In this paper, we use multi-point sources to illuminate the sample in digital holographic microscopy. The resolution of digital holographic microscopy is enhanced without shifting the CCD camera. The specimen is illuminated from many directions by using multi-point sources which are easily created by a lens-array. The high frequency information of specimen can be captured at a fixed position of CCD camera. All information is then synthesized to increase the resolution.

In this paper we propose a dark-field digital holographic microscopy system based on 360-degree oblique illumination. This setup is constructed without using a dark-field objective. The principle of 360-degree oblique illumination of vortex beam and dark-field digital holographic microscopy are introduced theoretically and experimentally. By analyzing the reconstructed image of a dark-field digital hologram of a USAF 1951 target, it is proved that the imaging resolution can be improved by this method. And also, comparison and analysis are made on the reconstructed image of a bright-dark field digital hologram of a pumpkin stem slice, the result shows that the imaging contrast is also enhanced with this method, and it is effective for dark-field digital holographic microscopy imaging of large transparent biological samples.
Digital holography (DH) is a potentially disruptive new technology for many areas of imaging science, especially in microscopy and metrology. DH offers a number of significant advantages such as the ability to acquire holograms rapidly, availability of complete amplitude and phase information of the optical field, and versatility of the interferometric and image processing techniques. This article provides a review of the digital holography, with an emphasis on its applications in biomedical microscopy. The quantitative phase microscopy by DH is described including some of the special techniques such as optical phase unwrapping and holography of total internal reflection. Tomographic imaging by digital interference holography (DIH) and related methods is described, as well as its applications in ophthalmic imaging and in biometry. Holographic manipulation and monitoring of cells and cellular components is another exciting new area of research. We discuss some of the current issues, trends, and potentials.

We propose a new method for improving the phase contrast of a multiphase digital holographic microscope using a spatial light modulator (SLM). Using the SLM as the annulus, our method improves the light contrast of the object edge to achieve higher accuracy. We demonstrate a digital holographic microscopy technique that provides a 30% improvement in the phase contrast compared to conventional microscopy, which utilizes a mechanical annulus. The phase-contrast improvement allows the 3D reconstructed hologram to be determined more precisely.
In this paper, an off-axis digital holographic microscopy compensated with self-hologram rotation is presented. The process is implemented via subtracting the unwrapped phase maps of the off-axis parabolic hologram and its rotation $180^{\circ}$ to eliminate the tilt induced by the angle between the spherical object wave O and the plane reference wave R. Merit of the proposed method is that it can be done without prior knowledge of physical parameters and hence can reconstruct a parabolic hologram of $1024{\times}768$ pixels within tens of milliseconds since it doesn't require a digital reference wave. The method is applied to characterize rough gold bumps and the obtained results were compared with those extracted from the conventional reconstruction method. The comparison showed that the proposed method can characterize rough surfaces with excellent contrast and in realtime. Merit of the proposed method is that it can be used for monitoring smaller biological cells and micro-fluidic devices.
Digital holographic microscopy allows optical path difference measurement. Optical path difference depends on the both refractive index and morphology of sample. We developed a dual-wavelength in-line digital holographic microscope that can measure simultaneously the refractive index and morphology of a sample, providing highly precise three-dimensional information. Here we propose theoretical and experimental methods for dual-wavelength in-line digital holographic microscopy. The measured data were reasonable, although there was data error. By improving the experimental method, we could measure the refractive index more precisely and obtain more accurate three-dimensional information on samples.
In this study digital holographic microscopy system for measurements of 3-D velocities of particles in MR fluid is developed. Holograms are recorded using either a CCD camera with a double pulse laser or a high-speed camera with a continuous laser. To process recorded holograms, the correlation coefficient method is used for focal plane determination of particles. To remove noise and improve the quality of holograms and reconstructed images, a Wiener filter is adopted. The two-threshold and image segmentation methods are used for binary image transformation. For particle pairing, the match probability method is adopted. The developed system will be applied to measurements of the characteristics of unsteady 3-D particle velocities in MR fluids through the next stage of this study.
This paper describes in detail the processing path leading to successful phase images stitching in digital holographic microscope for the extension of the field of view. It applies FIJI Grid/Collection Stitching Plugin, which is a general tool for images stitching, non-specific for phase images. The FIJI plugin is extensively supported by aberration and phase offset correction. Comparative analysis of different aberration correction methods and data processing strategies is presented, together with the critical analysis of their applicability. The proposed processing path provides good background for statistical phase analysis of cell cultures and digital phase pathology.
본 논문에서는 홀로그램정보의 손실이 없고, CCD 화소면의 최대 활용이 가능한 in-line 형태의 광학계를 구성하고, 보다 향상된 위상 천이법을 토대로 디지털 in-line 홀로그래피 마이크로스코피를 구현하였다 이러한 구성방식으로 디지털 홀로그래피의 최대 문제인 0차 회절 항에 의한 DC잡음과 in-line 홀로그래피에서 나타나는 트윈 이미지가 효율적으로 제거됨을 실험적으로 보였고, 위상 맵핑을 통해 계량적으로 분석하였다. 또한 광학장치 컨트롤과 영상처리가 연동하여 실시간적으로 이루어지므로, 본 논문에서 연구한 DHM 방식은 앞으로 미세분석을 위한 삼차원 현미경법에 많은 응용이 이루어 질것으로 기대된다.
In this paper, we present a method to automatically quantify the three-dimensional (3D) volume of red blood cells (RBCs) using off-axis digital holographic microscopy. The RBCs digital holograms are recorded via a CCD camera using an off-axis interferometry setup. The RBCs' phase image is reconstructed from the recorded off-axis digital hologram by a computational reconstruction algorithm. The watershed segmentation algorithm is applied to the reconstructed phase image to remove background parts and obtain clear targets in the phase image with many single RBCs. After segmenting the reconstructed RBCs' phase image, all single RBCs are extracted, and the 3D volume of each single RBC is then measured with the surface area and the phase values of the corresponding RBC. In order to demonstrate the feasibility of the proposed method to automatically calculate the 3D volume of RBC, two typical shapes of RBCs, i.e., stomatocyte/discocyte, are tested via experiments. Statistical distributions of 3D volume for each class of RBC are generated by using our algorithm. Statistical hypothesis testing is conducted to investigate the difference between the statistical distributions for the two typical shapes of RBCs. Our experimental results illustrate that our study opens the possibility of automated quantitative analysis of 3D volume in various types of RBCs.
