A Study on Industrial Media for Production of Lactic acid in Batch and Continuous Fermentations (회분식 및 연속배양에 있어서 고농도 젖산의 생산을 위한 공업용 배지연구)

  • We have investigated industrial media for lactic acid fermentation to reduce the cost of nitrogen sources. Corn steep liquor (CSL) was successfully used at 5% (v/v) in batch fermentations. Use of soluble CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL improved the productivity about 20% with an advantage of clearer fermentation broth. Yeast extract-complemented CSL media further increased the increased the productivity. It was found that 3.1 g/L yeast extract and 5% CSL could be an effective substitute for 15 g/L yeast extract in 10% glucose medium. Brewing yeast was also used as a sole nitrogen source equivalent to 5% CSL. A continuous culture coupled with cell-recycle by microfiltration at the dilution rate of 0.05-0.065 h-1 led to the highest lactic acid productivity. Lactic acid was recovered by electrodialysis from the cell free broth. Depleted cell free broth supplemented with 5-10 g/L of yeast extract performed reasonably in batch and continuous cultures. Reuse of the fermentation broth may reduce the cost of raw materials as well as minimize the fermentation wastes.

Ampicillin and co-trimoxazole susceptibility of Shigella isolated in 1978-80 (Shigella $1978{\sim}80$년 분리주의 Ampicillin 및 Co-trimoxazole 감수성)

  • Shigella remains to be an important enteric pathogen in this country for the moment. Moreover, since 1978, most of the isolates have become resistant to ampicillin and co-trimoxazole, which used to be the drugs of choice for shigellosis. Since a disc diffusion technique alone has been used in our routine susceptibility test, the minimum inhibitory concentrations(MIC) of both ampicillin and co-trimoxazole to Shigella have never been known. In order to determine these, 195 isolates were tested by an agar dilution method, all of which were isolated at Yonsei Medical Center during the period of June 1978 to July 1980. The following results were obtained. 1. Sixty cultures(29.7%) were susceptible to ampicillin, being the MIC of 8 ${\mu}g/ml$ or less and 53(27.2%) were susceptible to co-trimoxazole, being the MIC of TMP/SMZ 4/76 ${\mu}g/ml$ or less. S. flexneri type 2 was often resistant to both antimicrobic agents. 2. An increasing rate of resistant isolates was noted, particularly in the year of 1979. 3. Many isolates were resistant to both agents. Somewhat more cultures. were ampicillin susceptible and co-trimoxazole resistant than the other way around. It seems that the determination of species or even serotypes might be of help sometimes to select proper antimicrobic agent to control the infection. A routine antimicrobic susceptibility test of Shigella to both ampicillin and co-trimoxazole would be advisable for a better selection of chemotherapeutic agent.

Electrospray-Mass Spectrometric Analysis of Plasma Pyrophosphates Separated on a Multi-Modal Liquid Chromatographic Column

  • Pyrophosphates are the key intermediates in the biosynthesis of isoprenoids, and their concentrations could reveal the benefits of statins in cardiovascular diseases. Quantitative analysis of five pyrophosphates, including isopentenyl pyrophosphate (IPP), dimethylallyl pyrophosphate (DMAPP), geranyl pyrophosphate (GPP), farnesyl pyrophosphate (FPP), and geranylgeranyl pyrophosphate (GGPP), was performed using liquid chromatography-tandem mass spectrometry (LC-MS/MS) in negative ionization mode. After dilution with methanol, samples were separated on a 3 ${\mu}m$ particle multi-modal $C_{18}$ column ($50{\times}2$ mm) and quantified within 10 min. The gradient elution consists of 10 mM ammonium bicarbonate and 0.5% triethylamine (TEA) in water and 0.1% TEA in 80% acetonitrile was used at the flow rate of 0.4 mL/min. Overall recoveries were 51.4-106.6%, while the limit of quantification was 0.05 ${\mu}g$/mL for GPP and FPP and 0.1 ${\mu}g$/mL for IPP, DMAPP, and GGPP. The precision (% CV) and accuracy (% bias) of the assay were 1.9-12.3% and 89.6-111.8%, respectively, in 0.05-10 ${\mu}g$/mL calibration ranges ($R^2$ > 0.993). The devised LC-MS/MS technique with the multi-modal $C_{18}$ column can be used to estimate the biological activity of pyrophosphates in plasma and may be applicable to cardiovascular events with cholesterol metabolism as well as the drug efficacy of statins.

  • To determine the concentration of Lactoperoxidase (LPO), an indirect enzyme-linked immunosorbant assay(ELISA) was developed. Anti-LPO egg yolk immunoglobulin(IgY) was transferred to egg yolk by immunizing of Brown hens with LPO. The titer of purified anti-LPO IgY was 1: 520,000. The immunological response of anti- LPO IgY with ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin, casein and lysozyme were evaluated, resulting that the anti-LPO IgY found to be a specific antibody toward LPO and no cross-reaction was observed against ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin, casein, and lysozyme in double immunodiffusion test and ELISA test. In indirect ELISA method, coating concentration of LPO and dilution rate of anti-LPO IgY was 0.25$\mu\textrm{g}$/mL and 1:8,000 respectively. Sensitivity in the standard curve of LPO was ranged from 0.01 to 1$\mu\textrm{g}$/mL using anti-LPO IgY.

A Study on the Development of Vinegar Beverage Using Yacon Roots(Smallanthus sonchifolius) and Analysis of Components Changes During the Fermentation (야콘 뿌리를 이용한 발효 초음료 개발 및 발효 중 성분 변화)

  • The objective of this study was to develop a functional vinegar beverage using yacon roots, and examine the components' changes during fermentation. The alcohol fermentation was conducted by inoculation of Saccharomyces cerevisiae into yacon root juice with sugars. After 10 days of fermentation at $27^{\circ}C$, yacon wine contained 13% alcohol and $7.8^{\circ}Brix$ sugar contents. The yacon wine at 5% alcohol was fermented by Acetobacter aceti(KCTC1010) with 250 rpm agitation rate at $29^{\circ}C$ for 12 days. The acidity of yacon vinegar reached 4.4% after 12 days' fermentation. Statistically (p<0.05), the amount of Acetobacter aceti, pH, alcohol, with total polyphenol content of samples decreased with acidity, whereas individual sugar contents increased. Furthermore organic acids, total sugar, phenolic acids content, total antioxidant capacity, and color were not different among the five samples. The yacon vinegar beverage was prepared with high fructose syrup through the dilution of yacon vinegar. After making the yacon vinegar beverage, the pH, acidity, total sugar, and total microbial content were examined every week over 4 weeks' storage. There were no changes in the samples during that time.

Optimization of liquid phase enzyme immunoassay for determining of progesterone (Progesterone 측정을 위한 액상(液相) 효소면역측정법(酵素免疫測定法)의 최적조건에 관한 연구)

  • This study was carried out to develop an effective liquid-phase double antibody enzyme immunoassay for determining of progesterone. The optimum conditions of assay system, 1st and 2nd antibodies, enzyme conjugate, and time reaction were invested. The bovine plasma progesterone level in dairy cattle and korean native bulls were also analyzed. The results obtained were as follows; 1. The reproducibility of petroleum ether was superior to that of ethyl ether as extract solvent of progesterone in plasma. 2. The optimum dilution rate of 1st and 2nd antibody was 30,000 and 10 times, respectively. Affer the reaction of enzyme conjugate to progesterone 1st antibody, and then 2nd antibody competition reaction was enough for over 1hr. 3. Average plasma progesterone level in 4 pregnant and 9 nonpregnant Holstein was $2.5{\pm}0.5$ and $0.7{\pm}0.2ng/m{\ell}$, respectively. Average plasma progesterone level of 10 Korean native bulls was $0.1{\pm}0.001ng/m{\ell}$ From these results, by using liquid phase double antibody enzyme immunoassay for progesterone is applicable to detect of early pregnancy diagnosis, factorial analysis of reproductive disorder, and also reproductive physiological function such as monitoring of cyclicity during the post-partum period.

Synthesis of Graphene Using Thermal Chemical Vapor Deposition and Application as a Grid Membrane for Transmission Electron Microscope Observation (열화학증기증착법을 이용한 그래핀의 합성 및 투과전자현미경 관찰용 그리드 멤브레인으로의 응용)

  • We present a method of graphene synthesis with high thickness uniformity using the thermal chemical vapor deposition (TCVD) technique; we demonstrate its application to a grid supporting membrane using transmission electron microscope (TEM) observation, particularly for nanomaterials that have smaller dimensions than the pitch of commercial grid mesh. Graphene was synthesized on electron-beam-evaporated Ni catalytic thin films. Methane and hydrogen gases were used as carbon feedstock and dilution gas, respectively. The effects of synthesis temperature and flow rate of feedstock on graphene structures have been investigated. The most effective condition for large area growth synthesis and high thickness uniformity was found to be $1000^{\circ}C$ and 5 sccm of methane. Among the various applications of the synthesized graphenes, their use as a supporting membrane of a TEM grid has been demonstrated; such a grid is useful for high resolution TEM imaging of nanoscale materials because it preserves the same focal plane over the whole grid mesh. After the graphene synthesis, we were able successfully to transfer the graphenes from the Ni substrates to the TEM grid without a polymeric mediator, so that we were able to preserve the clean surface of the as-synthesized graphene. Then, a drop of carbon nanotube (CNT) suspension was deposited onto the graphene-covered TEM grid. Finally, we performed high resolution TEM observation and obtained clear image of the carbon nanotubes, which were deposited on the graphene supporting membrane.

Inference of Sequencing Batch Reactor Process using Oxidation Reduction Potential (ORP profile을 이용한 연속 회분식 반응기(Sequencing Batch Reactor)에서 무산소공정 추론)

  • The SBR(Sequencing Batch Reactor) process is ideally suited to treat high loading wastewater due to its high dilution rate. SBR operates by a cycle of periods consisting of filling, reacting, settling, decanting and idling. The react phases such as aeration or non-aeration, organic oxidation, nitrification, denitrification and other bio-logical reactions can be achieved in a reactor. Although the whole reactions can be achieved in a SBR with time distributing, it is hard to manage the SBR as a normal condition without recognizing a present state. The present state can be observed with nutrient sensors such as ${NH_{4}}^{+}-N$, ${NO_{2}}^{-}-N$, ${NO_{3}}^{-}-N} and ${PO_{4}}^{ 3-}-P.$ However, there is still a disadvantage to use the nutrient sensors because of their high expense and inconvenience to manage. Therefore, it is very useful to use common on-line sensors such as DO, ORP and pH, which are less expensive and more convient. Moreover, the present states and unexpected changes of SBR might be predicted by using of them. This study was conducted to get basic materials for making an inference of SBR process from ORP(oxidation reduction potential) of synthetic wastewater. The profiles of ORP, DO, and pH were under normal nitrification and denitrification were obtained to compare abnormal condition. And also, nitrite and nitrate accumulation were investigated during reaction of SBR. The bending point on ORP profile was not entirely in the low COD/NOx ratio condition. In this case, NOx was not entirely removed, and minimum ORP value was presented over -300mV. Under suitable COD/NOx ratio which complete denitrification was achieved, ORP bending point was observed and minimum ORP value was under -300m V. Under high COD/NOx ratio, ORP bending point was not detected at the first subcycle because of the fast denitrification and minimum ORP value was under -300mV at the time.

  • Some of the cultural conditions were improved in order to obtain the higher biomass of Lactobacillus bulgaricus NLS-4 which has the higher lactic acid producing activity as well. Among eight media including 11% non-fat milk medium as a control, the TIP medium was selected. By a batch experiment, the maximum cell concentration could be increased to 1.0$\times$10$^{9}$ cells per $m\ell$ when the organism was grown at 38$^{\circ}C$ for 18 hours with agitation speea of 200 rpm and under the constant level of pH 6.5 con-trolled with 1 N KOH solution in the selected medium. The cell concentration was further increased to 2.3$\times$10$^{9}$ cells per me in the steady state of continuous culture at the dilution rate of 0.17 hr$^{-1}$ for 18 hours.

Isolation and Characterization of Chlorothalonil-dissipating Bacteria from Soil. (토양으로부터 Chlorothalonil 전환 미생물의 분리 및 특성)

  • Chlorothalonil is a wide-spectrum fungicide that is widely used in the world. Chlorothalonil is known as a potential toxic pollutant due to its high application rate, persistence, and toxicity to humans and other species. With the Increase of necessity of bioremediation, this study was conducted to isolate the chlorothalonil dissipation bacteria from soil. Soil samples were collected from 184 sites of farmland and wastewater disposal soil.661 strains resistant to chlorothalonil were isolated by dilution method from chlorothalonil-containing enrichment culture. After incubating at $30^{\circ}C$ in 1/10 LB media containing 10 ppm of chlorothalonil for a week, dissipation ability of chlorothalonil was investigated by HPLC. Finally, a strain SH35B, capable of dissipating chlorothalonil efficiently, was selected. The strain SH35B was identified as Ochrobactrum sp. Ten ppm of chlorothalonil In 1/10 LB media were completely dissipated by the growth of Ochrobactrum sp. SH35B for 30 h at $30^{\circ}C$. In the isolated strain, the content of glutathione and the activity of glutathione S-transferase were supposed to be ones of the Important factors for chlorothalonil dissipation and were higher than those of control strains, Escherichia coli and Bacillus subtilis.
