Dietary restriction (caloric restriction) is the only intervention which has been reliably shown to extend the maximum life span of warm-blooded animals and delay the many phenomena associated with aging. It is also one of the most effective modulators of toxicity, especially cancer endpoints. In spite of the known modulator effects of caloric restriction, the biological mechanisms responsible for these effects had not been in vestigated until recently. The National Center for Toxicological Research (NCTR), in a collaborative effort with the National Institute of Aging (NIA), initiated a project whereby nine (9) combinations of rodent species/strains and diets were fed both restricted and ad libitum. The NIA's initiative was to identify biomarkers of aging whereas NCTR's initiative was to identify the biological effects associated with the profound effects caloric restriction has in protecting against both spontaneous (age-related) and chemically-induced toxic endpoints. Independent of sex or species, caloric restriction has similar effects on body temperature, oxygen consumption and $CO_2$production. Caloric restriction also decreased lipid glycolysis and metabolism in rats and mice, which suggest decreased production of metabolites which could lead to fatty acid epoxide formation. The age-associated loss of ciradian regulation of intermediate enzymes is also significantly reduced. Moreover, caloric restriction reduced the age-associated feminization of sexually dimorphic liver isozymes, increased several glucocorticoid responsive isozymes, elevated glucagon/insulin ratios, produced less microsomal superoxide and enhanced the capacity for utilzing detoxicating metabolic pathways. Calorically restricted rats have less than half the number of aflatoxin ($AFB_1$)-DNA adducts than ad libitum animals and urinary excretion of $AFB_1$ was increased significantly. Finally, DNA repair mechanisms are enhanced and oncogene expression is decreased in calorically restricted animals.
This experiment was conducted for the development of oriental feed additives in broiler chicks. It conducted experiment to investigate the effect of $1\%$ dietary supplementation of oriental feed additives containing oriental herbal medicines such as Saururus Chnensis B.(S. chnensis) and Carthamus Tinctorius L.(C. tinctorius) on growth performance, and physicochemical properties in broiler chicks. The results of this study demonstrated that chicks feed $1\%$ oriental feed additives decreased feed gain, and death rates compared with control treatment. Weight gain and meat color were higher in the $1\%$ supplement group than control. The supplementation of $1\%$ oriental feed additives increased unsaturated fatty acid, and reduced crude fat in the broiler chicks compared with those of control. In conclusion, this study implied that oriental feed additives containing oriental herbal medicines such as S. Chnensis and C. Tinctorius were effective in improving broiler chicks on growth performance, physical-chemical properties and is highly recommended for increasing economic returns.
The anti-obesity effects of a hot water extract from wasabi (Wasabia japonica Matsum.) leaves (WLE), without its specific pungent constituents, such as allyl-isothiocyanate, were investigated in high fat-diet induced mice. C57J/BL mice were fed a high-fat diet (control group) or a high-fat diet supplemented with 5% WLE (WLE group). Physical parameters and blood profiles were determined. Gene expression associated with lipid metabolism in liver and white adipose tissue were analyzed. After 120 days of feeding, significantly lower body weight gain, liver weight and epididymal white adipose tissue weight was observed in the WLE group compared to the control group. In liver gene expression within the WLE group, PPAR${\alpha}$ was significantly enhanced and SREBP-1c was significantly suppressed. Subsequent downstream genes controlled by these regulators were significantly suppressed. In epididymal white adipose tissue of the WLE group, expression of leptin, PPAR${\gamma}$, and C/EBP${\alpha}$ were significantly suppressed and adiponectin was significantly enhanced. Acox, related to fatty acid oxidization in adipocytes, was also enhanced. Our results demonstrate that the WLE dietary supplement induces mild suppression of obesity in a high-fat diet induced mice, possibly due to suppression of lipid accumulation in liver and white adipose tissue.

This study was conducted to investigate the effect of the total digestible nutrients (TDN) level of commercial concentrates on growth performance, carcass characteristics, and meat composition of late fattening Hanwoo steers. A total of 28 steers were randomly assigned to one of four dietary groups; T1 (73.30% TDN), T2 (74.50% TDN), T3 (76.40% TDN), and T4 (77.10% TDN). Average daily gain (ADG) was slightly but not significantly higher in the T2 than in the other treatments. Dry matter intake (DMI) and feed conversion ratio (FCR) were higher in the T2 than in the other treatments; however, the differences were not statistically significant. Carcass back fat thickness was thicker in the T4 and marbling score was higher in the T2 than in the other treatments; however, the differences were not statistically significant. The TDN level of concentrates had no effect on the physicochemical characteristics and fatty acid composition of the longissimus muscle. The finding of this study indicate that less than 74% or greater than 75% TDN in the commercial concentrate did not contribute to improve ADG, FCR, marbling score; therefore, in the present study, the recommendable TDN level in the commercial concentrate for late fattening period was 74% to 75% in terms of growth performance and marbling score of Hanwoo steer.
Objective: The objective of current study was to investigate the lactation performance and rumen fermentation characteristics of dairy cows fed a diet with alfalfa hay replaced by corn stover but supplemented with molasses. Methods: Sixteen Holstein cows in mid-lactation were randomly assigned to 1 of 2 dietary treatments: i) alfalfa based diet (AH), and ii) corn stover based diet supplemented with molasses (CSM). The experiment was conducted according to a $2{\times}2$ crossover design with 22-d each period, consisting of 17 d for adaptation and 5 d for data and samples collection. Results: Dry matter intake and milk yield were higher for cows fed AH than CSM (p<0.01). Milk protein content and nitrogen conversion were higher (p<0.05), while milk urea nitrogen was lower (p<0.01) for cows fed AH than CSM-fed cows. Contents of milk total solids, fat and lactose were not different between two groups (p>0.10). Total rumen volatile fatty acid concentration tended to be higher (p = 0.06) for cows fed AH than CSM-fed cows. Molar proportion of acetate was lower (p = 0.04), but valerate was higher (p = 0.02) in cows fed AH than CSM-fed cows. Rumen concentration of propionate, and isobutyrate, and ratio of acetate to propionate tended to be different (p<0.10) between two groups. The feed cost per kilogram of milk was lower in CSM than AH (p<0.01). No differences were found in feed efficiency and most plasma parameters tested (p>0.10). Conclusion: In comparison with AH diet, CSM diet could be fed to dairy cows without negative effect on feed efficiency, ruminal fermentation, but economically beneficial, indicating that CSM could be an alternative choice for dairy farms instead of AH to feed midlactation dairy cows.
Objective: This study aimed at studying the potential use of Flemingia (Flemingia macrophylla) as a protein source fodder to improve nutrients digestibility and ruminal fermentation efficiency in beef cattle. Methods: Four, Thai native beef cattle were randomly assigned in a 4×4 Latin square design. Four levels of Flemingia hay meal (FHM) were used to replace soybean meal (SBM) in the concentrate mixtures in four dietary treatments replacing levels at 0%, 30%, 60%, and 100% of SBM. Results: The experimental findings revealed that replacements did not effect on intake of rice straw, concentrate and total dry matter (DM) intake (p>0.05). However, the apparent digestibilities of DM, organic matter, crude protein, acid detergent fiber, and neutral detergent fiber were linearly increased up to 100% replacement levels. Moreover, the production of total volatile fatty acids, and propionate concentration were enhanced (p<0.05) whereas the concentration of acetate was reduced in all replacement groups. Consequently, the CH4 production was significantly lower when increasing levels of FHM for SBM (p<0.05). Furthermore, rumen bacterial population was additionally increased (p<0.05) while protozoal population was clearly decreased (p<0.05) in all replacement groups up to 100%. In addition, microbial nitrogen supply and efficiency of microbial nitrogen synthesis were enhanced (p<0.05), as affected by FHM replacements. Conclusion: The findings under this experiment suggest that 100% FHM replacement in concentrate mixture enhanced rumen fermentation efficiency, nutrients digestibilities, bacterial population, microbial protein synthesis, and subsequently reduced CH4 production in beef cattle fed on rice straw.
Objective: An experiment was conducted to assess the effect of dragon fruit peel pellet (DFPP) as a rumen enhancer of dry matter consumption, nutrient digestibilities, ruminal ecology, microbial protein synthesis and rumimal methane production in Holstein crossbred bulls. Methods: Four animals, with an average live-weight of 200±20 kg were randomly assigned in a 4×4 Latin square design to investigate the influence of DFPP supplementation. There were four different dietary treatments: without DFPP, and with 200, 300, and 400 g/h/d, respectively. Results: Results revealed that dry matter consumption of total intake, rice straw and concentrate were not significantly different among treatments (p>0.05). It was also found that ruminal pH was not different among treatments (p>0.05), whilst protozoal group was reduced when DFPP increased (p<0.01). Blood urea nitrogen and NH3-N concentrations were increased at 400 g of DFPP supplementation (p<0.01). Additionally, volatile fatty acid production of propionate was significantly enhanced by the DFPP supplementation (p<0.05), while production of methane was consequently decreased (p<0.05). Furthermore, microbial protein synthesis and urinary purine derivatives were remarkably increased especially at 400 g of DFPP supplementation (p<0.05). Conclusion: Plant secondary compounds or phytonutrients (PTN) containing saponins (SP) and condensed tannins (CT) have been reported to influence rumen fermentation. DFPP contains both CT and SP as a PTN. The addition of 400 g of DFPP resulted in improved rumen fermentation end-products especially propionate (C3) and microbial protein synthesis. Therefore, DFPP is a promising rumen enhancer and indicated a significant potential of DFPP as feedstuff for ruminant feed to mitigate rumen methane production.

BACKGROUND/OBJECTIVES: Aster yomena (Kitam.) Honda (AY) has remarkable bioactivities, such as antioxidant, anti-inflammation, and anti-cancer activities. On the other hand, the effects of AY against obesity-induced insulin resistance have not been reported. Therefore, this study examined the potential of AY against obesity-associated insulin resistance in high-fat diet (HFD)-fed mice. MATERIALS/METHODS: An obesity model was established by feeding C57BL/6J mice a 60% HFD for 16 weeks. The C57BL6/When ethyl acetate fraction from AY (EFAY) at doses of 100 and 200 mg/kg/day was administered orally to mice fed a HFD for the last 4 weeks. Normal and control groups were administered water orally. The body weight and fasting blood glucose were measured every week. Dietary intake was measured every other day. After dissection, blood and tissues were collected from the mice. RESULTS: The administration of EFAY reduced body and organ weights significantly compared to HFD-fed control mice. The EFAY-administered groups also improved the serum lipid profile by decreasing the triglyceride, total cholesterol, and low-density lipoprotein compared to the control group. In addition, EFAY ameliorated the insulin resistance-related metabolic dysfunctions, including the fasting blood glucose and serum insulin level, compared to the HFD-fed control mice. The EFAY inhibited lipid synthesis and insulin resistance by down-regulation of hepatic fatty acid synthase and up-regulation of the AMP-activated protein kinase pathway. EFAY also reduced lipid peroxidation in the liver, indicating that EFAY protected hepatic injury induced by obesity. CONCLUSIONS: These results suggest that EFAY improved obesity-associated insulin resistance by regulating the lipid and glucose metabolism, suggesting that AY could be used as a functional food to prevent obesity and insulin resistance.
BACKGROUND/OBJECTIVES: Hyperglycemia is a major cause of diabetes and diabetesrelated diseases. Sodium butyrate (NaB) is a short-chain fatty acid derivative that produces dietary fiber by anaerobic bacterial fermentation in the large intestine and occurs in foods, such as Parmesan cheese and butter. Butyrate has been shown to prevent obesity, improve insulin sensitivity, and ameliorate dyslipidemia in diet-induced obese mice. Therefore, this study examined the effects and mechanism of NaB on the secretion of inflammatory cytokines induced by high glucose (HG) in THP-1 cells. MATERIALS/METHODS: THP-1 cells were used as an in vitro model for HG-induced inflammation. The cells were cultured under normal glycemic or hyperglycemic conditions with or without NaB (0-25 μM). Western blotting and quantitative polymerase chain reaction were used to evaluate the protein and mRNA levels of nuclear factor-κB (NF-κB), interleukin-6, tumor necrosis factor-α, acetylated p65, acetyl CREB-binding protein/p300 (CBP/p300), and p300 using THP-1 cells. Histone acetyltransferase (HAT), histone deacetylase (HDAC), and pro-inflammatory cytokine secretion activity were analyzed using an enzyme-linked immunosorbent assay. RESULTS: HG significantly upregulated histone acetylation, acetylation levels of p300, NF-κB activation, and inflammatory cytokine release in THP-1 cells. Conversely, the NaB treatment reduced cytokine release and NF-κB activation in HG-treated cells. It also significantly reduced p65 acetylation, CBP/p300 HAT activity, and CBP/p300 gene expression. In addition, NaB decreased the interaction of p300 in acetylated NF-κB and TNF-α. CONCLUSIONS: These results suggest that NaB suppresses HG-induced inflammatory cytokine production through HAT/HDAC regulation in monocytes. NaB has the potential for preventing and treating diabetes and its related complications.

Bariatric surgery is the most effective treatment for sustained weight reduction, and it can result in substantial improvements in the severity of type 2 diabetes, metabolic syndrome, nonalcoholic fatty liver disease, and quality of life. However, sleeve gastrectomy, a weight loss surgery that removes two-thirds of the stomach, reduces appetite and nutrient absorption, impairing digestion and the absorption of nutrients like iron, vitamin B12, and protein-bound nutrients. This case study aims to demonstrate that patients undergoing sleeve gastrectomy require long-term and periodic monitoring of biochemical data, weight changes, and caloric and protein intake by a professional nutritionist to prevent malnutrition and nutritional deficiencies. In this case study, a 48-year-old woman was diagnosed with morbid obesity, hypertension, sleep apnea syndrome, and chronic gastritis. At initial evaluation, she was 160 cm tall and weighed 89 kg, with a body mass index of 34.8 kg/m2. At 1 postoperative year, she consumed 650 kcal and 25 g of protein per day, the percentage of excess weight loss was 141.1%, and body mass index was 21 kg/m2. Compared to preoperative levels, calcium and folic acid levels did not decrease after 1 postoperative year, but hemoglobin, ferritin, and vitamin B12 levels decreased. In conclusion, when patients experience rapid weight loss after sleeve gastrectomy, follow-up should be frequent and long. Dietary education should be conducted according to digestive symptoms, and oral nutritional supplements, including vitamins and minerals.
