Enhanced Degradation of Residual Cadusafos in Soils by the Microbial Agent of Cadusafos-degrading Sphingobium sp. Cam5-1 (미생물제(Sphingobium sp. Cam5-1) 처리에 따른 토양 중 카두사포스의 분해효과)

  • Cadusafos, an organophosphorus insecticide, has been commonly used against various pests worldwide. Organophosphorus pesticides have shorter half-lives and lower toxicities than organochlorine pesticides. However, excessive use of Cadusafos can increase pest resistance and issues with acetylcholine biomagnification, potentially resulting in human toxicity. In this study, we investigated the effect of a Cadusafos-degrading microbial agent (CDMA) prepared using Sphingobium sp. Cam5-1, which was previously reported to effectively degrade residual Cadusafos in soil. Experiments were conducted under both controlled laboratory and greenhouse field conditions. Under laboratory conditions, CDMA (106 cfu/g soil application rate) decomposed 97% of Cadusafos in the soil in the untreated control after 21 days. Additionally, when CDMA (106 cfu/g soil) was mixed with quicklime, 99% of Cadusafos was decomposed within 3 days. Under greenhouse field conditions, the combined effect of CDMA (106 cfu/g soil) and quicklime was not observed. However, CDMA (106 cfu/g soil) application alone was capable of decomposing 91% of Cadusafos after 3 days. These results indicate that CDMA can effectively decompose high residual levels of Cadusafos in soils under field conditions using a low inoculum rate.

Behavior of Intrinsic Viscosity and Moisture Content of Antistatic Polyethyleneterephthalate by Thermal Stabilizer (열안정제에 의한 제전성 폴리에틸렌테레프탈레이트의 고유점도 및 수분율의 거동 변화)

  • Moisture content and intrinsic viscosity of antistatic polyethyleneterephthalate(PET) depending on the thermal stabilizer content was studied. The terminal moisture content of antistatic PET was a function of drying temperature rather than drying time. Intrinsic viscosity drop of antistatic PET after melt spinning increased with increasing moisture content of it. After melt spinning, intrinsic viscosity of antistatic PET was decreased due to the thermal degradation of polyoxyalkyleneglycol(POAG) component of antistatic agent. Triphenylphosphate(TPP) was more effective as a thermal stabilizer than trimethylphosphate(TMP). A little intrinsic viscosity drop after melt spinning was found in PET containing 300 ppm of TPP as a thermal stabilizer.

Isolation and its effect of a second organism for single cell protein(SCP) production (세균 단세포단백질(SCP) 생산을 위한 보조균주의 분리와 그 효과)

  • Experiments were carried out to find the possibility of an economic production of single cell protein(SCP) in mixed culture by Cellulomonas sp. KL-6 and a second organism. The second organism, strain LI-10, was isolated from the large intestines of a mouse. 1. When these strains were mixed, cell growth and carboxymethyl cellulase (CMCase) activity were increased to about 63% and 161%, respectively compared with that of single culture of strain KL-6. We found the mixed culture as a proper method of degradation of cellulose in our study. 2. Strain LI-10 was identified as E. coli. 3. This strain produced trace amounts of cellobiose, but glucose was not found in detectable amounts in the filter paper(FP) medium. 4. $CaCO_3$ injected in the medium at the ratio of 0.1% not only enhanced cell growth but also was effective as an acid neutralizing agent. 5. When this organism was cultured under the optimal medium (glucose 0.1%, $NH_4Cl$ 0.1%, yeast extract 2.0%, $KH_2PO_4$ 0.1%, KCl 0.05%, pH 7.2 and a temperature 30$\circ$C) for 5 days, a cell mass produced 1.18 g/l. The results showed the increase of cell mass up to 300% compared to 0.28 g/l produced in CMC medium.

Inhibition of the Induction of Nitric Oxide Synthase by Kobusin

  • We isolated a lignan, kobusin from Geranium thunbergii and studied its effect on the expression of inducible nitric oxide synthase (iNOS) gene in a monocyte/macrophage cell line, RAW264.7 cells. Kobusin inhibited lipopolysaccharide (LPS)-stimulated NO production and the expression of iNOS in a concentration-dependent manner. To identify the mechanistic basis for its inhibition of iNOS induction, we examined the effect of kobusin on both the luciferase reporter activity using $NF-{\kappa}B$ minimal promoter and the nuclear translocation of p65. Kobusin suppressed the reporter gene activity and the LPS-induced movement of p65 in to nucleus. $NF-{\kappa}B$ activation is controlled by the phosphorylation and subsequent degradation of $I-{\kappa}B{\alpha}$, and in the present study, we found that $I-{\kappa}B{\alpha}$ phosphorylation was also inhibited by kobusin. Our findings indicate that kobusin may provide a developmental basis for an agent against inflammatory diseases.

Photocatalytic Degradation of Benzene in the Gas Phase using TiO2 Coated on Ceramic and Glass Beads (세라믹과 유리에 코팅한 TiO2 광촉매를 이용한 가스상 벤젠의 제거)

  • TiO$_2$ sol was prepared by sol-gel method, and this sol was coated in ceramic and glass bead by dip-coating method. The coated catalyst was applied to degrade benzene in the gas phase by exposing to UV -lamp (365 nm) in a batch reactor. The removal efficiency of the benzene was compared by changing various conditions such as the kind of chemical additives, the coating beads (ceramic and glass), solution pH, the initial concentration of TiO$_2$ sol, UV intensity, and benzene concentration. The physical structure of TiO$_2$ sol used in this study was found to be pu-rely anatase type from XRD analysis. The results showed that ceramic bead was effective as the coating agent rath-er than glass bead. The significant change in the benzene removal efficiency of benzene did not occur with chang-ing coating frequency and the initial concentration of TiO$_2$ sol. The removal efficiency of benzene increased with increasing UV intensity, and with acidic treatment of TiO$_2$-coated ceramic bead.

The Anticoagulant Fraction from the Leaves of Diospyros Kaki L. Has an Antithrombotic Activity

  • The leaves of Persimmon (Diospyros kaki L.) has long been used for tea in Korea since it was thought to be effective against hypertension. An anticoagulant fraction was purified through gel filtration G-100, hydrophobic, gel filtration G-150, and FPLC, Phenyl superpose column chromatographies. The purified fraction was homogenous and its Mr was estimated 10,000 Da by gel filtration and SDS-PAGE. The purified fraction was sensitive to treatment of subtilisin B, but not to heat and its activity was not changed after periodate oxidation, indicating that the activity was not due to carbohydrates. It delayed thrombin time (TT), activated partial thromboplastin time (APTT), and prothrombin time (PT) using human plasma. TT was more sensitive than APTT and PT, suggesting that the anticoagulant activity may be caused by a degradation or a defect of fibrin or thrombin. It did not cause the hydrolysis of fibrin after incubation. However, it inhibited thrombin-catalyzed fibrin formation with a competitive inhibition pattern. These results indicate that it may be an antithrombotic agent and that it is bound to fibrinogen binding sites of thrombin.

The investigation of conservation methods and destruction regions of papers under the various temperature and humidity changes (온.습도 변화에 따른 양지의 손상원인 및 보존방안 연구)

  • To examine the method for preservation of papers, 9 kinds of papers were purchased and pretreated for 6 months with various artificial conditions which were controlled as 3 temperature classes($20^{\circ}C$, $80^{\circ}C$, $-20^{\circ}C$&$20^{\circ}C$) and 4 humidity classes(40%, 60%, 80%, 100%).Whiteness, acidity and folding endurance were measured to examine the state of papers pretreated under each condition. From the resultexamining the state, all the papers were best condition at low temperature($20^{\circ}C$) and low humidity(40%) and damaged at high temperature($80^{\circ}C$) and high humidity(100%).The values of whiteness, acidity, and folding endurance were measure din relation with deterioration of papers. According to damage of papers caused by the chemical agent, papers became to be acidic, increased color difference and reduced the force of folding endurance. The effect of change of temperature($-20^{\circ}C$&$20^{\circ}C$) on degradation of papers was not well acquired. Because the period of investigation was not long enough to acquire the data. So the more studies are needed to find the cause of damage of papers.

Automatic Electronic Cleansing in Computed Tomography Colonography Images using Domain Knowledge

  • Electronic cleansing is an image post processing technique in which the tagged colonic content is subtracted from colon using CTC images. There are post processing artefacts, like: 1) soft tissue degradation; 2) incomplete cleansing; 3) misclassification of polyp due to pseudo enhanced voxels; and 4) pseudo soft tissue structures. The objective of the study was to subtract the tagged colonic content without losing the soft tissue structures. This paper proposes a novel adaptive method to solve the first three problems using a multi-step algorithm. It uses a new edge model-based method which involves colon segmentation, priori information of Hounsfield units (HU) of different colonic contents at specific tube voltages, subtracting the tagging materials, restoring the soft tissue structures based on selective HU, removing boundary between air-contrast, and applying a filter to clean minute particles due to improperly tagged endoluminal fluids which appear as noise. The main finding of the study was submerged soft tissue structures were absolutely preserved and the pseudo enhanced intensities were corrected without any artifact. The method was implemented with multithreading for parallel processing in a high performance computer. The technique was applied on a fecal tagged dataset (30 patients) where the tagging agent was not completely removed from colon. The results were then qualitatively validated by radiologists for any image processing artifacts.

Complete genome sequence of Bacillus thuringiensis C25, a potential biocontrol agent for sclerotia-forming fungal phytopathogens (생물학적방제 효과가 뛰어난 Bacillus thuringiensis C25 균주의 유전체 분석)

  • We here provide the complete genome sequence of Bacillus thuringiensis C25, the strain showing antagonistic effects on fungal phytopathogens. The genome comprised of 5,308,062 bp with 35.32% G+C content of a circular chromosome and a plasmid containing 308,946 bp with 32.23% G+C content. The chromosome and plasmid genome included 5,683 protein coding DNA sequences, 107 tRNA and 42 rRNA genes.

YAC tripeptide of epidermal growth factor promotes the proliferation of HaCaT keratinocytes through activation of EGFR

  • Epidermal growth factor (EGF) is known to play key roles in skin regeneration and wound-healing. Here, we demonstrate that Pep2-YAC, a tripeptide covering residues 29-31 in the B loop of EGF, promotes the proliferation of HaCaT keratinocytes with activity comparable to EGF. The treatment of HaCaT cells with Pep2-YAC induced phosphorylation, internalization, and degradation of EGFR and organization of signaling complexes, which consist of Grb2, Gab1, SHP2, and PI3K. In addition, it stimulated the phosphorylation of ERK1/2 at Thr 202/Tyr 204 and of Akt1 at Ser 473 and the nuclear translocation of EGFR, STAT3, c-Jun, and c-Fos. These results suggest that Pep2-YAC may be useful as a therapeutic agent for skin regeneration and wound-healing as an EGFR agonist.
