어성초 분획이 흰쥐 장기내 카드뮴 축적과 Metallothionein 생성에 미치는 영향(VI) (The Effects of Fractions of Houttuynia cordata THUNB on the Accumulation of Cadmium and Induction of Metallothionein in Rats(IV))

  • This study was conducted to investigate the antitoxic effects of Houttuynia cordata THUNB with chloroform and ethyl acetate fractions. The results were as follows: 1. Detoxication effects of chloroform and ethyl acetate fractions of Houttuynia cordata THUNB were increased in proportion to the dosages. Detoxication effects of ethyl acetate fraction of Houttuynia cordata THUNB were higher than chloroform fraction of Houttuynia cordata THUNB's results and detoxication effects in the kidneys were higher than results for the liver. 2. Metallothionein concentrations in the liver were higher than concentrations in the kidneys, and ethyl acetate fraction of Houttuynia cordata THUNB was higher than chloroform fraction of Houttuynia cordata THUNB in induction of metallothionein. 3. After the administration of chloroform and ethyl acetate fractions of Houttuynia cordata THUNB, body weights increased in proportion to chloroform and ethyl acetate fraction s dosage of Houttuynia cordata THUNB but changes of body weight dminished after 3 weeks. From the above results, this study suggests that chloroform and ethyl acetate fraction of Houttuynia cordata THUNB increased metallothionein induction to cadmium intoxication in rat s kidney and liver and decreased the toxicity of cadmium in rats.

식이 단백질과 Ca 수준이 흰쥐의 Cd 해독에 미치는 영향 (The Effect of Dietary Protein and Calcium Levels on the Cadmium Detoxication on Rats)

  • This study was performed to investigate the effect of dietary protein and calcium levels on cadmium detoxication in rats. Seventy male Sprague-Dawley rats weighing 208 $\pm$ 19 g were blocked into 10 groups of 7 animals according to body weight. Five groups were fed 15% protein-0.6% calcium diet with 100ppm cadmium in drinking water for first 15days and the other 5groups fed same diet without cadmium in drinking water for same period and served as controls. After this 15-day intoxication period, each one of cadmium intoxication and control groups were fed each of 4 kinds of detoxifying diets different with protein(40%, 15%) and calcium(1.3%, 0.6%) levels without cadmimum in drinking water for following 15 days of detoxifying period. Results were summarized as follows: 1) Food intake, body weight gain, F.E.R. and weights of liver, kidney and femur were increased by detoxifying diets and high protein diet was most effective in weight gains of liver and kidney. 2) When cadmium and metallothionein contents of initial intoxication group and those of all detoxication groups were compared, cadmium and metallothionein contents in the liver were not changed, but those in kidney increased, and those in intestine decreased markedly. 3) Only dietary protein level affected cadmium and metallothionein distribution among organs, and cadmium contents of whole blood, liver, kidney and femur were lower in high protein diet, but metallothionein contents in liver and kidney were higher in high protein diet. 4) Gel filtration chromatogram showed that most of cadmium in the cytosol was bound to metallothionein fractions in high protein-high calcium group. Results obtained indicated that high protein diet was effective in cadmium detoxication by increasing the induction of metallothionein synthesis. But high calcium diet did not play a role in cadmium detoxication.

  • This study was conducted to investigate the metallothionein (MT) induction by Lonicerae rios in cadmium chloride intoxication. The results were as follows: Generally, detoxication effects by Lonicerae fios extract increased to the increase of extract concentrations. When 90 mg/kg dosage of Lonicerae fios extract was administered, it showed the highest antitoxic effects in metallothionein induction. From the above results, Lonicerae rios extract increased metallothionein concentration and decreased the toxicity of cadmium in liver.

  • From the study of the definition of and intoxication, detoxication poisonous drug, the symptom and treatment of intoxication, andantidotes with DONG YUI BO GAM etc, it can be concluded as follows. 1. In ancint times, the poisonous drug was a drug which could treat diseases but after that it has been a drug which can injure one's health because it has severe property, poison or side effects 2. Intoxication means that poison goes and works in the body. Detoxication means treatment which clears the symptom of intoxication and removes the toxin in the body or on the body 3. At the intoxication's symptom, vomiting, madness, diarrhea, abdomianl pain, consumption are frequent in order 4. At the intoxication's treatment, vomiting method is used when poison is in the stomach, purgative method is used when poison is the intestine. 5. Mostly Hei-dou is used in drugs and Gan-kao. Qing-yan are used in order. 6. Mostly drugs of reliving fever are used in drugs. 7. Mostly cold drugs are used in drugs and warm, even, hot, cool drugs are used in order and mostly mild drugs are used in drugs and bitter, hot, salty, sore drugs are used in order.

제독유황에 대한 3개월간 랫드에서 반복경구투여 독성에 관한 연구 (Toxicity Study of Detoxication Sulphur at 3 Months Post-treatment in Rats)

  • 본 연구는 8주령 Sprague-Dawley계통의 암컷 랫드에서 제독유황을 반복경구투여 독성평가를 하기위해 시행하였다. 랫드에 처치한 물질은 사료섭취량의 0.2%, 1%, 5% 제독유황과 사료섭취량의 1% 유황으로 13주 경구투여를 하였다. 제독유황의 안전성을 확인하기 위해 다음과 같은 관찰 및 검사를 하였다. 검사항목으로는 체중과 사료 섭취를 측정하였고 임상증상, 안과학적 검사, 혈액학적 검사, 혈청 생화학적 검사를 관찰하였다. 또한 랫드에 간과 신장에서의 병리조직학적 변화를 관찰하였다. 그 결과 체중, 사료섭취량, 혈액학적 및 병리조직검사 소견에서 대조군과 제독유황 그룹 간의 유의적인 차이가 보이지 않았다. 혈청생화학적 검사소견에서는 0.2% 및 1% 투여군에서는 대조군과 비교시 용량의존적으로 유의적인 차이를 보이는 항목은 보이지 않았으나, 고용량군에서는 glucose 감소에 의한 저칼륨혈증, 인슐린 과잉분비의 독성을 의심할 수 있었으며, 또한 ALT와 ALP의 증가는 간 독성이나 간암, 황달의 유발이 의심되었다. 한편, 병리조직학적인 검사에서는 시험 전체군에서 대조군과 비교시 유의할만한 병리조직이상이 관찰되지 않았다. 따라서 본 연구에서는 사료섭취량 1%의 제독유황까지 투여는 랫드에서 독성을 나타내지 않는 것으로 사료된다.

청간산(淸肝散)이 흰쥐의 알코올 대사(代謝) 효소(酵素) 활성도(活性度)에 미치는 영향(影響) (Effects of Chunggansan on Detoxication of Alcohol by Activity of Enzyme in Rats)

  • Chunggansan was tested for the effects on detoxication mechanism of alcohol. Chunggansan was treated firstly into samples, and then ethanol intoxicated animal models were set with them. The administration of Chunggansan to the rats increased proportionally in alcohol dehydrogenase activities in liver in relation to the level of concentration and days of treatment. Especially, the alcohol dehydrogenase was the most active when the concentration of extract was 200mg/kg and it was 7th day. The enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase in liver highly increased in Chunggansan pre-medicating group compared to that of ethanol treated group. Also, the blood ethanol concentration in rats was considerably decreased. In conclusion, Chunggansan recovers the damage of liver due to acute alcohol intoxication by the increased enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase.

  • This study was conducted to investigate the antitoxic component in methanol fraction of Lonicerae fios. The results were as follows: 1. When a 500 ppm of water soluble fraction of Lonicerae flos was administered, it showed the highest antitoxic effect. 2. Generally, detoxication effects of methanol fraction of Lonicerae fios increased. 'When the ethyl acetate soluble fraction of Lonicerae fios was administered, it exhibited the highest antitoxic effect against the toxicity of cadmium in spleen.

  • The results were as follows ; 1. The treatment method is most used reinforcing the spleen and strengthening of Ki(氣) detoxication and draining of pus on each stage. In first stage, the method of actirating blood and aeration of Lag(絡) nourishing blood and moisturizing skin is used. In ulcer stage, the method of expelling blood stasis and detoxication, promoting pus and drainage is used. In chronic stage, strengthening of Ki(氣) and blood, invigiration and dispersion is used. 2. In the frequency of prescription, the most numerous prescription is Paljintang(八珍湯) and the next are Insamyangyoungtang(人蔘養榮湯). Taglisodongum(托裏消耗飮) ect. In the frequency of medicine the most numerous medicine is Angelicae Gigantis Radix(當歸) and the next are Glycyrrhizae Radix (甘草), Astragali Radix(황기) etc. 3. In the frequency of prescription of exteral therapy. the most numerous prescription is Saengioghongo(生肌玉紅膏), Saengisan(生肌散) and the next are hongsungdan(紅升丹), guildan(九一丹) etc. In the frequency of medicine of exteral therapy, the most numerous medicine is Glycyrrhizae Radix (甘草) and the next are Calomelas(輕粉), Draconis Resina(血竭) etc.

Effects of Panax Ginseng on the Development of Morphine Tolerance and Dependence

  • The present study was undertaken to determine the inhibitory effects of orally administered ginseng saponins (GS), protopanaxadiol saponins(PD) and protopanaxatriol saponins(PT) on the development of morphine induced tolerance and physical dependence in mice, and to determine the increases in the loss of morphine tolerance and dependence. The study also sought to determine the hepatic glutathione contents, which are closely related to the degree of detoxication of morphinone, a novel metabolite of morphine, and the effects of ginseng saponins on morphine 6-dehydrogenase. The results of the present study showed that GS, PD and PT administered orally inhibited the development of morphine-induced tolerance and dependence. GS, PD and PT, however, increased the loss of morphine tolerance and dependence. GS, PD and PT inhibited the reduction of hepatic glutathione concentration in mice treated chronically with morphine, and the activity of morphine 6-dehydrogenase. So we hypothesized that these results were partially due to the dual action of the test drugs, the inhibition of morphine production and the activation in morphine-glutathione conjugation due to the increased glutathione level for detoxication.

메틸수은(水銀)으로 손상(損傷)된 생쥐 간장(肝臟)에 대한 홍삼(紅蔘) 추출물(抽出物)의 해독효과에 관한 전자현미경적(電子顯微鏡的) 연구(硏究) (Electron Microscopic Study on Detoxication Effect of Red Ginseng Extract on Mouse Liver Injury induced by Methyl Mercury)

  • 생쥐 간세포(肝細胞)의 미세구조(微細構造)에 미치는 메틸수은(水銀)의 독성적(毒性的) 영향(影響)과 이에 대한 홍삼(紅蔘) 추출물(抽出物)의 해독효과를 전자현미경적(電子顯微鏡的)으로 연구하였다. 대조군(對照群)의 간세포(肝細胞)에 비하여 메틸수은(水銀)만을 투여(投與)한 군(群)에서는 핵(核)의 일부가 붕괴(崩壞)되고, mitochondria의 팽대(膨大), cristae의 수축(收縮) 내지 손실(損失) 및 다수의 대형 강포(腔胞)의 출현(出現)이 일어나고 glycogen의 양(量)이 감소되었다. 메틸수은(水銀)과 홍삼(紅蔘) 추출물(抽出物)을 병행(倂行) 투여(投與)한 군(群)에서는 메틸수은(水銀)만을 투여(投與)한 군(群)에서보다 mitochondria의 팽대(膨大) 정도(程度)가 적고 강포(腔胞)의 크기와 수가 감소되었으며 정상(正常) 세포(細胞)와 비슷한 상태로 나타났다.
