A numerical study was conducted to have the effect of $CO_2$ addition to fuel on the chemical reaction mechanism with the change of the initial concentration of $CO_2$ and the axial velocity gradient. From this study, it was found that there were two serious effects of $CO_2$ addition on a non-premixed flame ; a diluent effect by the reactive species reduction and chemical effect of the breakdown of $CO_2$ by the third-body collision and thermal dissociation. Especially, the chemical effect was serious at the lower velocity gradient of the axial flow. It was certain that the mole fraction profile of $CO_2$ was deflected and CO was increased with the initial concentration of $CO_2$. It was also ascertained that the breakdown of $CO_2$ would cause the increasing of CO mole fraction at the reaction region. It was also found that the addition of $CO_2$ did not alter the basic skeleton of $H_2-O_2$ reaction mechanism, but contributed to the formation and destruction of hydrocarbon products such as HCO. The conversion of CO was also suppressed and $CO_2$ played a role of a dilution in the reaction zone at the higher axial velocity gradient.
To investigate the effect of several bactericides and fungicides against fruit & vegetable decay grapefruit seed extract(GFSE) was tested in a dilution solution. GFSE was shown to be effective against decay and rind breakdown and to extend the shelf-life of fruits and vegetables during storage and tiansport. Antimicrobial efficiency of GFSE on a wide spectra of gram + & - bacteria, moulds and yeasts was demonstrated by the measurement of minimal inhibitory concentrations performed (Bacteria 10-500ppm, Fungi:250-1,000ppm, Yeasts:100-250ppm). GFSE was separated and extracted into water-soluble fraction, water-insoluble and non-dialyzed fraction to isolate the antimicrobial substances. The water-soluble fraction showed the most active antimicrobial effect. The antimicrobial substances were isolated by gas chromatography. As the result of the isolation using GC, Peak-D was found to be the antimicrobial compound in GFSE. The identification of the most antimicrobial substance was carried out by using GC-MS.
Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium responsible for a number of infections in humans that are difficult to treat, and as a result, is a substantial contributor to morbidity and mortality. In the present study, in search of natural products capable of inhibiting this multidrug-resistant bacterium, we investigated the antimicrobial activity of Lysimachia clethroides Duby root. The antibacterial activities of EtOH extract of Lysimachia clethroides Duby root and its n-hexane, EtOAc, n-BuOH and water fractions were evaluated against 15 strains of methicillin-resistant staphylococcus aureus (MRSA) and 1 standard methicillin-susceptible S. aureus (MSSA) strain by using the minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test. Antimicrobial activity of n-hexane fraction of Lysimachia clethroides Duby root was remarkable. Against the 16 strains, the minimum inhibitory concentrations (MICs) were in the range of $31.25-62.5{\mu}g/ml$ and FICI values for n-hexane fraction of Lysimachia clethroides Duby root+AM and n-hexane fraction of Lysimachia clethroides Duby root+OX were checkerboard method performed using the MRSA, MSSA and one clinical isolate strains via MICI 0.12-1 and 0.25-0.75, showing the increase of synergistic effect. When combined together, these antibiotic effects were dramatically increased. These effective combinations could be new promising agents in the management of MRSA.
The influence of oxidant addition on soot formation is investigated experimentally with ethylene, propane and mixture fuel co-flow diffusion flames. Oxidant addition into fuel shows the increase of integrated soot volume fractions for ethylene, ethylene/ethane and ethylene/methane mixture flames. However, the increase of integrated soot volume fraction with oxidant addition was not significant for propane and ethylene/propane mixture flames. This discrepancy is explained with $C_2\;and\;C_3$ chemistry at the early stage of soot formation process. The oxidant addition increases the concentration of $C_3H_3$ in the soot formation region, and therefore, enhances soot formation process. A new soot formation rate model that includes both dilution effect and chemical effect of oxygen is suggested to interpret the increase of integrated soot volume fractions with oxidant addition into ethylene. Also, the role of adiabatic flame temperature for the chemical effect of oxygen addition into fuel was reviewed. The influence of oxidant or diluent addition into fuel on soot formation process are the fuel dilution effect, the adiabatic flame temperature altering effect and/or the chemical effect of oxygen. Their relative importance could change with fuel structure and adiabatic flame temperature.

Methicillin-resistant Staphylococcus aureus (MRSA) is a serious clinical and an urgent problem worldwide. Few new drugs are available against MRSA, because MRSA has the ability to acquire resistance to most antibiotics, which consequently increases the cost of medication. In the present study, the antibacterial activity of Hydnocarpi Semen was investigated. The most effective method is to develop antibiotics from the natural products without having any toxic or side effects. Therefore, there is a need to develop alternative antibacterial drugs for the treatment of infectious diseases. Five Clinical isolates (MRSA) were obtained from five different patients at Wonkwang University Hospital (Iksan, South Korea). The Other 2 strains were ATCC 33591 (Methicillin-resistant strain) and ATCC 25923 (Methicillin-susceptible strain). Antibacterial activity (Minimal Inhibitory Concentrations, MICs) was determined by broth dilution method, disk diffusion method, MTT test, and checkerboard dilution test. Antibacterial activity of n-hexane fraction was remarkable, and had a MICs ranging from $31.25-125{\mu}g/m{\ell}$. FICI values for HFH+AM and HFH+OX were 0.13-0.19 and 0.04-0.29, showing the increase of synergistic effect. When combined together, these antibacterial effects were dramatically increased.
Enzyme-linked immunosorbent assay (ELISA) for detection of antibodies to reticuloendotheliosis virus (REV) at single serum dilution was standardized. REV HI, one of the Korean field isolates, was inoculated into chicken embryo fibroblast (CEF) cells and was harvested from the culture fluids and cells after 10 to 12 days. Viruses were purified by centrifugation at the $107,000{\times}g$ for 12 hours on 20, 30, 45% (W/V) sucrose gradient. Virus specific fraction was collected and used as ELISA antigen. To standardize ELISA, the optimal concentration of coating antigen ($1{\mu}g/well$) and conjugate (1/1000) was determined by corrected OD (OD value of positive serum-OD value of negative serum) and P/N ratio (OD value of positive serum/OD value of negative serum). To calculate ELISA titer by measuring absorbance at 1/400 single serum dilution, serum titrations were carried out for various sample sera together with standard positive and negative sera. The observed titers of serum samples were plotted against sample/positive (s/p) ratios at 1/400 serum dilution. From the above data, the ELISA titers could be calculated by the equation of $log_{10}$ ELISA titer = 2.2763 ($log_{10}$ s/p) + 3.482 (r = 0.93). For evaluating the sensitivity, the standardized method were compared with conventional agar gel immunodiffusion (AGID) test method using serum samples collected from REV infected field chicken flocks. Fifty seven of 60 samples (95%) were positive for REV by ELISA, whereas only 11 (18.3%) samples were positive by AGID test. This results suggested that the ELISA tests developed in this study could be used for detection of antibodies to REV with high sensitivity.
This study was performed to investigate the biological activities of Hericium erinaceus fruiting body, including antioxidative, fibrinolytic, thrombin inhibitory and ${\alpha}$-glucosidase inhibitory activities. The water extract of H. erinaceus was fractionated into hexane, chloroform, ethyl acetate, butanol and water fractions, and each of these fractions was individually assayed. The water extract showed high antioxidative activity at 87.78%. The water fraction showed the highest extraction yield at 35.68% (w/w). In the fraction activity tests, butanol and water fractions showed strong antioxidative activities at 87.91% and 92.27%, respectively. Assays for fibrinolytic activity indicated that only the ethyl acetate fraction has significant efficacy at 2.96 plasmin units/mL. The 10-fold dilution of hexane fraction showed high thrombin inhibitory activity, and ${\alpha}$-glucosidase inhibitory activity at 77.67% and 90.53%, respectively. In conclusion, solvent fractions of H. erinaceus can be used as materials for the development of biofunctional foods to prevent cardiovascular diseases.
The aim of this study was to investigate the antifungal activitiy of extracts from different parts of Cryptomeria japonica against Trichophyton mentagrophytes, Trichophyton rubrum and Microsporum gypseum. Disc diffusion and agar dilution methods were used for determining the antifungal activity of extracts, and their major constituents were analyzed by GC/MS. Black heartwood extract among the extracts showed the highest antifungal activity against dermatophytes. TLC was used for fractionating the effective fraction from the black heartwood extract with n-hexane and chloroform as developing solvents, and then antifungal activity of each fraction was examined against dermatophytes. As a result, seventh fraction showed the highest antifungal activity among nine fractions. The major constituent of the seventh fraction determined by GC/MS was expected to be sesquiterpene, damascone with 2 more carbon atoms. The other constituents were also identified as elemol, eudesmol and hinokione.
Antimicrobial activities were assayed through the hot-water extracts from 41 species of medicinal plants against Helicobacter pylori which is known as the ulcerogenic phathogen. Opuntia ficus-indica, Houttuynia cordata, Sinomenium acutum, and Coptis japonica showed the MIC at the concentrations less than 100 ppm, Pulsatilla koreana, Forsythia koreana, Rheum undulatum, and Perilla frutescens less than 200 ppm, Belamcanda chinensis, Arctium lappa, Cassia tora, Citrus tachibana, Siegesbeckia orientalis, and Caesalpinia sappan less than 300 ppm by the 2-fold dilution method. In disc method only three of them were confirmed to have antimicrobial activities which were increased in the order Perilla frutescens, Coptis japonica, Caesalpinia sappan. Three extracts were partitioned with chloroform, ethyl acetate and butanol in sequence and examined for the activity to inhibit H. pylori. The major ativities were observed in ethyl acetate fraction of Caesalpinia sappan, butanol fraction of Perilla frutescens, butanol and chloroform fraction of Coptis japonica. The partitioned fractions were found to have increased antimicrobial activities in all extracts. The experiments in which the extracts were added into urea R broth containing the crude urease derived from H. pylori resulted in the increase of pH and optical density at 560 nm to 8.15 and 1.7 respectively. Urease activity of H. pylori was inhibited over 80% by Caesalpinia sappan, Perilla frutescens, and Coptis japonica, of which Caesalpinia sappan suppressed up to 95%.
This study was attempted to see more clear relationships among the enterobacteria, especially between the intestinal normal flora and pathogenic bacteria. It has been known that some intestinal normal flora produce the bactrial metabolites that are harmful to other enteric bacteria. One of the metabolites is known as colicin, the protein fraction, which possesses certain degree of inhibitory effect against other bacterial growth fraction, whih possesses certain degree of inhibitory effect against other bacterial growth. As a preliminary study for a colicin purification, the antagonistic effect of E, coli to groups of Salmonella and Shigella has been studied by means of in vitro quantitative culture method. 1. E.coli showed definite inhibitory effects aganist both Salmonella and Shigella groups in the mixture of two organisms. 2. The inhibitory effects of E.coli in the E.coli-Salmonella and the E.coli-Shigella mixture occurred from 4 hours incubation following the inoculation. 3. Even the complete inhibition of pathogenic enteric bacterial growth was noticed in the E.coli-Salmonella mixture at overnight incubation. 4. Among the diluted mixtures, 1:100, 1:1,000, and 1:10,000, survival rate of pathogenic enteric bacteria in the mixtures with E.coli showed least affected at the 1:1,000 dilution. 5. It was found that the antagonistic effect aganist groups of Salmonella-shigella was depending upon the groups of the genera.
