Purified Polysaccharide Activating the Complement System from Leaves of Diospyos kaki L. (감잎(Diospyos kaki L.)으로부터 정제한 보체계 활성화 다당류)

  • Cold and hot water fractions of Diospyros kaki were screened to determine its anti-complementary activity. Flour of Diospyros kaki leaf (250 g) was boiled at $100^{\circ}C$ for 3 h and passed through a membrane of 10 kDa molecular weight (DK-0). DK-0 was precipitated with ethanol and refluxed with methanol to obtain the crude polysaccharide (DKC). DKC-1 was isolated by ion exchange chromatography on DEAE-Toyopearl 650C, and DKC-1c was purified from DKC-1 by size exclusion chromatography on Bio gel P-60. The anti-complementary activities of DKC-1c at $1000\;{\mu}g/mL$ were 85.4 and 61.1% via whole and alternative pathways, respectively. DKC-1c was determined as a neutral polysaccharide composed of glucose (29.0 mol.%), arabinose (24.3 mol.%), and galactose (16.2 mol.%) with the molecular weight of 66.6 kDa. Results of agarose gel immunoelectrophoresis revealed DKC-1c, as a complement activator, cleaved C3 into C3a and C3b via both pathways.

  • In order to inspect safety and function regarding the reduction of cadmium poisoning by polyphenol compounds prepared from persimmon leaves (Diospyros kaki folium), animal test was done. Rats, group P-1 and P-2, were treated with cadmium and polyphenols of persimmon leaves. A control was just treated with cadmium. GOT activities in P-1 and in P-2 were lower than that of the control without considerable difference, whereas GPT activities decreased in P-1 and P-2. The SOD activity of liver tissue increased in P-l and P-2 compared with that of the control. GST activity was higher than that of the control without considerable difference in P-1; however, it increased in P-2. Lipid peroxide value considerably increased in both P-1 and P-2.

Antioxidant and Anticancer Activities of Methanol Extracts Prepared from Different Parts of Jangseong Daebong Persimmon (Diospyros kaki cv. Hachiya) (장성 대봉감의 부위별 메탄올 추출물의 항산화 활성 및 항암 활성)

  • The antioxidant activity of methanol extracts from five different parts (flesh, peel, core, seed, calyx) of Jangseong Daebong persimmon (Diospyros kaki cv. Hachiya) were evaluated by determining total phenol content (TPC), DPPH radical scavenging activity (RSA), ABTS RSA, and reducing power (RP). The flesh extract gave the highest yield (92.93%) while the lowest yield was obtained from the seed (5.17%). The seed extract showed the highest total phenolic content ($76.47\pm0.009$ mg GAE/g extract), DPPH RSA ($IC_{50}=52.05\pm1.61\;{\mu}g/mL$), ABTS RSA ($IC_{50}=30.94\pm0.41\;{\mu}g/mL$) and RP ($IC_{50}=87.94\pm0.37\;{\mu}g/mL$). In addition, the calyx extract also showed high antioxidant activity. On the other hand, the core extract gave the lowest TPC and all antioxidant assays. In particular, HT-29 cells showed extensive cell death when treated with $500\;{\mu}g/mL$ of calyx extracts. Thus, these results suggest that methanolic extracts of Jangseong Daebong persimmon seed and calyx may serve as a potential source of natural antioxidant for food and nutraceutical application.

Antioxidant activity and neuroprotective effects of ethanol extracts from the core of Diospyros kaki (감 심지 에탄올 추출물의 항산화 활성 및 신경세포 보호 효과)

  • This study examined the antioxidant activity and neuroprotective effects of ethanol extracts obtained from Diospyros kaki core (DCE). The total polyphenol and flavonoid contents in DCE was 786.47±15.27 and 31.14±0.82 mg/g, respectively. In addition, DCE exhibited a dose-dependent induction of radical scavenging activity, determined by 1,1-diphenyl-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonicacid) (ABTS), ferric reducing antioxidant power (FRAP), and reducing power assays. The viability of HT22 hippocampal cells was examined to investigate the neuroprotective effect of DCE. DCE treatment did not induce cytotoxicity at concentrations below 1,000 ㎍/mL. Additionally, DCE treatment in the background of H2O2 induce oxidative stress revealed a significant increase in the survival rat, indicated by increased SOD activity and decreased levels of MDA, a lipid peroxidation product. Therefore, the results suggest that DCE can be used as a source of natural antioxidants source and a therapeutic agent for the treatment of brain disorders induced by oxidative stress and neuronal damage.

Calyx-End Browning in Various Persimmon (Diospyros kaki) Cultivars and Anatomical Observations in Flesh Tissues (단감의 품종별 과정부 갈변률과 과육조직의 해부학적 관찰)

  • Calyx-end browning in sweet persimmon (Diospyros kaki) fruits is the postharvest disorder during the storage and shows different proportions by the cultivars. This study was to evaluate fruit texture characteristics at harvest and to learn how browning in fruits affects the cell structures in different cultivars. Persimmon cultivars included 'Fuyu', 'Jiro', 'Uenishiwase', 'Daiandangam', and 'Ro-19', which were harvested at the end of October in 2003 and investigated after 100 days storage. Fruit texture varied with different cultivars. 'Jiro' and 'Ro-19' fruits did not have browning symptoms while 'Daiandangam' fruits had approximately 80% browning of them. There were no visual differences for the cell structure in fruit peels between fruits without browning, such as 'Jiro' and 'Ro-19', and fruits with browning, such as 'Fuyu', 'Uenishiwase', and 'Daiandangam'. The most outer layers in a 'Jiro' fruit peel arranged one to two epidermis which could not induce browning in the tissues, while 'Fuyu' had two to three layers, inducing a browning symptom. Although there were no differences for the tissue structure between browning and normal fruits, browning fruits did not have apparent cell organelle and proceeded degradation of cell walls in the flesh.

Establishment of Dyeing Data for Silk Fabrics and Cells Using Diospyros kaki Thunb (감나무 열매를 이용한 실크 및 세포에 대한 염색 데이터 확립)

  • In this study, it was analyzed with the dyeing pattern of Diospyros kaki Thunb (persimmon) and was tried to numerically evaluate how the dyeing pattern in silk fabrics and cells was changed by different mordants. When the dyed silk fabrics were sufficiently dried, the silk fabrics were found to have a pale yellow color. Interestingly, iron II sulfate mordant changed the color change the most, silk fabrics were dyed with a color close to brown or dark purple. For numerical analysis, 19% and 62.5% color changes could be induced by sodium tartrate plus citric acid and copper acetate, respectively. Iron II sulfate induced the greatest difference than that of untreated mordants at 88%. About 5% and 10% of Chinese hamster ovary (CHO) cells were stained by sodium tartrate plus citric acid and copper acetate, respectively. The staining effect induced by iron II sulfate was about 2.4 times higher than the staining effect by sodium tartrate plus citric acid. In previous studies, staining results have been visually confirmed. However, this results not only visually confirmed the dyeing, but also quantified the color change. In particular, if numerical results are continuously integrated into big data, any researcher will be able to easily obtain similar results even if the method, time, volume, etc. are changed. In addition, the numerical data of this study is considered to be an important basis for building a database for IoT construction and computer analysis.

In Vitro Propagation of Persimmon(Diospyros kaki) by Embryo Culture (감나무의 배 배양법에 의한 기내 증식)

  • The embryos(6-8mm) isolated from seeds of Diospyros kaki were cultured on Murashlge-Skoog(MS), Woody Plant Medium(WPM), Campbell Durzen(CD), Lictvay's Medium(LM), Kao-Michaluk(KM), Nitsch, White, Heller, Wolter-Skoog(WS) media. The results showed that MS and WPM media were most suitable to the development of embryos into plantlets with length of $5.4{\pm}1.2 cm$ and 5-6 leaves. However, when LM and KM media were used, the addition of 1 to $2{\mu} moles/\ell GA_3$ was required for the germination of the embryos. Superoxide dismutase (SOD) activities, one of the changing factors in leaves according to physiological status displayed to be exceptionally significant in the leaves of plantlets germinated from seeds in potting sand soil contrary to those of cultured embryos specially around germination period.
