7 cells were decreased by 30% and 19% at 200 ug/ml, respectively, showing dose-dependent decrease. The prodcution of IL-1beta, IL-6 and TNF-alpha in RAW 264.7 cells were decreased dose-dependently by 81%, 67%, and 20% at 200 ug/ml, respectively. Atotang was reduced LPS-stimulated production of $PGE_2$ by 33%. Atotang on C. albicans, S. aureus and S. epidermis was selected by a disc diffusion method and inhibition effect of the Atotang on the growth of S. epidermis was the greatest. Conclusion : The results indicated that Atotang showed biological activities showing anti-oxidant, anti-inflammatory and antibacterial effects. Based on these results, it is concluded that Atotang can be applied to the prescription on atopic dermatitis.

Comparative Analysis of Chemical Compositions and Antimicrobial Activities of Essential Oils from Abies holophylla and Abies koreana

  • The chemical compositions and antibacterial and antifungal activities of essential oils extracted from Abies holophylla and A. koreana were investigated. GC-MS analysis revealed that 38 compounds comprised 95.88% of the A. holophylla essential oil, with the main components being bicyclo[2.2.1]heptan-2-ol(28.05%), ${\delta}3$-carene(13.85%), $\alpha$-pinene(11.68%), camphene(10.41%), dl-limonene(7.61%), $\beta$-myrcene(7.11%), trans-caryophyllene(5.36%), and $\alpha$-bisabolol(3.67%). In the essential oil from A. koreana, 36 compounds comprised 98.67% of the oil, and the main compounds were bornyl ester(41.79%), camphene(15.31%), $\alpha$-pinene(11.19%), dl-limonene(8.58%), fenchyl acetate(5.55%), and $\alpha$-terpinene(2.29%). Both essential oils showed great potential of antibacterial activity against several bacteria tested, in the range of 2.2-$8.8{\mu}g$ per disc by the agar disc diffusion method, and minimal inhibitory concentration(MIC) values of 5.5-21.8 mg/ml by the microdilution method. Both oils showed very effective antifungal activities toward all pathogenic strains tested, including Candida glabrata, with MIC values in the range of 0.5-2.2 mg/ml. As a whole, A. koreana oil showed better antibacterial and antifungal properties than A. holophylla oil.
