Induction of 90K-specific Cytotoxic T Lymphocytes for Colon Cancer Immunotherapy

  • Background: Dendritic cell (DC)-based tumor vaccine is an attractive modality for the treatment of colon cancer because it has been recurred and produced few side effects in patients. Secretory glycoprotein 90K has been found at elevated level in various cancer tissues and sera. We investigated to establish a more effective DC vaccine for the treatment of colon cancer in which the levels of 90K are elevated. Methods: We obtained the concentrated 90K from 293T cells stably expressing 90K. DCs were cultured from peripheral blood monocytes, and a DC vaccine pulsed with tumor lysate was compared with a DC vaccine pulsed with 90K. We measured the functional activity for CTLs by using IFN-${\gamma}$-enzyme linked immunoabsorbent spot (ELISPOT) assay. Results: DCs pulsed with tumor lysate+90K exhibited the enhanced T cell stimulation, polarization of $\ddot{i}$ T cell toward Th1. The CTLs generated by DCs pulsed with 90K efficiently lysed HCT116 cells. The results indicate that 90K-speicifc-CTLs can recognize 90K proteins naturally presented by colon cancer cells. Conclusion: Our study suggests that 90K-specific CTLs generated by 90K-pulsed DCs could be useful effector cells for immunotherapy in colon cancer.

Fucoidan Enhances the Survival and Sustains the Number of Splenic Dendritic Cells in Mouse Endotoxemia

  • Fucoidan is a sulfated polysaccharide derived from brown algae that has been reported to perform multiple biological activities, including immunostimulation. In this study, we investigated whether fucoidan has beneficial effects on endotoxemia induced by LPS, a septic model in mice. The focus of this study was on survival rates and spleen function of the mice upon treatment. We found that fucoidan had prophylactic effects on the survival rate of mice with endotoxemia. Flow cytometric analysis using antibodies for subset-specific markers revealed that fucoidan profoundly reversed the depleted population of dendritic cells in mice with endotoxemia. According to Western blot analysis, the spleen cells of LPS/fucoidan-treated mice showed a higher expression of anti-apoptotic molecules compared to those of LPS-treated mice. Also, fucoidan-treated spleen cells were more responsive to mitogens. Taken together, these results demonstrate that fucoidan pre-treatment has beneficial effects on the survival rate and function of the spleen in mice with endotoxemia. This study may broaden the use of fucoidan in clinical fields, especially endotoxemia.

Upregulation by KCI Treatment of Eukaryotic Translation Elongation Factor 1A (eEF1A) mRNA in the Dendrites of Cultured Rat Hippocampal Neurons

  • Activity-dependent local translation in the dendrites of brain neurons plays an important role in the synapse-specific provision of proteins necessary for strengthening synaptic connections. In this study we carried out combined fluorescence in situ hybridization (FISH) and immunocytochemistry (IC) and showed that more than half of the eukaryotic elongation factor 1A (eEF1A) mRNA clusters overlapped with or were immediately adjacent to clusters of PSD-95, a postsynaptic marker, in the dendrites of cultured rat hippocampal neurons. Treatment of the neurons with KCl increased the density of the dendritic eEF1A mRNA clusters more than two-fold. FISH combined with IC revealed that the KCl treatment increased the density of eEF1A mRNA clusters that overlapped with or were immediately adjacent to PSD-95 clusters. These results indicate that KCl treatment increases both the density of eEF1A mRNA clusters and their synaptic association in dendrites of cultured neurons.

  • The effect of welding condition on the microstructures of the weld metal in A7N01 welded by $CO_2$ laser was investigated. The number of ripples was increased with decreasing power and increasing welding speed. In the bead without ripple lines, the subgrain microstructures distribution from the fusion line toward the center of the bead were in the order of cellular, dendritic and equiaxed dendrite. However, in the bead with ripple lines, cellular and dendritic were formed between the fusion boundary and the ripple line. Inaddition, those structures were also observed between the ripple line. Equiaxed dendrites were formed only at the center line region. Cellular and dendritics formed near the ripple line were larger than those formed near the fusion boundary. The cooling rates estimated by the dendrite arm spacing were in the range of 200 to 1150oC/s. Cooling rate was increased with decreasing the power and increasing the welding speed. Mg and Zn segregated at the boundaries of cellulars and dendritics, Mg was segregated more than Zn. The segregation of Mg and Zn decreased with increasing cooling rate. Hardness of the weld metal was lower than that of the base metal in all welding conditions and increased as the cooling rate increased.

Unsteady Flow Analysis on Flood Characteristics in KEUM River Downstream (금강 하류 홍수의 부정류 해석)

  • KEUM River downstream has some characteristics in which the dowunstream is affected with tidal motion, the several tributaries are forming a dendritic river system, and the channel cross-sections are irregular. The flood in this downstream can now be analyzed by the hydrological flood routing methods and under the assumption regarding the dendritic river system as a single reach. In this study the river system was used for the flood routing. The flood records which were measured in 1978 and 1987 were applied for calibration and verification of the unsteady flow model respectively. The results show that the flood at KANG-KYONG station was not affected with the tidal motion when the discharge at KONG-JU station exceeded about 5, 000$m^3$/sec, and that the bottle neck at IP-PO station intercepted the tidal influences.

  • Comparative study on the synaptic types of the nuclei accumbens septi and fundus striati of the chick and the rat was carried out. Basic synaptic types were established according to the size of synaptic vesicles, development of synaptic vesicles, development of synaptic thickening, kind of postsynaptic structures, etc. Comparing the synaptic types and appearance-ratio within the neuropils, the following results were obtained (see the data in the Table 1). 1. In the nucleus accumbens of the rat, the axo-spinous synapses are far less than those in the nucleus fundus striati (13.7%/68.2%). 2. In the nucleus accumbens, there found much more axo-dendritic types(II, III and V) than those appearing in the nucleus fundus striati(79.1%/27.1%). 3. In the nucleus accumbens of the chick, on the contrary to the case of the rat, more axo-spinous types(70.1%/31.4%) and less axo-dendritic types(12.3%/60.0%) were found as compared to those appearing in the nucleus fundus striati. 4. There found no large-sized synaptic vesicles in the chick nuclei accumbens septi and the fundus striati.

Effect of the Casting Conditions on the Globulization of Primary Al of $AlSi_7Mg$ Alloy (($AlSi_7Mg$알루미늄 합금의 초정 구형화에 대한 주조조건의 영향)

  • Semisolid forming requires alloys with non-dendritic microstructure of the thixotropy. Recently, low pouring temperture method without stirring, i.e. liquidus casting has been found out new fabrication method of the semisolid metals. Effects of melt superheat and mold conditions on the globulization of primary Al of $AlSi_7Mg$ alloy were investigated in gravity casting process without stirring. The microstructures of primary Al as function of melt superheat and mold temperature show globular, rosette and dendritic shapes. The conditions for globular microstructure of primary Al were low melt superheat < 35 K and low mold temperature < 500 K. The thermal conditions for globular microstructure of primary Al were undercooled melt at early solidification stages and slow cooling < 0.6 K/s. It was found that the initial microstructure was maintained throughout the solidification and the globules of primary Al can be obtained by high nucleation of fine and spherical nuclei due to enhanced undercooling of melt.

Porphyrin-Cored Arylether Dendrimers with Vinyl Groups in the Periphery

  • Benzyl arylether dendrimers with zinc porphyrin core and terminal vinyl groups have been synthesized and their photophysical properties and the influence of dendritic environments were investigated. Free base porphyrin-cored benzyl arylether dendrimers 1a-1c and 3a-3c, and their zinc derivatives 2a-2c and 4a-4c have been prepared. Absorption spectra are similar for all porphyrin-cored benzyl arylether dendrimers, except that absorption intensity at 280 nm increases in the higher generation of dendrimer. Fluorescence spectra are similar with two bands for all free base porphyrin dendrimers 1a-1c and 3a-3c, although fluorescence intensity ratio of shorter wavelength emission band to longer wavelength band varies with the generation of dendrimer. Emission efficiencies of 1a-1c and 3a-3c are lower than that of TTP. Emission efficiencies of 2a-2c and 4a-4c are higher than that of ZnTTP. Absorption and emission properties of 1a-1c, 2a-2c, 3a-3c, and 4a-4c were affected negligibly with dendritic environments.

Electrocatalytic Oxidation of HCOOH on an Electrodeposited AuPt Electrode: its Possible Application in Fuel Cells

  • Controlled electrodeposition of dendritic nano-structured gold-platinum (AuPt) alloy onto an electrochemically pretreated carbon paper substrate was conducted in an attempt to improve catalyst utilization and to secure an electronic percolation network toward formic acid (FA) fuel cell application. The AuPt catalysts were obtained by potentiostatic deposition. AuPt catalysts synthesized as bimetallic alloys with 60% Au content exhibited the highest catalytic activity towards formic acid electro-oxidation. The origin of this high activity and the role of Au were evaluated, in particular, by XPS analysis. Polarization and stability measurements with 1 mg $cm^{-2}$ AuPt catalyst (only 0.4 mg $cm^{-2}$ Pt) showed 52 mW $cm^{-2}$ and sustainable performance using 3M formic acid and dry air at $40^{\circ}C$.

Expression of CD320 in human B cells in addition to follicular dendritic cells

  • CD320 has been recently discovered and reported as a follicular dendritic cell (FDC) protein. Although CD320 is known to enhance proliferation of germinal center (GC) B cells, little other information is available. In this study, we investigated its cellular distribution in the GC. Confocal microscopy of human tonsil sections revealed co-localization of CD320 with CD19 and CD38 but not with CD3 indicating that GC B cells expressed CD320 in addition to FDC. In purified GC B cells, CD320 expression was inhibited in the nucleus, membrane and cytoplasm. Reverse transcriptase-polymerase chain reaction confirmed CD320 mRNA expression in B cells. These finding indicate that CD320 is expressed in B cells in addition to FDC, and that its GC activity may be more complicated than previously thought.
