  • Trimorphomyces papilionaceus dikaryon 으로부터 자영적인 돌연변이에 의해 분리한 이배체를 대상으로 생장속도, U. V. 광 조사 후의 생존율, U. V. 분광광도계와 Hoechst 33258 핵염색에 의한 DNA 함량등을 monokaryon 및 dikaryon 과 비교하여 결정하였다. 이배체는 dikaryon 핵의 DNA 양과 비슷하였고 monokaryon 핵의 DNA 양의 2배 정도가 되었다. Glyoxalase II 의 band 양상에서 monokayon은 한 개의 isozyme band 만을 보인 반면, 이배체와 dikaryon은 같은 위치에서 두개의 isozyme band 가 관찰되었다. 이원전개한 전기영동실험에서 dikaryon 특이 단백질은 분자량이 66.000 보다 컸으며 분자량 24,000-29,000 사이에서 이배체 특이 단백질이 존재한다.

Effects of Butachlor on the Growth of PurpleNnon-sulfur Bacteria (홍색 비유황광합성 세균에 미치는 제초제 Butachlor의 영향)

  • The effects of a herbicide butachlor[2-chloro-2', 6'-diethyl-N-(butoxymethyl) acetanilide] on the growth of the purple non-sulfur bacteria were investigated. The butachlor inhibited the growth of all species tested by 18-51%, except Rhodospirillum rubrum at concentrations of M, which would be field capacity. The photosynthetic growth rate of the species in the presence of butachlor was influenced by the nitrogen source. Cultures supplied with (NH&S04 showed a somewhat higher growth rate than those fixing dinitrogen, but they were more susceptible to butachlor (26-51%). On the contrary, butachlor enhanced the growth rate of Rhodospirillum rubrum in nitrogen gas conditions. When the culture was performed in medium with butachlor as the carbon source, the cells of fixing dinitrogen showed a higher exhaustion of butachlor than those supplemented with (N&)2S04, which exhaustion was examined by a decrease of the major absorbance at 213 nm and 260 nm. The exhaustion of butachlor as the carbon source had relation with the growth of the cells. The alkalization of culture supplemented with nitrogen gas was found in the cells treated with butachlor or untreated. The butachlor affected the carotenoid region but bacteriochlorophyll remained unaffected.

Genetic Variability of Pleurotus ostreatus Monospore Isolates by Random Amplified Polymorphic DNA Analysis (DNA 다형성 분석에 의한 느타리버섯 단포자 분리주의 유전적 변이성)

  • This study was carried out to obtain data concerning the genetic variability of Pleurotus ostreatus. Monospores of P. ostreatus were isolated and cultured to estimate differences in the rate of mycelial growth and genetic similarity among the isolates. Although the overall growth rates were slow compared to their parental dikaryon except 2-MI 4, significant differences in the rate of mycelial growth were observed among isolates. Random amplified polymorphic DNA (RAPD) analysis using twenty six random primers showed 345 polymorphic DNA bands from 35 monospore isolates and their parental dikaryon. DNA bands ranged from 0.1 to 4.0 Kb in size. Most various polymorphic DNA bands within monospore isolates were obtained when we used J (OPA-01) or W (OPB-04). The 36-MI 103 showed totally different band patterns from those of the others. RAPD analysis revealed that there is approximatly 75% genetic similarity between monospore isolates and their parental dikaryon except 36-MI 103, which showed only 49% genetic similarity. In addition, genetic similarity degrees were classified into four groups: I (parental dikaryon), II (fast growing type), III (moderate growing type), and IV (slow growing type). However, there is no correlation between mating type and mycelial growth rates.

  • Neohaplonts were obtained to improve mushroom strain from dikaryon strains of Flammulina Velutips, Ganoderma lucidum, Lyophyllum ulmarium, Pleurotus cornucopiae and Pleurotus spodoleucus by protoplast reversion. The noehaplont frequency from reversion colonies of protoplasts was 4.47-47.7%. Four more types of morphological characteristics were detected from neohaplonts of protoplast reversion in L. ulmarium, P. cornucopiae and P. spodoleucus, but one or two types were neohaplonts in F.velutipes and G. lucidum. Growth rate of neohaplonts was slow compared with dikaryon strains.

Comparison of Characteristics of Ganoderma lucidum According to Geographical Origins(III): Classification between Species of Genus Ganoderma Using Dikaryon-Monokaryon Mating

  • A Monokaryotic strain G8M without clamp connections was isolated from germinated basidiospore that was obtained from cultivated fruit body. Strain G8M was used as a tester isolate for 'dikaryon-monokaryon mating'(di-mon mating) with the strains of Ganoderma lucidum, G6 and G35(Korean wild strains), G3(Taiwan), G4(Canada), G15(America), G. oregonense G24, G. resinaceum G28, G. oerstedii G23, and G. subamboinense G29. Isolate G8M was compatible to Korean strains G6 and G35, but was incompatible to foreign strains G3, G4, or G15. Compatible reactions between strains were readily observed macroscopically. Clear barrage lines formed between incompatible strains. These clear lines were not apparent in compatible di-mon matings. The Korean strains were morphologically distinct; they did not form any chlamydospores, and stopped growth at $35^{\circ}C$. The strains of G. lucidum from Korea may be considered as different species from Taiwan, Canadian and American cultures.

Nuclear DNA inheritance of intra-specific somatic hybrids by di-mono cross in Pleurotus ostreatus based on URP-PCR analysis (URP-PCR 분석에 의한 느타리 이핵-단핵 계통간 교잡주의 핵 DNA 유전)

  • The primary objective of the present study is the characterization of the somatic hybrids of dikaryon-monokaryon (di-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus ostreatus strains with 48 intraspecific hybrids of 12 combinations between six P. ostreatus strains and one P. florida strain. The results on the experiments of hybridization rate, nuclear DNA patterns, and colors and morphology of fruit-bodies, are presented. In di-mono crosses, somatic hybrids among Pleurotus strains showed 100% of crossability as seen in those between P. ostreatus and P. florida strains indicating that the nuclei of a dikaryon is inferred to be migrated to a recipient. 87.5% of the somatic hybrids among Pleurotus strains were similar to the donor dikaryons, and 12.5% of the somatic hybrids presented DNA patterns of both parents. In 16.6% of di-mono crosses between P. ostreatus and P. florida, the nuclear DNA patterns of all hybrids showed the same or similar patterns compared to the donor dikaryons. 70.9% of the hybrids between P. ostreatus and P. ostreatus were similar to the donor dikaryons and 12.5% of them presented the DNA patterns of both parents. 79.2% of fruiting body morphology of the hybrids among Pleurotus strains were similar to the dikaryons and 20.8% of them were similar to both parents. Interestingly, the morphology of all dikaryons were dissimilar each other. All hybrid strains between dikaryon P. florida and monokaryon P. ostreatus showed the fruiting body of which colors were similar to those of the dikaryon, while the hybrids between dikaryon P. ostreatus and monokaryon P. florida were showed the combined colors of both parents. Therefore, the fruiting body color of P. florida tends to be generally dominant. In conclusion, the present study provides a way to find out and suggest superior hybrid strains using the nuclear DNA patterns of hybrids between Pleurotus strains as well as the characteristics of their fruiting bodies. The advantages of the di-mono crossing are needs to be fully utilized in mushroom breeding because it is an ideal way to develop the superior strains of Pleurotus.

The Genetic Variations of Pleurotus spp. on Subculture (느타리버섯 속(Pleurotus spp.)의 계대배양에 따른 유전적 변이)

  • The genetic variations and the rate of mycelial growth in the dikaryon and the monokaryon stages of Pleurotus spp. were surveyed during their subcultures. The highest growth rate was observed on both the 3rd and the 4th subcultures. The remarkably rapid growth rate was detected in P. ostreatus dikaryon. Genetic similarities in the dikaryon and the monokaryon of P. ostreatus were more than 57.5% and 85.7%, respectively, and those of P. eryngii were more than 85.2% and 84.8%, respectively. The genetic similarities of monokaryotic P. ostreatus were higher than those of dikaryotic. The topology of phylogenetic trees showed that the divergence and the clustering patterns of branch did not correlated with the number of subcultures. This suggests that genetic variations occur very randomly on mycelial cultures. These results suggest that the monokaryotic mycelia is genetically more stable than the dikaryotic in subcultures, and that it is very useful to stock monokaryotic mycelia for making spawns and breeding of Pleurotus spp.

Pholiota adiposa and its Related Species Collected from the Wild Forestry (야생에서 채집된 검은비늘버섯(Pholiota adiposa)균에 관한 연구)

  • Five basidiocarps of Pholiota species have been collected from the areas of BubJu Temple for last two years, and identified to those of P. adiposa or Pholiota species. The taxonomy of these basidiocarps with the morphological aspects was compared with the analysis obtained from the polymorphisms of PCR-RAPD bands made after reacted with the random primers. The polymorphic variations were observed within the species of the basidiocarps, but not between genomic DNA's of the mycelia obtained and the basidiocarps. Several different bands made from the primers (28 and 36) in PCR-RAPD reactions were identified within the genus of Pholiota and speculated to be specific for the individual basidiocarp of P. adiposa collected. The primers employed here were considered to be very useful for distinguishing the individual isolates or basidiocarps collected from the fields. Also, the basidiospores were obtained from the sporeprints of the above basidiocarps as a simple agar and confirmed with observations of clamp connection under microscopes. The matings of them indicated the 'tetrapolar' type, being different from the 'bipolar' type reported by Japanese basidiocarps of P. adiposa or P. nameko. Based on our work, the edible fungi collected were speculated to be a new breeding resource for those of Pholiota commercialized in Japan.

Studies on the Inheritance of fruitbody color in Flammulina velutipes (팽이버섯 자실체 색택의 유전연구)

  • Esterase isozyme band patterns were compared between the wild strain and commercial strain of Flammulina velutipes. Monospores were isolated from wild strain. ASI4019 and their mating types were determined. We investigated the relationship between pigmentation on the plate and fruitbody color to understand genetic relationship among F. velutipes strains. Dikaryotic strains mated between nonpigmenting strains produced white fruitbodies. However dikaryon obtained from mating between nonpigmenting monokaryon and brown pigmenting monokaryon produced brown fruitbody as the dikaryon obtained from mating of brown pigmenting monokaryons. The white fruitbody from wild strain was distinguished from that of commercial strain. When the nonpigmenting wild monokaryon was mated with commercial monokaryon, pale brown mushroom was produced. The BC1F1 was obtained by mating the above mentioned $F_1$ with commercial monokaryon. Fruitbody color of BC1F1 shared two types; one strain with all pale brown fruitbodies, and the other strain with separated eight pale brown and two mixed type involving pale brown and white fruitbodies.

Stain improvement in the white button mushroom 'Seolgang' and its varietal characteristics in Agaricus bisporus

  • The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, 'Seolgang' was developed by crossing two monokaryons 'CM020913-27' and 'SSU423-31'. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of 'Seolgang' on CDA was better at $20^{\circ}C$ and $25^{\circ}C$ when it was compared with that of '505 Ho'. The mature cap shape of new strain 'Seolgang' is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain '505 Ho', the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of '505 Ho'. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of '505 Ho'. Genetic analysis of the new strain 'Seolgang' showed different profiles compared to '505 Ho', CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.
