Effects of Alcohol Consumption and Fat Content in Diet on Chemical Composition and Morphology of Liver in Rat (알콜과 식이지방량이 흰쥐의 간 지질조성과 간조직형태에 미치는 영향)

  • Effects of alchohol and fat content in a balanced diet on chemical composition and morphology of liver were investigated in growing rats. Fourth eight male rats of Sprague-Dawley strain weighing about 160g were divided into 4 groups ; high fat diet group, alcohol-administered high fat diet group, low fat diet group and alcohol-administered high fat diet group, low fat diet group and alcohol-administered low fat diet group. High and low fat diets supplied 30% and 12%, respectively, of total calorie intake from fat, and alcohol was given by adding ethanol in drinking waster at 10%. Diets contained adequate amounts of all nutrients required for rats, including lipotrpoic agents(choline and methionine) to minimize effects of factors other than alcohol on liver damage. Ratios of liver weight to body weight were statistically different among groups. Liver/dody weight ratios alcohol-administered rats were significantly higher than those of non-alcohol groups after 6 weeks treatment. Although total lipid and triglyceride per gram liver were increased in alcohol-administered rats, especially low fat diet fed rats, the values were not significantly different. Opticmicroscopical observation revealed increase in cell size and no change in morphology of liver. Examination of hepatocytes by electron microscopy showed that fat droplets were observed in all groups but enlarged in the alcohol-administered low fat diet fed rat. Contents of protein, cholesterol and phospholipid were not affected by alcohol consumption. The level of lipid peroxide was significantly lower in the livers of alcohol-administered rats than in the livers of non-alcohol groups. The results of this study indicate that even moderate alcohol drinking and dietary fat content did not affect any significant change in composition and morphology of liver until 6 week treatment but that even moderate alcohol drinking caused some signs of steatosis of liver.

Histopathological Studies on Effect of Dropwort on Cadmium Poisoning in Mice (미나리가 마우스의 카드뮴 중독에 미치는 병리조직학적 관찰)

  • This study was undertaken to find out the effect of dropwort on histopathological changes of cadmium toxicity in mice. Thirty two mice of BALB/c strain were divided into a control group (A) and three experimental groups (B, C, D). Each group was consisted of four males and four females : group A received tap water and basal diet, group B received tap water and diet supplemented with 100 mg/kg of cadmium, group C received dropwort juice and diet supplemented with 100 mg/kg of cadmium, group D received dropwort juice and basal diet. All mice were dissected on the 35th day. Pathological changes in liver, kidney, spleen, heart, lung, stomach and muscle were observed. Group D showed no significant change as the control group. But group B showed many cytoplasmic vacuoles and necrosis of hepatocytes, glomerular swelling, and degeneration and necrosis of convoluted tubules in kidney, marked congestion and hemorrhage, and a large number of variably sized alveolar macrophages appearance in lung and swelling in some part of muscle fiber. On the other hand, group C showed a little convalescent changes and maintained their normal architectures in liver, kidney, lung and muscle.

The Effects of Isocaloric High-Fat Diet and Endurance Exercise on Insulin Resistance and Mitochondria Biogenesis (Isocaloric high-fat diet와 지구성 운동이 인슐린 저항성과 미토콘드리아 생합성에 미치는 영향)

  • The purpose of this research was to find effects of high fat diet and endurance exercise on mitochondria biosynthesis and insulin resistance in male Wister rat. Th rats were randomly separated into 4 groups: A, Isocaloric high fat diet group (50% of calories from fat), B. Chow group, C. Isocaloric high fat diet with endurance exercise (treadmill running, slop 8%, 23 m/min, 120 min/day, 5 days per week), D. Chow group with endurance exercise. Both Isocaloric high fat diet group and chow group were given an equal caloric composition with 3.2kcal/g. equal amount of food were checked every day and given to both groups. 4 weeks of high fat diet did not show any change in body weight and amount of body fat. Further the level of glucose and insulin in blood, and insulin-stimuilated glucose transport rate in epitrochlearis muscle was not affected by Isocaloric high fat diet. However, the endurance exercise showed statistically significant change in the level of insulin in blood. Although either Isocaloric high fat diet or endurance exercise alone did not change on mitochondria biogenesis marker, Isocaloric high fat diet with endurance exercise could induce the increased level of marker (p<0.05). Also, plasma free fatty acids were increased in this group. From this investigation, Isocaloric high fat diet with moderate-intensity endurance exercise is effective way to induce mitochondrial biogenesis.

The Inhibitory Effects of Bimanbang-1(肥滿1號方) on the Obese-mouse Fed High-fat Diet (비만1호방(肥滿1號方)이 고지방식이(高脂肪食餌)로 비만(肥滿) 유도(誘導)된 백서(白鼠)에 미치는 영향)

  • Objective: This study investigated the effects of Bimanbang-1 (肥滿1號方: here in after referred to BMB-1) on the obese gene and obese inhibitory. Methods: C57BL/6 mice were induced by high fat diet. Mice were divided into three groups (normal, high fat diet with control, high fat diet with BMB-1 extract) and fed for 15 weeks. Items of this experimental study are as follows: body weight change, final body weight, the weight change of the adipocytes in body, the level change of LFT, NEFA and creatinine, the expression of ${\beta}3AR$ and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}3AR$, leptin and $TNF-{\alpha}$ in adipocytes tissue. Result: 1. All experimental groups showedthat the weight change decreased considerably and the high density group showedthat the final weight decreased considerably. 2. The high density group showed that the amount of the adipocyte in weight decreased considerably. 3. All experimental groups showedthat the amount of ALT decreased considerably, and AST decreased in the high density group. However, the amount of creatinine and glucose did not increase considerably. 4. All experimental groups showed that the amount of total cholesterol, LDL-cholesterol, triglyceride, and NEFA decreased, and HDL-cholesterol increased considerably. 5. The high density groups showedthat the amount of leptin decreased considerably. 6. All experimental groups showed that the revelation of ${\beta}3AR$ in primary adipose cell and 3T3-L1 cell increased considerably, and that the revelation of leptin and $TNF-{\alpha}$ in primary adipose cellsand 3T3-L1 cells decreased considerably. 7. All experimental groups showed that the size of adipocyte in adipocytes tissue decreased. 8. All experimental groups showed that the adipose vacuoles in liver tissue decreased considerably. Conclusion: The findings suggest that Bimanbang-1 causes weight loss and histological change, thus it may be effective to treat obesity.

THE EFFECTS OF CALCIUM-DEFICIENT DIET AFTER IRRADIATION ON THE PERIODONTAL TISSUE FORMATION IN RAT PUPS (방사선조사와 저칼슘식이가 성장기 백서의 치주조직에 미치는 영향에 관한 실험적 연구)

  • The present study was designed to elucidate the effects of the Co-60 γ irradiation and/or calcium-deficient diet on the periodontal tissue formation in rat pups. The pregnant three-week old Sprague-Dawley rats were used for the study. The experimental group was divided into two groups, irradiation/normal diet group (Group 2) and irradiation/calcium-deficient diet group (Group 3). The control group was non-irradiation/normal diet group (Group 1). The abdomen of the rats at the 19th day of pregnancy were irradiated with single absorbed dose of 350 cGy. The rat pups were sacrificed on the 14th day after delivery, and the maxillae including molar tooth germ were taken. The specimens including the 1st molar tooth germ were prepared to make tissue sections for light and transmission electron microscopy. Some of tissue sections for light microscopy were stained immunohistochemically with anti-fibronectin and anti-osteonectin antibodies. The results were as follows; 1. In the periodontal ligament forming area, the fibroblasts of Group Z showed irregular arrangement and low activity. The immunoreactivity between the fibroblasts and collagen fibers was decreased, compared with Group 1. The fibroblasts of Group 3 showed atrophic change and clumped nucleus. The collagen fibers showed cystic change and low immunoreactivity to the fibronectin. 2. In the cementum forming area, the cementoblasts of Group 2 showed decrease of number and atrophic change. The cementoblasts of Group 3 showed edematous change, atrophy of cytoplasm, and clumping of nucleus. 3. In the alveolar bone forming area, the bone of Group 2 was thin and various degree of immunoreactivity to the osteonectin. Group 3 showed edematous osteoblasts, fibrous degeneration of bone marrow, and weak immunoreactivity to the osteonectin.

The Inhibitory Effects of So-yangin Biman-bang on the Obese-Mice Induced by High-Fat Diet (소양인(少陽人) 비만방(肥滿方)이 생쥐의 비만 억제에 미치는 영향)

  • Objectives : In order to investigate the effects of So-yangin Biman-bang(here in after referred to BM) on the obese gene and obese inhibitory, C57BL/6 mice were distinctively induced by high fat diet. Methods : C57BL/6 mice were divided into four groups(normal, high fat diet with control, high fat diet with Reductil(here in after referred to RDT), high fat diet with BM extract) and have been fed for 8 weeks. Through observing the process, the weight change of the adipocytes in body and liver, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, glucose, leptin change in the serum, the expression of ${\beta}3AR$ and leptin gene in 3T3-L1 cell and primary adipocyte cell. Results : The following results have been found in this study. 1. Losing weight have significantly been shown. 2. The weight change of the body adipocyte and liver have been decreased but no significance has been found. 3. With close observation on the change of blood, HDL-cholesterol has positively been increased in numbers but the level of glucose, LDL-cholesterol, triglyceride have distinctively been decreased. 4. The manifestation of ${\beta}3AR$ in 3T3-L1 cell and primary adipocyte cell have shown a definite increase in numbers but the manifestation of leptin has shown certain decrease in its amount. Conclusions : As results of this, So-yangin Biman-bang is thought to be effective in metabolic syndrom and lipid metabolism disorder that can possibly be occurred by obesity control and treatment. The clinical research will be needed in order to apply to So-yangin constitution in the future.

Effects of Bangpoongtongsungsankamibang extract on the obese SD rats induced by 2% cholesterol diet (방풍통성산가미방(防風通聖散加味方)이 비만유도(肥滿誘導) 백서(白鼠)에 미치는 영향(影響))

  • Objectives : These experimental studies were designed to investigate the effects of Bangpoongtongsungsankamibang on the weight and the lipid metabolism of the obese rats induced by 2% cholesterol diet. They were designed to change values in the serum and leptin levels in the rats. Materials and Methods : The serum and leptin levels were measured for a period of 8 weeks for obese SD rats induced by 2% cholesterol diet. Results : The body weight showed a tendency to decreased significantly in sample (p<0.05). The Total cholesterol and Triglyceride level significantly decreased in sample (p<0.05). The HDL Cholesterol levels significantly increased in sample (p<0.05). The LDH levels did not show a significant change in sample. The ALP levels significantly decreased in sample (p<0.05). The SGOT and SGPT level did not show a significant change in sample. The Leptin levels significantly decreased in sample (p<0.05). The average size of the epididymal fat cell significantly decreased in sample (p<0.05).

  • This study was attempted to observe the effect of Job's tear diet on change of body weight and tissue in rat. The changes of body weight were higher in Job's tear non-diet group(A) and seemed to lower in Job's tear diet group(c) as expected. The weight of each organ appeared to heavy more C group than A,Bgroup in Brain, Spleen. Lungs and Kidney. Liver was heavier in Job's tear to diet one day among six day group(B) and ovary was lighter in C group, The levels of each organ TBA-value were higher in Brain, Liver of A group and showed to higher Spleen, Lungs, Ovary of C group TBA-value of Kidney was revealed to lower more C group than A,B group.

Effect of ST36.ST44 and ST25.ST36 Electroacupuncture Stimulation on High Fat Diet Induced Obese Mouse (고지방 식이 유도 비만에 대한 족삼리.내정, 천추.족삼리 전침 자극의 효과)

  • The aim of this study is to learn the effects of ST36 ST44 and ST25 ST36 electroacupuncture stimulation on high fat diet-induced obese mouse. The control group was provided with high fat diet without electroacupuncture stimulation, while the experimental group received a electroacupuncture stimulation on ST36 ST44 and ST25 ST36 with three frequencies (10Hz, 100Hz, 200Hz) for 5 times a week during five week periods with the high fat diet. We investigated the body weight, weight gain, and food intake. After the electroacupuncture stimulation, a significant inhibition of increase in weight was found in the experimental group compared with the high fat diet group. But there were no significant differences made by the change of acupoint combinations and frequencies. After the electroacupuncture stimulation, no significant change was found in the food intakes, but the weight gain a week was decreased compared with the high fat diet group. Electroacupuncture stimulation may be effective in treatments of obesity to high fat diet, But no significant difference was found between three frequencies.

The Effect on the Change of Liver and Serum Cholesterol Contents of Albino Rats by the Feeding of the Suerose and Glucose (蔗糖과 葡萄糖이 白鼠肝臟 및 血淸 Cholesterol의 含量에 미치는 影響)

  • This experiment was made in order to observe the change in contents of the total cholesterol, free cholesterol and ester cholesterol in the liver and serum by the feeding of glucose and sucrose. The animals used for the experiment were adult male albino rat from a pure strain, weighing 285-332g. The animals were divided into standard, glucose and sucrose diet groups. The glucose and sucrose diet groups were redivided into 10%, 25% and 40% diet groups. Their liver and serum were used as sample after they were fed with the corresponding diets, respectively, for two months. 1. In the liver, the total cholesterol and ester cholesterol contents in the 40% glucose diet group were significantly increased and the free cholesterol contents in all diets were respectively increased, compared with the standard diet group. 2. In the serum, the total cholesterol contents in both diet groups, the 10% glucose and 10% sucrose, were decreased but the content in the 40% sucrose diet group was increased, compared with the standard diet group. The free cholesterol contents in the 10 and 40% glucose diet and 10% sucrose diet group were decreased, compared with the standard diet group. The ester cholesterol contents in the 10% glucose and 10% sucrose diet groups were decreased but the content in the 40% sucrose diet group was increased, compared with the standard diet group. Being taken into consideration of the above facts it was acquired that the cholesterol contents are affected by the amounts and kinds of dietary carbohydrate when the protein contents in each diet were constant.
