Compounds from the Seeds of Myristica fragrans and Their Cytotoxic Activity

  • Six lignan compounds, 1-(17,21-dihydroxyphenyl)-9-(12,13-dihydroxyphenyl)-1-nonanone (malabaricone C) (1), 7'-(3',4'-methylenedioxyphenyl)-8,8'-dimethyl-7-(3,4-dihydroxyphenyl)-butane (2), 7'-(3',4'-dimethoxyphenyl)-8,8'-dimethyl-7-(3-methoxy-4-hydroxyphenyl)-butane (3), 7-(4-hydroxy-3-methoxyphenyl)-7'-(3',4'-methylenedioxyphenyl)-8,8'-lignan-7-methyl ether (4), (+)-erythro-(7S,8R)-${\Delta}^{8^'}$-7-hydroxy-3,4,3',5'-tetramethoxy-8-O-4'-neolignan (5), and (+)-erythro-(7S,8R)-${\Delta}^{8^'}$-7-acetoxy-3,4,3',5'-tetramethoxy-8-O-4'-neolignan (6), were isolated from the seeds of Myristica fragrans. The chemical structures of these compounds were determined on the basis of spectroscopic analyses including 2D NMR. Compounds 1 - 6 were evaluated for their cytotoxic activity against the HL-60, MCF-7, and A549 cancer cell lines in in vitro.

Inabenfide-Induced Alleviation of Salt Stress in Rice as Linked to Changes in Salicylic Acid Content and Catalase Activity

  • The effect of inabenfide was investigated in rice seedlings subjected to salt stress in relation to changes in chlorophyll fluorescence(${\Delta}F/Fm'$), lipid peroxidation, salicylic acid(SA) content, and catalase(CAT) activity. A reduction in shoot growth of rice seedlings by 120 mM NaCl treatment was significantly alleviated by pretreatment with 30 ${\mu}M$ inabenfide. Sodium ion content was not affected by pretreatment with inabenfide, suggesting that alleviation was not due to a reduction in sodium ion uptake by the rice seedlings. At three days after NaCl treatment, the rice seedlings pretreated with inabenfide showed a higher ${\Delta}F/Fm'$(30%) and lower lipid peroxidation(28%) compared with the rice seedlings treated with NaCl alone. After NaCl treatment, CAT activity in the third leaf of rice seedlings decreased significantly but alleviated by pretreatment with inabenfide. Furthermore, pretreatment with inabenfide also reduced the level of SA which accumulated drastically in the third leaf of rice seedlings within a day after exposure to salt stress. These results suggest that inabenfide prevents SA accumulation in rice seedlings under salt stress which eventually induces the alleviation of salt stress damage.

  • To enhance application of FPC in food industry, salmon FPC hydrolysates were produced and their functional properties were investigated. Hydrolysates of salmon FPC showed high solubilities ranged from approximately $80\~95\%$. Emulsifying activity index (EAI) values of hydrolysates were higher than that of egg white, however sugar ester showed the highest EAI value of 32.5. Though sugar ester had the best emulsifying stability value $({\Delta}EAI)$ of 9.65, hydrolysates showed better ${\Delta}EAI$ value than that of egg white. Foam Activity of hydrolysates was predominantly better than those of controls and also foam stability value showed similiar trend. Osmolality of hydrolysates was increased with the increase of degree of hydrolysis (DH) and concentration of protein, but it showed very lower osmolality than that of NaCl. Flow property of hydrolysates showed different upward-downward curves, and hysteresis loop increased with the increase of DH. ACE inhibitory activity showed similiar value ranged from $21.1\~24\%$ at all DH values.

The Effect of Lactose and Calcium on the Acute Lead Poisoning in Rats (白鼠에서 乳糖과 칼슘이 急性 납중독에 미치는 영향)

  • This study was performed to investigate the effect of lactose in 4 different concentrations against the protective effect of calcium on the acute lead poisoning in rats after 4 weeks treatment. In this animal experiment, 70 albino male weanling rats (50-70g of body weight) of Sprague-Dawley strain were used. Lead was dissolved in the distilled water and intubated at the dose of 400mg lead (as acetate)/ kg of body weight/day. Calcium and lactose were administered in drinking water ad libiturn dissolved with the solution of 0.7% calcium gluconate mixed with 40, 80, 160 and 320mM lacotse respectively. The results obtained were summarized as follows: 1. The rate of body weight gain in all treated groups turned out to be lower than that in the control group during 4 weeks treatment. The slow-down of body weight gain was the most significantly observed in the group treated with lead only ( p < 0.05). 2. The relative spleen weight in lead only treated group was significantly lower than that of lead + calcium, lead + calcium + 80mM lactose treated group ( p < 0.05). 3. The value of RBC, WBC, Hb and Hct showed a decreasing tendency in the group treated with lead only ( p < 0.05), however, a significant decrease was not observed in the group treated with lead + calcium. On the other hand, the protective effect of calcium was deteriorated in the group treated with lead + calcium + lactose. 4. The activity of $\delta$-aminolevulinic acid dehydratase ($\delta$-ALAD activity) showed the same tendency as No. 2. 5. The lead concentration in the blood (PbB) showed an increasing tendency and the interrelation among the different groups was also identical with No. 2. 6. With a statistical approach, it was found out that the activity of $\delta$-ALAD and the lead concentration in the blood show a relation of inverse proportion(r=-0.7301). The diagram was interpreted with the logarithmic equation InY = 5.5357-0.0251X (X:PbB, Y:$\delta$-ALAD activity). 7. In the histopathological findings of the kidney, the protective effect of calcium was observed. However, the protective effect of calcium was restricted in the group treated with lead + calcium + lactose. As a conclusion, the intensity of the acute ingested lead poisoning was obviously reduced by calcium, however, the protective effect of calcium was deteriorated in proportion with the concentration of the lactose to be administered. On the other hand, it was also noted that the deterioration was lightly restrained in the group treated with the physiological concentration of 80mM lactose than the results shown in the groups treated with lactose of other concentrations.

Breeding of Ethanol-producing and Ethanol-tolerant Saccharomyces cerevisiae using Genome Shuffling (Genome shuffling을 이용한 에탄올 생산 및 내성 효모 균주의 육종)

  • To improve yeast strains for bioethanol production, yeasts with ethanol tolerance, thermotolerance, and ${\beta}$-1,3-glucanase activity were bred using yeast genome shuffling. Saccharomyces cerevisiae $BY4742{\Delta}exg1$/pAInu-exgA, which has extracellular ${\beta}$-1,3-glucanase activity, and the Aspergillus oryzae and S. cerevisiae YKY020 strains, which exhibit ethanol tolerance and thermotolerance, were fused by yeast protoplast fusion. Following cell fusion, four candidate cells (No. 3, 9, 11, and 12 strains) showing thermotolerance at $40^{\circ}C$ were selected, and their ethanol tolerance (7% ethanol concentration) and ${\beta}$-1,3-glucanase activity were subsequently analyzed. All the phenotypes of the two parent cells were simultaneously expressed in one (No. 11) of the four candidate cells, and this strain was called BYK-F11. The BYK-F11 fused cell showed enhanced cell growth, ethanol tolerance, ${\beta}$-1,3-glucanase activity, and ethanol productivity compared with the $BY4742{\Delta}exg1$/pAInu-exgA and YKY020 strains. The results prove that a new yeast strain with different characters and the same mating type can be easily bred by protoplast fusion of yeasts.

Expression System for Optimal Production of Xylitol Dehydrogenase (XYL2) in Saccharomyces cerevisiae (출아효모에서 xylitol dehydrogenase (XYL2)의 최적 생산을 위한 발현 시스템 구축)

  • In this study, the xylitol dehydrogenase (XYL2) gene was expressed in Saccharomyces cerevisiae as a host cell for ease of use in the degradation of lignocellulosic biomass (xylose). To select suitable expression systems for the S.XYL2 gene from S. cerevisiae and the P.XYL2 gene from Pichia stipitis, $pGMF{\alpha}-S.XYL2$, $pGMF{\alpha}-P.XYL2$, $pAMF{\alpha}-S.XYL2$ and $pAMF{\alpha}-P.XYL2$ plasmids with the GAL10 promoter and ADH1 promoter, respectively, were constructed. The mating factor ${\alpha}$ ($MF{\alpha}$) signal sequence was also connected to each promoter to allow secretion. Each plasmid was transformed into S. cerevisiae $SEY2102{\Delta}trp1$ strain and the xylitol dehydrogenase activity was investigated. The GAL10 promoter proved more suitable than the ADH1 promoter for expression of the XYL2 gene, and the xylitol dehydrogenase activity from P. stipitis was twice that from S. cerevisiae. The xylitol dehydrogenase showed $NAD^+$-dependent activity and about 77% of the recombinant xylitol dehydrogenase was secreted into the periplasmic space of the $SEY2102{\Delta}trp1/pGMF{\alpha}-P.XYL2$ strain. The xylitol dehydrogenase activity was increased by up to 41% when a glucose/xylose mixture was supplied as a carbon source, rather than glucose alone. The expression system and culture conditions optimized in this study resulted in large amounts of xylitol dehydrogenase using S. cerevisiae as the host strain, indicating the potential of this expression system for use in bioethanol production and industrial applications.

Synthesis and Evaluation of Antitumor Activity of 2- and 6-[(1,3- Benzothiazol-2-yl)aminomethyl]-5,8-dimethoxy-1,4-naphthoquinone Derivatives

  • 2- or 6-Substituted BZT-N derivatives were synthesized, and their cytotoxic activity against can-cer L1210 and SNU-1 cells was examined. The antitumor action was also assessed in mice bearing S-180 cells in peritoneal cavity. In a comparison, it was found that 6-substituted BZT-N derivatives exhibited higher potencies in both bioactivities than 2-substituted BZT-N derivatives against L1210 cells in in vitro and S-180 in vitro tests exception of compound 36. Interestingly, it was observed that 2-substituted compound 36, which has methyl group at RI position, exhib-ited a better antitumor activity than 6-substituted compounds against L1210 and SNU-1 in vitro. The EDso value of 2-substituted compound 36 against L1210 was found to be comparable to the EDso value of adriamycin and was even better against the solid cancer cell line SNU-1. It was also observed that 2-substituted compound 36 showed better antitumor activity in mice bearing S-180 cells in the peritoneal cavity. The T/C (%) value of 2-substituted compound 36 was simi-lar to that of adriamycin. Quantitative structure-activity relationship (QSAR) tests reveal that the experimental E $D_{50}$ values against SNU-1 closely correlate with both the calculated HOMO ener-gies ( $E_{HOMO}$) and the measured H-NMR chemical shift of 3-H ($\delta$$_{H}$). The results suggests that a compound having higher $E_{HOMO}$ and $\delta$$_{H}$ values usually should have a lower E $D_{50}$ (SNU-1) value.lue.lue.lue.

  • The effects of different concentrations of tocopherols on the oxidation stability of evening primrose oil (EPO) in the dark $(35{\pm}2^{\circ}C)$ were evaluated by some physicochemical characteristics such as peroxide, conjugated diene value and fatty acid composition. The results of the present study were as follows; Alpha-tocopherol at all concentrations (0.02%, 0.05%, 0.10%) showed a prooxidant effect during autoxidation of EPO. This prooxidant activity of alpha-tocopherol increased as its concentration increased rapidly. Delta-tocopherol showed antioxidant activity as its concentration increases at an early stage of storage. But the activity was fairly poor antioxidant activity and was not greater than antioxidant activity of BHT. However, the antioxidant effect of delta-tocopherol and BHT showed no apparent differences as compared with result of none.

A Study on Correlations between EEG and Personality (성격과 EEG의 상관에 관한 연구)

  • This study is an experimental study about the relationship between EEG and personality dimensions. Subjects were 70 healthy undergraduate students (32 males, 38 females) The Korean version of Eysencks Personality Questionnaire-Revised (EPQ-R) were used. EEGs were recorded in two different conditions (eyes open and eyes closed). To obtain more tellable data, EEG recordings of subjects were replicated 6 weeks later. The analytical methods used in the present study rendered two coincident tendencies: Theta with neuroticism relationship in eyes closed condition, and alpha-delta with psychoticism relationship in eyes closed condition. These results generally support the theory of Robinson (2001) that delta, theta, and alpha waves are associated with activity of brainstem, limbic system, and thalamo-cortical arousal system, and that thalamo-cortical alpha system has inhibitory effects on brainstem delta system.

Study of 3-Ketosteroid Dehydrogenase System Using Whole-cell-enzyme from Arthrobacter simplex

  • A new assay method for delta-l-dehydrogenated-3-ketoco-rticosteroid in the presence of proteinous material or whole-cell-enzyme and 3-ketocorticosteroid has been developed. This method makes use of the linear relationship between the ratio of absorbances at 265 nm and at 242 nm and the fractional concentration of delta-1-3-ketosteroid. Theoretical values were calculated based on the absorbances of proteinous material at fixed concentrations of the 3-ketosteroid and delta-1-dehydrogenated-3-ketosteroid. The values obtained experimentally showed good agreement with the values obtained experimentally showed good agreement with the values theoretically predicted. The new assay method developed for the steroid mixtiure containing proteinous material is of some practical importance. The use of such assay method enables one to determine the enzyme activity and the rate of enzyme reaction or conversion rather quickly, easily and accurately. By the use of this assay method, the reaction kinetics of whole-cell-enzyme has also been studied. It was found that it followed the simple Michaelis-Menten type enzyme kinetics. Also the reversibility of this reaction with actively metabolizing cell was examined. It was found that delta-l-dehydrogenated-3-ketosteroid could not be hydrogenated reversibly to 3-ketosteroid by this enzyme system.
