Screening of Medicinal Plants containing Lipase Inhibitor and Optimal Extraction Conditions (지질분해효소 저해물질 함유 약용식물의 선발 및 저해물질 추출조건)

  • This study was performed to develop new anti-obesity agents for functional foods from medicinal plants and herbs. Methanol extracts from 560 kinds of medicinal plants and herbs, and their lipase inhibitory activities were investigated. Methanol extract from Desmodium oxyphyllum Dc showed the highest lipase inhibitory activity at 74.2%. This lipase inhibitory activity was maximally extracted when powder of Desmodium oxyphyllum Dc was treated with 80% methanol (1 : 20) for 48 h.

Nutritional Characteristics and Physiological Functionality of Lipase Inhibitor-Containing Desmodium oxyphyllum DC. Extracts (지질분해효소 저해물질 함유 도둑놈의 갈고리 추출물의 영양특성 및 생리기능성)

  • This study was performed to develop new anti-obesity agents from medicinal plants for functional food industry. We prepared lipase inhibitor-containing ethanol extracts from Desmodium oxyphyllu DC. and its nutritional characteristics, stability of temperature and pH and physiological functionalities were investigated. Crude lipid content and curde protein content of ethanol extract from Desmodium oxyphyllum DC. were 26.5% and 5.7%, respectively and its asparagine content was 4,860 mg/100 g, dry basis. The ethanol extracts were also stable from pH 3.0 to pH 9.0 and below $80^{\circ}C$. The ethanol extract were showed high lipase inhibitory activity of 74.2% and acetylcholinesterase inhibitory activity of 68.4%.

Purification and Characterization of Lipase Inhibitor from Desmodium oxyphyllum DC. (도둑놈의 갈고리의 지질분해효소 저해물질의 정제 및 특성)

  • A lipase inhibitor from Desmodium oxyphyllum DC. was purified by methanol extraction, systematic solvent extraction, silica gel column chromatography, $C_{18}$ solid phase extraction chromatography and RP-HPLC. We obtained the purified lipase inhibitor with 182 ng($IC_{50}$) of lipase inhibitory activity for a 0.06% yield. Its molecular weight was estimated to be 655.37 Da from an instrumental analysis of MALDI-TOF-MS and it was identified copper-3,5-dibromo-2-hydroxybenzoic acid ($C_{14}H_8Br_4CuO_6$) by $^1H$, $^{13}C$ NMR analysis.
