Utilization and Isolation of new active substances from Sericulture Related MaterialsI.Potentiation of Mulberry leaves for diseases attacking aged population

  • As the average span of human life is continuously increasing, especially the old aged groups, are suffering from various chronic and critical diseases e.g. cardiovascular diseases, coronary heart disease, diabetes, atherosclerosis and alzheimer's etc. However, effective and safe treatment methods have not yet been investigated threathening old aged groups. This research was planned to isolate compounds with the therapeutic potential for the above mentioned chronic diseases from the mulberry leaves. Biological screenings were carried out for the following categories; anti-atherogenic, anti-diabetic and antihypertensive effects. The results were as follows; Mulberry leaves, 20% $\alpha$-treated Gaelyrangppong showed significant 81% of blood glucose lowering effects in alloxan-induced hyperglycemic mice. Particularly, butanol-soluble fractions of mulberry leaves showed the more significant hypoglycemic activity than other fractions in alloxan induced hyperhlycemic mice. Also in the group given 1g/kg doses of extract of mulberry leaves, total cholesterol level was decreased significantly by as much as 49% in hyperlipidemia-induced rats. In Mulberry leaves post-treated group, the atheroscelosis index, HDL-cholesterol/Total-cholesterol, was increased significantly by as much as 91%.

Four months of magnetized water supplementation improves glycemic control, antioxidant status, and cellualr DNA damage in db/db mice (제2형 당뇨 모델 db/db 마우스에서 4개월의 자화수 섭취 후 혈당, 항산화 상태 및 세포 DNA 손상 개선 효과)

  • Purpose: Water is magnetically charged upon contact with a magnet. Although magnetic water products have been promoted since the 1930's, they have not received wide acceptance since their effectiveness is still in question; however, some have reported their therapeutic effects on the body, especially the digestive, nervous, and urinary systems. Methods: In this study, the effect of magnetized water on glycemic control of 14 diabetic mice (CB57BK/KsJ-db/db) in comparison with 10 control mice (CB57BK/KsJ-db/+(db/+)) was investigated. Seven diabetic control (DMC) mice and seven diabetic mice + magnetized water (DM+MW) were kept for 16 weeks, followed by intraperitoneal glucose tolerance test (IPGTT). Weekly blood glucose was measured from tail veins. Blood obtained from heart puncture was used for HbA1c analysis. Results: Blood glucose level showed a significant difference starting from the $10^{th}$ week of study ($496.1{\pm}10.2mg/dl$ in DMC vs. $437.9{\pm}76.9mg/dl$ in DM+MW). Blood glucose followed by IPGTT showed no significant difference between groups at 0, 30, 60, 90, and 120 min, although glucose level at 180 min was significantly reduced in DM+MW mice. Plasma insulin level in DM+MW groups was only 39.5% of that of DMC groups ($5.97{\pm}1.69ng/ml$ in DMC vs. $2.36{\pm}0.94ng/ml$ in DM+MW). Levels of HbA1c were 12.4% and 9.7% in DMC and DM+MW groups, respectively. Conclusion: These results show the promising therapeutic effect of magnetized water in regulating blood glucose homeostasis; however, long-term supplementation or mechanistic study is necessary.

Analysis of the Physiological Healing Effects by Forest Types - Focused on Hypertensive and Diabetic - (숲의 종류에 따른 생리적 치유효과 분석 - 고혈압과 당뇨병 환자를 중심으로 -)

  • This study analyzes the physiological healing effects on users according to forest types. Forest types are classified into Chamaecyparis obtusa, Quercus serrata and Pinus densiflora and the subjects of this study, 64 hypertensives, 59 diabetics and 59 ordinary people, were tested to observe their physiological effects. The index of physiological reaction comprises electroencephalogram(EEG), heart rate variability(HRV), blood pressure and cortisol level. An analysis of the stress-buffering effect from the physiological reactions of the overall subjects indicates that Quercus serrata forests have a higher stress-buffering effect than others in terms of central nervous, endocrine and autonomic nervous systems. Chamaecyparis obtusa forests are contributory to soothing stress in the central nervous and endocrine systems. Pinus densiflora forests contribute in some indexes of the endocrine and autonomic nervous systems. In contrast, Pinus densiflora forests are less influential on a stress-buffering effect than the other two. In the level of the effect for lowering blood pressure, Chamaecyparis obtusa and Quercus serrata forests are more effective than Pinus densiflora forests. Therefore, staying in the first two forests can heighten the healing effect of the lowering of blood pressure for hypertensive patients. Every forest is efficacious to lowering diabetic blood glucose levels. Spending time in Quercus serrata forests is more effective for hypoglycemic.

Effects of ${\beta}-glucan$ from Lentinus edodes and Hordeum vulgare on Blood Glucose and Lipid Composition in Alloxan-induced Diabetic Mice (표고버섯과 보리에서 추출한 ${\beta}-glucan$이 Alloxan 유발 당뇨 마우스의 혈당 및 지질 성분에 미치는 영향)

  • Effects of ${\beta}-glucan$ from Lentinus edodes and hordeum vulgare on blood glucose and lipid composition were investigated. Diabetes mellitus was induced in male ICR mice by the injection of alloxan into the tail vein at a dose of 75 mg/kg. The ${\beta}-glucan$ were administered orally for 10 days and the normal and alloxan-control group were orally administered with saline. The body weight gain and food intake were monitored every day and plasma levels of glucose, triglyceride, total cholesterol, LDL-cholesterol were determined at last day. Also the weight of liver, heart, spleen and kidney were determined. The ${\beta}-glucan$ from Lentinus edodes and hordeum vulgure lowered significantly body weight gain in alloxan-induced diatetic mice (p<0.05) and plasma glucose levels compared to that of alloxan-control group. Plasma triglyceride level in B500 was lowered in alloxan-induced diabetic mice. The ${\beta}-glucan$ of hordeum vulgare lowered weight of liver significantly (p<0.05). In conclusion, it was assumed that ${\beta}-glucan$ from hordeum vulgare have anti-hyperglycemic and anti-obese effects by reducing body weight gain and decreasing serum glucose and triglyceride level.

Cardiovascular Autonomic Neuropathy Predicts Higher HbA1c Variability in Subjects with Type 2 Diabetes Mellitus

  • Background: This study aimed to investigate the association between the presence and severity of cardiovascular autonomic neuropathy (CAN) and development of long-term glucose fluctuation in subjects with type 2 diabetes mellitus. Methods: In this retrospective cohort study, subjects with type 2 diabetes mellitus who received cardiovascular autonomic reflex tests (CARTs) at baseline and at least 4-year of follow-up with ${\geq}6$ measures of glycosylated hemoglobin (HbA1c) were included. The severity of CAN was categorized as normal, early, or severe CAN according to the CARTs score. HbA1c variability was measured as the standard deviation (SD), coefficient of variation, and adjusted SD of serial HbA1c measurements. Results: A total of 681 subjects were analyzed (294 normal, 318 early, and 69 severe CAN). The HbA1c variability index values showed a positive relationship with the severity of CAN. Multivariable logistic regression analysis showed that CAN was significantly associated with the risk of developing higher HbA1c variability (SD) after adjusting for age, sex, body mass index, diabetes duration, mean HbA1c, heart rate, glomerular filtration rate, diabetic retinopathy, coronary artery disease, insulin use, and anti-hypertensive medication (early CAN: odds ratio [OR], 1.65; 95% confidence interval [CI], 1.12 to 2.43) (severe CAN: OR, 2.86; 95% CI, 1.47 to 5.56). This association was more prominent in subjects who had a longer duration of diabetes (>10 years) and lower mean HbA1c (<7%). Conclusion: CAN is an independent risk factor for future higher HbA1c variability in subjects with type 2 diabetes mellitus. Tailored therapy for stabilizing glucose fluctuation should be emphasized in subjects with CAN.

Preparation and Characterization of Pioglitazone Loaded PLGA Nanospheres for the Treatment of Type 2 Diabetes (제2형 당뇨병 치료제인 Pioglitazone을 봉입하기 위한 PLGA 나노입자 제조 및 분석)

  • The pioglitazone loaded poly(lactide-co-glycolide)(PLGA) nanospheres were prepared by emulsion-evaporation method and optimized for particle size and entrapment efficiency. The optimized particles were 125~170 nm in size with narrow size distribution and showed above 85% entrapment efficiency at 30% of pioglitazone loading when prepared with 3% w/v of poly(vinyl alcohol) (PVA) as a surfactant. These particulate carriers exhibited a controlled in vitro release of pioglitazone for 40 days at a nearly constant rate. The pioglitazone loaded PLGA nanospheres were not only effective to reduce the blood sugar level of diabetic rats but also non-toxic for the animal body, in particular for sensitive organs like kidney, liver, heart, lung and spleen. These results indicate that PLGA nanospheres have a great potential for oral delivery of pioglitazone.

Exercise training and selenium or a combined treatment ameliorates aberrant expression of glucose and lactate metabolic proteins in skeletal muscle in a rodent model of diabetes

  • Exercise training (ET) and selenium (SEL) were evaluated either individually or in combination (COMBI) for their effects on expression of glucose (AMPK, PGC- $1{\alpha}$, GLUT-4) and lactate metabolic proteins (LDH, MCT-1, MCT-4, COX-IV) in heart and skeletal muscles in a rodent model (Goto-Kakisaki, GK) of diabetes. Forty GK rats either remained sedentary (SED), performed ET, received SEL, ($5\;{\mu}mol{\cdot}kg$ body $wt^{-1}{\cdot}day^{-1}$) or underwent both ET and SEL treatment for 6 wk. ET alone, SEL alone, or COMBI resulted in a significant lowering of lactate, glucose, and insulin levels as well as a reduction in HOMA-IR and AUC for glucose relative to SED. Additionally, ET alone, SEL alone, or COMBI increased glycogen content and citrate synthase (CS) activities in liver and muscles. However, their effects on glycogen content and CS activity were tissue-specific. In particular, ET alone, SEL alone, or COMBI induced upregulation of glucose (AMPK, PGC-la, GLUT-4) and lactate (LDH, MCT-1, MCT-4, COX-IV) metabolic proteins relative to SED. However, their effects on glucose and lactate metabolic proteins also appeared to be tissue-specific. It seemed that glucose and lactate metabolic protein expression was not further enhanced with COMBI compared to that of ET alone or SEL alone. These data suggest that ET alone or SEL alone or COMBI represent a practical strategy for ameliorating aberrant expression of glucose and lactate metabolic proteins in diabetic GK rats.

  • Mucormycosis is the common name given to several different diseases caused by fungi of the order Mucorales. The mucoraceae are ubiquitous fungi and are common inhabitants of decaying matter. In contrast to the widespread distribution of these fungi, disease in humans is limited, in most cases, to people with severe immunocompromised, diabetes mellitus, or trauma. 1be fungus gains entry to the body through the respiratory tract. The spores are presumably deposited in the nasal turbinates and may be inhaled into the pulmonary alveoli. The manifestations of mucormycosis are rhinocerebral, pulmonary, cutaneous, gastrointestinal, central nervous system, and miscellaneous. Sporadic reports can be found of mucormycosis involving other areas : heart, bones, kidney, bladder, mediastinum, and trachea. However, isolated tracheal mucormycosis is very rare. Therefore, we report a 57-year old, noninsulin dependent diabetic woman who presented with acute, severe degree of upper airway obstruction due to isolated mucormycosis of the trachea.

Atherosclerotic Cardiovascular Disease Risk and Statin Prescription Status in Korean Adult Patients (국내 성인환자에서 죽상동맥경화성 심혈관질환 위험도와 스타틴 처방 현황)

  • Background: Cardiovascular (CV) disease is known as one of the major causes of death from disease worldwide. Statin therapy plays a pivotal role in atherosclerotic cardiovascular disease (ASCVD) lowering the LDL-cholesterol level effectively. The purpose of this study was to evaluate the association of the intensity of statin therapy in adult patients of Korea and the risk of ASCVD of the patient group. Methods: We used data from sample of patients from the Health Insurance Review and Assessment Service (HIRA-NPS-2018). We analyzed the patterns of prescribing statins including types of statin, statin intensity, and number of patients with ASCVD or risk of ASCVD. Results: 155,512 patients were included in the analysis, and 27,950 patients (18.0%) was over 75 years. High-intensity statin usage was increased in ASCVD patients compared with the low-intensity statin use. The OR (odds ratio) of high-intensity statin were increased in myocardial infarction patients compared with low-intensity statin use showing the highest OR; 12.40 (95% CI; 9.48-16.22). At patient groups of angina, ischemic heart disease and carotid disease, high-intensity statin prescription rate was increased compared with low-intensity statin. However, there was no statistical significance between both statin prescription rates in patients of peripheral arterial disease, abdominal aneurysm, diabetic mellitus and atherosclerosis. Conclusion: The statin prescription rate showed intensity increasing tendency according to the risk of ASCVD. More aggressive statin therapy might be beneficial for the ASCVD patients based on the recent guidelines of dyslipidemia.

Comparison of the efficacy of the herbs for upper medication on glucose tolerance induced by high fat/high sucrose feeding-induced mice (본초 귀경에 따른 상소한약의 당내성 유발 마우스에서의 개선 효능 비교 연구)

  • Objectives : To prove the channel-tropism theory of herbal medicines on diabetes mellitus as emaciation-thirst disease in Korean Medicine Theory, we investigated the selective therapeutic effects of Mori Cortex Radidus (MCR), Schisandrae Fructus (SF), Anemarrhenae Rhizoma (AR) for the upper emaciation on different organs in high fat and high sucrose (HF/HS) feeding-induced prediabetic mice. Methods : Diabetes in C57BL/6 mice was induced by the administration of high fat (45 kal%) and high sucrose (32 kal%) for 8 weeks, and them treated with each extract at 250 or 500 mg/kg body weight for 4 weeks (once a day). Oral glucose tolerance test and body weight was measured once a week. Insulin, total cholesterol, triglyceride, ${\gamma}GTP$, GOT and GPT were measured in the sera of all mice. Histopathological changes of different organs, lung, heart, pancreas, stomach, liver, and kidney were observed by H&E staining. Results : The results revealed that MCR extract inhibited the impaired glucose tolerance and lung damage, and increased serum insulin levels in HF/HS-induced prediabetic mice. SF extract inhibited the impaired glucose tolerance and lung damage, increased serum insulin levels, and decreased serum triglycerige levels. Meanwhile, AR extract inhibited the impaired glucose tolerance and lung damage, and decreased serum levels of insulin, total cholesterol and triglycerige levels. Conclusions : These results demonstrated that MCR, SF, and AR extract as the upper emaciation herbal medicines were followed their channel-tropism theory like a lung, and may have a selective therapeutic potential for control of diabetic stage.
