Studies on the Preparation of Digestive Enzyme Tablest (IV) (소화효소제(消化酵素劑)의 제조(製造)에 관(關)한 연구(硏究) 제4보(第4報))

  • Since the active center of digestive enzymetic preparations, while under storage, lose their activity and potency by the exposure to moisture, colorization, solidifying and other physical changes. It is more important than beautiful package form that protected packaging form from moisture to get a pharmaceutical safety preparations and to maintain a definite potency. Then, in order to get a desirable conditions of storage and packaging, we used shellac and $AEA^{\circledR}$ as a coating base, and blister package, foil and bottle container as a packaging material. Temperature were set on room temperature and $37^{\circ}C$, moisture was adjusted to 40% RH and 80 % RH as a accelerated conditions. Accelerated test was carried out 6 times. The results are as follows: 1) The effect of packaging conditions give great influence on the maintenace of the stable potency. 2) Best result was produced with bottle container package. 3) $AEA^{\circledR}$ is more useful than shellac as a coating base of prevention from moisture. 4) Absorption of moisture gave considerable effects on potency and it has a limited point. 5) Difference in potency between optimal and worst condition is 472 u/2T, and difference in effective period is about 44 months.

Characteristics of Digestion Dynamics of Rice and Oat Straw Relating to Microbial Digestion in the Rumen of Sheep Given High-Concentrate Diets

  • Rumination behavior, in vivo digestibility of cell wall constituents, particle size reduction in the rumen, and retention time in the digestive tract of sheep were examined using rice and oat straw as roughage sources. The in sacco digestibility, rumen fermentation, and microbial population and internal adenosine 5-triphosphate (ATP) content were also determined under feeding conditions of high-roughage and high-concentrate diets. Chewing number and time in rumination behavior were higher with rice straw than with oat straw, while the in sacco and in vivo DMD of rice straw were consistently lower than those of oat straw. Rice straw also showed higher frequency of thinner and longer particles in the rumen contents and lower retention time in the whole digestive tract as compared to those of oat straw. Rice straw was more effective to maintain the ruminal pH than oat straw, being reflected in higher internal ATP content of large-type protozoa on the high- concentrate diet. Changes in the ruminal microflora by shifting from the low- to the high- concentrate diet were also different between rice and oat straw.

  • Dansam, the root of Salvia miltiorrhiza Bge, (Labiatae), has a bitter taste and a slightly 'cold' property, and is nontoxic. In the present study, effect of Dansam on nitric oxide (NO) generation from peritoneal macrophags was examined. Dansam had no effect on NO generation by itself, whereas recombinant interferon-${\gamma}\;(rIFN-{\gamma})$ alone had modest activity. When Dansam was used in combination with $rIFN-{\gamma}$, there was a marked cooperative induction of NO generation in a dose-dependent manner, The optimal effect of Dansam on NO generation was shown at 6 hr after treatment with $rIFN-{\gamma}$. Furthermore, the effect of Dansam was mainly dependent on Dansam-induced tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ secretion. These results suggest that Dansam induces NO generation from macrophages by the result of Dansam-induced $TNF-{\alpha}$ secretion.

Effect of Decreasing Body Weight with Plant Extracts Containing Rubi Fructus (복분자 함유 생약추출물의 체중 조절 효과에 관한 연구)

  • Obesity is one of causes of the all adult diseases. We investigated the body weight decrease effect of the selected plants by digestive enzyme activity inhibition test. In a preliminary test, Inonotus obliquus and Rubus coreanus Miq. were found to be effective. Based on this result & previous result, we manufactured the mixture of plant extracts named as Misol$^{TM}$. Misol is applied anti-obesity beverage and rice. When we administered Miso/ sub TM/ to rats, it was found to be effective in body weight decrease. Result that is experimented during 20 weeks, is effective body weight and total cholesterol, triglyceride decrease. In case of human, anti-obesity beverage administered group showed 5.65% body weight decrease, 4% waist length decrease and 6% abdomen length decrease respectively. And anti-obesity rice administered group showed 2.3 kg body weight decrease, 2.9 cm waist length decrease and 3.74 cm abdomen length decrease respectively. This body control effect was supported by plant extracts in the Misol, anti-obesity beverage and rice - administered group. From these results, we suggest that this functional food could be helpful for body weight controlling obesity.

Fermented noodles with degraded gluten (FNDG) improved digestion and gut motility in enteritis-induced mice

  • Gluten proteins are key to developing a unique quality of flour because they confer viscosity, cohesiveness, and elasticity in the dough. However, gluten can impede digestion and absorption in gluten-sensitive individuals. In this study, enteritis was induced in mice with dextran sulfate sodium (DSS) salt. The mice later received a diet consisting of 3%, 12%, or 30% fermented noodles with degraded gluten (FNDG) or 30% normal noodle flour for 8 weeks. FNDG did not alter the growth performance or body composition. However, FNDG resulted in increased amylase activity in a dose-dependent manner (p<0.001), and it also improved the digestive capacity. FNDG at 30% concentration increased the level of gastrin (p<0.01) implying increased gut motility. The serotonin receptor levels were significantly reduced by FNDG at 12% (p<0.05) and 30% (p<0.01) concentrations. These findings indicate that a diet containing FNDG could help in the recovery from intestinal inflammation with improving digestive ability and gut motility. Overall, the inclusion of degraded gluten in the diet was found to enhance digestion, gut motility, and absorption in mice.

Protective Effect of Nypa fruticans Wurmb. Water Extract on Acute Pancreatitis (해죽순 물 추출물의 급성 췌장염 억제 효과)

  • Nypa fruticans Wurmb. (NF) has been used as a folk remedy to treat inflammatory diseases in Asia. In this study, we aimed to investigate the protective effect of NF water extract on cerulein-induced acute pancreatitis (AP). To measure the protective effects of NF on AP, AP was induced via intraperitoneal injection of cerulein (50 ㎍/kg) hourly for 6 h in mice. NF water extract (100, 250, or 500 mg/kg) or saline was administrated to intraperitoneal injection 1 h before the first injection of cerulein. The mice were sacrificed at 6 h after the final injection of the cerulein. Pancreas, and blood sample were taken for further analysis. Administration of NF water extract inhibited the pancreatic injury, the elevation of pancreatic weight/ body weight ratio, and the elevation of serum digestive enzymes such as amylase and lipase during cerulein-induced AP in mice. Also, pancreas MPO activity, which represents neutrophil infiltration, was inhibited by administration of NF water extract. Taken together, administration of NF water extract reduces the severity of cerulein-induced AP, which suggests a clinical basis that NF could be a potential agent to prevent AP.

The Influence of Dietary Factors on the Incidence of Non-Insulin-Dependent Diabetes Mellitus (당뇨병 유발에 영향을 미치는 식이 요인)

  • The purpose of this study was to identify dietary factors related to the incidence of diabetes mellitus in Korea. The study consisted of 165 diabetic patients, male and female, aged 30 to 70 years and 198 healthy persons as controls. Diabetic patients who had been diagnosed with diabetes mellitus for less than five years before the study period were recruited from eight different hospitals located in Seoul, Korea. Socioeconomic status, state of illness, physical activity, food habits and food intake were assessed. Food intake was assessed by food frequency questionnaire method using a 105-food frequency questionnaire developed for diabetic patients. The stress and activity indices of diabetic patients were not significantly different from control, but alcohol consumption and smoking levels were higher in diabetics than controls. Other diseases of male diabetic patients included liver diseases, digestive system diseases, and hypertension, while those of female diabetics were hypertension, neuralgia, arthritis and digestive system diseases. These disease patterns are different from Western countries whose most common complications are hypertension and hyperlipidemia. More irregular and less varied meals were found in the diabetic group compared to the control group, suggesting that diabetic patients have generally undesirable food habits. Otherwise, food and nutrient intake of diabetics did not differ greatly from the control group. It was found that diabetic patients consumed more cereals and less fruit than the control group, and also that male diabetics consumed more alcohol. The carbohydrate : protein : fat energy distribution ratio was 61.7 : 15.8 : 22.5 in male patients and 65.1 : 14.9 : 19.7 in female patients. Discriminant analysis showed that diabetes risk factors differed with sex. In male patients, the important factors were body mass index(BMI), vitamin C intake, family diabetic history and vegetable intake, while in female patients they were BMI, cereals intake, carbohydrate intake, vitamin C intake, stress, food habits and Ca intake. These results show that excessive intake of energy and fat are not the major causal factors in Korean NIDDM. Therefore, the diabetes risk factors of Western countries may not directly apply to Koreans. Mors study is needed to clarify the risk factors of Korean NIDDM.

Changes in Esterase Isozyme Activity After Pesticides Treatment in Digestive Juice of Monochamus saltuarius (Gebler) Adult (북방수염하늘소(Monochamus saltuarius) 성충의 살충제 처리에 따른 소화 효소의 활성 변화)

  • Esterase isozymes were investigated from digestive juice of M. saltuarius adults after pesticide treatment. Twelve esterase isozymes were separated on 12% native-PAGE gel and stained with three different substrates(${\alpha}$-naphthyl acetate, ${\beta}$-naphthyl acetate, and ${\alpha}$-naphthyl butyrate). Interestingly, the isozyme of Est1(${\alpha}$-naphthyl acetate) was strongly inhibited by the carbofuran and methomyl. The Est1 activity was completely inhibited by the chlorpyrifos and partially inhibited by methidation about 70 %. In addition, eserine suppressed esterase isozyme activities of Est1 about 70% and isozyme activities of Est2, Est3, and Est4 were weakly inhibited. ${\alpha}$-pinene did not suppressed esterase isozyme activities but activities of esterases were very weakly inhibited in camphor and bornyl acetate.

Dietary Exogenous α-Amylase Modulates the Nutrient Digestibility, Digestive Enzyme Activity, Growth-Related Gene Expression, and Diet Degradation Rate of Olive Flounder (Paralichthys olivaceus)

  • In this study, a 12-week feeding experiment was conducted to characterize the effects of exogenous α-amylase on the growth, feed utilization, digestibility, plasma α-amylase activity, feed degradation rate, and fecal particle size of olive flounder (Paralichthys olivaceus). Diet was supplemented with 0 (AA0; control), 100 (AA100), 200 (AA200), or 400 (AA400) mg/kg of α-amylase, respectively. Fish (273.1 ± 2.3 g) were stocked into 12 tanks (25 fish/1,000-L tank) and 3 tanks were randomly selected for each diet group. As a result, α-amylase was found to have no significant effects (p ≥ 0.05) on the growth, feed utilization parameters, and whole-body proximate compositions. α-Amylase-treated fish exhibited only a significant increase in the apparent digestibility coefficient of carbohydrates compared to the controls. In addition, in vitro analyses revealed that α-amylase dose-dependently increased (p < 0.05) the feed degradation rate, while photographs of the intestinal content after 2, 4, and 8 h of feeding demonstrated an improved degradation rate in the α-amylase-treated groups. Plasma α-amylase content was higher in the AA200 and AA400 groups, whereas the control group produced significantly larger-sized fecal particles (90% size class) than these two groups. In the intestine, no changes were observed in the expression levels of the immune-related TNF-α, IL-1β, IL-2, immunoglobulin-M, HSP-70, lysozyme, and amylase alpha-2A. However, growth-related genes IGF-1, IGF-2, TGF-β3, and growth hormone genes were upregulated in muscle tissues. Collectively, exogenous α-amylase has positive roles in the modulation of the digestibility coefficient, blood α-amylase concentration, growth-related gene expression, and diet degradation for improved digestion in olive flounder.

The Optimal Enrichment Condition of Rotifer Brachionus rotundiformis (소화효소 활성으로 본 rotifer Brachionus rotundiformis의 적정 영양강화 조건)

  • The purpose of the study was to suggest the optimal lipid enrichment conditions used digestive enzyme activity of rotifer changing due to water temperature and salinity. The high population growth appeared at the experiment temperature more than 28 degrees highly on the culture temperature(maximum 32 degrees, 1,453 individual/mL). The fecundity was low at high temperature, and the egg ratio was high at low temperature. Population growth of 10 and 15 ppt appeared in most highly, but the fecundity and the egg ratio were high most significantly appeared in natural seawater(32 psu). The digestive enzyme activity by the culture environment mainly showed high activity in natural seawater(amylase exclusion, 15 psu). However, the TAP activity by the water temperature showed highly at the more high temperature, but the amylase and the lipase appeared at low temperature. We carried out the lipid enrichment at 20 degrees and 26 degrees in a condition of the natural seawater. Total protein, the total essential amino acids differed not significantly. The methionine content that was essential amino acids, a total lipid content, unsaturated index of fatty acids, DHA and the DHA/EPA ratio were high significantly each in $20^{\circ}C$ enrichment trial. Therefore, we could suggest the $20^{\circ}C$ and natural seawater for the optimal lipid enrichment condition in aquaculture, because methionine contents, several indexes by the lipid, TG-lipase activity, fecundity and egg ratio are high.
