  • After establishment of Banwol industrial complex in 1987, Ansan city becomes the largest industrial sector development in Gyeonggi-do, Korea. As the population and industrial activity grow over this region, toxic air pollutants, particularly POPs (Persistent Organic Pollutants) from various emission sources have been major public concerns. Air samples for POPs monitoring were collected at the industrial sites ($A_2$), residential sites ($B_1$, $B_2$), commercial site (C), and rural/remote site (D) of the area of Ansan during 2008 with a prolonged industrial sampling site $A_1$ from 2001 to 2008. All samples were analysed for 2,3,7,8 substituted-polychlorinated dibenzo-p-dioxin and dibenzofurans (PCDD/Fs) and dioxin like polychlorinatd diphenyls (dl-PCBs). In site $A_1$, a steady decline of their concentrations from 2003 to 2008 was observed due to the reinforced emission guideline from waste incinerators. The average concentration of the PCDD/Fs and dl-PCBs ranged between 0.118 pg-TEQ/$m^3$ (rural/remote site D) and 0.532 pg-TEQ/$m^3$ (industrial area $A_2$). These level were generally consistent with previous studies in Gyeonggi-do, while higher than other places. Most of PCDD/Fs congener were partitioned into particle phase, whereas dl-PCBs were partitioned into gas phase. The logarithm of gas-particle partition coefficient $K_P$ of dl-PCBs and PCDD/Fs were well correlated with sub-cooled liquid vapor pressure $P_L$. The slope $m_T$ of log $K_P$ versus log $P_L$ for PCDD/Fs (-1.22) and dl-PCBs (-1.02) in industrial area ($A_2$) were high compared to other residential/commercial area. It suggests that this area was likely influenced by the direct emission source of PCDD/Fs and dl-PCBs. To simulate the partition of PCDD/Fs and dl-PCBs between gas and particle phase, Junge-Pankow model ($P_L$-base) and $K_{oa}$ model were applied. It was found that J-P model was more suitable than the $K_{oa}$ model in this study.

  • It is it well known fact that the most relevant toxic emissions such as polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), dioxin-1ike polychlorinated biphenyls (PCBs) are produced from incinerators. Induction of cytochrome P450IA1 catalyzed 7-ethoxyresorufin 0-deethylase (EROD) activity in mammalian cell culture (EROD-microbioassay) is a selective and sensitive parameter used for the quantification of dioxin-like compounds. • The liver progenitor cells could form a potential target cell population fore both tumor-initiating and -promoting chemicals. Induction of drug-metabolizing and antioxidant enzymes, including AhR-dependent CYP1A1, NQO-1 and AKR1C9, was detected in the rat liver epithelial WB-F344 "stem-like" cells. Additionally, WB-F344 cells express a functional, wild-type form of p53 protein, a biomarker of genotoxic events, and connexin 43, a basic structural unit of gap junctions forming an important type of intercellular communication. In this cellular model, two complementary assays have been established for detection of the modes of action associated with tumor promotion: inhibition of gap junctional intercellular communication (GJIC) and proliferative activity in confluent cells. We found that the PAHs and PCBs, which are AhR agonists, released WB-F344 cells from contact inhibition, increasing both DNA synthesis and cell numbers. Genotoxic effects of some PAHs that lead to apoptosis and cell cycle delay might interfere with the proliferative activity of PAHs. Contrary to that, the nongenotoxic low-molecular-weight PAHs and non-dioxin-like PCB congeners, abundant in the environment, did not significantly affect cell cycle and cell proliferation; however both groups of compounds inhibited GJIC in WB-F344 cells. The release from contact inhibiton by a mechanism that possibly involves the AhR activation, inhibition of GJIC and genotoxic events induced by environmental contaminants are three important modes of action that could play an important role in carcinogenic effects of toxic compounds. The relative potencies to inhibit GJIC, to induce AhR-mediated activity, and to release cells from contact inhibition were determined for a large series of PAHs and PCBs and their metabolites. In vitro bioassays based on detection of events on cellular level (deregulation of GJIC and/or proliferation) or determination of receptor-mediated activities in both ?$stem-like^{\circ}{\times}$ and hepatocyte-like liver cellular models are valuable tools for detection of modes of action of polyaromatic hydrocarbons. They may serve, together with concentration data, as a first step in their risk assessment.

  • Dioxin contamination on agroproducts is one of the most important issue in food safety. Herein, we measured concentration of 17 dioxins polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) and 14 dioxin like-polychlorinated biphenyls (DL-PCBs) in the main grain (rice and barley) that were produced in Korea. The 36 rice samples were collected from rice processing complex at eight provinces, and the average concentrations of each province were ranged from 2.98 pg/g wet weight (w.w) to 4.98 pg/g w.w. as total PCDD/Fs and DL-PCBs. The seven barley samples were collected from Jeon-nam and Jeon-buk provinces, and their average total concentrations of PCDD/Fs and DL-PCBs were 3.00 pg/g w.w. and 3.24 pg/g w.w. respectively. The residual amounts and toxic equivalent (TEQ) of DL-PCBs in rice and barley were higher than PCDD/Fs. The average total TEQ of rice and barley were 0.0056 pg-TEQ/g and 0.0092 pg-TEQ/g on lower bound estimation respectively. Estimated daily intake of dioxins from rice and barley were calculated 1.03 pg-TEQ/day/person and 0.0534 pg-TEQ/day/person respectively. These were estimated 0.46% and 0.03% of Korean TDI based on 55 kg body weight.

  • Objectives: Persistent organic pollutants (POPs) are known to be the detrimental chemicals in the body, even at low levels, and are stored in adipose tissue. Recently, POPs have been reported to be associated with chronic diseases, including cancer and cardiovascular disease, and aging and obesity are reported as common factors in chronic disease. However, there have been only a few studies on the associations of POPs with age and body mass index (BMI) in Korea. Therefore, we analyzed the associations of serum POPs levels with age and BMI in Korea. Methods: This cross-sectional study includes 444 subjects (253 men and 191 women) from the Korean Cancer Prevention Study-II (2004-2011). Serum levels of 33 polychlorinated biphenyls (PCBs) and 19 organochlorine pesticides (OCPs) were measured by a gas chromatographer (Agilent 6890) coupled to high resolution mass spectrometer (JEOL JMS-800D). Results: Concentrations of PCB 153 (men: 12.26 ng/g lipid, women: 10.50 ng/g lipid) and p,p'-DDE (men: 94.66 ng/g lipid, women: 96.66 ng/g lipid) were the highest among serum PCBs and OCPs, respectively. PCBs and OCPs were significantly positively correlated with age in both sexes. After adjustment for age, non-dioxin like PCBs were significantly negatively correlated with body mass in women. However, cis-heptachlor epoxide was significantly positively correlated with body mass index in both sexes. When analyzed by sex, stronger associations were shown between POPs and age in men and POPs and BMI in women. Conclusion: These results may provide baseline data for the study of POPs and for the health management field in Korea.

  • We investigated the concentrations and distribution patterns of 17 polychlorinated dibenzo-p-dioxins/dibenzofurans(PCDD/Fs), 12 dioxin-like polychlorinated biphenyls(DL-PCBs) and 24 polybrominated diphenyl ethers(PBDEs) in sediments from Ulsan Bay in Korea. The concentrations of PCDD/Fs, DL-PCBs, and PBDEs in 33 sediment samples ranged from 0.11 to 4.86($1.81{\pm}1.04$) pg $WHO_{2005}$-TEQ $g^{-1}$ dry weight(dw), 0.06 to 44.2($4.02{\pm}7.99$) pg $WHO_{2005}$-TEQ $g^{-1}$ dw, and 2.81 to 63.8($19.4{\pm}13.9$) ng $g^{-1}$ dw, respectively. DL-PCBs had dominant contributions(mean, 88%) of total TEQ concentrations in sediment. The concentrations of target compounds in inner locations were higher than those in outer locations in Ulsan Bay (p<0.05). The dominant contribution of highly chlorinated DD/Fs in sediment was associated with combustion process from industrial complexes. Distribution pattern of DL-PCBs was similar with those of commercial PCB products. BDE209 was a dominant congener in sediment, suggesting high use amount of commercial deca-BDE product in surrounding areas.

  • Samples of water, soil, and sediment were taken from 10 streams near Andong, Korea in May 2004. To assess the degree of environmental pollution of each stream, chemical pollutants such as total notrogen (T-N), total phosphorus (T-P), chemical oxygen demand (COD), heavy metals, organophosphorus pesticides, organochlorine pesticides, and dioxin-like PCB congeners were analyzed by standard process tests or U.S. EPA methods. In addition, biomarkers originated from insect immune systems of beet armyworm, Spodoptera exigua, were used to analysis of the environmental samples. Except Waya-chun stream showing T-N content of 9.12 mg/L, most streams were contaminated with relatively low levels of overall pollutants in terms of T-N, T-P, and COD, compared to their acceptable environmental levels designated by the Ministry of Environment. Contents of Pb and Cd in samples of each stream were much lower than environmentally permissible levels. However, several times higherconcentrations of Pb and Cd were found in locations at Mi-chun, Kilan-chun, and Hyunha-chun streams, in comparison with other streams. Diazinon, parathion, and phenthoate compounds among organophosphorus pesticides were detected as concentrations of 0.19, 0.40, and $1.13\;{\mu}g/g$, respectively, from soil sample collected in the vicinity of Mi-chun stream. On the other hand, 16 organochlorine pesticides and 12 dioxin-like PCB congeners, known as endocrine disrupting chemicals, selected in this study were not found above the limit of detection. Biomarker analyses using insect immune responses indicated that Waya-chun stream was suspected as exposure to environmental pollutants. Limitation and compensation of both environmental analysis techniques are discussed.

  • Polychlorinated biphenyls (PCBs) were once used throughout various industries; however, because of their persistence in the environment, exposure remains a global threat to the environment and human health. The Kv1.3 and Kv1.5 channels have been implicated in the immunotoxicity and cardiotoxicity of PCBs, respectively. We determined whether 3,3',4,4'-tetrachlorobiphenyl (PCB77), a dioxin-like PCB, alters human Kv1.3 and Kv1.5 currents using the Xenopus oocyte expression system. Exposure to 10 nM PCB77 for 15 min enhanced the Kv1.3 current by approximately 30.6%, whereas PCB77 did not affect the Kv1.5 current at concentrations up to 10 nM. This increase in the Kv1.3 current was associated with slower activation and inactivation kinetics as well as right-shifting of the steady-state activation curve. Pretreatment with PCB77 significantly suppressed tumor necrosis factor-α and interleukin-10 production in lipopolysaccharide-stimulated Raw264.7 macrophages. Overall, these data suggest that acute exposure to trace concentrations of PCB77 impairs immune function, possibly by enhancing Kv1.3 currents.
