In order to propose a optimum method increasing accuracy and reproducibility in analysis of urine lead levels, we compared matrix modifier addition method and microwave digestion method. The results were as follows ; 1. Where the concentration of $Pd(NO_3)_2$(matrix modifier) was $50mg/{\ell}$, and ashing temperature was $900^{\circ}C$, the results were optimum. In case of matrix modifier addition method, the average coefficient of variation (CV) of normal man was 24.1%, and lead worker was 7.9%. The average urine lead levels of standard were $10.42\mu{g}/{\ell}$ and $19.89\mu{g}/{\ell}$ , the accuracies compared to reference values were 97.0% and 92.6%, respectively. 2. Microwave digestion temperature($160~180^{\circ}C$), time(15~25min) and the ratio of urine/ashing acid(1:1~4:1) did not significantly affect lead absorbance and background absorbance. Therefore we set up a optimum analytical conditions as follows: temperature, $160^{\circ}C$; time, 15min; the ratio of urine/ashing acid, 4:1. after samples were digested by the above analytical conditions, lead absorbance and background absorbance was measured at $450^{\circ}C$ as ashing temperature. The average coefficient of variation (CV) of normal man was 12.4%, and lead worker was 6.2%. The average urine lead levels of standard urine were $10.66\mu{g}/{\ell}$ and $23.31\mu{g}/{\ell}$, the accuracies compared to reference values were 99.3% and 103.9%, respectively. From the results, we suggest that microwave digestion method is a more favorable method than matrix modifier addition method because of easiness to reduce organic matter, possibility to analysis at low temperature and accuracy.
The performance of elutriated acid fermentation with slurry-type piggery waste was investigated, especially to evaluate the effects of temperature and pre-treatment. In the first phase, the acid elutriation reactor with piggery waste after centrifugation operated at both mesophilic and thermophilic conditions to evaluate the effect of temperature. Solubilization yield($gVFAs/gSCOD_{prod.}$) and acidification rate($gVFAs/gSCOD_{prod.}$) in the thermophilic digestion were 0.45 and 0.55, which were higher than those of the mesophilic digestion, 0.25 and 0.45. In addition, the acid elutriation reactor at thermophilic temperature is more effective in removing e-coli. In the second phase, the acid elutriation reactor was fed with piggery waste before centrifugation. With piggery wastes before centrifugation, the solubilization yield and the acidificaton rate were 0.40 and 0.80, respectively, which were higher than the rates using piggery waste after centrifugation at both mesophilic and thermophilic conditions. The higher sludge volume reduction of 80% benefits sludge management. Furthermore, economical advantages can be achieved by removing the pre-treatment process, such as centrifugation. Consequently, the treatment with piggery waste before centrifugation proved to be effective. Also, the optimum temperature condition was estimated at mesophilic or thermophilic conditions, considering solubilization yields and acidification rates, though the system should be heated.
본 연구는 인체의 골중 납 농도를 측정하는데 필요한 시료 전처리법과 기기 분석법을 평가하기 위하여 수행되었다. 절단한 인간 사체 다리의 뼈를 상온에서의 산 분해법과 마이크로웨이브 분해법 (microwave oven method)을이용하여 납을 추출하였으며, 유도결합 플라즈마 질량분석법 (ICP-MS)과 흑연로 원자 흡수 분광 광도법 (GF-AAS)으로 납 농도를 측정하였다. 표준시료를 이용하여 회수율을 조사한 결과, 산분해 후 ICP-MS로 측정한 경우 높은 회수율을 보였으나, 산분해 후 GF-AAS로 측정한 것은 표준 농도보다 낮은 값을 보였으며, 마이크로웨이브 분해한 경우는 두 측정기기 모두 표준 농도보다 높은 값을 보였다. 뼈 시료의 경우, 산분해 후 ICP-MS로 측정한 납 농도는 GF-AAS의 값과 높은 상관성을 보였으나 (상관계수 = 0.983), GF-AAS가 ICP-MS보다 계통적으로 높은 값을 측정하였다. 마이크로웨이브 분해 역시 두 분석방법이 높은 상관성을 보였으나 (상관계수 = 0.950), 대체로 산분해에 의한 값보다 높은 농도를 보였다. 결론적으로, 상온에서 질산으로 분해 후 ICP-MS를 이용하여 측정한 것이 골중 납 농도를 결정하는데 편이성과 정확성 면에서 가장 효율적인 방법으로 보인다.
The objective of this study was to find an optimum methane yield condition in anaerobic digestion of sewage sludge with microwave pretreatment. The pretreatment process was carried out using a lab scale industrial microwave unit (2,450 MHz frequency). The digestion efficiency of pretreated sludge was evaluated by biochemical methane potential (BMP) test. Box-Behnken design and Response Surface Analysis (RSA) were applied to determine the optimal combination of sludge mixing ratio (0 to 100%), power (400 to 1600 W), holding time (0 to 10 min) and pretreatment temperature (60 to 100℃). BMP test results showed that Volatile Solid (VS) removal efficiency was up to 48% at a condition of 0% for mixing ratio, 1600 W for power, 5 min for holding time, and 80℃ for pretreatment temperature. Methane production was up to 832.3 mL/g VSremoved at a condition of 50% for mixing ratio, 1000 W for power, 5 min for holding time, and 80℃ for pretreatment temperature. The results of the variance analysis (ANOVA) showed that the p-value of the power and pretreatment temperature among the independent variables were significant (p<0.05), and in particular, the pretreatment temperature significantly affected on the solubilization and methane production. The optimum condition for the maximum methane yield (847 mL/g VSremoved) was consist of 38.4% of mixing ratio, 909.1 W of power, 4.1 min of holding time, and 80℃ of temperature within the design boundaries.
In experiment 1, eighty crossbred steers ($239{\pm}15kg$) were used in a 229-d experiment to evaluate the effects of increasing levels of enzymatically hydrolyzed yeast (EHY) cell wall in diets on growth performance feedlot cattle during periods of elevated ambient temperature. Treatments consisted of steam-flaked corn-based diets supplemented to provide 0, 1, 2, or 3 g EHY/hd/d. There were no effects on growth performance during the initial 139-d period. However, from d 139 to harvest, when 24-h temperature humidity index averaged 80, EHY increased dry matter intake (DMI) (linear effect, p<0.01) and average daily gain (ADG) (linear effect, p = 0.01). There were no treatment effects (p>0.10) on carcass characteristics. In experiment 2, four Holstein steers ($292{\pm}5kg$) with cannulas in the rumen and proximal duodenum were used in a $4{\times}4$ Latin Square design experiment to evaluate treatments effects on characteristics of ruminal and total tract digestion in steers. There were no treatment effects (p>0.10) on ruminal pH, total volatile fatty acid, molar proportions of acetate, butyrate, or estimated methane production. Supplemental EHY decreased ruminal molar proportion of acetate (p = 0.08), increased molar proportion of propionate (p = 0.09), and decreased acetate:propionate molar ratio (p = 0.07) and estimated ruminal methane production (p = 0.09). It is concluded that supplemental EHY may enhance DMI and ADG of feedlot steers during periods of high ambient temperature. Supplemental EHY may also enhance ruminal fiber digestion and decrease ruminal acetate:propionate molar ratios in feedlot steers fed steam-flaked corn-based finishing diets.
본 연구에서는 인산염계 모나자이트정광을 수산화나트륨 용액에 의한 분해와 영산 침출공정을 이용하여 최적의 희토류 침출조건을 확립하고자 하였다. 모나자이트정광의 hot digestion 분해 및 염산침출 실험용 수행결과 알카리 분해는 NaOH/TREO 몰비 15, 분해반응온도 $140^{\circ}C$, 분해반응시간 2시간 이상의 조건에서 분해율 90% 이상을 나타내었다. 그리고 알카리 분해 산물의 염산침출 반응 최적조건은 염산농도 6N, 침출반응 온도 $70^{\circ}C$, 침출반응시간 2시간 및 광액농도 15% 내외가 가장 효과적이었고, 이 때 회토류 침출율은 90% 이상 이었다.

AD has been one of the leading technologies that can make a large contribution to producing renewable energy and to reducing $CO_2$ and other GHG emission, it is becoming a key method for both waste treatment and recovery of a renewable fuel and other valuable co-products. A classification of the basic AD technologies for the production of biogas can be made according to the dry matter of biowaste and digestion temperature, which divide the AD process in wet and dry, mesophilic and thermophilic. The biogas produced from AD plant can be utilized as an alternative energy source, for lighting and cooking in case of small-scale, for CHP and vehicle fuel or fuel in industrials in case of large-scale. This paper provides an overview of the status of biogas production and utilization technologies.

Organic waste used as a feedstock in the anaerobic digestion (AD), it includes carbon and nitrogen. Carbon and nitrogen have an effect on the various digestive characteristics during AD, however, the study is rare about those of the interaction. This study investigates the influence of carbon type and carbon to nitrogens (C/N ratios) on the AD characteristics of organic waste. Experimental treatments involved a combination of three carbon types with three C/N ratios. The AD tests were carried out using a 125-mL serum bottle at a constant temperature of 37℃ and moisture 95% for 18 days. Degradation pattern shows the difference among three-carbon treatments, the starch group was faster than other groups. Maximum methane production date was similar between starch (9.96 ± 0.05 day) and xylan group (10.0 ± 0.52 day), those of the cellulose group (14.6 ± 1.80 day) was slower than other groups (p < 0.05). The lag phase was only affected by the carbon type (p < 0.05). Ammonia nitrogen was mainly affected by nitrogen concentration regardless of carbon type (p < 0.05). This study showed that xylan is useful as feedstock in order to decrease the lag phase, and it showed that ammonia was independently affected by the nitrogen concentration.
The carbon and nitrogen removal process using PVA-gel (Polyvinyl Alcohol) immobilized microorganisms was studied. The process has been operated under diverse process conditions for 12 months. The process consists of denitrification with internal recycle of 300%, nitrification, aerobic digestion reactors and settling tank. Nitrificatoin and nitrogen removal efficiency according to gel packing ratio and temperature were studied. Yield for Heterotrophs ($Y_H$), decay coefficient for Heterotrophs($b_H$) in aerobic digestion reactor were determined to seize sludge reduction mechanism and compared with typical data of activated sludge process. Then SRT in aerobic digestion reactor was determined on an experimental basis and sludge reduction efficiency was calculated. The process was implemented successfully with sludge reduction efficiency of 92.0~98.5% on a basis of biomass.
