Experimental Research on the Effect of the Number of Layers by Overlay Welding of Monel-Clad Pipe on Weldability (모넬(Monel)-Clad 파이프의 오버레이 용접 적층수가 용접성에 미치는 영향에 관한 실험적 연구)

  • Overlay welding affects the chemical components and weld hardness by dilution of the lamination layer thickness, which determines the surface properties. This study experimentally investigates different numbers of layers for overlay welding monel materials, which are anti-corrosion materials. The Fe content, weldability of the base metal and monel materials, hardness, and surface flatness were examined. Each evaluation was carried out after overlay welding with three layers on the base material and pipe base material of the plate. The Fe content was evaluated by analyzing the constituents of each layer. The Fe content was satisfactory in the three layers. The weldability of the laminate specimens was evaluated by a bending test. The hardness and bead flatness of the laminate specimens were evaluated by micro Vickers and 3D measurements. The hardness was highest in the heat-affected zone with one layer, and it decreased with increasing lamination. In the case of bead flatness, there is a sharp difference in the deviation with increasing numbers of laminations, which should be considered carefully.

Extension of Storage Time of Chicon using Nanoparticle of Machilus thunbergii Extracts (후박추출물의 나노입자화를 통한 치콘의 저장기간 연장)

  • Machilus thunbergii has been showed relation to antimicrobial activity on minimal inhibitoty concentration (MIC) and colony forming inhibitory activity (CFIA) test, so that can be used to food preservatives for green vegetable. Nanoparticles has been made of edible materials. 80% of the nanoparticles has been characterized by image analyser and electron microscopy, showing in the range under 300 nm diameter. The sprayed nanoparticles remained on the surface of chicon even after washing by dilution water, then activate biological activities for storage of chicon with storing and releasement system of extracts. Chicon treated nanoparticle has been kept fresh condition about 2 months longer than 3 weeks of the non-treated control. It can be tell that treatment with nanoparticle of M. thunbergii extracts extends storage time of chicon possibly by inhibition of ethylene production through efficiency control on cell breathing.

Characterization of Cucumver mosaic virus Isolated from Hydrangea macrophylla for. otaksa (Sieb. et Zucc) Wils. (수국에서 분리한 Cucumber mosaic virus의 특성)

  • An isolate of Cucumber mosaic cucumovirus(CMV) was isolated from Hydrangea macrophylla for. otaksa(Sieb. et Zucc. ) Wils. showing mosaic symptoms, and designated as Hm-CMV. Hm-CMV was characterized by the tests of host range, physical properties, serological properties, RNA and coat protein compositions, and reverse transcription and polymerase chain reaction (RT-PCR) analysis. Twelve species in 4 families were used in the host range test of Hm-CMV and could be differentiated from Y-CMV used as a control CMV by the ringspot and line pattern on inoculated leaves of several tobacco plants. Thevirus produced local lesions on inoculated leaves of Chenopodium amarticolor, C. quinoa and Vigna unguiculata. The physical properties of the virus were as follows; thermal inactivation point(TIP) was 60$\^{C}$, dilution end point (DEP) was 10$\^$-3/, and longevity in vitro (LIP) was 3∼4 days. Hm-CMV was serologically identical to Y-CMV. SDS-polyaciylamide gel electrophoresis(SDS-PAGE) showed one major protein band of about 28 kDa. In RNA or dsRNA analysis, Hm-CMV consisted of four RNA or dsRNA species, but satellite RNA was not detected. In RT-PCR using CMV-common primer and CMV subgroup I-specific primer, bothe amplified expected size of about 490 bp and 200 bp DNA fragments from Hm-CMV, respectively. Restriction enzyme analysis of the 490 bp RT-PCR products using EcoR I and Msp I showed that Hm-CMV belonged to CMV subgroup I. However, Hm-CMV could be differentiated from other CMV subgroup I isolates by RNA fingerprinting by arbitrarily primed polymerase chain reaction (RAP-PCR).

Antifungal Activity of Bacillus vallismortis 1A against Phytopathogen (식물병원균에 대한 Bacillus vallismortis 1A 균주의 항진균 활성)

  • In order to isolate novel oligotrophic bacteria exhibiting antifungal activities, soils were collected from pepper-cultivated fields of Yeongyang, Jecheon, Nonsan, Eumsong and Goesan area in Korea. From soils in pepper cultivated area, a total of 9,354 strains were isolated as oligotrophic bacteria by the R2A dilution method. Among 9,354 oligotrohic bacteria candidates, 1A strain was selected by screening against Phytophthora capsici causing phytophthora blight of hot pepper in the greenhouse and field. The strain was identified as Bacillus vallismortis based on its 16S rDNA sequence and key characteristics as compared with those of authentic cultures of B. vallismortis(KACC 12149) and B. mojavensis(KACC 12096). The strain showed broad spectrum of antibiotic activity in vitro test, as revealed in its strong inhibitory activity to the genera Phytophthora, Collectotrichum, Botrytis and Fusarium, but not to Rhizoctonia and Magnaporthe. In pot experiments, infection rate of hot pepper in the non-treated pots was about 89%, while it was only 29% in the pots treated with 1A strain. The result indicated B. vallismortis 1A is a potential biocontrol agent for phytophthora blight of hot pepper

Forsythia suspensa Extract Has the Potential to Substitute Antibiotic in Broiler Chicken

  • Two experiments were conducted to investigate the potential for Forsythia suspensa extract (FSE) to substitute for antibiotic in broiler chicken. First, a well-diffusion assay procedure and a 2-fold dilution method were used to determine the bacteriostatic activity of FSE on Escherichia coli K88, staphylococcus aureus, and salmonella was assayed. An inhibitory effect of FSE was observed on the growth of these bacteria. This effect seems to be dose depended, which disappeared after 25.00, 12.50, 1.56 mg/ml. Second, a 42-d trial with 252 broiler chickens (d 1, $38.7{\pm}1.1$ g BW) was conducted to evaluate the effect of dietary supplementation of FSE in broiler chicken. The feeding program consisted of a starter diet from d 1 to 21 and a finisher diet from d 22 to 42. Dietary treatments included were: i) NC: negative control fed a corn-soybean meal based diet; ii) PC: positive control group fed based diet with chlortetracycline; and iii) FC: a test group fed with 100 mg FSE/kg diet. In this study growth performance did not differ among treatments during the starter period. However, dietary supplemental chlortetracycline and FSE increased (p<0.05) average daily gain (ADG), average daily feed intake (ADFI) compared with NC during the finisher and overall phase. Apparent digestibility of calcium on d 21, digestibility of energy and calcium on d 42 of FC was greater (p<0.05) than NC. Moreover, cecal Escherichia coli counts for birds from FC were lower (p<0.05) than NC. Dietary FSE supplementation also improved (p<0.05) villus height and villus height to crypt depth ratios in both duodenum and ileum and decreased (p<0.05) crypt depth in the duodenum. Duodenum villus height and villus height to crypt depth ratio in both duodenum and ileum from the FC group were also greater (p<0.05). Serum growth hormone and IGF-1 were not influenced by different treatments. Apparently, FSE has the potential to substitute for antibiotic in broiler chicken.

Investigation of Hydraulic Flow Properties around the Mouths of Deep Intake and Discharge Structures at Nuclear Power Plant by Numerical Model (수치모의를 통한 원자력 발전소 심층 취·배수 구조물 유·출입구 주변에서의 수리학적 흐름특성 고찰)

  • A cooling system is indispensable for the fossil and nuclear power plants which produce electricity by rotating the turbines with hot steam. A cycle of the typical cooling system includes pumping of seawater at the intake pump house, exchange of heat at the condenser, and discharge of hot water to the sea. The cooling type of the nuclear power plants in Korea recently evolves from the conventional surface intake/discharge systems to the submerged intake/discharge systems that minimize effectively an intake temperature rise of the existing plants and that are beneficial to the marine environment by reducing the high temperature region with an intensive dilution due to a high velocity jet and density differential at the mixing zone. It is highly anticipated that the future nuclear power plants in Korea will accommodate the submerged cooling system in credit of supplying the lower temperature water in the summer season. This study investigates the approach flow patterns at the velocity caps and discharge flow patterns from diffusers using the 3-D computational fluid dynamics code of $FLOW-3D^{(R)}$. The approach flow test has been conducted at the velocity caps with and without a cap. The discharge flow from the diffuser was simulated for the single-port diffuser and multi-ports diffuser. The flow characteristics to the velocity cap with a cap demonstrate that fish entrainment can significantly be minimized on account of the low vertical flow component around the cap. The flow pattern around the diffuser is well agreed with the schematic diagram by Jirka and Harleman.

Optimization of Electrolysis Using Sacrificial Electrode for the Treatment of Electroless Nickel Plating Wastewater (희생전극을 이용한 무전해 니켈 도금 폐수의 전기분해처리 최적화)

  • The effluent limit of nickel from electroplating wastewater has been strengthened from 5 mg/L to 3 mg/L from 2014. However, currently applied treatment process for nickel plating wastewater is unable to meet the effluent limit, most of the treatment concept conducted by treatment plant is dilution with other metal bearing wastewater. This can cause very significant impact to the environment of nickel contamination. With this connection, the feasibility test has been conducted with the use of electrolysis by using sacrificial electrodes. Experiments were conducted in synthetic and electroless nickel plating wastewater. Optimal condition of current density, pH were derived from the synthetic wastewater. It was found that the removal efficiency of nickel exceeded 94% at the operation condition of at pH 9 and the current density of $1{\sim}2mA/cm^2$. At this conditions, the iron sludge was generated very low amount. However, it was unsuccessful to meet the effluent limit by applying these treatment conditions to the real electroplating wastewater. This can be explained due to the matrix effect of other metals and anions contained real electroplating wastewater. From the result of further study, the optimal conditions for the real wastewater treatment were found out to be at pH 9, current density $6{\sim}7mA/cm^2$, for 5 minutes of operating time. At these conditions, 88% removal of nickel was achieved, which results the residual nickel concentration was below 3 mg/L.

Effect of Medium and Small Scale Shipyard Operations on Environmental Pollutions Related with Civil Appeal (중.소규모 조선소 조업이 주요 민원 유발성 환경오염에 미치는 영향)

  • The effect of small and medium scale shipyard operations on environmental pollutions related with civil appeal of nearby residential areas was estimated. PM-10(particles with an aerodynamic diameter less than or equal to 10 $\mu$m) concentrations were higher, as the sampling sites were nearer from shipyards. PM-10 concentrations of sampling sites closer to shipbuilding companies were higher during the normal operation periods than on vacation at which only small works were done. The larger amount of dry deposition was observed as the sampling sites were closer to shipyards. The odor intensity was highly influenced by ambient temperature. Almost all odor intensities measured by air dilution sensory test went over the emission standard in summer. Odor properties measured by instrumental analysis were observed to exceed the emission standard enforced to the shipyards. Odor compounds such as ammonia, sulfur compounds and trimethylamine went over the standard. Concentrations of styrene and aldehydes were lower than the olfactory threshold. All equivalent sound levels measured at daytime except the value of SP-3 pont were in excess of the environmental standard. Almost all sound levels at nighttime in all measuring sites were higher than the environmental standard. The results of this study shows counterplans need to be established for reducing the civil appeal related pollutions in the research areas.

Persistence of Chlorfenapyr in Paprika Leaf and Its Residual Biological Activity to Two Spotted Spider Mite, Tetranychus urticae (파프리카 잎 중 Chlorfenapyr의 잔류량 변화와 점박이응애에 대한 잔효 생물 활성)

  • A comparative experiment was conducted to investigate the persistence of chlorfenapyr residue and its biological activity to two spotted spider mite, Tetranychus urticae, after treatment of chlorfenapyr suspension concentrate (10% SC) on paprika plants at recommended (2000 times dilution) and double dosage. Biological half-lives of chlorpfenapyr were estimated as 8 and 5 days in leaves and fruits of paprika plants, respectively. While initial deposits of chlorfenapyr residues in leaves at recommended and double dosages were 22.22 and 37.75 ${\mu}g\;g^{-1}$ at the time of application, its residue decreased to 1.56 ${\mu}g\;g^{-1}$ and 3.62 ${\mu}g\;g^{-1}$ at 29 days after treatment, respectively. Residual biological activity of chlorfenapyr SC to T. urticae at recommended dosage lasted for 7 or 15 days on the basis of 24 or 48-hrs mortality test assayed with feeding on excised leaf chlorfenapyr-treated. The control efficacy to the mite showed a good agreement with the persistence of chlorfenapyr residues in leaves.

Application of Soil Washing Technology for Arsenic Contaminated Soil (비소로 오염된 토양에 대한 토양세척기법의 적용성 연구)

  • Several tests were conducted to optimize design parameters of soil washing technique for arsenic contaminated tailings and soils. Arsenic contaminated tailings and soils have been sampled from the N nine, Kwangwondo and the K mine, Kyungsangbukdo, respectively. According to the result of sequential extraction procedure, total arsenic concentrations were 21,028 $\pm$ 190, 443$\pm$7, and 37$\pm$3 mg/kg, for mine tailings, dry field, and river sedimentary soil, respectively. The subtotal of weakly bonded and easily releasable arsenic concentrations which were 2,284$\pm$100 (10.9%), 151$\pm$5 (34.0%), 15$\pm$3 (39.5%)mg/kg for mine tailings, dry field, and river sedimentary soil, respectively. Kinetics of arsenic extraction using NaOH showed that arsenic was extracted more than 90% after 6 hours for all samples. The optimized concentration of NaOH were 200 mM for all samples while the optimized dilution ratio were different to have 1:10 (mine tailings) and 1:5 (dry field, river sedimentary soil), respectively. Results of sequential soil washing tests using NaOH showed that arsenic concentrations obtained by Korean Standard Test Procedure were decreased to meet the regulation for both river sedimentary soil and dry field while they were not decreased largely for mine tailings, even though NaOH had much higher efficiencies of arsenic extraction than other extractants.
