Response Surface Analysis of Dietary n-3/n-6 and P/S Ratio on Reduction of Plasma Lipids in Rats (흰쥐현장지질 감소에 관한 n-3/n-6 와 P/S 섭취비율의 반응표면분석)

  • Response surface analysis was used to study dietary ratios of n-3/n-6 fatty acid and P/S to minimize plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C levels and maximize plasma HDL ${\cdot}$ C levels of rats. Because the dietary components were not statistically independent, they were studied in combinations of two variables. The two-variable combinations were the most useful in locating the desired maximum or minimum plasma triglycerides, total cholesterol and LDL ${\cdot}$ VLDL-C response in terms of the proportions of the dietary components. Response surface contours and three dimensional plots were developed for each plasma lipid response. The contours and three dimensional plots were used to help determine those combinations of the dietary fatty acid ratios that would produce the desired minimum or maximum lpid responses. The statistical analyses indicated that the minimized plasma cholesterol response levels could be attained with a diet consisting of 2.26 n-3/n-6 fatty acid and 2.15 P/S ratios.

The Effects of Combined Obesity Management Program on Blood Lipids and Dietary Habit in Obese Women (통합비만관리 프로그램이 여성의 혈중 지질대사 및 식습관 양상에 미치는 영향)

  • Purpose: This study was to determine the effects of a combined obesity management program in obese women. The study also attempted to measure the effects of the program on blood lipid, body mass index, dietary calory and dietary habit. Method: This research employed a one group pretest-posttest design. A total of 251 obese women(BMI: over 25) were selected for this research. The subjects in a combined obesity management program (combined exercise therapy and dietary therapy) for 8weeks. Results: After performing the program for 8 weeks, the result were follows. 1) There were significant reduction in total cholesterol(t=4.10, p= .00), triglyceride(t=5.09, p= .00) but no significant reduction BMI. 2) There were reduction in dietary calory, but no significantly. 3) There were significant increase in dietary habit. Conclusion: From these findings, it was confirmed that a combined obesity management program for obese women can decrease TC, TG, BMI and increase dietary habit. Therefore, this program can be utilized for various population groups including women, and further researches are required on program for men, elderly.

Dietary Intakes, Plasma Levels and Urinary Excretions of Taurine in Adolescents and Adults Residing in Seoul Area (서울지역 청소년 및 성인의 타우린 섭취량, 혈중 농도 및 소변내 배설량에 관한 연구)

  • As diverse physiological functions of taurine have been reported, taurine-containing health drinks and products are marketed worldwide for the treatment of various conditions such as improvements of liver, heart and circulatory functions or as an aid to athletic performance. Although animal studies have shown that taurine is fairly safe when supplemented in the diet for an extended period, the effective dose range of taurine for dietary supplements is in controversy. Reports on dietary taurine intakes have been sparse, and would serve as a guideline for determining an appropriate taurine dosage. The present study was aimed to estimate dietary intake level of taurine using the taurine content database of commonly used food stuffs established recently in our laboratory, and also to evaluate plasma concentration and urinary excretion of taurine in adolescents and adults residing in Seoul area. Dietary taurine intakes of the subjects were 219$\pm$16.9mg/day for 16-19 years old(n=123), 177$\pm$18.1mg/day for adults older than 20 years old(n=123). Male subjects(n=115) consumed 216$\pm$21.1mg of taurine/day, while female subjects(n=131) consumed 181$\pm$14.3mg of taurine/day(p<0.05). The level of dietary taurine intake was positively correlated with the levels of dietary intakes of energy, carbohydrate, total lipids, cholesterol, vitamin A, vitamin B$_1$, niacin, vitamin C, calcium, phosphorous, sodium and potassium at p<0.01, and with dietary intakes of iron and animal lipids at p<0.05, respectively. Plasma taurine concentration of subjects were 135$\pm$5.9$\mu$mol/L, which is considered to be within a normal range for healthy subjects. The subjects excreted 1158$\pm$72.7nmol/ of tarine mg creatinine in their urine, which is approximated as 150-170mg of taurine/24hr urine based on the assumption that 18mg creatinine/kg/day is excreted in the urine of healthy adults, and this would be about 80% of the daily taurine intake observed in the same subjects. Dietary taurine intake level was positively correlated with plasma taurine concentration, as well as with urinary taurine excretion corrected by creatinine excretion at p<0.05. The present study was the first report of taurine intake, and plasma concentration and urinary excretion of taurine in a Korean population so far, and these results would serve as an index for the future study evaluating taurine status in a diverse population within and outside Korea. Relationship between Dietary, Blood and Urinary Levels of Lead, Blood Pressure and Serum Lipids in Korean Rural People on Self-Selected Diet (일부 농촌 성인 남녀의 식이, 혈액 및 뇨중의 납수준과 혈압, 혈청지질과의 관계)

  • This study was performed to evaluate the relationship between dietary, blood and urinary levels of lead and blood pressure and serum lipids in 30 healthy adult living in rural area of Korea. Analysis for the nutritional status of subjects were performed by 3-day dietary intake record, duplicated diet collection, 24-hour urine collection, and venous blood sampling before anthropometry. The mean daily intakes of lead estimated for 3 days was $120.1{\pm}22.0\mu\textrm{g}$. The blood levels and 24-hour urinary excretion of lead were $10.8{\pm}3.6\mu\textrm{g}/dl\;and\;36.5{\pm}9.5\mu\textrm{g}$, respectively. The systolic and diastolic blood pressure were $113.0{\pm}16.9mmHg\;and\;76.7{\pm}12.1\;mmHg$. The serum levels of total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol and ${\beta}-lipoprotein$ were $158.8{\pm}32.9mg/dl,\;104.6{\pm}48.8mg/dl,\;45.7{\pm}9.9mg/dl,\;92.2{\pm}28.5mg/dl\;and\;426.4{\pm}141.5mg/dl$, respectively. There was no significance in the relation between lead and blood pressure. In the relation between lead and serum lipids, it showed negative correlation with lead intake and HDL-choleterol at the level of significance of p<0.01. But there was no significance in the relation between lead and serum levels of otehr lipids.

  • Genetically selected beef cattle are fed high-energy diets in intensive production systems developed in industrial countries. This type of feeding can induce rumen dysfunctions that have to be corrected by farmers to optimise cost-effectiveness. The risk of rumen acidosis can be reduced by using slowly degradable starch, which partly escapes rumen fermentation and goes on to be digested in the small intestine. Additives are proposed to stabilise the rumen pH and restrict lactate accumulation, thus favouring the growth of cellulolytic bacteria and stimulating the digestion of the dietary plant cell wall fraction. This enhances the energy value of feeds when animals are fed maize silage for example. Supplementation of lipids to increase energy intake is known to influence the population of rumen protozoa and some associated rumen functions such as cellulolysis and proteolysis. The end products of rumen fermentation are also changed. Lipolysis and hydrogenation by rumen microbes alter the form of fatty acids supplied to animals. This effect is discussed in relation with the quality of lipids in beef and the implications for human health. Conditions for optimising the amount of amino acids from microbial proteins and dietary by-pass proteins flowing to the duodenum of ruminants, and their impact on beef production, are also examined.

Effect of Dietary Lipids and Stress on Neurotransmitters in Rats (식이지방과 스트레스가 신경전달물질의 수준에 미치는 영향)

  • To investigate the effect of dietary lipids and stress on brain catecholamine and serotonin concentration, sixty three weanling male Sprague-Dawley rats(mean body weight$\pm$SD : 171$\pm$3g) were fed a diet containing fish oil, soybean oil or beef tallow and than, each was exposed to three different types of stress, isolated, grouped or cold, respectively. Cold stress seemed to be most severe and living together in a large cage with some playing equipments is more stressful than living alone in a classical small cage evidenced by plasma corticosterone level. Average food intake and body weight gain were not significantly different among exprimental groups. In adrenal catecholamines, norepinephrine was significantly affected by diet and stress and dopamine was by stress. Norepinephrine concentration of the fish oil group was lowest among diet groups. Adrenal epinephrine, however, was not. It was also shown than the cold stress significantly increased the brain norepinephrine concentration. The cold stress significantly induced higher content of brain serotonin than the grouped stress. However, the concentratin of 5-hydroxyindoleacetic acid(5-HIAA), the metabolite of serotonin, was not significantly different among groups. Therefore, this results suggest that stress affects sympathetic neuronal activity, and fish oil might lighten the burden of stress.

Effect of Dietary Conjugated Linoleic Acid on Plasma Levels of Glucose and Lipids and Hepatic Lipogenic Enzyme Activity in Otsuka Long Evans Tokushima Fatty Rats (OLETF 비만쥐에서 CLA첨가 식이가 혈장의 포도당과 지질농도 및 간조직의 Lipogenic Enzyme 활성도에 미치는 영향)

  • The study was designed to observe whether the conjugated linoleic acid supplemented to diet could reduce plasma levels of glucose and lipids which were increased in 27-weeks old Otsuka Long Evans Tokushima Fatty(OLETF) rats. Twenty male OLETF rats of 7 weeks old were fed an experimental diet containing 4.5%(w/w) total fat including 1% CLA and six of twenty rats were sacrified at 6 weeks feeding. The rest of OLETF rats was divided into 2 groups, one group was continuously fed for 14 weeks more the same experimental diet containing 1% CLA and the other group was fed control diet which eliminated CLA. CLA did not significantly reduce food intake and body weight gain in OLETF obese rats. Plasma triglyceride and total cholesterol level were significantly increased at older age of OLETF obese rats, but CLA could significantly reduce plasma cholesterol and triglyceride increased in obese rats. However, CLA was not strong enough to reduce the increased plasma glucose level and hepatic lipogenic enzyme acitivies. CLA was mostly deposited in epididymal fat pad and could be incorporated into hepatic microsomal membrane and did interfere the conversion of C18 : 0 into C18 : 1 in liver. In conclusion, CLA could have anti-atherogenic effect by reducing plasma cholesterol and triglyceride which was increased in genetically obese rats, but CLA(1%) was not good source of dietary fatty acid to reduce body fatness and plasma glucose which was increased by obese gene in older rats.

Effects of excessive dietary methionine on oxidative stress and dyslipidemia in chronic ethanol-treated rats

  • BACKGROUND/OBJECTIVE: The aim of this study was to examine the effect of high dietary methionine (Met) consumption on plasma and hepatic oxidative stress and dyslipidemia in chronic ethanol fed rats. MATERIALS/METHODS: Male Wistar rats were fed control or ethanol-containing liquid diets supplemented without (E group) or with DL-Met at 0.6% (EM1 group) or 0.8% (EM2 group) for five weeks. Plasma aminothiols, lipids, malondialdehyde (MDA), alanine aminotransferase (ALT), and aspartate aminotransferase were measured. Hepatic folate, S-adenosylmethionine (SAM), and S-adenosylhomocysteine (SAH) were measured. RESULTS: DL-Met supplementation was found to increase plasma levels of homocysteine (Hcy), triglyceride (TG), total cholesterol (TC), and MDA compared to rats fed ethanol alone and decrease plasma ALT. However, DL-Met supplementation did not significantly change plasma levels of HDL-cholesterol, cysteine, cysteinylglycine, and glutathione. In addition, DL-Met supplementation increased hepatic levels of folate, SAM, SAH, and SAM:SAH ratio. Our data showed that DL-Met supplementation can increase plasma oxidative stress and atherogenic effects by elevating plasma Hcy, TG, and TC in ethanol-fed rats. CONCLUSION: The present results demonstrate that Met supplementation increases plasma oxidative stress and atherogenic effects by inducing dyslipidemia and hyperhomocysteinemia in ethanol-fed rats.
