  • B cell has been identified as the sensitive cellular target responsible for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) -induced immune suppression. In isolated cell systems, the differentiation of B cells into antibody secreting plasma cells is believed to be inhibited by TCDD. We also have previously demonstrated IgM secretion was suppressed by TCDD in LPS-activated murine B cell line, CH12.LX.(omitted)

  • It is it well known fact that the most relevant toxic emissions such as polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), dioxin-1ike polychlorinated biphenyls (PCBs) are produced from incinerators. Induction of cytochrome P450IA1 catalyzed 7-ethoxyresorufin 0-deethylase (EROD) activity in mammalian cell culture (EROD-microbioassay) is a selective and sensitive parameter used for the quantification of dioxin-like compounds.   • The equilibrium geometry, adiabatic electron affinity, harmonic vibrational frequencies, and infrared intensities of 1,4,6,9-Tetra chlorinated dibenzo-$\rho$-dioxin anion have been studied using B3LYP method. The local minimum conformation is found to be twisted form with $D_2$ symmetry. The predicted adiabatic electron affinity is 0.54 eV. The predicted inversion barrier between twisted and planar conformers was 0.9 kcal/mol, thus we expect that experimentally observed structure seems to be planar. the characteristic IR band of 1,4,6,9-TCDD anion appears at 1510 $cm^4$, which is a benzene ring skeleton vibration mode related with CaCb and C2C3 bond. this characteristic band could be sued for identication of 1,4,6,9-TCDD anion.

  • 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) is known to induce cytochrome p450 1A1 and to activate c-Src kinase and p21 Ras. This study examined the molecular interactions of adaptor proteins including Shc, Grb2, and Sos in rat primary hepatocytes and their relationship to the induction of CYP1A1 by TCDD. TCDD induced CYP1A1 level and EROD activity in a dose-dependent mode. Sos/Grb2 association isincreased by TCDDㅑㅜ a dose dependent mode. Tyrosine phosphorylated Shc, mainly p152, onloads to Grb2/Sos complex upon TCDD stimulation. The electrophoretic mobility shift of Sos is showed by TCDD. These results indicate that TCDD modulated the molecular interaction features of adaptor compoes proteins including Shc, Grb2, and Cnl in early signaling pathway of TCDD-mediated CYP 1A1 induction of rat primary hepatocyte.

  • In Korea, the number of municipal and hazardous waste incinerators have been increasing since the 1980s. In addition, municipal waste incinerators (MWI) are almost always located in residential areas. Therefore, the human health risk caused by dioxin has become a growing public concern in Korea. In this report, we describe the results of study in which serum levels of PCDDs/PCDFs in a group of workers at the MWI were compared to those in a comparison group of community residents who had never worked at the MWI.
