사육수온에 따른 흰다리새우 Litopenaeus vannamei 소화기관의 trypsin 활성과 배설률 (Trypsin Activity in the Digestive Organs and Gastric Evacuation Rate of Litopenaeus vannamei at the Different Rearing Water Temperatures)

  • 흰다리새우 Litopenaeus vannamei 적정 사육기법을 개발하고자 사육수온(20, 23과 $26^{\circ}C$)에 따른 간췌장, 전장과 중장에서의 trypsin 활성 및 먹이섭취량을 조사하였다. $26^{\circ}C$에서의 흰다리새우 간췌장내의 효소활성은 $23^{\circ}C$ 구간보다 2배, $20^{\circ}C$ 구간보다 3배의 활성을 보였다. 전장에서의 효소활성은 사육수온에 관계없이 300 nmol/mg/min 과 340 nmol/mg/min로 유사한 최대 활성을 보였으나 먹이섭취 후 최대 활성이 나타나는 시간은 수온이 높을수록 짧아지는 것으로 나타났다. 중장에서의 효소활성은 먹이섭취량과 관계없이 $26^{\circ}C$ 구간에서 가장 높게 기록되어 수온이 높을수록 중장에서의 효소활성 또한 높아지는 것으로 나타났다. 체중에 대한 전장과 중장의 무게 비율의 변화, 즉 섭취된 먹이량에 의한 비율변화를 조사한 결과 전장에서의 최대 먹이섭취는 $23^{\circ}C$ 구간으로 체중의 0.6%를 섭취하였으며, $26^{\circ}C$는 0.5% 그리고 $20^{\circ}C$에서는 0.4%의 먹이를 섭취한 것으로 나타났다. 본 연구에서 최대먹이섭취는 $23^{\circ}C$에서 일어났지만 수온이 높은 구간인 $26^{\circ}C$에서 효소의 활성은 가장 높았고 배설률 또한 높아서 먹이의 잔류시간이 가장 짧은 것으로 나타났다. $20^{\circ}C$ 구간에서는 가장 적은 양의 먹이를 섭취하였고 효소활성 또한 가장 낮았으나 소화관내 먹이 잔류율이 실험기간 동안 가장 높은 것으로 나타났다. 각 수온에서 먹이섭취량에 따른 소화효소의 활성변화와 배설률에 따른 먹이 잔류시간을 고려하여 먹이공급량과 시간을 조절한다면 최적의 성장을 유도할 수 있는 방법으로 제시될 수 있다는 가능성을 확인하였다.

Chronic Administration of Monosodium Glutamate under Chronic Variable Stress Impaired Hypothalamic-Pituitary-Adrenal Axis Function in Rats

  • The hypothalamic-pituitary-adrenal (HPA) axis is the primary endocrine system to respond to stress. The HPA axis may be affected by increased level of corticotrophin-releasing factors under chronic stress and by chronic administration of monosodium glutamate (MSG). The purpose of this study was to investigate whether chronic MSG administration aggravates chronic variable stress (CVS)-induced behavioral and hormonal changes. Twenty-four adult male Sprague-Dawley rats, weighing 200~220 g, were divided into 4 groups as follows: water administration (CON), MSG (3 g/kg) administration (MSG), CVS, and CVS with MSG (3 g/kg) administration (CVS+MSG). In addition, for the purpose of comparing the effect on plasma corticosterone levels between chronic stress and daily care or acute stress, 2 groups were added at the end of the experiment; the 2 new groups were as follows: naive mice (n=7) and mice exposed to restraint stress for 2 h just before decapitation (A-Str, n=7). In an open field test performed after the experiment, the CVS+MSG group significant decrease in activity. The increase in relative adrenal weights in the CVS and CVS+MSG group was significantly greater than those in the CON and/or MSG groups. In spite of the increase in the relative adrenal weight, there was a significant decrease in the plasma corticosterone levels in the CVS+MSG group as compared to all other groups, except the naive group. These results suggest that impaired HPA axis function as well as the decrease in the behavioral activity in adult rats can be induced by chronic MSG administration under CVS rather than CVS alone.

  • Histochemical, cytochemical, and immmunocytochemical investigations were conducted to find out the cellulase activities in the accessory glands of the digestive system in the oriental land snail Nesiohelix samarangae under the LM, SEM, and TEM. It was found that the Type 1 and Type 3 cells out of five types of the epithelial cells of the digestive gland were labeled with the immunogold(protein-A gold) particles. Otherwise, none of epithelial cells of the mucus gland and the salivary gland were not labeled with the immunogold particles.

Reduction of Interlukin-8 by Peptides from Digestive Enzyme Hydrolysis of Hen Egg Lysozyme

  • Lysozyme was treated with digestive enzymes and the production of interleukin 8 (IL-8) was measured in Caco-2 cell with the peptides from lysozyme upon stimulating with lipopolysaccharide (LPS) to investigate the overall anti-inflammatory activity of lysozyme when it is in digestive tracts. Lysozyme reduced IL-8 production, and the peptides from pepsin hydrolysis of lysozyme had the similar effect. The products of trypsin digestion of lysozyme had no effect on the reduction of IL-8 production while those of pepsin-trypsin hydrolysis did. The effectiveness of lowering IL-8 production was not different by time of the peptide addition. When Caco-2 cells were pre-incubated with peptides for 24 hr, the reduction effects were observed from the peptides from pepsin hydrolysis, indicating that some of the peptides are still remaining in the cells. Therefore, it can be concluded that the IL-8 reduction effect of lysozyme against LPS still remained even after the pepsin and trypsin hydrolysis.

Effects of Supplementing Different Levels of a Commercial Enzyme Complex on Performance, Nutrient Availability, Enzyme Activity and Gut Morphology of Broilers

  • A trial was conducted to study the influence of different levels of a commercial enzyme complex on performance, nutrient availability, blood parameters, digestive tract measurements, amylase and trypsin activity of the digestive tract and gut morphology in broilers fed the typical diets in north China. There were four treatments: the control diet and the other three enzyme complex supplemented diets which were 180 mg/kg, 360 mg/kg and 720 mg/kg enzyme complex supplemented to the control diet, respectively. The birds fed the diets supplemented with 180 mg/kg and 360 mg/kg enzyme complex had better performance and nutrient availability, the activities of amylase and trypsin in the digestive tract in the two treatments were improved, the villus height and surface area of villus in the small intestine increased and the crypt depth and epithelial thickness of small intestine decreased. Relative weights of pancreas and relative weights and lengths of small intestine decreased. However, the addition of 720 mg/kg enzyme complex had no effects on these parameters and increased crypt depth and epithelial thickness of the small intestine. The data suggested that suitable supplementation of enzyme complex was beneficial for the birds, while excess enzyme complex inhibited secretion of endogenous enzyme and destroyed the structure of the small intestine.

  • The present study was conducted to find out effect of PFOS on survival, activity, growth and organ structure of the melania snail, Semisulcospira gottschei. Experimental groups were composed of one control condition and four PFOS exposure condition (0.1, 0.5, 1.0 and 5.0 mg/L). After 16 week exposure, survival rate and activity were not significantly influenced at the two lower exposure groups, 0.1 and 0.5 mg/L, but they were significantly reduced in 1.0 and 5.0 mg/L groups. Total weight and meat weight rate (MWR) were reduced in PFOS exposure group in comparison to control group. Also, histological degenerations such as acidification of mucous, necrosis and split of muscular fiber bundle, atrophy of anterior pedal gland were recognized in the foot. Hepatopancreas showed the atrophy and degeneration of the digestive cell, vacuolation of digestive gland and closure of lumen in digestive gland.

Effects of Different Levels of Supplementary Alpha-amylase on Digestive Enzyme Activities and Pancreatic Amylase mRNA Expression of Young Broilers

  • Four hundred and forty 1-day-old Arbor Acre broilers were fed commercial starter diets with 0, 250, 750 and 2,250 mg/kg of an alpha-amylase preparation from 1 to 21 days of age to investigate the effects of an exogenous enzyme on growth performance, activities of digestive enzymes in the pancreas and anterior intestinal contents and pancreatic amylase mRNA expression. Body weight gain (BWG) and average daily gain (ADG) increased linearly (p<0.01) with increasing levels of supplementary amylase but feed conversion ratio (FCR) was not affected. There was a negative quadratic change of protease and amylase in the small intestinal contents with the increase of supplementary amylase level. The activity of intestinal trypsin was also increased (p<0.05). Lipase was unaffected by amylase supplementation (p>0.05). The pancreatic protease, trypsin, and lipase were not affected by exogenous amylase levels. Consistent with the tendency for a linear depression of amylase activity, pancreatic ${\alpha}$-amylase mRNA was down-regulated by dietary amylase supplementation. The present study suggested that oral administration of exogenous amylase affected activities of intestinal enzymes and the production of pancreatic digestive enzymes in a dose-dependent manner.

White Leghorn Chick의 초기 성장단계에서 소화기관의 발달, 소장의 소화 효소 및 간 조직의 항산화 방어작용 (Post-Hatching Development of Digestive Organs, Intestinal Digestive Enzymes and Hepatic Antioxidant Defense System in White Leghorn Chicks)

  • 본 연구는 White Leghorn에서 부화 후 일령(0-d, 3-d, 7-d, 14-d 및 21-d)에 따른 소화기관의 발달과 기능 및 항산화 방어작용에 대한 변화를 조사하기 위해 실시하였다. 일령에 따른 체중은 부화 후 0, 3 및 7일령까지는 변화가 적었지만, 7일령 이후에는 유의하게(P<0.05) 증가하였다. 체중에 따른 소화 장기의 상대적 무게(g/100 g BW)에서, 간의 상대적 무게는 0일령에서 현저히 낮았지만 3~7일령에 현저히 증가한 후(P<0.05) 21일령까지 유의차 없이 점차 감소하였다. 상대적 췌장 무게 역시 간 무게의 변화와 유사한 경향을 보이며, 7일령에서 최대 발달을 보였지만(P<0.05) 그 이후에는 점차 감소하는 경향을 보였다. 상대적 소장 무게와 길이의 변화는 부화 후 급격히 증가하여 7일령에서 최대 성장을 보이고(P<0.05) 점차 감소하여 21일령 이후에는 현저히 낮아졌다. 소장 점막세포의 무게는 부화 후 현저히 증가하여(P<0.05) 14일령에 최대 발달률을 보였으나 21일령에는 감소하였다(P<0.05). 이당류 분해효소의 특이적 활성도는 0일령에 가장 낮았으나, 3일령에서 유의적으로 높은 최대 활성도 증가를 보였다(P<0.05). Sucrase 활성도는 14일령 이후 유의하게 감소하였으나(P<0.05), maltase는 지속적으로 높은 활성도를 유지하는 것으로 나타났다. Leucine aminopeptiase 특이적 활성도는 0일령부터 높게 나타나 일령에 상관없이 일정하게 유지되는 것으로 나타났다. SOD와 GST 특이적 활성도는 0일령에서 가장 낮았으나, 7일령 이후 유의적으로 높은 활성도를 보였다(P<0.05). GPX 활성도는 0, 7일령과 비교 시 14일령 이후 활성도가 현저하게 증가하였다(P<0.05). 지질 과산화(MDA) 함량은 모든 일령에서 매우 낮게 나타나 일령에 따른 차이가 없었다. 이상의 결과로 보아, White Leghorn 병아리는 부화 후 3~7일령에서 소화 장기 발달과 소화 효소 활성도가 급격히 증가하며, 7일령 이후 체 성장과 간 조직의 대사 작용이 활발해지면서 항산화 효소의 활성도가 함께 증가하는 것으로 나타났다.

선씀바귀 추출물의 항산화 및 항당뇨 효과 (Antioxidant and anti-diabetic effects of Ixeris strigosa extract)

  • 본 연구는 선씀바귀의 항산화 및 탄수화물 소화효소 억제 효과를 평가하여 제 2형 당뇨환자에게 중요한 항산화제 및 혈당 상승 억제제로서의 가능성을 타진하고자 하였다. 선씀바귀 추출물의 총 폴리페놀, 플라보노이드 함량과 in vitro 항산화 활성 및 소화효소 저해 활성을 조사한 결과 ISE가 ISW보다 총페놀과 플라보노이드 함량이 높았고 항산화 활성 및 소화효소 저해 활성 또한 높았다. ISE의 계통분획물 중에 에틸아세테이트 분획물과 부탄올 분획물의 소화효소 저해 활성이 가장 우수하고, 추출물의 항산화활성과 탄수화물 소화효소 억제효과가 총 페놀 및 플라보노이드함량과 높은 상관성을 가지는 것으로 볼 때, ISW에 비해 상대적으로 높은 ISE의 항산화 및 소화효소 저해 활성은 페놀 화합물, 특히 플라보노이드 화합물 때문임을 알 수 있었다. 따라서 ISE와 에틸아세테이트 그리고 부탄올 분획물은 우수한 항당뇨 기능성 소재라고 판단된다.
