콩 추출물 투여가 흰쥐 혈청의 항산화효소 활성에 미치는 영향 (Activities of Antioxidant Enzymes in Serum of Rats Feeding Soybean Extract)

  • 본 시험은 콩 추출물의 흰쥐 투여에 따른 항산화 효소의 활성을 분석하고, 추출물의 원료인 콩 품종간, 추출물의 투여량 및 지방식이조성에 따른 효소활성도를 측정하였다. 시험결과를 요약하면 아래와 같다. 1. 콩추출물을 투여한 처리구에서 SOD와 GSHpx의 활성이 대조구에 비하여 높게 나타났으나, 통계적 유의성은 인정되지 않았다. 2. 지방식이 및 콜레스테롤을 흰쥐에 투여한 결과 콩추출물 섭취에 따른 항산화계 효소 SOD와 GSHpx의 활성은 검정콩 1호가 다원콩 및 황금콩에 비하여 높았다. 3. 지방식이조성 및 추출물의 투여량에 따른 항산화계 효소 활성도는 통계적으로 유의성이 인정되지 않았다. 4. 각 지방식이에 따른 콩추출물의 SOD와 GSHpx활성도에서 품종과 콩추출물 농도간에서는 상호작용효과가 인정되었다. 5. GSHpx 활성도에서 품종, 콩추출물 농도 및 지방식이간에서 상호작용효과가 인정되었다.

결명자 에탄올 추출물 및 식이섬유의 이화학적 특성 (Physicochemical Properties of Ethanol Extracts and Dietary Fiber from Cassia tora L. Seed)

  • This study was conducted to investigate the physicochemical properties of the ethanol extracts and soluble dietary fiber from Cassia tora L. seed. The proximate composition of Cassia tora, soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract, molecular mass distribution, sugar contents and viscosity of soluble fiber from Cassia tora were analyzed. Cassia tora contains 12.6% of moisture, 5.2% of ash, 13.4% of crude protein, 7.2% of crude fat, 8.8% of insoluble fiber and 48.3% of soluble fiber. The effects of extract condition on soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract were investigated. The soluble solid contents were higher in 70% or 50% ethanol extracts than those in 100% ethanol extracts and showed highest value in grind sample extracts. In Hunter's color value, 100% ethanol extracts and whole Cassia tora sample extracts were higher in L and b value, but on the contrary, were lower in a value, than those of the other. The highest emodin and rhein contents were observed in 70% and 50% ethanol extracts, respectively, and showed higher value in room temperature extracts than in heating extracts. The molecular mass of soluble fiber from Cassia tora seed was estimated by gel filtration chromatography. Most soluble fiber(80%) exhibited a molecular mass range of between 50~2000 kDa. The major sugars of soluble fiber from Cassia tora seed were identified as xylose, mannose and galactose. The apparent viscosity of 0.5% soluble fiber from Cassia tora seed was 33 mPas showing a higher value than pectin or xanthan gum.

EFFECTS OF LYSINE AND SODIUM ON THE GROWTH PERFORMANCE, BONE PARAMETER, SERUM COMPOSITION AND LYSINE-ARGININE ANTAGONISM IN BROILER CHICKS

  • An experiment with completely randomized design was performed to investigate the effects of lysine and supplemented sodium on growth performance, nutrients utilization, acid-base balance and lysine arginine antagonism in broiler chicks. The experiment was carried out with 3 levels of dietary lysine (0.6, 1.2 and 1.8%) and 3 levels of sodium(0.4, 0.8 and 1.2%) for an experimental period of 7 weeks. Body weight gain of 1.2% lysine group was significantly (p<0.01) higher than that of low or high lysine group. The highest feed consumption was obtained at 1.2% lysine and 0.4% sodium supplemented level (ML-1.2) and the lowest at LL-1.2. The best feed efficiency was obtained at ML-0.8 level and the worst at LL-1.2 level. Mortalities of high (1.8%) and low (0.6%) lysine groups were significantly (p<0.05) higher than medium lysine (1.2%) group. Among the sodium levels, the mortality at 1.2% sodium supplemented level was significantly (p<0.01) different by the levels of dietary lysine. Lysine-arginine antagonism was observed in high lysine diet. Among the lysine levels, the lowest none weight and length were shown in low lysine group. Interactions between lysine and sodium were significantly (p<0.05) shown in femur weight. The levels of sodium and lysine affected significantly (p<0.01) the utilization of nitrogen, ether extract, total carbohydrate and energy.

REPULSIVE EFFECT AND PALATABILITY OF DIETARY PHENYLALANINE IN LAYING HENS

  • Food intake of birds can be affected by particle size as well as diet composition, In order to investigate whether food intake of diets including excessive amount of phenyalanine(Phe) was influenced by diet types. a series of experiments were conducted in growing chicks and laying hens. Growing chicks significantly decreased food intake in a semipurified excessive Phe diet compared with a semipurified control diet, while laying hens fed a practical diet including excess Phe did not significantly reduce their food intake over a corn starch-substituted control diet. In an attempt to find out whether diet type affects food intake in layers, the semipurified type diet with excess Phe greatly reduced food ingestion, but the effect was delayed in the practical type diet. Moreover, under choice feeding regimes between the Phe and either starch, tyrosine(Tyr) or fiber diets in order to investigate whether the decreased food intake in the presence of an excess of dieary Phe in laying hens is involved in the palatability for the diet, there was no significant difference between Phe and starch diet while a preference for the Phe diet tended to be increased when birds were offered Tyr. Laying hens ingested significantly more the Phe diet than the fiber diet within 1 h after feeding. For supporting the idea that preference for the Phe diet may be affected by manipulating taste sense, an anaesthetic or saline was intramuscularly administered under the tongue just before a choice feeding. Preference for the Phe diet was not significantly different from that for the fiber reference diet within 1 h in the anaesthetized birds while the birds preferred the Phe diet in the saline treated group. It is suggested that because birds are able to select a diet, the decreased food intake induced by dietary excess Phe may be due to the repulsive effect of Phe after ingestion but not the taste of Phe.

Effects of Lysine Intake during Middle to Late Gestation (Day 30 to 110) on Reproductive Performance, Colostrum Composition, Blood Metabolites and Hormones of Multiparous Sows

  • Lysine intake during gestation has a major impact on subsequent reproductive performance. The objective of this experiment was to determine the effect of lysine intake from mid-gestation until farrowing on the reproductive performance of multiparous sows. On day 30 of gestation, 200 Landrace${\times}$Large White sows were randomly assigned to one of four groups based on body weight and parity (n = 50). The gestation diets contained 0.46, 0.56, 0.65 or 0.74% lysine. Increasing dietary lysine concentration improved sow body condition at farrowing and increased litter weights (p<0.05). Dietary lysine level also had a significant effect on the dry matter (p<0.05) and protein content (p<0.05) of colostrum. Increased lysine intake increased serum insulin concentration (p<0.05) and there was a trend towards increased serum prolactin content (linear, p = 0.07). However, increased lysine tended to decrease blood urea N (quadratic, p = 0.05). These results suggest that higher lysine levels (0.65-0.75%) than those recommended by the National Research Council improved reproductive performance for multiparous gestating sows and this increase may be partially mediated through blood metabolites or metabolic hormone levels.

Effects of dietary valine:lysine ratio on the performance, amino acid composition of tissues and mRNA expression of genes involved in branched-chain amino acid metabolism of weaned piglets

  • Objective: The goal of this study was to investigate the effects of dietary standard ileal digestible (SID) valine:lysine ratios on performance, intestinal morphology, amino acids of liver and muscle, plasma indices and mRNA expression of branched-chain amino acid (BCAA) metabolism enzymes. Methods: A total of 144 crossbred pigs (Duroc${\times}$Landrace${\times}$Large White) weaned at $28{\pm}4days$ of age ($8.79{\pm}0.02kg$ body weight) were randomly allotted to 1 of 4 diets formulated to provide SID valine:lysine ratios of 50%, 60%, 70%, or 80%. Each diet was fed to 6 pens of pigs with 6 pigs per pen (3 gilts and 3 barrows) for 28 days. Results: Average daily gain increased quadratically (p<0.05), the villous height of the duodenum, jejunum and ileum increased linearly (p<0.05) as the SID valine:lysine ratio increased. The concentrations of plasma ${\alpha}-keto$ isovaleric and valine increased linearly (p<0.05), plasma aspartate, asparagine and cysteine decreased (p<0.05) as the SID valine:lysine ratio increased. An increase in SID lysine:valine levels increased mRNA expression levels of mitochondrial BCAA transaminase and branched-chain ${\alpha}-keto$ acid dehydrogenase in the longissimus dorsi muscle (p<0.05). Conclusion: Using a quadratic model, a SID valine:lysine ratio of 68% was shown to maximize the growth of weaned pigs which is slightly higher than the level recommended by the National Research Council.

성숙한 쥐의 대퇴골과 요추의 조성과 골격관련 인자에 대한 난소절제와 칼슘섭취 효과 (Effect of Ovariectomy and Calcium Intake on Femur and Lumbar Compositions and Bone Related Factors in Adult Rats)

  • This study explored the effect of calcium levels and/or ovariectomy on bone composition and its related factors using the female Sprague-Dawley rats which achieved peak bone mass in normal(0.5%) calcium intake during growth period. The rats were randomly divided into six groups and fed 0.1%, 0.5% and 1.5% calcium diets for 8 weeks after ovariectomized and sham operation. The results indicated that body weight gain was higher in ovariectomized groups than sham groups, regardless of dietary calcium levels and food intakes. Serum Ca and P concentrations were normal level regardless of dietary calcium levels and operation. Estrogen concentration was decreased in ovariectomized rat groups. Serum alkaline phosphatase activity and urinary hydroxyproline were increased in ovariectomized groups. When rats were fed normal Ca diet during growing period, weight, length and breaking force of femur were not significantly different in all groups but increased the same level. Generally, lipid contents in lumbar and femur were lower in low calcium groups and this effect was more pronounced in femur. In case of femur, the significant decrease in Ca contents of bone was observed in a relatively short period of feeding low Ca diet, even though it appeared th desirable peak bone mass had achieved through the growing period by supplementing the adequate amount of calcium. The marked decrease of estrogen levels after ovariectomy did not seem to influence greatly bone parameters measured except bone wet weight. Therefore, this study indicated that lower intake of Ca might be more important determinating factor against osteoporosis than postmenopausal state stimulated by ovariectomy in female rats. With normal or high intake of Ca it appears possible to prevent bone loss in postmenopausal period, and this might apply only in case of achieving peak bone mass in the growing with the adequate intake of calcium. Assessment of Anthropometry, Nutritional Compositions and Contribution of School Meals to the Daily Nutrient Requirements of Primary School Children from Rural Communities

  • The study aims to evaluate the nutritional status and influence of school meal intakes on RDA of primary school children in Akure community, Ondo State, Nigeria. A cross-sectional study was conducted among 728 primary school children aged between 6 and 15 years. Data were collected using interviewer-administered semi-structured questionnaires. The questionnaires collect information on demographic characteristics and home dietary intake of the subjects. The heights and weights of the children were measured using a standard procedure and height-far-age and weight-far-height z-score were determined. The children's school meal intakes were weighed for 4 days and samples were collected for chemical analysis. The results showed that 37.8% of the children were not wasted, 35.7% mildly wasted, 18.7% moderately wasted and 7.8% severely wasted. Also, 57.8% were not stunted, 29.3% mildly stunted, 11.0% moderately stunted and 1.9% were severely stunted. The subjects' home dietary intakes showed that 73.6% ate starchy food only, 19.9% ate protein based food, while 11.6% and 11.5% consumed fruits/vegetables and snacks to complement home meals respectively. The chemical composition of school meal was energy 379 - 413kcal, moisture content 5.9 -7.3g, carbohydrate 56.5 - 69.4g, fat 4.6 - 12.7g, crude fiber 0.1 - 2.4g, ash content 3.6 - 8.5g and protein 14.9 - 22.3g. The mineral contents were calcium 45.9 - 59.2mg, sodium 5004 - 59.6mg, zinc 2.3 - 3.1mg, magnesium 55.0 - 61.6mg, potassium 55.3 - 69.3mg, copper 0.2 - 0.3 mg, while others 1.3 - 1.9mg, 243 - 659mg and 831 - 9,510mg were iron, phosphorous and vitamin-A respectively. The contribution of school meals to subjects' RDA was within 2.9% and 1540%. In summary, school meal intake contributed positively to the RDA and nutritional status of the school children.

죽초액의 첨가가 육성오리의 생산성 및 육질에 미치는 영향

  • 죽초액을 사료내 0%, 1%, 2% 및 4%로 첨가하여 5주 동안 급여했을때 육성오리의 생산성 및 육질에 미치는 영향을 조사한 결과는 다음과 같다. 증체량과 일당증체량은 죽초액 1%구에서 비교적 증가하였으나 유의성은 인정되지 않았다. 사료섭취량은 죽초액을 첨가함에 따라 약 간 증가하는 경향이였다. 도체중은 1%구가 비교적 증가하는 경향이였다. 죽초액 2%와 4%구에서 육성오리 가슴육의 단백질함량의 유의적인(P<0.05) 증가와 지방의 유의적인(P<0.05) 감소를 나타냈다. 콜레스테롤 함량과 TBA는 죽초액을 첨가함으로써 유의적(P<0.05)으로 감소하였다. 지방산 조성은 죽초액 2%구에서 콜레스테롤의 전구물질이며 포화지방산의 가장 높은 비율을 차지하는 palmitic acid가 죽초액을 급여함으로써 감소하는 경향인 반면에, 맛과 풍미에 영향을 주며 불포화지방산의 가장 높은 비율을 차지하는 oleic acid는 죽초액을 급여함으로써 증가하는 경향이었다. 관능평가 결과 맛과 풍미 그리고 외관에 있어 죽초액 1%와 2%구에서 유의적(P<0.05)으로 높게 나타났다. 장내 살모넬라균과 대장균의 수가 죽초액의 첨가수준의 증가에 따라 감소하는 경향으로 항균작용 효과가 있음을 알 수 있었다.

갑상선 호르몬의 경구투여가 육계의 사양성적, 체조성, 혈청 호르몬 농도 및 에너지 대사에 미치는 영향 (Effect of Dietary Thyroid Hormone on Growth Performance, Body Composition, Serum Thyroid Hormone Concentration and Energy Metabolism of Broiler Chicks)

  • This study was conducted to investigate the effect of dietary thyroid hormone (T$_3$and T$_4$) on growth, feed conversion ratio and serum T$_3$or T$_4$concentration of broiler chicks. Zero to six week-old broiler chicks were randomly allocated into seven treatment groups for feeding trials with three replication : control, T$_3$(0.1), T$_3$(1.0), T$_3$(5.0), T$_4$(0.1), T$_4$(1.0), T$_4$(5.0) ppm group. Concentrations of T$_3$and T$_4$in serum were analyzed. The weight gain of T$_3$(1.0), T$_3$(5.0) and T$_4$(5.0) groups were significantly lower than that of control. No statistically significant adverse effect was observed in other groups (p〉0.05). Feed intake was significantly lower in T$_3$(0.1, 1.0 and 5.0) and T$_4$(5.0) than in control group (p〈0.05), and the feed conversion ratio had a similar trend to the feed intake change. The contents of liquid and abdominal fat pad in carcass were significantly decreased in all T$_3$and T$_4$groups (p〈0.05). T$_3$and T$_4$concentration in serum was significantly increased at over 1.0ppm of the hormone supplementation level. As T$_3$addition level increased, T$_4$concentration in serum reduced ; however, T$_3$in serum was directly proportional during fasted were slightly increased when T$_3$or T$_4$was added to broiler diets.
