리그닌 이양체(二量體) 모델화합물(化合物)의 합성(合成) - Veratrylglycerol-β-vanillylalcohol ether 화합물(化合物) - (Synthesis of Dimeric Lignin Model Compounds - Veratrylglycerol-β-vanillylalcohol ether compounds -)

  • Veratrylglycerol-${\beta}$-vanillylalcohol ether 결합 (${\beta}$-O-4 linkage)을 갖는 이양체(二量體) 리그닌 모델화합물(化合物)의 합성(合成)하여 각종 리그닌 반응(反應)의 시료(試料)로서 활용(活用)하고자 3,5-dimethoxyacetophenone을 출발물질(出發物質)로 하여 ${\beta}$-ring에 측소(側銷)를 갖는 이양체(二量體) 리그닌 모델화합물(化合物)(I-IV)을 합성(合成)하였고, 융점측정, $^1H(^{13}C)$-NMR 및 Mass 스펙트라에 의하여 그 화학구조(化學構造)를 동정(同定)하였다. 이 합성화합물(合成化合物)중에서 1-(3,4-dimethoxyphenyl)-2-(2'-methoxy-4'-hydroxymethylphenoxy)ropanediol-1,3[IV]는 매우 중요한 리그닌 이양체(二量體) 화합물(化合物)이다. 이러한 리그닌 합성연구(合成硏究)는 기초적(基礎的)인 연구(硏究)의 중요성(重要性) 뿐만아니라 응용면(應用面)에서도 그 활용도(活用度)가 높을 것이다.

  • 일차원 수소결합형 배위고분자 $[Cu^{II}(C_{13}H_{17}N_3OBr)(C_8H_5O_4)]{\cdot}2H_2O.CH_3OH$을 합성하여 단결정 X-선 회절 연구로 특성을 규명하였다. 단량체 단위는 사각평면의 중심 $Cu^{II}$를 갖고 있다. 네개의 배위자리 중 세자리는 $N_2O$-주개 세트를 갖는 Schiff 염기형 리간드 (4-bromo-2-[(2-piperazin-1-yl-ethylimino)-methyl]-phenol)가 차지하고, 네 번째 자리는 옆에 있는 테레프탈레이트 단위의 산소 원자가 차지한다. 두개의 인접한 중성분자는 분자간 N-H---O 및 O-H---N 수소결합에 의해 연결되어 이합체 쌍을 형성한다. 각 이합체 쌍은 불연속적인 물 및 메탄올 분자에 의해 수소결합으로 다시 연결되어 일차원 초분자 네트워크를 형성한다.

우롱차로터 새로운 Polyphenol 분리 및 통풍 예방 효과 (Isolation of a Novel Polyphenol from Oolong Tea and Its Effective Prevention of the Gout)

  • 약용 식물로부터 여러 효소의 저해제를 탐색 하였다. 한국산 우롱차엽으로부터 80%의 아세톤 추출물을 Sephadex LH-20, MCI-gel, Fuji gel 등을 사용하여 분리하였고 이 화합물은 Anisaldehyde 및 $FeCl_3$에 붉은색과 청색을 나타내었다. 이 화합물의 유도체화에 의한 기기분석 결과에서 상부는 -(-)epicatechin로 하부는 -(-)epigallocatechin 3-O-gallate 결합된 dimeric proanthocyanidin 종류인, $epicatechin-(4{\beta}{\rightarrow}8)-epigallocatechin{\;}3-O-gallate$로 화학 구조가 결정되었다. Xanthin oxidase 저해 효과에서는 $50{\;}{\mu}mole$에서 62%의 저해를 보여주어 앞으로 통풍 예방 기능성 식품 신소재로서의 이용이 가능하다는 것을 확인하였다.

Novel Macrolide Actin-inhibitors Isolated from Sea Sponges

  • Several marine toxins with macrolide structure have been found to act on actin. One of these toxins is mycalolide B isolated from the genus Mycale. This compound belongs to macrolide antibiotics and consists of tris-oxazole with strong cytotoxic activity ($IC_{50}$: 10-50 nM for growth of L1210 murine leukemia cells). This compound was found to be an actin-depolymerizing agent with the mode of action distinct from that of the known actin inhibitor, cytochalasin D. Tolytoxin, a macrolide isolated from cyano-bacteria with similar chemical structure to mycalolide B, seems to have similar effect. Another macrolide compound, aplyronine A, showed the effects similar to those of mycalolide B. Although bistheonellide A, a dimeric macrolide, did not show a severing effect, it de polymerized F-actin and sequestered G-actin by forming 1 : 2 complex with G-actins. Swinholide A has a structure and effects similar to those of bistheonel-lide A. In conclusion, mycalolide B, tolytoxin, aplyronine A, bistheonellide A and swinholide A are the members of "actin de polymerizing macrolide" the mechanism of which is different from that of cytochalasin D.halasin D.

HP0902 from Helicobacter pylori is a thermostable, dimeric protein belonging to an all-β topology of the cupin superfamily

  • Here, we report the first biochemical and structural characterization of the hypothetical protein HP0902 from Helicobacter pylori, in terms of structural genomics. Gel-permeation chromatography and dynamic light scattering indicated that the protein behaves as a dimer in solution. Circular dichroism spectroscopy showed that HP0902 primarily adopts a $\beta$-structure and the protein was highly thermostable with a denaturing temperature higher than $70^{\circ}C$. Finally, the backbone NMR assignments were obtained on the [$^{13}C,^{15}N$]HP0902 and the secondary structure was determined using the chemical shift data. Additionally, the local flexibility was assessed via a heteronuclear $^1H-^{15}N$ steady state NOE experiment. The results revealed that HP0902 would adopt a compactly folded, all-$\beta$ topology with 11 $\beta$-strands. All of the results clearly support the notion that HP0902 belongs to the cupin superfamily of proteins.

Copper(II) Coordination Polymers Assembled from 2-[(Pyridin-3-ylmethyl)amino]ethanol: Structure and Magnetism

  • The one-dimensional coordination polymers, $[Cu^{II}(L)(NO_3)_2]_n$ (1) and {$[Cu^{II}(L)(NO_3)]{\cdot}2H_2O}_{2n} (2), were synthesized from $Cu(NO_3)_2{\cdot}3H_2O$ and 2-[(pyridin-3-ylmethyl)amino]ethanol (L, PMAE) in methanol by controlling the molar ratio of copper(II) salt. Copper(II) ion in 1 has one pyridine group of PMAE whose an aminoethanol group coordinates adjacent copper(II) ion. As the pyridine group is bonded to neighboring copper(II) ion, 1 becomes a one-dimensional chain. Contrary to 1, the structure of 2 shows that the oxygen atom of ethoxide group is bridged between two copper(II) ions, which forms a dinuclear complex. Additionally, the pyridine group of PMAE included one dinuclear unit is coordinated to the other dimeric one each other, which leads to a one-dimensional polymer. Due to the structural differences, 1 exhibits weak antiferromagnetic interaction, while 2 shows strong antiferromagnetic interaction. Due to direct spin exchange via oxygen of PMAE 2 has a much strong spin coupling than 1.

Self-assembly of Amphiphilic Zinc Chlorins in an Aqueous Medium as a Model for Chlorosome of Green Photosynthetic Bacteria

  • Synthetic zinc chlorins possessing a hydrophylic polyoxyethylene chain at the 17 -position were prepared. An amphiphilic zinc chlorin possessing a single chIorin moiety showed absorption maxima at 675 nm in an aqueous medium, indicating that the zinc chIorin did not form large aggregates but a dimeric structure. In contrast, amphiphilic zinc chlorin dyads in which two zinc chlorin moieties were connected with a hydrophilic polyoxyethylene linkage showed red-shifted absorption band around 720-740 nm in an aqueous medium. The result indicated that the amphiphilic zinc chlorin dyad self-aggregated to form chlorosome-like oligomer.

  • A ${\beta}-galactosidase$ gene of Bifidobacterium adolescentis Int57 (INT57) was cloned using the shotgun method. The sequence of the ${\beta}-galactosidase$ gene existing in the sequenced 3,260-bp fragment showed higher than 40% homology with other bacterial ${\beta}-galactosidase$ genes. The expression in Escherichia coli suggested that the ${\beta}-galactosidase$ might have a monomeric, dimeric, or tetrameric protein structure. This is probably the first peer-reviewed sequence analysis of the ${\beta}-galactosidase$ gene of the genus Bifidobacterium.

  • A new ardimerin (1) was isolated from the herb of Ardisia japonica. Its structure was determined by the elucidation of spectroscopic data and a chemical transformation. Compound (1) showed potent radical scavenging effect on DPPH radical $(IC_{50},\;0.32\;{\m}M)$.

  • Reaction of barbituric acid (BA), 1,3-dimethyl barbituric acid (DMBA) and 2-thiobarbituric acid (TBA) with cyanogen bromide and aldehydes in the presence of L-(+)-tartaric acid afforded a new route for the synthesis of stable heterocyclic 5-aryl-1H,1'H-spiro[furo[2,3-d]pyrimidine-6,5'-pyrimidine]2,2',4,4',6'(3H,3'H,5H)-pentaones which is a dimeric form of barbiturate (uracil and thiouracil derivative). In the reaction of 1,3-diethyl thiobarbituric acid (DETBA) the Knoevenagel condensation and then Michael adducts were obtained under the same condition. Structure elucidation is carried out by $^1H$ NMR, $^{13}C$ NMR, FT-IR and Mass analyses. Mechanism of the formation is discussed.
