  • Endocrine disruptors have been implicated in carcinogenesis in animal studies, but carcinogenetic effects on human remain controversial. In order to examine the genotoxicity of two common endocrine disruptors, Bisphenol A and Diethylstilbestrol, cytogenetic endpoints including chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) analyses and DNA damage by single cell gel electrophoresis (SCGE) were assessed. The effects of Bisphenol A and Diethylstilbestrol on the frequencies of CA and MN were increased in a dose-dependent manner and that of Bispheol A was more significant by Kendall'$\tau$test. Bisphenol A and Diethylstilbestrol also increased the frequency of SCE. Bisphenol A and Diethylstilbestrol induced DNA damage in a dose-dependent manner and the DNA damage induced by Diethylstilbestrol in human blood lymphocytes was more significant.

Iodocasein, Tapazole 및 Diethylstilbestrol이 닭의 갑상선기능(甲狀腺機能)에 미치는 영향(影響) (The Effects of Iodocasin, Tapazole, and Diethylstilbestrol in the Thyroidal Activities of Chicken)

  • The effects of iodocasein, tapazole, and diethylstilbestrol in the thyroidal activity of chicken were tested. Iodocasein and tapazole were mixed with basal ration at the levels of 0.17 and 0.01 per cent respectively and were fed to one-weeks-old chicken for two months. Six milligrams of diethylstilbestrol were implanted in the subcutaneous tissue. Subsequently 10 uci of radioactive iodine $I^{131}$ were injected intramuscularly into each of the chicken to measure the thyroidal activities of the chicken one month after the treatment was started. The results obtained were as follows: 1) The thyroidal uptake of $I^{131}$ was not observed in the iodocasein group 2) The highest thyroidal uptake rates of I were shown twelve hours after the injection in the all groups except the iodocasein group. 3) The higher thyroidal uptake rates of I were observed in the female chicken than in the male chicken(P<0.01) of the control and the tapazole groups. However, the significant difference between the two sexes was not found in the diethylstilbestrol group. 4) Tapazole at the level of 0.01 per cent of the basal ration of the chicken increased the thyroidal activity slightly.

Diethylstilbestrol의 단핵구의 세포간 유착과정 조절효과 (Modulatory Effect of Diethylstilbestrol on CD29-Mediated Cell-cell Adhesion in Monocytic U937 Cells)

  • Diethylstilbestrol (DESB) is a synthetic estrogen not only that routinely prescribed, but also that known to be a teratogen. In this study, we found a novel pharmacological feature that DESB is able to positively modulate CD29 $({\beta}1-integrin)$ function. Thus, DESB up-regulated homotypic cell-cell adhesion of monocytic U937 cells mediated by CD29. However, DESB did not increase the surface level of CD29 and its binding activity to ligand (fibronectin), according to flow cytometric analysis and cell-fibronectin adhesion assay. Instead, the DESB-mediated up-regulation of cell-cell adhesion was blocked by several signaling enzyme inhibitors. Treatment of U0126 [an extracellular signal-regulated kinase (ERK) inhibitor], SB20358 (a p38 inhibitor) or Rp-8-pCPT-cGMP (a protein kinase G inhibitor) clearly inhibited DESB-mediated up-regulation of cell-cell adhesion induced by CD29. However, estrogen receptor antagonist ICI 182,780 failed to abrogate DESB effect. Therefore, our data suggest that DESB may up-regulate CD29-mediated cell-cell adhesion via modulating intracellular signaling enzymes such as ERK, PKG, and p38, independent of estrogen receptor function.

  • This experiment was carried out for 42 days to investigate the effect of caponizing and diethylstilbestrol (DES) on body weight gain, rate of feed conversion, weight of carcass and percentage of dressing in young chickens. The chickens used were 128 male Hamphorn of 30 days old. In the body weight gain of chickens, control group was higher than the other groups (P<0.01); $T_1$ group was the lowest group. Rate of feed conversion of control group was higher than $T_2$ and $T_3$ groups.

Effects of Streptozotocin, Bisphenol A and Diethylstilbestrol on Production of Reactive Oxygen Species and Lipid Peroxidation in the Boar Sperm

  • Streptozotocin (STZ), bisphenol A (BPA), and diethylstilbestrol (DES) are known as endocrine disruptors, occurs oxidative stress in animal cells. Generally, oxidative stress induces reactive oxygen species (ROS) and lipid peroxidation of sperm and lead to decreased viability and fertility in pigs. Therefore, we investigated the influence of STZ, BPA and DES on ROS production and lipid peroxidation on boar sperm. Collected sperm were incubated with semen extender containing $10{\mu}M\;STZ$, $10{\mu}M\;BPA$ and $20{\mu}M\;DES$ for 3, 6 and 9 hours. Intracellular ROS and lipid peroxidation of sperm were analyzed by 2', 7'-dichlorofluorescein diacetate and malondialdehyde methods. The results show that, intracellular ROS was not significantly different among the all treatments, but lipid peroxidation was significantly increased in STZ group at 3 hour after incubation with boar sperm (P<0.05). These results suggest that STZ stimulates lipid peroxidation more than ROS production and may exert a negative effect on sperm fertility.

Estrogenicity of 4-nonylphenol and diethylstilbestrol on in vitro oocyte maturation of the dusky tripletooth goby, Tridentiger obscurus

  • This study investigated the estrogenicity of 4-nonylphenol (NP) and diethylstilbestrol (DES) in vitro during oocyte maturation in the marine fish, Tridentiger obscurus, using steroid hormone assays and GVBD assay. Vitellogenic (0.53mm diameter) and fully vitellogenic (0.75mm diameter) oocytes were in vitro exposed to NP (0.045 453.82 nM and DES (0.037 372.62 nM). In vitellogenic oocytes, 45.38 and 453.82nM NP and 3.73 372.62nM DES increas the estradiol-$17{\beta}$ (E2)/testosterone (T) ratio. In fully vitellogenic oocytes, 0.45, 45.38 and 453.82nM NP and 3.73nM DES increased E2/T. In the GVBD assay, 0.45 and 4.54nM NP and 0.037, 3.73 and 37.26nM DES inhibited GVBD. These results suggest that NP and DES have estrogen-agonistic effects in oocyte maturation in T. obscurus. In addition, NP and DES have different sensitivity according to the oocyte developmental stage, and the estrogenagonistic effects of DES were greater than were those of NP.

마우스에 있어서 Diethylstilbestrol의 면역독성에 미치는 홍삼 Ethanol 유출물의 영향 (The Effect of Red Ginseng Ethanol Extract on the Immunotoxicity of Diethylstilbestrol in ICR Mice)

  • The effect of red ginseng ethanol extract on the immunotoxicity of diethylstilbestrol (DES) was studied in ICR mice. ICR male mice were divided into S groups (10 mice/group), and red ginseng ethanol extract (50, 100 and 200 mg/kg body wt., respectively) and DES (1 mg/kg body wt.) were injected intraperitoneally (i.p.) to ICR mice once a day for 2 weeks. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune response were evaluated by humoral immunity, cell-mediated immunity, non-specific immunity, and circulating leukocyte counts. The results of this study were summarized as followings: 1. The DES-treated control group as compared with normal group showed the tendency to decrease body weight rate and relative liver weight, decreased both humoral and cellular immune responses, phagocyte activity, and circulating leukocyte counts, but increased the natural killer (NK) cell activity. 2. Compared with the DES-treated control group, DES plus red ginseng ethanol extract-treated groups significantly decreased the body weight rate (P<0.01). Relative liver weight was significantly decreased in DES plus red ginseng ethanol extract (50mg/kg)-treated group (P<0.01), but significantly increased in DES plus red ginseng ethanol extract (100mg/kg)-treated group (P<0.01). Relative spleen and thymus weights were significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (200 mg/kg)-treated group (P<0.01). 3. Both humoral and cellular immune responses were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. 4. Phagocyte activity and circulating leukocyte counts were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. NK cell activity was significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (50 and 200 mg/kg)-treated groups (P<0.01).

Physiological Effects of Diethylstilbestrol Exposure on the Development of the Chicken Oviduct

  • Estrogen has dramatic effects on the development and function of the reproductive tract in mammals. Although diethylstilbestrol (DES) triggers the development of reproductive organs in immature animals, continued exposure to DES induces dysfunction of the female reproductive tract in mice. To investigate the effects of neonatal estrogen exposure on the reproductive tract of female chickens, we implanted DES pellets into the abdominal region of immature female chicks and then examined the effects of DES on the oviducts of both immature chicks and sexually mature chickens (30 weeks old). DES induced mass growth and differentiation of the oviduct in immature chicks. The chick oviduct increased by 2.7- and 29-fold in length and weight, respectively, following primary DES stimulation. In secondary DES stimulation, the length and weight of the chick oviduct increased by 4.5- and 74-fold, respectively. Additionally, DES treatments caused abnormal development of the infundibulum and magnum in hen oviducts. Furthermore, infundibulum abnormality gave rise to unusual ovulation of follicles and resulted in infertility and dysfunction of the magnum, such as less production of egg white proteins. Our results indicate that DES exposure during early developmental stages in chickens has detrimental effects on the development and maintenance of the female reproductive tract after sexual maturation.
