Fatty acid compositions of Panax ginseng leaves (6 year) grown under different light intensity in field and of the detached leaves exposed to high temperature (20 hours) were investigated by gas chromatography. Linoleic, linolenic, palmitic and palmitoleic acid were the major components(80%) of leaf lipid. The higher the growth light intensity, the lower the percentage of unsaturated acids or bonds, indicating metabolic adaptation to high temperature. Pattern similarity of fatty acid composition was little changed until 20% light but significantly different at 30%, suggesting 20% as limitation light intensity. The close similarity of fatty acid composition between the loaves grown under 30% light and the one at harvest rises uncertainty between adaptation to high temperature and senescence. Total fatty acid content decreased with the increase of light intensity. Short term high temperature $(25^{\circ}C\;or\;35^{\circ}C)$ increased total fatty acid content, unsaturated acid percentage and insignificant difference in pattern similarity of composition.
Activity of senescence-induced antioxidant enzymes in the detached rice seedlings (Oryza sativa L. cv. Dongjin) was examined. The levels of $\textrm{H}_2\textrm{O}_2$ content and peroxidase (POD) activity were gradually increased during leaf senescence, whereas catalase activity was decreased. The activity of superoxide dismutase (SOD) was increased, and ascorbate peroxidase (APX) and glutathione reductase (GR) were slightly increased until 3d and 4d of dark induced-senescence, and thereafter were decreased. The activation of all SOD isoforms showed a significant decrease after 6d and 7d. After 4d to 7d of dark senescence, there was a significant effect in enhancing the activity of APX-12 and -13 isoforms as compared with light, despite similar levels in total APX activity. GR-8 and -10 isoforms were more effective in leaf senescence at 4d to 7d, particularly with respect to dark-induced senescence. These results suggest that the metabolism of active oxygen species such as $\textrm{H}_2\textrm{O}_2$ is dependent on various functionally interrelated antioxidant enzymes such as catalase, peroxidase, SOD, APX and GR.
A leaf blight caused by a species of Phytophthora was found on fatsia plants (Fatsia japonica Decne et Planch.) growing in an apartment garden in Daegu, Korea in late April to May, 2003. The species of Phytophthora isolated from the diseased plants produced sporangia and sex organs on V8 juice agar medium. Sporangia were papillate, ovoid to subspherical, and caducous with a pedicel. The dimensions of the sporangia were $31.2-46.8\times23.4-33.2{\mu}m$ in range, $39.6\pm4.1\times28.3\pm2.8{\mu}m$ in $average{\pm}standard$ deviation, I/b ratio approximately 1.40, with papillae about $3.6{\mu}m$ high, and pedicels $0.95.8{\mu}m$ long. Oogonia were spherical, $25.0-32.5{\mu}m$ in range with an average of $28.2\pm2.3{\mu}m$ in diameter. Antheridia were predominantly paragynous, globose to ovoid, $8.8-13.8\times7.5-10.0{\mu}m$ with an average of $10.9\pm1.2 \times9.2\pm1.1{\mu}m$. Oospores in the oogonia were aplerotic or plerotic, and $20.0-25.0{\mu}m$ in diameter with an average of $23.5\pm1.5{\mu}m$. Pathogenicity of the isolate was confirmed on detached leaves of fatsia. The disease was observed only in April and May of 2003 when the weather was unusually wet. It then diminished with increase of temperature in the year and did not appear again on the same plants in 2004. Thus, the fungus appeared to be a relatively weak pathogen of fatsia.

We have isolated more than a dozen cDNA clones corresponding to genes that were expressed in Arabidopsis leaves when they were kept in the dark. The nucleotide sequence analysis showed that some of the clones encoded proteins with significant homology to $\beta$-glucosidase (din2), branched-chain $\alpha$-keto acid dehydrogenase subunit E1$\beta$(din3), and another subunit E2 (din4), yeast RAD23 (din5), asparagine synthetase (din6), pre-mRNA splicing factor SRp35 (din7), phosphomannose isomerase (din9), seed imbibition protein (din10), and 2-oxoacid-dependent oxidase (din11). Accumulation of transcripts from din3,4,6 and 10 occurred rapidly after the plants were transferred to darkness. Transcripts from din2,9, and 11 could be detected only after 24 h of dark treatment. Inhibition of photo-synthesis by DCMU strongly induced the accumulation of transcripts from those genes, and application of sucrose to detached leaves suppressed the accumulation both in the dark and by DCMU. These observations indicate that expression of the genes is caused by sugar starvation resulted from the cessation of photosynthesis. We further showed that din2-encoded protein also accumulated in senescing leaves. Given these results, possible roles of din genes in leaves in the dark and senescing leaves are discussed.

In the present study we evaluated the efficacy of a bioformulation of Streptomyces cameroonensis for control of black pod disease in cocoa and enhancement of seedling growth. The formulation developed using talc powder and cassava starch as carriers showed high shelf-life of 1.07 × 106 CFU/g after six months storage at 4℃. The formulation was tested for inhibition of spore germination in Phytophthora megakarya and showed 100% inhibition at 10% (w/v) of formulation. To determine the efficacy of the formulation, we performed an in planta assay in the greenhouse on two hybrids of cocoa seedlings, the tolerant SNK413 × (♂) T79/467 and the susceptible UPA 134× (♂) SCA 12. Detached leaf assay showed a significant reduction in the disease severity index of about 67% for the tolerant hybrid and 55% for the susceptible hybrid compared to non-treated plants. A significant enhancement in stem length, leaf surface area and root weight was observed. Analysis of biochemical markers of defense showed a significant increase in total polyphenol, flavonoid, and total protein contents. There was also significant upregulation of PR-proteins such as chitinases, peroxidases and β-1, 3-glucanases following treatment of both tolerant and susceptible hybrids, though with a higher level of synthesis in the tolerant hybrids. A significant increase was also observed in polyphenol oxidase activities in plants treated with the formulation. This work demonstrated the stability and effectiveness of the S. cameroonensis powder formulation in suppressing black pod disease in cocoa and subsequently enhancing the growth of seedlings.

Though information exists regarding the pathogenesis of the shot-hole disease (SH) in flowering cherry (FC), there has been a lack of research focusing on SH management. Therefore, here, we investigated the inhibitory activities of antagonistic bacteria against SH pathogens both in vitro and in vivo as well as their biochemical characteristics and bioactive compounds. Two biosurfactant-producing bacterial antagonists, identified as Bacillus velezensis strains JCK-1618 and JCK-1696, exhibited the best effects against the growth of both bacterial and fungal SH pathogens in vitro through their cell-free culture filtrates (CFCFs). These two strains also strongly inhibited the growth of the pathogens via the action of their antimicrobial diffusible compounds and antimicrobial volatile organic compounds (VOCs). Crude enzymes, solvent extracts, and biosurfactants of the two strains exhibited antimicrobial activities. Liquid chromatography/electrospray ionization time-of-flight mass spectrometric analysis of the partially purified active fractions revealed that the two antagonists produced three cyclic lipopeptides, including iturin A, fengycin A, and surfactin, and a polyketide, oxydifficidin. In a detached leaf assay, pre-treatment and co-treatment of FC leaves with the CFCFs led to a large reduction in the severity of the leaf spots caused by Epicoccum tobaicum and Bukholderia contaminans, respectively. In addition, the two antagonists produced indole-3-acetic acid, siderophore, and a series of hydrolytic enzymes, along with the formation of a substantial biofilm. To our knowledge, this is the first report of the antimicrobial activities of the diffusible compounds and VOCs of B. velezensis against the SH pathogens and their efficiency in the biocontrol of SH.
This is the first study reporting the evaluation of transgenic lines of tomato harboring rice chitinase (RCG3) gene for resistance to two important fungal pathogens Fusarium oxysporum f. sp. lycopersici (Fol) causing fusarium wilt and Alternaria solani causing early blight (EB). In this study, three transgenic lines TL1, TL2 and TL3 of tomato Solanum lycopersicum Mill. cv. Riogrande genetically engineered with rice chitinase (RCG 3) gene and their R1 progeny was tested for resistance to Fol by root dip method and A. solani by detached leaf assay. All the R0 transgenic lines were highly resistant to these fungal pathogens compared to nontransgenic control plants. The pattern of segregation of three independent transformant for Fol and A. solani was also studied. Mendelian segregation was observed in transgenic lines 2 and 3 while it was not observed in transgenic line 1. It was concluded that introduction of chitinase gene in susceptible cultivar of tomato not only enhanced the resistance but was stably inherited in transgenic lines 2 and 3.
Sporulation potential and conidia release phase of Pyricularia oryzae on lesions under the natural conditions were measured in 1985 and 1986 leaf blast seasons. The amount of conidia produced in lesions on detached leaves and conidia released under the field condition were reached peak at $5{\sim}7$ days after lesion appearance. The maximum numbers of conidia produced and released were 16,200 and 15,900, respectively. Conidia release under the natural conditions lasted for 30 days.
Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.
A total of 55 Colletotrichum isolates from strawberry plants with anthracnose symptoms(crown rot) were inhibited in mycelial growth on potato-dextrose agar(PDA) amended with fungicides in variable degrees depending on the chemicals used, especially showing no growth on PDA with 1 mg/m/tricyclazole. However, in the detached leaf test by treating chemicals before or after inoculation of Colletotrichum sp., tricyclazole was little effective in controlling symptom development; instead azoxystrobin, which had low in vitro inhibition of mycelial growth, inhibited strongly the symptom development. Some chemicals were tested for the control of strawberry crown rot in greenhouse using three methods, sprays soil drenching and plant dipping. No or little control effect were made by chemical spray and soil drenching, but plant dipping in chemical solution, especially azoxystrobin: reduced crown rot development by about 50% in the greenhouse suggesting that the azoxystrobin treatment may be an effective control method of the crown rot of strawberry. No differences in the control efficacy were noted according to the dipping time and chemical concentration of azoxystrobin not less than 10 min and 250 mg/m/, respectively.
