Effect of Ethyl Acetate Extract of the Dried Prunus mume on the Growth of Staphylococcus aureus (오매의 에틸아세테이트 추출물이 Staphylococcus aureus의 증식에 미치는 영향)

  • The dried Prunus mume, an alkaline food abundant in organic acids (citric acid, malic acid and tartaric acid), has been largely used in both folklore remedies and Chinese herbal medicine for a long time. This study was performed to investigate the antimicrobial activity of the dried Prunus mume. The fractionation of the methanol extracts from Prunus mume was conducted using petroleum ether, chloroform, ethyl acetate and butanol, respectively. The antimicrobial activity of the Prunus mume extracts was then determined against food-borne pathogens using a paper disc method. The ethyl acetate extract showed the strongest antimicrobial activity against the eigth food-born pathogens used in this present study. Diaion HP 20 column chromatography was performed to remove some sugars that might inhibit the antimicrobial activity of Prunus mume. The strongest antimicrobial activity of ethyl acetate fraction of Prunus mume was shown against Staphylococus aureus. The growth inhibition curve was determined using ethyl acetate extracts of Prunus mume against Staphylococus aureus, which showed the growth inhibition up to 72 hours at 1,000 ppm concentration.

Meliasendanins E-J, Nor-neolignan Constituents from Melia toosendan and their Anti-inflammatory Activity

  • A phytochemical investigation of the fruits extract of Melia toosendan afforded the isolation of two new nor-neolignans, meliasendanins E (1) and F (2), as well as twelve known compounds (3 - 14) using various separation technique such as Diaion HP20, silica, RP-18 gel column chromatography and semi-preparative HPLC. Their chemical structures were elucidated by extensive NMR spectroscopic data including 2D-NMR, and HR-ESI-MS as well as ECD data. Among the twelve known compounds, the absolute structures of 3 - 6 were determined first, and given the trivial names as meliasendanins G-J (3 - 6). Based on the evaluation of anti-inflammatory activity, compounds 7 - 8 exhibited inhibitory effects on LPS-induced nitric oxide production in RAW 264.7 macrophages with IC50 values of 34.6 and 39.5 µM, respectively.

Isolation and Identification of a Novel Anticancer Compound from Solanum nigrum (용규(Solanum nigrum)에서 HT29 세포에 대한 신규 항암 활성 단일 물질 분리)

  • To identify and isolate anticancer active compounds from Solanum nigrum, S. nigrum was extracted with MeOH and then fractionated with various organic solvents ($CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$). The cytotoxic effects of the MeOH extracts from S. nigrum and its organic solvent-soluble fractions were also tested in HT29 cells. All the MeOH extracts of S. nigrum and its organic-solvent extracts induced cytotoxicity in the HT29 cells. Among the extracts, $H_2O$ was the most effective. The $H_2O$ extract was purified further by repeated silica gel, Sephadex LH-20, Diaion HP- 20, and RP-18 column chromatography. An active anticancer compound, Des-N-26-methylene-dihydrotomatidine, was isolated with a molecular weight of 416 and a molecular formula of $C_{28}H_{48}O_2$. Analysis of the cytotoxic effects of Des-N-26-methylene-dihydrotomatidine on the HT29 cells compared to those of tomatine and tomatidine are similar in its structure, is higher than tomatidine above the 40 ${\mu}g/ml$ concentration, but lower than tomatine. This is the first study to describe the anticancer activity of Des-N-26-methylene-dihydrotomatidin, isolated from S. nigrum. Des-N-26- methylene-dihydrotomatidine seems to have potential as a natural bioactive compound.

Production and Characteristics of Antidandruffy Compound from Chrysanthemum zawadskii (구절초(Chrysanthemum zawadskii)로부터 비듬균에 대한 항균물질의 생산 및 특성)

  • In order to develop a potent antidandruffy agent from medicinal plants, various extracts of medicinal plants and herbs were prepared and investigated its antifungal activity against Malassezia furfur. Among 85 kinds of the secondary screened medicinal plants and herbs, water extract of Chrysanthemum zawadskii had the highest antifungal activity against dandruffy-producing Malassezia furfur. The antifungal compound was also maximally extracted when the powder of Chrysanthemum zawadskii was treated with water at $50^{\circ}C$ for 6 hours. The antifungal compound from Chrysanthemum zawadskii was partially purified by silica gel column chromatography and the partial purified compound was soluble at $70^{\circ}C$ for 20 min and pH $3.0{\sim}9.0$.

  • Pata de Vaca (Bauhinia forficata) Is a tree which grows naturally in the rainforests and tropical parts of Peru and Brazil, as well as tropical zones of Asia, eastern Paraguay and northeastern Argentina. The active fraction (Pata-50) of the 70% ethanol extract from Pata de Vaca was sequentially fractionated by HP-20 Diaion column chromatography and C-18 column chromatography, and its characteristics were investigated. The growth of all cancer cells tested except for MCF-7 was Inhibited in a concentration-dependent manner by Pata-50. Its $IC_{50}$ values were estimated to be 40.4 $\mu\textrm{g}$/$m\ell$ on AGS, 51.3 $\mu\textrm{g}$/$m\ell$ on HT-29, 52.1$\mu\textrm{g}$/$m\ell$ on HepG2, 65.2$\mu\textrm{g}$/$m\ell$ on A549, and 77.5$\mu\textrm{g}$/$m\ell$ on HeLa cells. A flow cytometric analysis of HepG2 cells revealed induction of apoptosis, but cell cycle regulation was not affected. The HepG2 cell population of apoptosis region increased In a concentration-dependent manner by Pata-50.

Anti-Obesitic Effects of Cathepsin S Inhibitory Fraction Derived from Paecilomyces tenuipes in Mice Fed a High-Fat Diet (고지방식이를 섭취한 쥐에서 눈꽃동충하초 유래 cathepsin S 저해 분획의 비만 억제 효과)

  • In this study, cathepsin S (CTSS) inhibitory fraction was isolated from Paecilomyces tenuipes and anti-obesitic effects of the fraction were evaluated in mice, fed a high-fat diet. Hot water extract of P. tenuipes (DHW) was divided into 2 fractions, water eluate fraction (DHP1) and methanol eluate fraction (DHP2) using Diaion HP-20. $IC_{50}$ values for DHW, DHP1 and DHP2 against CTSS were 108.7, 890.3 and 2.3 ${\mu}g$/mL, respectively. To evaluate anti-obesitic effects of the fractions, each fraction was administrated orally to C57BL/6 mice for 4 weeks with a high-fat diet. DHP2 had the highest inhibitory effect on CTSS activity, causing serious reduction in weight gain, a reduction in the amount of adipose tissue and in serum lipids levels. These results suggest that the inhibition of CTSS by compounds derived from P. tenuipes may be effective in preventing and in ameliorating obesity.

Quantitative Changes of Polyphenolic Compounds in Mulberry (Morus alba L.) Leaves in Relation to Varieties, Harvest Period, and Heat Processing

  • Six polyphenolic compounds, such as chlorogenic acid (CA), rutin (RT), isoquercitrin (IQT), quercetin-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (QMG), astragalin (AG), kaempferol-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (KMG), were isolated from mulberry leaves by a series of isolation procedures, such as Diaion HP-20, silica-gel, Sephadex LH-20, and ODS-A column chromatographies. The chemical structures of the phenolic compounds were identified by UV and NMR spectral analyses. Levels of polyphenols in mulberry leaves from six different mulberry cultivars ranged from 1,042.16 to 1,871.97 mg% per dry weight; Guksang cultivar showed the highest levels of polyphenols, whereas Gaeryangdaehwa contained the least polyphenol contents. Generally, levels of polyphenols in mulberry leaves decreased with increasing harvest time, except for Yoolmok, but increased with heat processing time, except QMG and KMG. These results suggest that the heat processed mulberry leaves of Guksang cultivar harvested in early May can be potentially useful sources for production of high quality mulberry leaf teas.

MR304-1, A Melanin Synthesis Inhibitor Produced by Trichoderma harzianum (Trichoderma harzianum이 생산하는 melanin 생성 저해물질 MR304-1)

  • During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR304 which was capable of producing high level of an inhibitor was selected. Based on taxonomic studies, this fungus could be classified as Trichoderma harzianum. The active compound (MR304-1) was purified from culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatographv and HPLC. The inhibitor was identified as 3-(1,5-dihvdroxy-3-isocyanocyclopent-(E)-3-envl)prop-2-enoate by spectroscopic methods of UV, ESIMS, $^{1}$H-NMR, $^{13}$C-NMR, NOE, HMQC and HMBC. MR304-1 showed strong mushroom tyrosinase inhibitory activity with IC$_{50}$ value of 0.25 $\mu $g/ml. It inhibited melanin biosynthesis with 15 mm inhibition zone at 30 $\mu $g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work. It also inhibited melanin biosynthesis in B16 melanoma cells with a niinimum inhibitory concentration of 0.05 $\mu $g/ml.

Sterin C, a New Antioxidant from the Mycelial Culture of the Mushroom Stereum hirsutum

  • Structurally new antioxidative metabolite was isolated from mycelial culture of mushroom Stereum hirsutum. Culture broth was subjected to Diaion HP-20 column chromatography, and 70% aqueous MeOH eluent was extracted with EtOAc. EtOAc extract was purified through silica gel and Sephadex LH-20 column chromatographies, and reversed phase $C_{18}$ HPLC. Compound was revealed to be new dihydroxylated derivative of sterin B with molecular formula of $C_{12}H_{16}O_5$(MW 240) by MS and various NMR spectral data analyses, and designated as sterin C. Sterin C showed superoxide radical-scavenging activity with $EC_{50}$ value of 0.31 mM.

Isolation and Identification of bakkenolides and caffeoylquinic acids from the aerial parts of Petasites japonicus

  • The major aim of this work is the research of secondary metabolites isolated from the aerial parts of Petasites japonicus. The plant material is extracted with a polar solvent, which is 95% by volume methanol at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAC and n-BuOH fraction, two bakkenolides and two caffoylquinic acid were isolated using the Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies. According to the results of the results of physico-chemical and spectroscopic data including NMR, MS and UV. The chemical structures of the compounds were respectively determined as bakkenolide B (1), bakkenolide D (2), 1,5-dicaffeoylquinic acid (3), and 5-O-caffeoylquinic acid (4). These results suggest that the compounds isolated from the aerial parts of this plant were almost identical with known components of Petasites japonicus. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest area and time.
