BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a common metabolic disease triggered by epigenetic alterations, including lysine acetylation at histone or non-histone proteins, affecting the stability or transcription of lipogenic genes. Although various natural dietary compounds have anti-lipogenic effects, their effects on the acetylation status and lipid metabolism in the liver have not been thoroughly investigated. MATERIALS/METHODS: Following oleic-palmitic acid (OPA)-induced lipid accumulation in HepG2 cells, the acetylation status of histone and non-histone proteins, HAT activity, and mRNA expression of representative lipogenic genes, including $PPAR{\gamma}$, SREBP-1c, ACLY, and FASN, were evaluated. Furthermore, correlations between lipid accumulation and HAT activity for 22 representative natural food extracts (NExs) were evaluated. RESULTS: Non-histone protein acetylation increased following OPA treatment and the acetylation of histones H3K9, H4K8, and H4K16 was accelerated, accompanied by an increase in HAT activity. OPA-induced increases in the mRNA expression of lipogenic genes were down-regulated by C-646, a p300/CBP-specific inhibitor. Finally, we detected a positive correlation between HAT activity and lipid accumulation (Pearson's correlation coefficient = 0.604) using 22 NExs. CONCLUSIONS: Our results suggest that NExs have novel applications as nutraceutical agents with HAT inhibitor activity for the prevention and treatment of NAFLD.
Our nation is confronted with the situation that the rice, a principal food, short of some essential amino acids, leads to imbalanced meals insufficient in the nutrient of Protein, to bring many difficulties in the elevation of nutritional state in our nation. While. our country has been produced much amounts of Panax Ginseng roots which has a stimulating effects on the metabolism of protein, lipid and nucleic acids in the body. And the leaf and trunk of Panax Ginseng were also produced a considerable amounts as the by-products. Author believe that these by-products (leaf and trunk) of Panax Ginseng might have some components possessing simillar activity with Panax Ginseng root although the quantity and qualify of the functional components may more or less be different. Therefore, this study was demised to observe the supplemental effect of the Panax Ginseng-by-Products on the dietary protein efficiency and nutritional state of rats. The feeds used for this experiment were rice containing 30% barely, fish four, and the leaf, trunk and small root of the Panax Ginseng, and the contents of the general nutrients including protein, lipid and carbohydrate etc. in each feed were analyzed for the combination of each feed. And, being based on analytical values of Protein in food. fish Pour as Protein source was added were rice containing 30% barely to be include 8.6 to 8.7%, 12%, 15% and 18% of protein. Then 2% of the leaf, trunk or small reef of Panax Ginseng was supplemented into each of above protein diet group, ton 16 kinds of diets were Prepared. The male albino rats from a Pure strain, weighing 70g to 80g. were used for experimental animals. They were maintained with coresponding fist for f and 8 weeks, and the growth rate, consumption of diets and protein, efficiency of feed and Protein in animals were determined. The lipids, proteins and cholesterols in serum and liver were also determined quantitatively after they were sacrificed in coresponding term. The results obtained are summarized as follows: 1. Body weigh of diet group containing 8.6 to 8.7%,12%, and 15% of protein are increased remarkably by supplement of 2% of the leaf or small root of Panax Ginseng in comparison with each of controls. But this tendency could not observed in diet group containing 18eA Proteins. 2. Feed efficiency showed same tendency in comparison with changes of gained body weight. Specially, in each of diets containing 8.7%, 12%, 15% and 18% of Proteins, supplement of the leaf of Panax Ginseng showed the better feed efficiency than supplement of the trunk or small root. 3. In feeding group for 8 weeks, protein efficiency showed worst efficiency in diet containing 18% proteins and showed the best efficiency was the diet group containing 12% Proteins. And the efficiency was improved according to supplement of the leaf of Panax Ginseng. 4. Nitrogen contents in serum and liver did not show large differences each other in all diet groups. But contexts of total cholesterol and 1ipid were decreased markedly in diet groups containing 12%, 15% and 18% of proteins in comparison with diet group containing 8.6% to 8.8% of proteins.
This study was done to investigate the association of blood clinical parameters and dietary intakes with serum adiponectin level. Athropometric measurement, dietary intakes, serum lipids and adiponectin levels were examined in 160 overweight and obese women. The subjects were divided into 5 groups by quintile according to serum adiponectin level. Weight, BMI, waist circumferences and waist/hip ratio of the highest quintile group were significantly lower than those of the lowest quintile group. Serum lipid analysis showed a significant higher level of TG, LDL-cholesterol, LDL/HDL ratio, AI, and serum hs-CRP in the lowest quintile group. Similarly, correlation data also showed that serum adiponectin level was positively correlated with serum HDL-cholesterol level (p < 0.01) and was negatively correlated with BMI (p < 0.01), waist circumferences (p < 0.01), waist/hip ratio (p<0.01), systolic (p < 0.01) and diastolic blood pressure (p < 0.05), TG (p < 0.01), LDL-cholesterol (p < 0.05), LDL/HDL ratio (p < 0.05), AI (p < 0.01), Homa-IR (p < 0.01), hs-CRP (p < 0.05) and leptin (p < 0.05). Dietary intake data showed that protein intake was significantly lower in the highest quintile group compared to the lower quintile groups while intakes of vitamin C was significantly higher in highest quintile group after adjustment by BMI, waist and energy intake, In addition, the highest quintile group had higher fiber intakes than the lower quintile groups. These results might suggest that a diet high in fiber and vitamin C and low in protein for obese patients would better be recommended to improve adiponectin level. However, further research is needed to elucidate the association of dietary intakes or dietary patterns and serum adiponectin level.

This study was undertaken to dtermine whether dietary fibers had an effect on postprandial plasma lipoprotein and liver lipid composition in rats. Each experimental diet ocntained 10 % dietary fiber by weight. All animals were sacrificed in postprandial state. Sodium alginate-fed animals has significantly lower plasma cholesterol, VLDL-, LDL-cholesterol, TG, and protein levels than did the fiber-free control group. In addition, liver cholesterol and TG concentrations were the lowest in sodium alginate -fed animals. There was no significant change in HDL-cholesterol levels among experimental groups. Cellulose-fed animals also have lower plasma cholesterol and TG levels than fiber-free controls, but liver TG concentrations were not different from those in fiber-free controls. This study demonstrates that dietary fiber included in the diet of rats is able to alter postprandial lipoportein cholesterol and TG, and that sodium alginate, a soluble fiber, was the most effective in lowering plasma and liver cholesterol and TG.
This study was carried out to examine the association of age, nutrient intake, alcohol drinking and smoking on serum lipid and apolipoprotein levels in 100 healthy adults (54 males and 46 females). The serum total cholesterol, low density lipoprotein cholesterol (LDL-C) (p<0.05), apolipoprotein B (Apo B) levels, LDL-C:high density lipoprotein cholesterol (HDL-C) ratio, Apo B:apolipoprotein(Apo A-I) ratio and atherogenic index (AI) (p<0.001) were significantly higher in males than females, but HDL-cholesterol level was significantly lower (p<0.01) in males than females. The plasma ascorbic acid concentration had no difference in both genders. Most of nutrient intakes except Ca, vitamin A and vitamin $B_2$ were higher than RI (Recommended intake) in both males and females. The meal frequency per day was significantly higher (p<0.01) in males than females, while meal speed, the heaviest meal, meal quantity and meal regularity had no significant differences. The rates of drinking and smoking were significantly higher (p<0.01) in males (59.3%, 37.2%) than females (17.3%, 6.9%). The age was positively correlated with triglyceride level in males (p<0.05), and with total cholesterol (p<0.001), LDL-cholesterol (p<0.001), Apo B (p<0.01), LDL-C:HDL-C ratio (p<0.01), Apo B:Apo A-I ratio (p<0.05) and AI (p<0.01) in females. The levels of serum lipid and apolipoprotein were more correlated with protein intake than other macronutrient intakes in males, while those in females were more correlated energy, carbohydrate and lipid intakes than protein intake. The effect of drinking on serum lipid and apolipoprotein levels was bigger in males than females, while the effect of smoking on those was bigger in females than males. The plasma ascorbic acid concentration was positively correlated with HDL-cholesterol (p<0.05), Apo A-I levels (p<0.001), whereas negatively correlated with Apo B:Apo A-I ratio (p<0.05) in males. And that was positively correlated with HDL-cholesterol (p<0.05) in females. These results suggest that moderate macronutrient intake, less alcohol consumption and non-smoking were necessary to maintain healthy lipid profile with aging in adults.

This study investigated the effects of dietary lipid sources on growth performance and body composition of juvenile far eastern catfish, Silurus asotus. Three replicate groups of fish (average weight 3.6 g) were fed with one of the following experimental diets containing 10% beef tallow (BT), 5% BT plus 5% corn oil (CO), 5% BT plus 5% linseed oil (LO), or 5% BT plus 5% squid liver oil (SO) as the lipid source for 5 weeks. No significant difference was observed in the survival among groups. The weight gain of fish fed the LO (high in 18:3n-3) and SO (high in n-3 highly unsaturated fatty acid) diets was significantly higher than that of the fish fed the CO (high in 18:2n-6) and BT diets (P<0.05). The feed efficiency of fish fed LO and SO diets was significantly higher than that of the fish fed the BT diet (P<0.05), but not significantly different from that of the fish fed the CO diet. The protein efficiency ratio of fish fed the SO diet was significantly higher than that of fish fed the CO and BT diets (P<0.05), but not significantly different from that of fish fed the LO diet. The 18:1n-9 of whole-body polar lipid fraction in fish fed the BT diet increased compared to that of fish fed the other diets. Fish fed the CO and LO diets had significantly higher contents of 18:2n-6 and 20:4n-6, and 18:3n-3, than the fish fed the other diets in polar and non-polar lipid fractions, respectively (P<0.05). Significantly higher contents of 20:5n-3 and 22:6n-3 were observed in the whole-body polar lipid fraction of fish fed the SO diet compared with fish fed the other diets (P<0.05). The study results indicate that linseed oil and squid liver oil containing n-3 fatty acids are good dietary lipid sources for the growth of far eastern catfish.
This experiment was conducted to investigate the effect of dietary supplemental Astragalus membranaceus on performance and meat quality of broiler chicks for 5 weeks. Basal diets based on corn and soybean meal contained 21% crude protein for the first three weeks and 19% for the rest of two weeks. Four levels of dietary Astragalus membranaceus(0, 0.25, 0.50, 1.00%) were fed in a one way design. There were four replicates of 50 chicks per treatment. Weight gain, feed intake, feed efficiency, blood component, breast meat lipid and protein were measured at the end of experiment. No increased growth rate was observed in chicks fed the basal diet supplemented with Astragalus membranaceus. Chicks fed the diet containing 0.5 and 1.00% Astragalus membranaceus tended to depress the growth rate. Feed intake of 0.25% Astragalus membranaceus treatment was significantly lower than control group(P<0.05), Dietary supplemental Astragalus membranaceus improved the feed efficiency compared to the control group, but was not significantly different between them. However, Astragalus membranaceus treatment exhibited decreased serum cholesterol and AST compared to those of control group, but was not significantly different between supplemental groups. Serum hematocrit, triglyceride, phosphate and BUN decreased at 0.25% Asparagus membranaceous supplementation. Growth hormone and IGF-1 (ng/ml) of chicks fed more than 0.50% Astragalus membranaceus significantly decreased compared to those of control(P<0.05). The total lipid and protein content of chicks fed 0.25 and 0.50% Astragalus membranaceus tended to increase, but did not show consistency at more than 0.50% supplement. The results of this experiment indicated that optimum dietary Astragalus membranaceus level to improve the performance and meat quality of broiler chicks may be less than 0.25%.

Objective: We tested the hypothesis that increasing dietary copper (Cu) to gravid ewes would enhance brown adipose tissue (BAT) thermogenesis in their offspring. Methods: Twin-bearing ewes were assigned on d 70 of gestation to diets containing 3, 10, or 20 ppm dietary Cu (n = 8 per group). Twin lambs were assigned at birth to a cold (6℃) or warm (28℃) environmental chamber for 48 h. Blood was collected from ewes and from lambs and perirenal BAT was collected after 48 h in the environmental chambers. Results: Prenatal Cu exposure increased ewe plasma triiodothyronine (T3) and thyroxine concentration (T4) (p<0.01) but prenatal Cu exposure had no effect on lamb plasma concentrations of T3, T4, glucose, or nonesterified fatty acid concentration (p≥0.08). The high level of prenatal Cu exposure depressed 48-h rectal temperature (p = 0.03). Cold exposure decreased BAT norepinephrine (NE) and increased BAT dopamine (p≤0.01), but prenatal Cu exposure had no effect on BAT cytochrome C oxidase activity or BAT NE or dopamine (p≥0.07). However, BAT of lambs from high-Cu ewes maintained higher uncoupling protein-1 (UCP1) gene expression than BAT of lambs from low- and medium-Cu ewes following warm or cold exposure in environmental chambers (p = 0.02). Cold exposure caused near depletion of BAT lipid by 48 h (p<0.001), increased BAT cytochrome c oxidase activity (p<0.01), and depressed plasma fatty acid concentrations (p<0.001). Conclusion: Although prenatal Cu exposure increased BAT UCP1 expression during warm and cold exposure, prenatal cold Cu exposure depressed 48-h rectal temperature. Cold exposure decreased BAT lipid content by over 80% and decreased lamb plasma fatty acid concentration by over 40%, indicating that fuel reserves for thermogenesis were nearly depleted by 48 h of cold exposure.
This study determined the effects of dietary restriction on growth and the expression of lipid metabolism and growth hormone signaling genes in the longissimus dorsi muscle (LM) of Korean cattle. Thirty-one Korean cattle steers (average age 10.5 months) were allocated to normal (N; n = 16) or dietary restriction (DR; n = 15) groups. The feeding trial consisted of two stages: for the 8-month growing period, the DR group was fed 80% of the food intake of the normal diet, and for the 6-month growth-finishing period, the DR group was fed a DR total mixed ration with 78.4% of the crude protein and 64% of the net energy for gain of the normal diet. The LM was biopsied 5 months (period 1 [P1] at 15.5 months of age) and 14 months (period 2 [P2] at 24.5 months of age) after the start of feeding. The mRNA levels were determined using real-time polymerase chain reaction. Body weight, daily feed intake, average daily gain, and feed efficiency were lower in the DR group compared with the normal group at both P1 and P2. At P1, the lipogenic fatty acid synthase (FASN) mRNA levels were lower (p<0.05) in the DR group compared with the normal group. The DR group tended (p = 0.06) to have higher of levels of growth hormone receptor (GHR) mRNA than the normal group. At P2, the DR group tended to have lower (p = 0.06) androgen receptor (AR) mRNA levels than the normal group. In conclusion, our results demonstrate that dietary restriction partially decreases the transcription of lipogenic FASN and growth hormone signaling AR genes, but increases transcription of the GHR gene. These changes in gene transcription might affect body fat accumulation and the growth of the animals.

This study was done to investigate the effects of n-3, n-6 fatty acid and d-limonene on the hepatic membrane lipid composition, protein kinase C (PKC) and glutathione S-transferase (GST) activities in experimental rat hepatocarcinogenesis. Sprague-Dawley female rats were fed with two different types of dietary oil for 20 weeks. Corn oil (CO) and sardine oil (SO) were used at 15% by weight as a source of n-6 and n-3 fatty acid, respectively. One week after feeding, rats were intraperitoneally injected twice with a dose of diethylnitrosamine (DEN, 50 mg/kg body weight) and after 1 week 0.05% phenobarbital (PB) was provided with drinking water. Membrane fractional lipid composition showed that the content of cholesterol was higher in 50 group than CO group and also significantly decreased by d-limonene. The content of phospholipid was increased by carcinogen treatment but not affected by dietary oils or d-limonene. Membrane C/PL molar ratio was significantly decreased by d-limonene or carcinogen treatment in 50 groups but not in CO groups. Fatty acid composition was changed by dietary oils but not by carcinogen treatment or d-limonene. Cytosolic PKC activity was not significantly different by dietary oils, d-limonene or carcinogen treatment. However, membrane PKC activity was significantly increased by carcinogen treatment and decreased by d-limonene. Cytosolic GST activity was affected by d-limonene or carcinogen treatment in all dietary groups. These data indicate that dietary oils, d-limonene and carcinogen treatment can not change much membrane phospholipid composition. But membrane C/PL molar ratio was changed by carcinogen treatment and d -limonene although the effect was different between dietary oils. Therefore, it is suggested that different dietary oils and d-limonene can somewhat modulate the changes of membrane fluidity and activities of membrane bound enzymes like membrane associated PKC during carcinogenesis.
