Chemically-Induced Delayed Cutaneous Hypersensitivity in Guinea Pigs and Rabbits Experimentally Infected with Mycobacterium bovis (Mycobacterium bovis 감염(感染)이 화학적(化學的)으로 유발(誘發)된 지연형(遲延型) 피부과민증(皮膚過敏症)에 미치는 영향(影響))

  • In order to evaluate the effects of the cell-mediated immunity of the animal in the chronic diseases the guinea pigs and rabbits were inoculated with Mycobacterium bovis. After 6 weeks these animals were sensitized and challenged with 2,4-dinitrochlorobenzene. The cutaneous reactions observed in these animal species were similar each other. Macroscopic and microscopic responses in the animal experimentally infected with M. bovis were markedly reduced compared to those in the control animals. The results indicated that the cell-mediated immunity of the animals was depressed by infection of M. bovis.

  • Kinetic studies on the rates of reactions of 2, 4-dinitrochlorobenzene with p-toluidine, aniline and p-chloroaniline in $CH_3CH-CH_3OH$ binary solvent mixtures have been carried out. The experimental results have been explained in terms of the specific solvation by alcoholic hydrogen. It has been shown that the bond breaking step is rate determining in the solvent system studied and the energy barrier is getting low as the solvent changes from acetonitrile to methanol.

Effect of Cnidium officinale Mixture on Recovery Capability of DNCB-induced Allergic Contact Dermatitis in Rat

  • This study is aimed to investigate the skin thickness, plasma components, serum IgE level and antioxidant enzymes activities by mixtures of Cnidium officinale, Saururus chinensis, Houttuynia cordata and Glycyrrhiza uralensis on the allergic contact dermatitis of rat induced by 1-chloro-2,4-dinitrochlorobenzene (DNCB). Mixtures reduced the thickness of the skin and removed the dead skin cells compared to the skin of rats treated with DNCB alone. Also, these mixtures down-regulated the contents of lipid and IgE, and reduced the activities of superoxide dismutase and catalase. These results indicate that the mixtures significantly recovered the contact dermatitis induced by DNCB. In conclusion, it is thought that the mixtures could be useful for the allergic contact dermatitis.

  • Seungmagalgeuntang-gamibang has long been known to have anti-allergic effect. However, the mechanism of action of Seungmagalgeuntang-gamibang is not well investigated. The author analysed the effects of Seungmagalgeuntang-gamibang on the vascular permeability, delayed-type and contact hypersensitivities, and phagocytic function, the results obtained are as follows: 1. Administration of Seungmagalgeungtang-gamibang decreased the vascular permeability induced by serotonin in the mouse. 2. Administration of Seungmagalgeuntang-gamibang decreased the vascular permeability induced by histamine without statistical significant. 3. Administration of Seungmagalgeuntang-gamibang decreased the delayed-type hypersensitivity induced by sheep red blood cells. 4. Administration of Seungmagalgeungtang-gamibang decreased the contact hypersensitivity induced by dinitrochlorobenzene. 5. Seungmagalgeungtang-gamibang increased the phagocytic-activities of macrophages in vitro and in vivo. 6. Seungmagalgeungtang-gamibang enhanced the formation of reactive oxygen intermediates in vitro and in vivo. The above results demonstrate that Seungmagalgeuntang-gamibang suppresses the hypersensitivity reactions with increasing the phagocytic functions and formations of reactive oxygen intermediates from macrophages.

  • It seems like that the characteristics and drug-resistances of fungi are respectively different in various circumstances. Scores of chemicals were applicated to the leather-fungi in this study. M-dinitrobenzene, 2, 4-dinitrochlorobenzene and phenyl mercuric acetate inhibited the growth of Aspergilli which were isolated from Korean-leather. The minimum fungicidal limits of p-nitrophenol, 8-hydroxyquinoline and sodium-pentachlorophenolate against the Korean-originated strains are different from that of other country. In the mass-screening of fungicides, artificial "Leather-extracts media" have been designed and used, and the media contributed to screening-tests. Fat and oils which are the materials of fat-liquoring in leather manufacture affects the drugresistance of the leather-fungi. It is found that the accelerating-method on malt-agar plate is effective to determinate the fungicidal action of chemicals in short time.hort time.

  • N-Arylation reaction of nitrogen-containing heterocycles such as pyridine, nicotinamide and 4,4'-bipyridine was studied. We prepared N-2,4-dinitrophenyl derivatives initially by reacting the above heterocycles with 2,4-dinitrochlorobenzene in ethanol, and then treated the N-2,4-dinitrophenylated heterocycles with various aniline derivatives, $XC_6H_4NH_2$(X = -H, p-$CH_3$, p-$C_2H_5$, p-Cl, p-CN, p-OH, p-$OCH_3$, o-Cl, m-$CH_3$) to yield the corresponding N-arylated compounds in fairly good yields. $H^1$-nmr patterns and peak assignments of the N-arylated products were described.

The regulatory effect of AST cream on atopic dermatitis-like skin disease.

  • In this study, we investigated an inhibitory effect of AST cream on atopic dermatitis (AD) using a 2,4-dinitrochlorobenzene-induced AD murine model. Topical treatment with AST cream ameliorated the severity of AD-like lesional skin through decreases in infiltration of inflammatory cells and time of scratching behaviors. Also, AST cream reduced histamine and IgE levels in serum. The protein levels of IL-4 and IL-6 in AD-like lesional skin were suppressed by AST cream. These findings suggest that AST cream would be an alternative therapeutic agent for AD-like skin diseases.

  • Generally, berry fruits have various pharmacological activities such as anti-inflammation, anti-oxidation and anti-cancer effects. The effects of gooseberry, a berry fruits, on atopic dermatitis (AD) have not been widely examined. The aim of this present study is to investigate whether gooseberry modulates AD. We examined the pharmacological effects of gooseberry on 2, 4-dinitrochlorobenzene (DNCB)-induced AD symptoms in mice. To determine the anti-atopic mechanism of gooseberry, we investigated its effects on the production of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in PMA + ionophore -stimulated human mast cells (HMC-1). The results demonstrated that gooseberry attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, gooseberry suppressed the expression of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in stimulated HMC-1. These findings demonstrate that gooseberry is potential agent for treating AD and allergic inflammation.

Effects of Gammakdaejo-tang(GMD) on DNCB induced Atopic Dermatitis in Mice (감맥대조탕이 DNCB로 유발된 생쥐의 아토피피부염에 미치는 영향)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease with frequent relapses. This study was to investigate the effects of Gammakdaejo-tang(GMD) in DNCB induced atopic dermatitis mice. Methods : The study was divided into five comparion groups. 2,4-dinitrochlorobenzene(DNCB) solution was applied to Nc/Nga mice to induce atopic dermatitis, followed by normal group, negative control group with distilled water, positive control group with Dexamethasione and GMD 200mg/kg or 400mg/kg. The control group was orally administered 200㎕ once daily for 4 weeks. Visual skin condition, Immunoglobulin E, Histamine, Cytokine, Immune cells, Tissue biomarkers were observed. Results : As a result of the dermatitis score evaluation, it was confirmed that the GMD-administered group improved symptoms compared to the negative control group. As a result of measuring IgE, the GMD-administered group significantly decreased compared to the negative control group. As a result of measuring Histamine, GMD group except 200mg/kg of GMD significantly decreased compared to negative control group. As a result of measuring cytokine, GMD 200mg/kg significantly reduced IL-1β, IL-6 and TNF-α compared to the negative control. 400mg/kg significantly reduced IL-1β, IL-4, IL-5, IL-6, IL-10, TNF-α and significantly increased IL-2, IFNγ. As a result of confirming the immune cells, all experimental groups showed no difference in basophil, GMD group significantly reduced monocyte and eosinophil compared to negative control group, and GMD 400mg/kg group significantly reduced white blood cell and neutrophil. And significantly increased lymphocytes. As a result of measuring the gene expression level, all GMD group significantly increased TGF-β1 compared with the negative control group, and filaggrin, VEGF and EGF were significantly increased in GMD 400mg/kg group. Epidermis, dermis thickness, and eosinophil infiltration were found to be decreased in all GMD groups compared with the negative control group. Conclusions : GMD is effective in atopic dermatitis by reducing imbalance of immune response of T cells (Th1 / Th2) and reducing skin tissue damage and inflammatory response.

Inhibitory Effect of Sargassum fulvellum Water Extract on 2,4-Dinitrochlorobenzene-induced Atopic Dermatitis-like Skin Lesions in Mice (참모자반 물 추출물의 항아토피 효과)

  • This study was intended to evaluate the anti-atopic effect of Sargassum fulvellum water extract (SFWE). Atopic dermatitis (AD) was induced in BALB/c mice by spreading 2,4-dinitrochlorobenzene (DNCB) to the dorsal skin area. The production of IL-4 and total IgE of the SFWE treated group was significantly less than the DNCB only group. On the other hand, the production of the IFN-γ of SFWE treated group was greater than that of the DNCB only group. In addition, SFWE alleviated the AD symptoms when compared to the DNCB only group and reduced the epidermal thickness and the number of mast cells in histological analysis. In conclusion, these results suggest that the application of SFWE has an anti-atopic activity through the modulation of IL-4 and IFN-γ cytokines, and the total IgE in DNCB-induced BALB/c mice. Therefore, SFWE can be utilized with atopic disease therapies.
