This study was conducted to examine the effect of several saccharides on the induction of capacitation and acrosome reaction (AR) and to examine the effects of mono and polysaccharides on the penetration activity of boar spermatozoa. Spermatozoa were inseminated in medium with fucose, galactose and mannose as monosaccharide, and fucoicIan. galactan and marman as polysaccharide. The penetration rates were significantly (p<0.05) lower in medium with galactose (40.6%), mannose (38.1%), fucose (41.6%) and fucoidan (36.6%) compared with control (56.7%). The rates of AR were increased (40.7 to 59.8%) by the preincubation periods prolonged from 0 to 4 hr (p<0.05). Similar tendencies were observed in AR when spermatozoa were treated with monosaccharides, but not significantly differ among the groups treated with different time of preincubation with some exception of galactose. When spermatozoa were treated with polysaccharides, the rates of AR were significantly (p<0.05) increased by preincubation time prolonged from 0 to 4 hr with an exception of fucoidan. In conclusion, the present study suggests that penetration rate of spermatozoa is higher in presence of polysaccharides than monosaccharides. Also, it may resume that the comparing to control, the all saccharides (L-fucose, D-galactose, D-mannose, fucoidan. galactan and mannan)-treated groups slightly increase the AR pattern as preincubation time prolonged.

The objective of this study was two folds: to investigate the relationship between paternal identification rate and sperm quality parameters such as motility and sperm chromatin structure assay after heterospermic insemination; to see if mutual complement between tests and development of useful technique to enhance the fertility in artificial insemination. In individual boar's fertilizing ability, 3 high fertility boars showed significantly high fertility (p<0.05) compared to 3 low fertility boars, but there was no difference in litter size between two groups. Sperm motility test in pooled and individual semen using computer assisted sperm analysis (CASA) revealed that no significant difference among boars. The high fertile boar showed tendency of low %Red (High red fluorescence/green+red fluorescence) in sperm chromatin structure assay (SCSA) but paternal identification rate from piglets did not differ after heterospermic insemination. The correlation coefficient between individual or pooled semen function test and farrowing rates were well correlated as follows: %Red with litter size (r= - 0.53, p=0.03); %Red with paternal identification rates (r=-0.51, p=0.03); paternal identification rates with litter size (r=0.57, p=0.02). These results indicate that sperm chromatin structure assay and sperm quality parameter test in pooled semen are useful method to predict and evaluate the fertilizing capacity after heterospermic insemination in boars.
The objective of this study was to assess the developmental potential in vitro produced embryos frozen-thawed with the various containers, and also examined expression and localization of heat shock protein 70 at these embryos. For the vitrification, 2-cell, 8-cell and blastocyst stage embryos produced by in vitro fertilization (IVF) and nuclear transfer (NT) were exposed the ethylene glycol 5.5 M freezing solution (EC 5.5) for 30 sec, loaded on each containers such EM grid, straw and cryo-loop, and then immediately plunged into liquid nitrogen. Thawed embryos were serially diluted in sucrose solution, each for 1 min. and cultured in CRI-aa medium. Survival rates of the vitrification production were assessed by re-expanded, hatched blastocysts. There were no differences in the survival rates of IVF using EM grid and cryo-loop. However, survival rates by straw were relatively lower than other containers. The use of cryo-loop resulted in only survival of nuclear transferred embryos (43.7%). Also, there embryos after IVF or NT were analysed by semi-quantitive reverse transcription-polymerase chain reaction (RT- PCR) methods for hsp 70 mRNA expression. Results revealed the expression of hsp 70 mRNh were higher thawed embryos than control embryos. Immunocytochemistry used to localize the hsp 70 protein in embryos. Two and 8-cell embryos derived under control condition was evenly distributed in the cytoplasm but appeared as aggregates in some frozen-thawed embryos. However, in the control, blastocysts displayed aggregate signal while Hsp70 in frozen-thawed blastocysts appeared to be more uniform In distribution. Therefore, this result suggests that the exploiting Hsp 70 in the early embryos may be role for protection of stress condition for increase viability of embryos within IVF, NT and there frozen-thawed embryos.
The sterilization effects of methylene blue (MB), formalin, and iodine on the egg of marine medaka, Oryzias dancena, were investigated for disinfecting naididae worm, Chaetogaster diastrophus through sterilization. To determine harmfulness of MB, formalin, and iodine, lethal concentrations 50 ($LC_{50}$) of three chemicals were analyzed in the eggs of marine medaka. The sterilized periods of each chemical were set at 1 hr. Sterilized rates of naididae worm in each chemical were significantly affected and increased drastically as the concentration of each chemical increased (p<0.05). Sterilization abilities of naididae worm were most effective for formalin, but survival rates of egg and hatched rates for formalin were lowest among each chemical. The $LC_{50}$ of MB over 96 hrs were 185.26, 103.84, and 127.15 ppm for adults, juveniles, and eggs respectively. The toxic effects of MB were clearly dose dependent for each life stage (p<0.05). The toxicity sensitivity of juveniles to MB was dramatically higher than that of other groups. In 48 hrs after sterilization, cortisol and glucose concentrations of the adult group with MB treatment were significantly higher than those of the adult group with no treatment (p<0.05). This research provides useful data on sterilization effect of MB, formalin, and iodine, acute toxicity in marine medaka egg and toxicity, sensitivity of life stage of MB in marine medaka.
In this study, in vitro maturation system using fetal bovine serum (FBS) or porcine follicular fluid (pFF) was investigated to produce comparable oocytes to those derived from in vivo. Control group of oocytes was cultured in TCM 199 supplemented with 0.1% polyvinyl alcohol (PVA). Other three groups of oocytes were cultured in TCM 199 supplemented with 10% FBS, 10% pFF or 5% FBS + 5% pFF, respectively. After 44 h maturation, oocytes with the first polar body were activated with two electric pulses (DC) of 1.2 kv/cm for 30 ${\mu}sec$. Also, matured oocytes of four groups were reconstructed and fused. Reconstructed embryos were cultured in PZM-3 under 5% $CO_2$ in air at $38.5^{\circ}C$ for 6 days. The oocytes matured in the medium supplemented with FBS or/and pFF showed significantly higher maturation rates (64.0 vs. 73.9 to 85.2%). In PA embryos, cleavage rates (89.7 vs. 77.1 to 86.6%) and blastocysts rates (30.0 vs. 16.2 to 26.2%) were significantly higher in pFF group (p<0.05). In NT embryos, there was no difference among treatments in cleavage rate, but the blastocyst rates (28.5 vs. 15.5 to 24.6%) were significantly higher in pFF group (p<0.05). The apoptosis rate was significantly higher (p<0.05) in the control than other groups (10.8 vs. 4.9 to 8.2% for PA, 3.1 vs. 0.5 to 1.3% for NT). In order to select the comparable oocyte to in vivo oocytes, each group of oocytes was stained with Brilliant cresyl blue (BCB) after 42h maturation. The matured oocytes were separated according to color of cytoplasm; stained group (BCB+) and unstained group (BCB-). The oocytes matured in the presence of FBS or/and pFF showed significantly higher staining rates (70.3 to 72.7 vs. 35.1%) (p<0.05). To verify the fact that the supplementation of FBS or/and pFF can increase the maturation rates, cdc2 kinase activity, the catalytic subunit of MPF, was determined. The cdc2 kinase activity of the oocytes matured in the medium supplemented with FBS or/and pFF was significantly higher than control group (6.7 to 9.3 vs. 3.8). In conclusion, the supplementation of FBS or/and pFF can support in vitro maturation rate of porcine oocytes through the increment of cdc2 kinase activity level in the cytoplasm.
본 연구는 소 수정란의 동결에 있어서 완만 및 급속동결과 vitrification동결 후의 생존성을 비교하고 이울러, 내동제의 종류 및 배 발달단계가 vitrification동결 보존 후의 발생율을 조사하고자 수행 하였다. 1. 소 수정란의 vitrification 동결보존 후의 발생율과 부화율은 67.5%와 27.5%로서 급속동결 및 완만동결 운해 후의 발생율과 부화율 42.5% 와 20.0% 및 52.5%와 25.0%에 비해 높게 나타났으나 대조군(82.5%, 37.5%)에 비해서 낮게 나타났다. 2. EFS와 EDS내동제의 처리에 따른 소 수정란의 vitrification 동결보존 후의 발생율과 부화율은 47.5%와 22.5% 및 52.5%와 27.5%로서 대조군의 82.5%와 37.5%에 비해 낮게 나타났다. 3. 소 수정란의 2 cell, 8 cell, morulae, blastocyst stage별 vitrification 동결보존 후의 발생율과 부화율은 25.0%와 15.0%, 32.5%와 20.0%, 37.5%와 20.0%, 52.5%와 27.5% 및 47.5%와 25.0%로서 대조군(82.5%와 37.5%)에 비해 낮게 나타났다.
본 연구는 돼지 태아 섬유아세포유래 공여세포를 미세주입에 의해 주입 후 재 조합한 핵 이식 배에 대한 배양액, 세포주기의 동기화, 배양시간 및 난자의 활성화에 따른 융합율과 체외발생율에 대해 조사하였다. 핵 이식 배를 NCSU-23, TL Hepes 및 TZM-3 배양액으로 1시간 및 8시간 배양하였을 때 배반포로의 분할율은 각각 15.6%, 14.0%, 15.0% 및 13.9%, 10.5%, 13.3%로서 배양액 및 시간에 따른 분할율의 유의적인 차이는 없었다. 공여핵원용 세포를 0, 8, 15시간 배양했을 때 G2/M기로의 체외발달율은 12.0%, 18.0%, 48.0%였다(p<0.01). 공여핵원용 세포를 12-24시간 배양했을 때 G2/M기로의 체외발달율은 유의한 증가를 나타내지 않았다. 공여핵원용 세포를 10% FBS + NCSU-23 배양액으로 1-2, 6-8, 12-14일간 배양 후 핵 이식한 배의 융합율은 각각 60.0%, 73.3%, 62.5%였으며, 분할율은 각각 36.0%, 56.7%, 50.0%였다. 0.5% FBS + NCSU-23, 0.5% + TL-Heaps 및 0.5% + TZM-3 배양액으로 5% $O_2$조건 하에서 배양하였을 때 핵 이식배의 $\geq$2 cell 및 배반포로의 발생율은 각각 12.5$\pm$1.6%, 11.1$\pm$1.8%, 11.7$\pm$1.0%였으며, 10% $O_2$조건 하에서 배양하였을 때 핵 이식배의 $\geq$2 cell 및 배반포로의 발생율은 각각 10.5$\pm$1.5%, 9.8$\pm$1.4%, 10.0$\pm$0.8%였다 배양액과 $O_2$ 조건에 따른 유의한 발생율에 차이는 인정되지 않았다.
The purpose of this study is to explore the underlying causes of the development of private security in the United States. These include the increasing crime rates and fear of crime, the poor performance of public policing, the decentralization of political authority, the increase of mass private property, economic growth and increase of personal income, commodification of security, insurance companies' demand for tighter security and fear of litigation, and historical events such as World War I & II. Based on the findings of this study, it is suggested that no single factor would account for the growth of private security in the United States. In other words, the rapid growth of private security in the United States should be attributed to the interrelated influences of the factors given above. Finally, it might be the contribution of this study that the future development of private security in Korea would be guided by understanding the case of the United States because of the similarities of the developmental process in private security industry of two countries.
Thc chimeric morulae were produced following aggregation of the half embryos which were microsurgically bisected at 8-cell and early morula stage. Different phenotypic embryos were obtained by mating ICR female mice with ICR or CBA male mice. The early morula stage was thc desirable stage for the aggregation of mouse embryos after bisection. The post-thawed survival rates of bisected-aggregated embryos that developed into normal blastocyst after conventional freezing in DMSO and ethylene glycol were 30.5 and 32.896, respectively. One offspring was produced by transferring the 67 frozen-thawed bisected-aggregated embryos.

The identification of embryo-specific genes would provide insights into early embryonic development. However, the current methods employed to identify the genes that are expressed at a specific developmental stage are labor intensive and suffer from high rates of false positives. Here we employed a new and accurate reverse transcription-polymerase chain reaction (RT-PCR) technology that involves annealing control primers (ACPs) to identify the genes that are specifically or prominently expressed in bovine early blastocysts and hatched blastocysts produced in vitro.
