Ingredients of Antioxidant Activity from Calyx of Diospyros kaki Thunberg (감꼭지의 항산화 활성 성분)

  • In this study, in order to search for new functional materials from natural products, was carried out the study of antioxidant active ingredients in persimmon calyx(calyx of Diospyros kaki Thunberg). I have experimented with the effect of antioxidant activity of five different extract(MeOH, n-hexane, EtOAc, n-BuOH and $H_2O$ extract) obtained from persimmon calyx. As a result, the butanol extract, that is the main component fraction of antioxidant activity was found. Three compounds were isolated by silica gel column chromatography from the n-BuOH extract of persimmon calyx. Their structures of compound 1, 2 and 3 isolated from n-BuOH extract of persimmon calyx were identified as quercetin, (+)-catechin and gallic acid by using the TLC, $^1H$-NMR and $^{13}C$-NMR.

Differences in Tree Growth and Nutrient Absorption of Persimmon (Diospyros kaki) and Date Plum (D. lotus) Seedlings

  • D. kaki and D. lotus are used as rootstocks for astringent persimmons in Korea but characteristics of their seedlings have not been determined. In this experiment, their seeds were sown in 3-L pots on April 18 and the seedlings were grown until October 24. Growth and nutrient absorption were compared at the end of the season after destructively harvesting the seedlings. Seedling growth of D. lotus was much faster than that of D. kaki in terms of total stem length, stem diameter, and number of leaves. However, chlorophyll value and specific leaf weight were higher in D. kaki than in D. lotus. Dry weight of D. lotus was 3.6- and 3.7-fold higher than that of D. kaki in above-ground parts and the root, respectively. D. kaki seedlings were characterized by higher concentrations of N, P, K, Ca, and Mg in the leaves, stem, or the root. However, total contents of the elements were 1.8- to 3.7-fold higher in a D. lotus seedling due to its greater dry weight. Since D. lotus seedlings absorbed more inorganic elements on a tree basis and grew more vigorously than D. kaki seedlings, the level of fertilization for astringent persimmons should be adjusted depending on rootstocks to maintain the trees at the optimum vigor.

Identification of Anti-oxidant, Anti-aging, and Whitening Effects of Diospyros kaki Extracts (감 추출물의 항산화, 항노화 및 미백 기능 규명)

  • In this study, we prepared two extracts using 1,3-butylene glycol (DBG) and ethanol (DET), with Diospyros kaki and determined the anti-oxidant, anti-aging, and whitening effects in vitro. Anti-oxidant activity was measured by DPPH and superoxide dismutase (SOD) method, and as a result, both DBG and DET extracts confirmed their anti-oxidant properties by exhibiting significant DPPH scavenging and SOD-like activities. For anti-aging activity, we measured elastase and hyaluronidase inhibition, and the inhibition of MMP-1 expression. Both DBG and DET significantly inhibited elastase and hyaluronidase activities dose-dependently, and MMP-1 expression was also reduced in both extracts. We also measured the whitening effects with tyrosinase activity and melanin production, but only DBG showed a decrease in tyrosinase. In summary, D. kaki extract has strong antioxidant, anti-aging, and whitening functions, and it is believed that it can be used as a cosmetic material in the future.

Determination of the Boundary between Juvenile-Mature Wood of Diospyros kaki and Their Wood Anatomical Variations

  • Persimmon wood (Diospyros kaki) is a seasonal fruit-producing plant with a beautiful dark pattern in its wood that is suitable for high-quality furniture, sculptures and musical instruments. The utilization of persimmon wood can be improved by determining its anatomical characteristics, such as juvenile and mature wood. This study aimed to determine the boundaries between juvenile and mature wood and observe the anatomical properties of juvenile and mature wood and their variations in the axial direction. Three 30-year-old persimmon (D. kaki) trees grown in Karo, North Sumatra, Indonesia, were used in this study. The boundary between juvenile and mature wood was determined by measuring the fiber length and vessel element length from near the pith to near the bark. Anatomical observations were conducted in the juvenile and mature wood areas. The results showed that the average boundaries between juvenile and mature wood were 44.11 mm from the pith and were not significantly different in the axial direction of the trees. Furthermore, the wood anatomy categories of juvenile and mature wood differed significantly in terms of fiber diameter, fiber proportion, vessel proportion, and axial parenchyma proportion. In the axial direction, vessel diameter, ray parenchyma frequency, and ray parenchyma proportion at the base, middle, and top of the tree were significantly different.

Preparing Bi-component Dye of Unripe Diospyros kaki THUNB. Fruit and Ecklonia cava and Investigating Its Dyeing Propeties on Fabric (풋감과 감태의 이성분 복합염료 제조와 섬유 염색성 고찰)

  • This paper proposes a bi-component dye, including the unripe fruit of Diospyros kaki THUNB and Ecklonia cava, to substitute for traditional persimmon dyeing because fabrics dyed with persimmon juice become stiffer and natural persimmon is insufficient for dyeing. This study examined the color difference and fabric stiffness depending on the ratio of Ecklonia cava for in a one-bath dye solution with Diospyros kaki THUNB and showed that 6% of Ecklonia cava in the bi-component dye was the optimum for decreasing the fabric stiffness. Based on these results, a bi-component dye constituting of 94% Diospyros kaki THUNB and 6% Ecklonia cava was prepared. The particle size was found to be smaller than both single dyes and it maintained a similar amount of Catechin to Diospyros kaki THUNB dye. Finally, cotton fabric dyed with a bi-component dye was much improved in terms of the fabric hand and the surface color was similar to that of the traditional persimmon-dyed fabric. These results could help to develop the natural persimmon dyeing industry.

  • Asiacornococcus kaki (Kuwana) known as a serious pest on Diospyros kaki Thunb. has been reported erroneously as Eriococcus lagerstroemiae Kuwana. These two species were redescribed and illustrated for correct identification by the morphological characters of adult females with known host plants. Asiacornococcus Tang & Hao is a newly known genus from the Korean Peninsula.

Effects of Aqueous Extract of Diospyros Kaki Calyx on Anti-thrombotic Activity in vitro and in vivo (시체(柿蒂)의 in vitro와 in vivo 항혈전 효능 연구)

  • Objectives : The aim of this study is to research an anti-thrombus effect by Diospyros Kaki Calyx. Methods : The healthy human plasma were gained and used in vitro study such as factor X activity (FXa) inhibition, prothrombinase inhibition, prothrombin time (PT) and activated partial thromboplastin time. Fifteen SD rats were divided into three groups ; intact control group (orally administrated with distilled water 5ml/kg) and two experimental group treated with extract of diospyros kaki calyx (EKC). Experimental rats were orally 600 mg/kg concentration of EKC and 200 mg/kg concentration of EKC. After an hour from administration, we anesthetized rats and made arteriovenous (AV) shunt rat models to study weight of thrombus, took whole blood to study content of thromboxane B2 and blood clotting time. Results : In vitro, EKC significantly increased inhibitory activity of FXa, prothrombinase compared with intact control group ($^*P$ <0.05). PT and aPTT were increased in EKC treated (600 mg/kg) group compared with intact control group ($^*P$ <0.05). In vivo, blood clotting time of experiment group treated with EKC 600 mg/kg were significantly increased compare with that of intact control group (p<0.05) and content of thromboxane B2 was significantly decreased in group treated with EKC 600 mg/kg in serum. The weight of thrombus were significantly reduced in group treated with EKC 600 mg/kg compared with intact control group (p<0.05). But in vivo experiment study, those parameters of group treated with EKC 200 mg/kg were relatively decreased compared with those of intact control group without statistical significance. Conclusions : EKC has an antithrombic activity because of inhibition internal course such as FXa and prothrombin. And EKC inhibited a hole blood clotting in vivo experiment by low content of thromboxane B2.

Improving Effect on Aatopic Dermatitis with Treatment of Selected Herbs ; Polygoni Multiflori Radix, Diospyros Kaki, ilite and its Mixture in NCNga Mice (백하수오(白何首烏), 시엽(枾葉), 일라이트 조성물의 Atopy 병태모델 치료효과)

  • We observed the efficacy of natural herbs and mixture in treating atopic dermatitis using anti-human IgE treated Human HMC-I cell and NCNga mice model. First, we selected three herbs, Cynonchum witfordii, Diospyros kaki, Ilite which were used to treat skin disease in Traditional Korea Medicine. Using Human HMC-I cell treated with anti-human IgE, we investigate in vitro whether each herb effects on IL-4, IL-13, $TNF-{\alpha}$ expression and $TNF-{\alpha}$, Histamine secretion value. Finally, we conducted study whether the mixture of the selected herbs is more effective than each herb which consist the mixture and control group. The results show that the mixture is better in improving atopic dermatitis condition.

The Anticoagulant Fraction from the Leaves of Diospyros Kaki L. Has an Antithrombotic Activity

  • The leaves of Persimmon (Diospyros kaki L.) has long been used for tea in Korea since it was thought to be effective against hypertension. An anticoagulant fraction was purified through gel filtration G-100, hydrophobic, gel filtration G-150, and FPLC, Phenyl superpose column chromatographies. The purified fraction was homogenous and its Mr was estimated 10,000 Da by gel filtration and SDS-PAGE. The purified fraction was sensitive to treatment of subtilisin B, but not to heat and its activity was not changed after periodate oxidation, indicating that the activity was not due to carbohydrates. It delayed thrombin time (TT), activated partial thromboplastin time (APTT), and prothrombin time (PT) using human plasma. TT was more sensitive than APTT and PT, suggesting that the anticoagulant activity may be caused by a degradation or a defect of fibrin or thrombin. It did not cause the hydrolysis of fibrin after incubation. However, it inhibited thrombin-catalyzed fibrin formation with a competitive inhibition pattern. These results indicate that it may be an antithrombotic agent and that it is bound to fibrinogen binding sites of thrombin.
