Effects of simazine [2-chloro-4,6-bis(methylamino)-s-triazine] on the photochemical reactions of isolaed spinach chloroplasts and crude thylakoids of Anabaena inequalis UTEX B-381 were compared. Simazine inhibited photosynthetic O2 evolution and increased the chlorophyll fluorescence in whole cells of Anabaena. The electron transfer from diphenylcarbazide to 2,6-dichlorophenolindophenol was inhibited by simazine treatment in spinach chloroplasts, but not in crude thylakoids of Anabaena. In spinach chloroplasts, the chlorophyll fluorescence was increased by simazine treatment in the presence of diphenylcarbazide and ferricyanide, but not in the presence of diphenylcarbazide and silicomolybdate. In crude thylakoids of Anabaena, simazine treatment did not increase the chlorophyll fluorescence in the presence of either diphenylcarbazide and silicomolybdate, or diphenylcarbazide and ferricyanide. There results suggest that the inhibitory site of simazine on photosynthetic electron transport chain of anabaena is different from that of spinach chloroplasts. And there may be a possiblity that the inhibition site of simazine in Anabaena lies on the donor side of photosystem II, before the site of electron donation by diphenylcarbazide.
A hexavalent chromium (Cr (VI)) is one of the hazardous substances regulated by the RoHS. The determination of Cr (VI) in various polymers and printed circuit board (PCB) has been very important. In this study, the three different analytical methods were investigated for the determination of a hexavalent chromium in Acrylonitrile Butadiene Styrene copolymer (ABS) and PCB. The results by three analytical methods were obtained and compared. An analytical method by UV-Visible spectrometer has been generally used for the determination of Cr (VI) in a sample, but a hexavalent chromium should complex with diphenylcarbazide for the detection in the method. The complexation did make an adverse effect on the quantitative analysis of Cr (VI) in ABS. The analytical method using diphenylcarbazide was also not applicable to printed circuit board (PCB) because PCB contained lots of irons. The irons interfered with the analysis of hexavalent chromium because those also could complex with diphenylcarbazide. In this study, hexavalent chromiums in PCB have been separated by ion chromatography (IC), then directly and selectively detected by inductively coupled plasma atomic emission spectrometry (ICP-AES). The quantity of Cr (VI) in PCB was 0.1 mg/kg.

In this paper, a potentiometric sensor based on polyaniline conducting polymer for potentiometric determination of Cr (VI) ions is reported. Polyaniline was synthesized electrochemically (cyclic voltammetry method) onto a micro pencil graphite electrode (0.7 mm diameter) in the presence of HCl and diphenylcarbazide (termed as (PGE/PAni/DPC). Some initial experiments were performed in order to find out the optimized conditions for preparation of the introduced Cr (VI) sensor electrode. The plot of E vs. log [Cr (VI)], showed a linear response in the range from $1.0{\times}10^{-6}$ to $1.0{\times}10^{-1}$ M. High repeatability with the detection limit of $8.0{\times}10^{-7}$ M was obtained.

Inhibiting action of semicarbazide, thiosemicarbazide, sym. diphenylcarbazide towards corrosion of zinc in hydrochloric acid has been investigated. The rate of corrosion depends on the nature of the inhibitor and its concentration. The values of inhibition efficiency from, weight loss, thermometric measurements are in good agreement with those obtained from polarization studies. From the polarization studies, the inhibitors used act as mixed absorption type inhibitors, increased adsorption resulting from an increase in the electron density at the reactive C=S and C=O groups and N-atoms. The thermodynamic parameters of adsorption obtained using Bockris-Swinkels adsorption isotherm reveal a strong interaction of these carbazides on zinc surface.
The effect of $Cd^{2+}$ on the electron transport rate of PSI and PSII was investigated in isolated spinach chloroplasts. In photosystem II, the rate of electron transport was decreased as the concentration of $Cd^{2+}$ was increased from 1 to $100\;\mu\textrm{M}$. The inhibitory effect of $Cd^{2+}$ was reduced when diphenylcarbazide was added to the reaction medium, indicating that $Cd^{2+}$ affects primarily psn oxygen evolving complexes of thylakoid membrane. The inhibitory effect of $Cd^{2+}$ was reduced when $Mn^{2+}\;and\;Ca^{2+}$ were added to the reaction medium, but the inhibitory effect was not fully relieved. Although the activity of psn was decreased significantly by the treatment of $50\;\mu\textrm{M}\;Cd^{2+}$, Fv/Fm was decreased slightly. However, the treatment of $100\;\mu\textrm{M}\;Cd^{2+}$ resulted in the marked decrease of Fv/Fm. In photosystem I, the rate of electron transport decreased as the concentration of $Cd^{2+}$ was increased from 0.2 to 3.2 mM. The inhibitory effect of $Cd^{2+}$ was decreased when the chloroplast treated with $Cd^{2+}$ was washed by centrifugation.gation.
A simple semiquantitative procedure was developed for the determination of sub-ppm level of chromium(VI) in aquatic samples by using an analytical micro-column packed with diphenylcarbazide(DPC) gel beads. DPC gel beads were prepared by swelling XAD-2 resin(115∼150 mesh in dry condition) in ethanol for 10min, packing into a glass column(1.5 mm bore, 65nm length) and adsorbing 1ml of ethanol solution of $2{\times}10^{-3}M$ DPC for 20min at room temperature. When 0.5ml of ethanol solution containing chromium(VI) was passed through the DPC gel column for 40min, the original white color of the reagent gel turned to red-violet from the up-stream of the column. As the length of colored band was proportional to the total amount of chromium(VI) in the sample solution passed through the column, the concentration of chromium(VI) could be determined from the calibration line which had been prepared by using the standard solution. Chromium(VI) ion as small as from 0.1 to 0.8 ppm could be determined with ${\pm}5{\sim}{\pm}15{\%}$ relative errors. Since other interfering cations were few, 100-fold excess of Fe(III), 50-fold excess of Cu(II) could be masked with EDTA. This method was successfully applied to the analysis of chromium(VI) in industrial effluents.
An on-line flow injection analysis with dual pre-concentration method was developed to determine the ultra trace tri and hexavalent chromium in water. In this system, the cation and anion pre-concentration columns were combined with a 10-port injection valve and then used to separate and concentrate Cr (III) and Cr (VI) selectively. The two species of concentrated chromium were sequentially eluted and determined by using HCl-KCl buffer of pH 1.8 as an eluent. Cr (III) was oxidized by hydrogen peroxide to Cr (VI). It was detected spectrophotometrically at 548 nm by complexation with DPC (diphenylcarbazide). Several factors such as concentration of $H_2O_2$, DPC and coil length in reaction condition were optimized. The linear range for Cr (III) and Cr (VI) was 0.1-50 ${\mu}g$/L. The limit of detections ($3{\sigma}$) of Cr (III) and Cr (VI) were 52 ng/L and 44 ng/L under the optimized FIA system, and their recoveries 98% and 103%, respectively. This method was applied to analyze contamination level of chromium species in tap water, groundwater and bottled water.
The room temperature fluorescence induction of chloroplasts was utilized as a probe to locate the site of inhibition by mercury, copper and zinc on PS II by mercury. Inhibitory effect of Hg2+ on electron transport activity was notable as compared with Cu2+ and Zn2+. At concentrations of HgCl2 over 50 $\mu$M, activities of PS II and whole-chain electron transport decreased more than 70%, while that of PS I decreased about 10~30%. This suggests that PS II is more susceptible to Hg2+ than PS I is. In the presence of diphenylcarbazide (DPC), 50 $\mu$M HgCl2 inhibited the reduction of dichlorophenolindophenol (DCPIP) about 50%. Addition of heavy metals induced marked decrease in maximal variable fluorescence/initial fluorescence [(Fv)m/Fo], but no changes in Fo. With various concentrations of heavy metals, changes of chlorophyll a fluorescence emitted by PS II showed gradual decrease in photochemical quenching (qQ), which indicates an increase in reduced state of electron acceptor, QA. Especially, the addition of HgCl2 caused a notable decrease of qQ. In the presence of 50 $\mu$M CuCl2, energy-depended quenching (qE) was completely reduced, whereas in the presence of 50 $\mu$M CuCl2 and ZnCl2 it was still remained. The above results are discussed on the effects of mercury in relation to water-splitting system and plastoquinone (PQ) shuttle system.
A study was conducted to compare three analytical methods for determination of hexavalent chromium in the welding fume. Precision and accuracy arc documented for colorimetric, ion chromatographic, and inductively coupled plasma-optical emission spectroscopic method. Evidence is presented that welding fume can affect the oxidation of trivalent chromium. A simple sonication extraction method, proposed in this study, instead of hot alkaline extraction has the advantage of minimizing the potential for chromium oxidation.
Using a column of macrorecticular gel beads impregnated with alcohol solution of diphenylcarbarzide (DPC), the enrichment of Cr(Ⅵ) ion in 0.1M sulfuric acid was investigated. The column performance was compared with respect to the types of solid support such as Amberlite XAD and Diaion HP. Diaion HP-20 was found to be most suitable for this purpose. The break through point was increased when increasing the amount of DPC absorbed in the resin and the concentrated sample solution showed higher break through capacity than the dilute one. The extraction of Cr(Ⅵ) was not affected by the presence of 100 ppm Fe(Ⅲ) which amounted to ten times of 10 ppm Cr(Ⅵ) and the presence of other metal ions which amounted to 100 times of 10 ppm Cr(Ⅵ). Because the used gel particles were effectively regenerated with methanol, they were able to be used repeatedly.
