Dexamethasone Induces $Fc{\gamma}RIIb$ Expression in RBL-2H3 Cells

  • Mast cells are involved in allergic responses, protection against pathogens and autoimmune diseases. Dexamethasone (Dex) and other glucocorticoids suppress $Fc{\varepsilon}RI$-mediated release of inflammatory mediators from mast cells. The inhibition mechanisms were mainly investigated on the downstream signaling of Fc receptor activations. Here, we addressed the effects of Dex on Fc receptor expressions in rat mast cell line RBL-2H3. We measured mRNA levels of Fc receptors by real-time PCR. As expected, Dex decreased the mRNA levels of activating Fc receptor for IgE ($Fc{\varepsilon}R$) I and increased the mRNA levels of the inhibitory Fc receptor for IgG $Fc{\gamma}RIIb$. Interestingly, Dex stimulated transcriptions of other activating receptors such as Fc receptors for IgG ($Fc{\gamma}R$) I and $Fc{\gamma}RIII$. To investigate the mechanisms underlying transcriptional regulation, we employed a transcription inhibitor actinomycin D and a translation inhibitor cycloheximide. The inhibition of protein synthesis without Dex treatment enhanced $Fc{\gamma}RI$ and $Fc{\gamma}RIII$ mRNA levels potently, while $Fc{\varepsilon}RI$ and $Fc{\gamma}RIIb$ were minimally affected. Next, we examined expressions of the Fc receptors on cell surfaces by the flow cytometric method. Only $Fc{\gamma}RIIb$ protein expression was significantly enhanced by Dex treatment, while $Fc{\gamma}RI$, $Fc{\gamma}RIII$ and $Fc{\varepsilon}RI$ expression levels were marginally changed. Our data showed, for the first time, that Dex regulates Fc receptor expressions resulting in augmentation of the inhibitory receptor $Fc{\gamma}RIIb$.

Effects of Korean Red Ginseng Extract for the Treatment of Atopic Dermatitis-Like Skin Lesions in Mice

  • Atopic dermatitis (AD) is an allergic, inflammatory skin disease characterized by chronic eczema and mechanical injury to the skin, caused by scratching. Korean red ginseng (RG) has diverse biological activities, but the molecular effects of RG on allergic diseases, like AD, are unclear. The present study was designed to investigate whether RG inhibits 1-chloro-2,4-dinitrobenzene (DNCB)-induced AD in a mouse model. DNCB was applied topically on the dorsal surface of Balb/c mice to induce AD-like skin lesions. We observed the scratching behavior and examined the serum IgE level and interleukin (IL)-4 and IL-10 in splenocytes compared with dexamethasone. We also evaluated the DNCB-induced mitogen-activated protein kinases (MAPKs), NF-${\kappa}B$, and Ikaros activities after RG treatment using reverse transcriptase-polymerase chain reaction, Western blotting, and ELISA. Our data showed that the topical application of RG significantly improved the AD-like skin lesions and scratching behavior. RG decreased not only the mRNA expression of IL-4 and IL-10, but also the secretion of IL-4 protein and serum IgE in mice. Additionally, RG treatment decreased the DNCB-induced MAPKs activity and subsequent Ikaros translocation irrespective of NF-${\kappa}B$. We suggest that RG may be useful as a therapeutic nutrition for the treatment of AD.

황련감초 추출물의 상피지방장벽 생성을 통한 Th2 분화 조절 (Effect of Th2 Differentiation Control through Formation of Skin Fat Barrier on Coptidis Rhizoma & Glycyrrhiza Uralensis extract)

  • Objectives This study is conducted to evaluate Th2 skewed condition control through skin fat barrier formation from the treatment using Coptidis Rhizoma and Glycyrrhiza uralensis extract. Methods The 6-week-old NC/Nga mice were divided into 4 groups: Control group (Ctrl), lipid barrier eliminate treatment group (LBET), Coptidis Rhizoma and Glycyrrhiza uralensis feeding treatment after lipid barrier elimination group (CGFT), dexamethasone feeding treatment after lipid barrier elimination group (DxFT). After 3 days, differences in skin condition, improvement of skin fat barrier, and control of Th2 skewed condition of each group were observed. Results Pathologic skin damage and tissue changes were less in the CGFT group than in the LBET and DxFT groups, and Transepidermal water loss (TEWL) and pH were also significantly decreased (p < 0.05). The filaggrin intensity and positive response also increased significantly in the CGFT group (p < 0.05). Kallikrein-related peptidase (KLK) 7, Protease activated receptor (PAR)-2, Thymic stromal lymphopoietin (TSLP), Interleukin (IL)-4, and the products of the Th2 differentiation process also showed a significant decrease compared to the LBET and DxFT groups (all p < 0.05). Conclusions The Coptidis Rhizoma and Glycyrrhiza uralensis extract causes skin barrier recovery and function recovery through the formation of skin fat barrier. This leads to the conclusion that Coptidis Rhizoma and Glycyrrhiza uralensis extract can control Th2 differentiation through the formation of skin fat barrier.

치주인대세포의 골기질 단백질 유전자 발현에 대한 Dexamethasone의 영향 (The effect of dexamethasone on the gene expression of the bone matrix protein in the periodontal ligament cells)

  • 치주조직 재생을 위해서는 새로운 백악질과 치조골 그리고, 치주인대의 재생이 필요하며, 이러한 재생을 담당할 세포의 분화가 필수적이다. 이러한 분화를 담당하는 것은 치주인대세포이며, 이 중 골아세포의 분화가 중요하다. 본 실험의 목적은 치주조직 재생에 있어서 중요한 요소인 치주인대세포의 골아세포성 세포로의 분화를 관찰하며, Dex가 광물화에 미치는 영향과 농도에 따른 차이를 알아보고자 시행하였다. 또한, 광물화시 발현되는 여러 골기질 단백질 중 Matrix GlaProtein의 발현양상도 관찰하였다. 교정치료를 목적으로 내원한 환자의 제1소구치 부위의 정상치은을 절제하고, 건강한 제1소구치를 발거하여 치은섬유아세포와 치주인대세포를 분리, 배양하여, ascorbic acid와 ${\beta}$-glycerophosphate 투여군을 실험1군, ascorbic acid, ${\beta}$-glycerophosphate, Dex 100nM 투여군을 실험 2군, ascorbic acid, ${\beta}$-glycerophosphate, Dex $5{\mu}M$ 투여군을 실험3군, 그리고, 단순 배양만 시킨군을 대조군으로 하여 비교하였다. 시간경과에 따른 치주인대세포 형태의 변화 양상은 초기에 방추형 혹은 다각형의 단일층 형태에서 7일경에는 세포 크기와 수가 증가하여 복합층 형태로 변화했으며, 배양 14일 이후에는 세포들의 방향성이 없어지고, 더욱 치밀해 졌다. 골 결절형성은 치주인 대세포의 Dex 투여군에서만 21일째에 나타났으며, $5{\mu}M$ 투여군에서 100nM 투여군보다 더 많이 나타났다. ALP 활성도를 비교해보면 치주인대세포에서 0, 7일 경에는 활성도를 보이지 않았으며, 14일경에 높은 활성도롤 나타냈으며, 21일에도 비슷한 활성도를 유지하였다. MGP 유전자 발현 양상은 대조군과 실험군 모두에서 Matrix Gla Protein에 대한 유전자의 발현이 나타났으며,그 발현양상은 모든 시기에서 일정하였다. 이상의 결과로 보아 치주인대세포는 골아세포로의 분화가 가능하며, Dex는 농도의존적으로 광물화에 영향을 미치는 것으로 사료된다. 그리고, MGP는 치주인대세포에서 발현이 감지되었으며, 광물화에는 영향을 미치지 않는 것으로 사료된다.

Estrogen reinforces barrier formation and protects against tumor necrosis factor alpha-induced barrier dysfunction in oral epithelial cells

  • Purpose: Epithelial barrier dysfunction is involved in the pathophysiology of periodontitis and oral lichen planus. Estrogens have been shown to enhance the physical barrier function of intestinal and esophageal epithelia, and we aimed to investigate the effect of estradiol (E2) on the regulation of physical barrier and tight junction (TJ) proteins in human oral epithelial cell monolayers. Methods: HOK-16B cell monolayers cultured on transwells were treated with E2, an estrogen receptor (ER) antagonist (ICI 182,780), tumor necrosis factor alpha ($TNF{\alpha}$), or dexamethasone (Dexa), and the transepithelial electrical resistance (TER) was then measured. Cell proliferation was measured by the cell counting kit (CCK)-8 assay. The levels of TJ proteins and nuclear translocation of nuclear factor $(NF)-{\kappa}B$ were examined by confocal microscopy. Results: E2 treatment increased the TER and the levels of junctional adhesion molecule (JAM)-A and zonula occludens (ZO)-1 in a dose-dependent manner, without affecting cell proliferation during barrier formation. Treatment of the tight-junctioned cell monolayers with $TNF{\alpha}$ induced decreases in the TER and the levels of ZO-1 and nuclear translocation of $NF-{\kappa}B$. These $TNF{\alpha}-induced$ changes were inhibited by E2, and this effect was completely reversed by co-treatment with ICI 182,780. Furthermore, E2 and Dexa presented an additive effect on the epithelial barrier function. Conclusions: E2 reinforces the physical barrier of oral epithelial cells through the nuclear ER-dependent upregulation of TJ proteins. The protective effect of E2 on the $TNF{\alpha}-induced$ impairment of the epithelial barrier and its additive effect with Dexa suggest its potential use to treat oral inflammatory diseases involving epithelial barrier dysfunction.

Inhibition of Experimental Systemic Inflammation (Septic Inflammation) and Chronic Bronchitis by New Phytoformula BL Containing Broussonetia papyrifera and Lonicera japonica

  • Broussonetia papyrifera and Lonicera japonica have long been used in the treatment of inflammatory disorders in Chinese medicine, especially respiratory inflammation. Previously, a new phytoformula (BL) containing B. papyrifera and L. japonica was found to exert strong anti-inflammatory activity against several animal models of inflammation, especially against an animal model of acute bronchitis. In the present investigation, the effects of BL on animal models of septic inflammation and chronic bronchitis are examined. Against lipopolysaccharide (LPS)-induced septic inflammation in mice, BL (200-400 mg/kg) reduced the induction of some important proinflammatory cytokines. At 1 h after LPS treatment, BL was found to considerably inhibit TNF-${\alpha}$ production when measured by cytokine array. At 3 h after LPS treatment, BL inhibited the induction of several proinflammatory cytokines, including IFN-${\gamma}$ and IL-$1{\beta}$, although dexamethasone, which was used as a reference, showed a higher inhibitory action on these biomarkers. Against chronic bronchitis induced by LPS/elastase instillation in rats for 4 weeks, BL (200-400 mg/kg/day) significantly inhibited cell recruitment in bronchoalveolar lavage fluid. Furthermore, BL considerably reduced lung injury, as revealed by histological observation. Taken together, these results indicate that BL may have a potential to treat systemic septic inflammation as well as chronic bronchitis.

Clinical Features and Hearing Outcomes of Sudden Sensorineural Hearing Loss in Diabetic Patients

  • Background and Objectives: This study aimed to evaluate the clinical features and the clinical factors associated with prognosis of sudden sensorineural hearing loss (SSNHL) in diabetic patients. Subjects and Methods: Forty-nine diabetic with unilateral SSNHL were retrospectively included. All patients received systemic high dose steroid therapy within one month after onset and had more than one month of follow-up audiogram. The basic characteristics of the patients, initial and follow-up audiograms, laboratory data, and methods of steroid treatment were collected. Results: Compared to reference values in healthy subjects, 79%, 55%, and 45% of the patients had higher values of mean neutrophil-lymphocyte ratio (NLR), platelet-lymphocyte ratio (PLR), and lymphocyte-monocyte ratio (LMR), respectively. Older patients had significantly less degree of hearing loss, but they also had significantly worse hearing thresholds in the unaffected ear. After steroid treatment, less than half patients (47%) showed hearing recovery. Simultaneous intratympanic dexamethasone (ITD) injections with systemic steroid did not confer an additional hearing gain or an earlier recovery rate in diabetic patients with SSNHL. In the multivariate analysis, initial hearing thresholds of affected ear and timing of steroid treatment were significantly associated with hearing prognosis in diabetic patients with SSNHL. Conclusions: Diabetic patients with SSNHL tended to have increased NLR, LMR, and PLR, which are reported to be associated with microvascular angiopathy. Simultaneous ITD injections to improve hearing recovery in diabetic patients with SSNHL seems unnecessary.

적양파 추출물의 항비만 활성 (Anti-Obesity Effects of Red Onions Extract)

  • 비만은 에너지의 섭취와 소비의 불균형으로 지방조직이 비정상적으로 분화하면서 생기는 대사질환으로 알려져있다. 본 연구에서는 본 연구는 적양파 추출물 처리에 따른 Pancreatic lipase 억제, 지방세포분화 억제 활성을 확인하고자 하였다. 적양파추출물 처리에 따른 활성은 지방세포 분화 및 관련 유전자에 대한 평가는 3T3-L1 지방전구세포를 이용하고 Real-Time PCR을 통하여 확인하였다. 실험 결과, Pancreatic lipase 활성 억제 실험에서 적양파 추출물은 농도 의존적으로 lipase 활성을 억제하였다. 지방세포분화 실험을 수행한 결과, insulin, dexamethasone, 3-isobutyl-1-methylxanthine(MDI)등으로 분화 유도된 3T3-L1 세포에서 적양파 추출물은 지방전구세포의 분화를 억제하였으며 동시에 지방구 형성을 억제하는 것으로 나타났다. 또한, 지방전구세포의 분화 과정과 관련된 C/EBP-α, C/EBP-β, PPAR-γ의 발현을 억제하였다. 본 실험에서 적양파 추출물은 지방분해효소를 억제하며, 지방전구세포 분화와 관련된 유전자 발현을 억제함으로써 지방세포 분화 및 지방구형성을 억제할 수 있는 항비만 소재로의 개발 가능성이 높다고 판단된다.

Twelve-oxoeicosatetraenoic acid-induced fetal membrane release improves postpartum ovarian function, milk production, and blood plasma biochemical parameters in cows

  • Objective: We aimed to determine the effects of 12-oxoeicosatetraenoic acid (12-KETE)-induced placenta release on the performance of mother cows (milk yield, ovarian function, and blood plasma biochemical properties). Methods: Experimental treatments were as follows: i) natural delivery including natural placental release (control cows); ii) induced calf delivery with placental retention (RP cows); and iii) induced calf delivery and 12-KETE-induced placental release (KE cows). Delivery in pregnant KE cows was induced with dexamethasone and prostaglandin. These cows were injected with 12-KETE after calf discharge, resulting in the release of the fetal placenta. RP cows were not treated with 12-KETE after inducing delivery, resulting in placental retention. Results: The milk yield in RP cows during the first 50 days after delivery was significantly lower than that in control cows (p<0.05), whereas KE cows exhibited a similar milk yield to that of control cows. The postpartum plasma progesterone levels of control cows increased 14 days after delivery on average; however, its increase was delayed by 10 days in RP cows. Meanwhile, the 12-KETE treatment (KE cows) brought the timing of progesterone increase forward to the normal level (control cows). Among the 20 biochemical parameters examined, the total cholesterol levels in blood plasma 14 days after delivery were lower in RP cows than that in the other two treatment groups (control cows and KE cows) (p<0.05). In addition, the plasma level of haptoglobin tended to be low in cows that discharged their placentas shortly after delivery. Conclusion: These findings indicate that 12-KETE treatment can alleviate the disorder caused by placental retention.
