Comparison of Digestive Function Among Rabbits, Guinea-Pigs, Rats and Hamsters. II. Digestive Enzymes and Hindgut Fermentation

  • The aim of this trial was to study the response of laboratory animals including omnivores (rats) and herbivores (rabbits, guinea pigs and hamsters) to the same level of dietary fiber on their digestive enzymes and hindgut fermentation. Ten weanling animals of each of four species, rabbits, guinea-pigs, rats and Syrian hamster, were fed a basal diet of 18% crude protein and 10% crude fiber for six weeks. The digesta and tissue of each intestinal segment were collected to measure the activity of digestive enzymes. Rabbits contained the highest secreted pepsin activity in the stomach, whereas rats contained the highest protease and ${\alpha}-amylase$ activity in the small intestine, and lower fibrous hydrolases in the hindgut than rabbits, guinea pigs and hamsters. The total VFA productions in the caecum and colon were highest in rats, followed by hamsters and rabbits, while the guinea pigs contained the lowest VFA and a different pattern of VFA molar ratio from the other laboratory animals. The degree of hindgut fermentation in these laboratory animals was in reverse to the trend for their fiber digestion.

Digestive Enzyme Activity within Crystalline Style in Three Species of Bivalves (이매패류 3종의 당면체 소화효소 활성)

  • This study was examined digestive enzyme activity in the crystalline style of the three species of bivalves. Bivalves used in this study were Tegillarca granosa (n=61), Mytilus galloprovincialis (n=30) and Saxidomus purpuratus (n=30) and collected from southern coast of Korea on May 2010. Digestive enzymes activities in the crystalline style were assayed in spectrophotometer. Amylase and cellulase occupied approximately 90% of digestive enzyme in crystalline style of T. granosa, M. galloprovincialis, and S. purpuratus. And protease activity in crystalline style of T. granosa, M. galloprovincialis and S. purpuratus showed the lowest values to 0.02, 0 and 0.08%, respectively. Digestive enzyme activity in crystalline style of three species was measured in the order of cellulase > amylase > chitinase > laminarinase.

Electrophoretic Studies on Haemolymph Protein, Digestive Fluid Protein and Digestive Amylase Activity in the Wild silkworm, Theophila mandarina. (상잠의 형액과 소화액에 있어서 단백질과 Amylase 활성에 관한 전기영동적 연구)

  • The haemolymph protein, digestive fluid proteins and digestive fluid amylase activity of wild silkworm, Theophila mandarina those of the were studied by polyacrylamide gel electrophoresis. In addition, they was also compared with silkworm. 1. 6 main protein bands in female and 7 main protein bands in male were detected in the larval haemolymph of T. mandarina where as 8 and 7 main protein bands in female and male of B. mori were observed. Some differences in the haemolymph protein ands of T. mandarina and B. mori were observed. 2. 15 protein bands and 12 protein bands were found in the larval digestive fluid of T. mandarina and B. mori respectively. Some differences in the mobility of digestive fluid proteins of T. mandarina and B. mori were noticed. 3. Larval digestive fluid amylases were anionic and moved near the tracking dye in both T. mandarina and B. mori. Mobility of the digestive fluid amylases relative to bromophenol blue were 0.019 and 0.020 in T. mandarina and B. mori respectively.

The Changes of Digestive Enzyme Activity in Early Stages of the River Puffer, Takifugu obscurus (황복, Takifugu obscurus, 초기 단계의 소화효소 변화)

  • The digestive enzyme activities such as $\alpha-amylase$, trypsin and pepsin from the laboratory-reared river puffer Takifugu obscurus were measured from the time course of 1 day until the 65 day after hatching. In the case of $\alpha-amylase$, it was showed minimum activity of 0.0493 U/mg at the total length (TL) 10 mm, and showed maximum activity of 0.1480 U/mg at 19 mmTL. Trypsin and pepsin were showed their maximum activities of 0.0264, 0.0258 U/mg and 0.0178, 0.0201 U/mg when the total length of 16 and 24 mm, and represented remarkable correlations between the changes of enzyme activity and growth rate. The ontogenetic variations of digestive enzymes were represented clearly different patterns; i.e, the pepsin showed higher activity when the periods of larva ($4\~5\;mmTL$) and juvenile II ($19\~24\;mmTL$), however, the trypsin represented maximum activity at the stages of juvenile I ($11\~16\;mmTL$) and young fish (27 mmTL), respectively.

Effects of Replacement of Fish Meal by Soy Protein Isolate on the Growth, Digestive Enzyme Activity and Serum Biochemical Parameters for Juvenile Amur Sturgeon (Acipenser schrenckii)

  • An 8-wk experiment was conducted to evaluate the effect of replacing fish meal (FM) with soy protein isolate (SPI) on the growth, digestive enzyme activity and serum biochemical parameters of juvenile Amur sturgeon (Acipenser schrenckii). SPI was used to replace 0, 25, 50, 62.5, 75, 87.5, 100% of dietary FM and 100% replacement supplemented crystalline amino acid. Healthy sturgeon with an average initial weight of $26.38{\pm}0.24$ g were randomly assigned to 24 aquaria (8 treatments with triplicates each) at an initial stocking density of 11 fish per aquarium and cultured for 8 wks. The results showed that 75.00% or more substitution resulted in a poor weight gain rate, feed conversion ratio and survival rate compared to that of fish fed the control diet (p<0.05), whereas no significant differences were observed between diets of 25.00% to 62.50% substitution. Protease, lipase and amylase activity in foregut, mid-gut and hindgut were significantly (p<0.05) decreased by diets where SPI replacement levels were 62.50% or more. Levels of serum total protein (TP) and globulin decreased significantly from 21.03, 10.34 to 14.05, 5.63 g/L with the increasing dietary SPI (p<0.05), but alkaline phosphatase activity significantly increased (p<0.05). In addition, supplemental crystalline amino acid in the FM absence diet did not improve growth performance, intestine digestive enzyme activities and serum biochemical parameters. In conclusion, the results from this study showed adverse effects of inclusion of SPI in diets on growth performance, feed utilization and serum biochemical parameters in juvenile Amur sturgeon. Based on WGR and replacement ratio presented in this report, a 57.64% replacement level was recommended.

The Effects of DL-methionine and DL-methionine Hydroxy Analogue on Growth Performance, Contents of Serum Amino Acids and Activities of Digestive Proteases in Broilers

  • This study was conducted to determine the effect of DL-methionine (Met) and DL-methionine hydroxy analogue (MHA) supplementation on the growth performance, contents of serum amino acids and activities of digestive protease in broiler chicks from 0 to 3 weeks old . There were three treatments in this study: (1) control (basal diet), (2) 0.24% Met supplementation and (3) 0.368% MHA supplementation. The results showed that Met and MHA supplementation did not significantly (p>0.05) improve feed efficiency and body weight gain for broilers from 0-1, 0-2 and 0-3 weeks of age. The serum levels of homocystine, methionine and taurine were significantly (p<0.05) higher with supplementation of Met or MHA than with control. The pepsin activity of proventriculus was increased with (p<0.05) Met supplementation at 21 days of age and with MHA supplementation at 7 and 14 days of age. The trypsin activity was also increased (p<0.05) with MHA supplementation at 7 days of age. The chymotrypsin activity in pancreas and the dipeptidase activity in small intestinal mucosa and content were not affected (p>0.05) by Met or MHA supplementation.

  • his study has been carried out to investigate amylase activity of digestive juice in the 5th day of the 5th instar influenced by the dietary composition (quantitative ratio between carbohydrate and protein) and the rearing temperature during the 4th moulting period. The larvae grew on three kinds of semi-synthetic diets. The A-diet has more carbohydrate than the others, the B-diet has carbohydrate in 1 : 2 with protein, and the C-diet has more protein than the others. All the diets were kept at 16$^{\circ}C$, 25$^{\circ}C$ and 32$^{\circ}C$ during the 4th moulting period. Amylase activity of digestive juice at the 5th day of the 5th instar was analyzed by Fuwa's method. The results were as follows. 1. Both A and C-diets were worse than B-diet in the larval growth and development. 2. The dietary composition influencing amylase activity of digestive juice was not related to the rearing temperature during the 4th moulting period. Amylase activity was stronger in C-diet, B-diet and A-diet order. 3. It was found that amylase activity at 32$^{\circ}C$ was stronger than that at 16$^{\circ}C$ in all kinds of diets. 4. There was an inter-action in amylase activity of male larval digestive juice between the dietary composition and the rearing temperature during 4th the moulting period.

  • This study has been carried out to investigate the effect of protein components of a diet on the increasing of larval body weight and diet efficiency, as well as the amylase activity of larval digestive juices during the 5th instar. Defending on the amounts of soybean meal as a protein source, the six different kind of artificial diets containing mulberry leaf powder fed to the larvae. The results obtained may be summarized as follows: 1. As the protein components of the diet were increased, the amount of increased larval body weight was also increased. 2. As the protein components of the diet were increased, both the cocoon weight and cocoon layer weight were also increased. 3. As the protein components of the diet were increased, both the amount of diet digested and coefficient of digestibility were also increased. 4. As the protein components of the diet were increased, the diet efficiency of larvae were also increased. 5. But the experimenters could not observe any correlation between the increase of protein components of a diet and the amylase activity of the digestive juices.

Effect of Dietary Xylooligosaccharide on Indigestion and Retarding Effect of Bile Acid Movement Across a Dialysis Membrane (식이 Xylooligo당의 난소화성과 담즙산 흡수 지연효과)

  • The digestibility of xylooligosaccharide(XO) by juices of the digestive tract and retardation effect of XO on the adsorption of bile acids were compared with fructooligosaccharide(FO) and isomaltooligosaccharide(IO). In vitro digestion experiments showed that any hydrolyzed products of FO, IO and XO were not detected by HPLC after reaction with saliva, pancreatic, artifical intesteinal, and large intestinal luices, and artifical sera for 4 hours at 37$^{\circ}C$. However, IO were mostly digested by the small intestinal juice, and some quantity of FO were digested. XO were not digested at all by any enzyme of digestive tract. In order to investigate retardation effect of XO on the bile acid absorption. In vitro, permeability of bile acid against dialysis membrane was determined in the mixture which contained guar gum instead of XO was set 100%. The premeability of bile acid showed about 50% in the FO and IO mixture and 43% in the XO mixture. The activity of lactase in FO group and activity of sucrase and maltase in XO group in rat small intestinal mucosa were significantly decreased. Consequently, the present results indicate that XO is indigestible in digestive tract and has retarding effect of adsorption of bile acid compared with the other oligosaccharides. The disaccharidase activity of the XO dietary group was lower than that of the other oligosaccharides dietary group. Furthermore, it was suggested that hydrolysis of sugar may be retarded in digestive tract and glucose level in blood may be controlled effectively by the XO.

Changes in Antioxidant Enzyme Activity and Physiological Responses to Cadmium and Tributyltin Exposure in the Ark Shell, Scapharca Broughtonii

  • Cadmium (Cd) and tributyltin (TBT) are common contaminants of marine and freshwater ecosystems, and can induce the formation of reactive oxygen species (ROS). These ROS can, in turn, cause oxidative stress. In the present study, we investigated time-related effects of Cd (0.05 and 0.1 ppm) and TBT (5 and 10 ppb) treatment on antioxidant enzyme activity, i.e., the activity of superoxide dismutase (SOD) and catalase (CAT) in the gills and digestive glands of the ark shell, Scapharca broughtonii. In addition, hydrogen peroxide ($H_2O_2$) concentrations, lysozyme activity, and glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) levels were measured in the hemolymph. We found that Cd and TBT treatment significantly increased antioxidant enzyme mRNA expression and activity in the digestive glands and gills in a time-dependent manner. In response to the Cd and TBT treatments, antioxidant enzymes mRNA expression and activity increased up to day 5 in the digestive glands and then decreased by day 7. In the gills, antioxidant enzymes mRNA expression and activity increased up to day 3 and then decreased by day 5. Likewise, $H_2O_2$ concentrations significantly increased up to day 5 and then decreased by day 7. Finally, lysozyme activity decreased during the experimental period, whereas GOT and GPT levels were significantly increased in a time-dependent manner. These results suggest that antioxidant enzymes play an important role in decreasing ROS levels and oxidative stress in ark shells exposed to Cd and TBT.
