Differences of Antioxidative Level in the Leaves and Seed Germinability among Five Natural Populations of Hibiscus hamabo

  • Variations in antioxidant capacity of leaves and characteristics of seed germination among five Hibiscus hamabo populations in Korea were evaluated. While the leaves of H. hamabo from Shincheon contained highest Na concentration, those from Hado and Ohzo does lowest. MDA content and SOD activity didn't show significant difference among populations, but total protein and anthocyanin content were significantly different among five populations. The filled seed rates of five populations were ranged from 92.0 to 98.7%, and the moisture contents of seeds from Shinyang and Soando were lower than that of R. syriacus (5.18%). The fresh weights of H. hamabo from Hado and Ohzo were the highest and those of seeds from Shinyang and Soando were the lowest. Significant variations among H. hamabo populations were detected for all seed germination characters: percentage of germination, average germination time and germination rate and uniformity. SOD activities of seeds from Onpyoung and Soando were the highest of five populations, and total protein concentrations of seeds in Ohzo and Shinyang populations were higher than those of other populations. H. hamabo is considered exposed to salt stress and all characteristics of seed germinations were inferior to other species.

Genetic Diversity of Wild and Cultivated Populations of American Ginseng (Panax Quinquefolium) from Eastern North America Analyzed by RAPD Markers

  • The objective of this study was to assess genetic diversity among 6 different wild ginseng populations from New York, Kentucky, North Carolina, Pennsylvania, Tennessee and Virginia, and to compare these wild populations to one cultivated population. RAPD markers were used to estimate the genetic difference among samples from the 7 populations. The 64 random primers were screened, and 15 primers were selected which exhibited the 124 highly reproducible polymorphic markers. The ratio of discordant bands to total bands scored was used to estimate the genetic distance within and among populations. Multidimensional scaling (MDS) of the relation matrix showed distinctive separation between wild and cultivated populations. The MDS result was confirmed using pooled chi-square tests for fragment homogeneity. This study suggests that RAPD markers can be used as population-specific markers for American ginseng.

  • Three buffalo populations viz. Bhadawari, Tarai and local buffaloes of Kerala were genotyped using 24 heterologous polymorphic microsatellite loci. A total of 140 alleles were observed with an average observed heterozygosity of 0.63. All the loci were neutral and 18 out of the 24 loci were in Hardy Weinberg Equilibrium. The $F_{IS}$ values (estimate of inbreeding) for 16 loci in all the three populations were negative. This indicated lack of population structure in the three populations. The effective number of immigrants was 5.88 per generation between the Tarai and Bhadawari populations which was quite high suggesting substantial gene flow. The genetic distances revealed closeness between the Tarai and Bhadawari populations which was expected from geographical contiguity. The FST values were not significantly different from zero showing no population differentiation. The Correspondence Analysis based on the allelic frequency data clustered the majority of the Tarai and Bhadawari individuals as an admixture.

  • An enzyme analysis of the liver fluke, Clonorchis sinensis from Kimhae, Korea and from Shenyang, China was conducted using a horizontal. starch gel electrophoresis in order to elucidate their genetic relationships. A total of eight enzymes was employed from two different kinds of buffer systems. Two loci from each enzyme of aconitase and esterase (${\alpha}-Na{\;}and{\;}{\beta}-Na$) : and only one locus each from six enzymes, gluucose-6-phosphate dehydrogenase (G6PD), ${\alpha}-glycerophosphate$ dehydrogenase (GPD), 3-hydroxybutyrate dehydrogenase (HBDH), malate dehydrogenase (MDH), phosphoglucose isomerase (PGI), and phosphoglucomutase (PGM) were detected. Most of loci in two populations of C. sinensis showed homozygous monomorphic banding patterns and one of them, GPD was specific as genetic markers between two different populations. However, esterase (${\alpha}-Na$), GPD, HBDH and PGI loci showed polymorphic banding patterns. Two populations of C. sinensis were more closely clustered within the range of genetic identity value of 0.998-1.0. In summarizing the above results, two populations of C. sinensis employed in this study showed mostly monomorphic enzyme protein banding patterns, and genetic differences specific between two populations.

Morphological and genetic variability among Ecklonia cava (Laminariales, Phaeophyceae) populations in Korea

  • Ecklonia cava Kjellman is a common kelp found in shallow subtidal in warm-temperate waters in the northwest Pacific Ocean. This species has shown substantial morphological variation along with subsistence in different locations and local environments. We quantified the magnitude of morphological variation of E. cava from six populations along ~700 km of coastline from Jeju Island to Dokdo in Korea. In addition, we examined genetic distance among the populations using random amplified polymorphic DNA (RAPD) analysis. Most morphological characteristics investigated were significantly different among locations. Multivariate analyses indicated two phenetically distinct groups (nearshore, sheltered vs. offshore, exposed), indicating wave exposure with turbidity are presumably major factors for the separation. With RAPD data, results of Nei's diversity (H) and AMOVA showed considerable variations in within- and between-populations. Pairwise ${\Phi}_{ST}$ and $N_m$ values indicated moderate gene flow between the six locations. Results of Nei's analysis revealed three genetically distinct groups, not consistent with the morphological groupings, indicating that a time gap may exist between morphological and genetic variations. This study also suggests dispersal distance of this kelp may be longer than what is commonly thought and genetic similarity in the populations was largely reflected by the direction of ocean current rather than just geographical distance.

  • 유기성 폐기물 처리를 위해 강원도 홍천과 충청도 영동에서 사육되고 있는 두 지렁이 집단의 생식력 차이, 생식적 격리 정도 및 CO I 유전자의 서열을 비교하였다. 두 집단의 생식력에 큰 차이가 없었으며, 두 집단 간에 생식적 격리도 전혀 이루어지지 않았다. CO I 유전자 염기 서열을 분석하여 두 집단의 종을 동정한 결과 붉은줄지렁이 또는 줄지렁이로 확인되었고, 두 집단의 유연관계가 매우 가까운 것으로 나타났다. 따라서 두 집단은 같은 지렁이 종임을 강력히 시사하였다.

Factors Affecting the Vineyard Populational Diversity of Plasmopara viticola

  • Vitis vinifera is very susceptible to downy mildew (Plasmopara viticola). A number of authors have suggested different genetic populations of this fungus exist in Europe, each showing a different degree of virulence. Work performed to date indicates this diversity to be the result of different factors. In areas where gene flow is greater and recombination more frequent, the diversity of P. viticola appears to be wider. In vineyards isolated by geographic barriers, a race may become dominant and produce clonal epidemics driven by asexual reproduction. The aim of the present work was to identify the conditions that influence the genetic diversity of P. viticola populations in the vineyards of northwestern Spain, where the climatic conditions for the growth of this fungus are very good. Vineyards situated in a closed, narrow valley of the interior, in more open valleys, and on the coast were sampled and the populations of P. viticola detected were differentiated at the molecular level through the examination of microsatellite markers. The populations of P. viticola represented in primary and secondary infections were investigated in the same way. The concentration of airborne sporangia in the vegetative cycle was also examined, as was the virulence of the different P. viticola populations detected. The epidemiological characteristics of the fungus differed depending on the degree of isolation of the vineyard, the airborne spore concentration, and on whether the attack was primary or secondary. Strong isolation was associated with the appearance of dominant fungal races and, therefore, reduced populational diversity.

분흥성게의 지역별 개체군 성장에 관한 연구 (Growth Variation among the Different Populations of Sea Urchin, Pseudocentrotus depressus)

  • 분흥성게의 양식군, 방류군, 야생군에 대하여 생식판에 나타나는 색소대를 연륜으로 하여, cluster 분석에 의한 지역별 성장 양상을 파악하고, 군집상호의 성장특성을 비교검토 하였다. 1. 단일연급군으로 구성된 양식군은 매년 9월 25일에서 10월 5일 사이에 출생한 개체들로서, 약 9-10개월후의 표본군에서는 $r_1$은 전혀 나타나지 않았고, 출생한지 1년 10-11개월의 표본군에서는 $r_1,$ 2년 9-10개월생에서는 $r_1$$r_2$가 형성되어있었으므로, $r_1$$r_2$의 윤문을 각각 연륜으로 취급하였다. 2. 양식군의 성장이 방류군 및 야생군보다 월등히 우수하였다. 또한 양식하는 개체군과 동일한 8-12mm의 어린성게를, 인접한 장소에 방류한 방류군은 자연산의 성장과 유사한 성장을 하였다. 3. 동일 연령에서 각장분포의 범위가 큰 것은 산란기가 긴 원인도 무시할 수 없지만, 양식군 및 야생군의 각 연급간의 overlap현상이 큰 것으로 보아 개체간의 성장 차가 큰 것 같다. 4. Yamaguchi, Cheju를 제외하고는 Kasiwazima에서 Nomozaki에 이르기까지 거리상으로 인접할수록 성장의 유사경향이 높다. 그리고, Cheju는 지리적으로 고립되어 있음에도 불구하고, 보다 가까운 Kasiwazima보다 Aba, Mogi 및 Toishi와 더욱 유사한 성장을 하였고, 위도상으로 가장 북쪽에 위치한 Yamaguchi가 가장 남쪽에 위치한 Nomozaki와 유사한 성장을 나타내고 있었다.

Bandsharing Values and Genetic Distances of Two Wild Shortnecked Clam, Ruditapes philippinarum Populations from the Yellow Sea Assessed by Random Amplified Polymorphic DNAs-Polymerase Chain Reaction

  • Genomic DNAs were extracted from the muscle of twenty-two specimens of two shortnecked clam, Ruditapes phifippinarum populations collected in Anmyeondo and Seocheon. Genetic differences within and between populations were analysed by random amplified polymorphic DNAs-polymerase chain reaction (RAPD-PCR) using twenty arbitrary decamer primers. Out of 20 primers, 6 generated a total of 1,111 major and minor RAPD bands from individuals of two sites, producing approximately 4.2 average polymorphic bands per primer in individuals from Anmyeondo and ranging in size from less than 50 to larger than 1,500 base pairs (bp). The electrophoretic analysis of RAPD products amplified showed moderate levels of similarity among the different individuals in Seo-cheon population. The average bandsharing values (BS value) of the samples within population from Anmyeondo ranged from 0.155 to 0.684, whereas it was 0.143∼0.782 within population from Seocheon. The average BS value between individuals No. 13 and No. 14 from Seocheon was 0.782 which was higher than that of those from Anmyeondo. The single linkage dendrogram resulted from three primers (OPA-08, -09 and -20), indicating six genetic groupings composed of group 1 (No.4, 8 and 10), group 2 (No. 18), group 3 (No.2, 5 and 7), group 4 (No. 1, 3, 6, 9, 11, 12, 13, 14, 15 and 17), group 5 (16, 19 and 20) and group 6 (No. 21 and 22). In the Seocheon population, the individual No. 18 clustered distinctly from the others of this population. The observed genetic distance between the two populations from Anmyeondo and Seocheon was more than 0.209 (0.247 and 0.275). The shortest genetic distance (0.094) displaying significant molecular differences was between individuals No. 13 and No. 14. Especially, the genetic distance between individuals No. 22 and the remnants among individuals in two geographical populations was highest (0.275). This result illustrated that individual No.22 is distinct from other individuals within two shortnecked populations. The different geographical features of two sites may have caused the genetic diversity in two shortnecked clam populations.

난질과 유생발생을 이용한 4개 멍게(Halocynthia roretzi) 어미계군 특성 비교 (Differences in Egg Quality and Larval Development among Four Populations of Sea Squirt Halocynthia roretzi Adults)

  • To compare four populations of sea squirt Halocynthia roretzi adults, their egg quality and larval development were investigated in the hatchery. The populations were: south sea wild (SSW), south sea cultured (SSC), east sea wild (ESW) and Iwate Japan cultured (IJC). Egg quality and larval development were compared using 13 factors (fertilization rate and diameter, proximate composition, amino acid and fatty acid contents of eggs, hatching rate and various sizes of tadpole larvae) which were obtained from each population. Fertilized egg diameter, hatching rate and size of tadpole larvae were significantly different among the four populations (P<0.05). Eggs were produced with higher crude protein ($5.20{\pm}0.00%$ SSW, $4.71{\pm}0.01%$ ESW, $4.66{\pm}0.01%$ SSC and $3.96{\pm}0.01%$ IJC) and lipid ($1.22{\pm}0.01%$ ESW, $1.01{\pm}0.00%$ SSW, $0.77{\pm}0.01%$ SSC and $0.69{\pm}0.00%$ IJC,) contents from domestic wild populations than from Japanese or cultured populations. Also amino acid and fatty acid contents were different. The extent of similarity between domestic and Japanese populations (30.5% IJC:SSW, 34.3% IJC:SSC and 40.7% IJC:ESW) was relatively low but was very high between SSW and SSC (73.9%). These results may have been due to differences in the abundance of food types and environmental conditions in the four localities and consequent differences in the diets of the sea squirts.
