Acetaminophen (APAP) overdose is known to cause severe hepatotoxicity mainly through the depletion of glutathione. In this study, we compared the cytotoxic effects of APAP on both a normal murine hepatic cell line, BNL CL.2, and its SV40-transformed cell line, BNL SV A.8. Gene expression profiles for APAP-treated cells were also obtained using microarray and analyzed to identify differences in genes or profiles that may explain the differences of susceptibility to APAP in these cell lines. These two cell lines exhibited different susceptibilities to APAP (0-$5,000{\mu}M$); BNL SV A.8 cells were more susceptible to APAP treatment compared to BNL CL.2 cells. A dose of $625{\mu}M$ APAP, which produced significant differences in cytotoxicity in these cell lines, was tested. Microarray analysis was performed to identify significant differentially expressed genes (DEGs) irrespective of APAP treatment. Genes up-regulated in BNL SV A.8 cells were associated with immune response, defense response, and apoptosis, while down-regulated genes were associated with catalytic activity, cell adhesion and the cytochrome P450 family. Consistent with the cytotoxicity data, no significant DEGs were found in BNL CL.2 cells after treatment with $625{\mu}M$ APAP, while cell cycle arrest and apoptosis-related genes were up-regulated in BNL SV A.8 cells. Based on the significant fold-changes in their expression, a genes were selected and their expressions were confirmed by quantitative real-time RT-PCR; there was a high correlation between them. These results suggest that gene expression profiles may provide a useful method for evaluating drug sensitivity of cell lines and eliciting the underlying molecular mechanism. We further compared the genes identified from our current in vitro studies to the genes previously identified in our lab as regulated by APAP in both C57BL/6 and ICR mice in vivo. We found that a few genes are regulated in a similar pattern both in vivo and in vitro. These genes might be useful to develop as in vitro biomarkers for predicting in vivo hepatotoxicity. Based on our results, we suggest that gene expression profiles may provide useful information for elucidating the underlying molecular mechanisms of drug susceptibility and for evaluating drug sensitivity in vitro for extrapolation to in vivo.
Human skin is the first line of defense for the protection of the internal organs of the body from different stimuli. Ultraviolet B (UVB) irradiation induces skin damage and inflammation through the secretion of various cytokines, which are immune regulators produced by cells. To prevent the initiation of skin inflammation, keratinocytes that have been irreversibly damaged by radiation must be removed through the apoptotic mechanism. Ixeris dentata (family: Asteraceae) is a perennial medicinal herb indigenous to Korea. It has been used in Korea, China, and Japan to treat in digestion, pneumonia, diabetes, hepatitis, and tumors. To gain insight into the anti-inflammatory effects of I. dentata, we examined its influence on UVB-induced pro-inflammatory cytokine production in human keratinocytes (HaCaT cells), by observing cells that were stimulated with UVB in the presence or absence of I. dentata. In the present study, pro-inflammatory cytokine production was determined by performing enzyme-linked immunosorbent assay, reverse transcription polymerase chain reaction, and western blot analysis to measure the activation of mitogen-activated protein kinase (MAPKs). I. dentata inhibited UVBinduced production of the pro-inflammatory cytokine interleukin (IL)-6 in a dose-dependent manner. Further, I. dentata inhibited the UVB-induced expression of cyclooxygenase (COX)-2. Furthermore, I. dentata inhibited the phosphorylation of c-Jun NH2-terminal kinase and p38 MAPKs, suggesting that it inhibits the secretion of the pro-inflammatory cytokines IL-6 and IL-8, and COX-2 expression, by blocking MAPK phosphorylation. These results suggest that I. dentate can potentially protect against UVB-induced skin inflammation.
친환경 고체산화제 GDN (guanidine dinitramide, $NH_2C(NH_2)NH_2N(NO_2)_2$)을 ADN (ammonium dinitramide, $NH_4N(NO_2)_2$)과 구아니딘 카보네이트(guanidine carbonate, $NH_2C(=NH)NH_2{\cdot}1/2H_2CO_3$)를 이용하여 부산물을 최소화시킨 고순도, 고수율로 합성하였으며, 이때 결정화 용매와 합성 과정에 따라 ${\alpha}$-form과 ${\beta}$-form을 각각 얻었다. 이들의 화학적 조성과 함량은 동일하지만, 구조적인 형태가 다르다는 것을 Raman-IR과 TGA-DSC 분석을 통해 관찰하였다. 특히 열특성(TGA-DSC)에 있어서 ${\alpha}$-form과 ${\beta}$-form 결정은 매우 큰 차이를 보였다. 흡열온도는 $100^{\circ}C$ 부근으로 유사한데 비하여, 발열최고온도에서는 ${\alpha}$-form은 $155.7^{\circ}C$이었으며, ${\beta}$-form은 $191.6^{\circ}C$로 나타났다. 또한 발열량도 ${\alpha}$-form은 536.4 J/g이었으나, ${\beta}$-form은 1310 J/g으로 2.5배 가까이 증가하는 형태를 나타냈다. 특히 열분해되는 패턴이 ${\alpha}$-form은 1단계 분해과정을 거치면서 급격히 질량이 감소하는 반면, ${\beta}$-form은 2단계 분해과정을 보이며 서서히 질량이 감소하는 특성이 관찰되었다.
To perform the engagement level simulation between the underwater vehicle model and the surface model those are constituted with various systems/ sub-systems, we implemented four different federates as a federation according to the IEEE 1516 HLA (High Level Architecture) protocol that is the international standard in the distributed simulation. Those are CFCS (Command and Fire Control System) federate, motion federate, external entities (torpedos, countermeasure and surfaceship) federate, and visualization federate that interacts with OSG (Open Scene Graph)-based visualization rendering module. In this paper, we present the detailed method about the model constitution for discrete event simulation in the distributed environment. For the sake of this purpose, we introduce the DEVS (Discrete Event System Specification)-HLA-based modeling method of the CFCS federate that reflects not only the interations between models, but also commands from user and tactics manager that is separated from the model. The CFCS federate makes decisions in various missions such as the normal diving, the barrier misision, the target motion analysis, the torpedo launch, and the torpedo evasion. In the perspective of DEVS modeling, the CFCS federate is the coupled model that has the tactical data process model, command model and fire control model as an atomic model. The message passing and time synchronization with other three federates are settled by the $m\ddot{a}k$ RTI (Runtime Infrastructure) that supports IEEE 1516. In this paper, we provides the detailed modeling method of the complicated model that has hierarchical relationship such as the CFCS system in the submarine and that satisfies both of DEVS modeling method for the discrete event simulation and HLA modeling method for the distributed simulation.
Background: Rhizobacteria play an important role in plant defense and could be promising sources of biocontrol agents. This study aimed to screen antagonistic bacteria and develop a biocontrol system for root rot complex of Panax notoginseng. Methods: Pure-culture methods were used to isolate bacteria from the rhizosphere soil of notoginseng plants. The identification of isolates was based on the analysis of 16S ribosomal RNA (rRNA) sequences. Results: A total of 279 bacteria were obtained from rhizosphere soils of healthy and root-rot notoginseng plants, and uncultivated soil. Among all the isolates, 88 showed antagonistic activity to at least one of three phytopathogenic fungi, Fusarium oxysporum, Fusarium solani, and Phoma herbarum mainly causing root rot disease of P. notoginseng. Based on the 16S rRNA sequencing, the antagonistic bacteria were characterized into four clusters, Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetesi. The genus Bacillus was the most frequently isolated, and Bacillus siamensis (Hs02), Bacillus atrophaeus (Hs09) showed strong antagonistic activity to the three pathogens. The distribution pattern differed in soil types, genera Achromobacter, Acidovorax, Brevibacterium, Brevundimonas, Flavimonas, and Streptomyces were only found in rhizosphere of healthy plants, while Delftia, Leclercia, Brevibacillus, Microbacterium, Pantoea, Rhizobium, and Stenotrophomonas only exist in soil of diseased plant, and Acinetobacter only exist in uncultivated soil. Conclusion: The results suggest that diverse bacteria exist in the P. notoginseng rhizosphere soil, with differences in community in the same field, and antagonistic isolates may be good potential biological control agent for the notoginseng root-rot diseases caused by F. oxysporum, Fusarium solani, and Panax herbarum.
괄목할만한 과학기술성과는 이종분야간 초융합 또는 융합의 융합에서 주로 나타나고 있으며, 기술경쟁국들은 고위험 고수익 연구생태계를 추진하고 있다. 구글은 실패를 두려워하지 않는 관점에서 수많은 새로운 도전을 수행하고 있고, 미국 고등연구계획국(DARPA)의 혁신적인 연구추진은 지금까지 인터넷, GPS, 반도체, 마우스, 자율주행차, 드론 등과 같은 획기적인 성과물을 만들었다. 중국은 달기지 개척, 세계 최대 규모의 핵융합 에너지와 초대형 입자가속기 프로젝트를 추진 중이며, 일본도 '문샷형 기술개발연구제도'를 착수하여 파괴적 혁신창출을 추진하려 한다. 우리나라도 글로벌 과학난제의 도전과 해결을 위해 신규 연구프로그램을 준비하고 있다. 따라서 지질자원기술 연구자가 중심이 되어 도전하고 해결하며 공감할 수 있는 과학난제의 주제 발굴과 이들 주제에 관한 사전연구 등이 필요하다. 이를 위해서는 신진연구자 중심의 장기 창의연구사업, 우수한 과제책임자 선정, 선행연구나 참고문헌도 없는 획기적 주제 도출, 과제제안절차 간소화, 아이디어 중점 선정시스템 혁신, 컨퍼런스 형태의 집단지성에 의한 결과평가 등이 필요하다고 사료된다.
결정화/응집(drowning-out/agglomeration, D/A) 공정을 이용하여 평균 입도 $550{\mu}m$인 구형 Al/RDX/AP 에너지 복합체를 제조하였다. SEM과 X-선 분광분석을 이용해 복합체의 표면 구조와 Al의 분포를 분석하였다. 열분석 결과 D/A 공정에 의해 제조된 Al/RDX/AP 복합체는 물리적 혼합에 의한 복합체와 비교하여 분해 개시 온도가 약 $50^{\circ}C$ 정도 낮아졌으며, 동시에 활성화 에너지의 증가에 의해 열적 안정성도 상승하는 것으로 확인되었다. AP의 1차 분해 구간에서는 물리적 혼합과 D/A 공정에 의한 복합체 모두 Prout-Tompkins 모델에 의해 잘 모사되었다. 그러나 AP의 2차 분해 구간에서는 물리적 혼합에 의해 제조된 복합체는 zero-order 모델로 해석되는 반면, D/A 공정에 의해 제조된 복합체는 contracting volume 모델로 해석됨을 알 수 있었다.
본 연구는 군의 보안문제가 국방과학기술의 발전과 더불어 사이버보안 등 기술적인 분야에 치중되어 왔던 점을 감안하여 조직문화 관점에서 군의 보안문제에 대해 접근하였다. 이러한 배경에서 선행연구를 통해 군의 보안문제에 영향을 미치는 변수를 추출 후 변수 간의 인과적 관계를 구조방정식모형 분석을 통해 검증하였다. 연구 결과의 의의는 다음과 같다. 첫째, 군 조직문화는 군인들의 가치관(국가관, 안보관, 군인정신)을 확립하는데 기여함은 물론 보안준수의지와 군 조직의 보안수준을 향상시키는데 직/간접적인 영향을 주고 있었다. 이것은 현재 군의 전반적인 조직문화가 장병들의 보안의식과 조직 보안성과 달성에 영향을 주고 있다는 점을 알려줌과 동시에 올바른 조직문화 조성을 위해 각고의 노력이 필요함을 나타낸다. 둘째, 군인의 가치관은 개인의 보안준수의지와 보안성과에 긍정적인 영향을 나타내고 있었다. 가치관은 조직문화를 이해하는 것에서 출발하며, 올바른 가치관이 형성되었을 때 조직의 보안태세 확립에 노력을 기울일 수 있다는 점을 알려준다. 셋째, 한국군의 맥락에 부합된 변수들을 추출하여 인과적 관계를 검증함으로써 연구의 완성도를 높였다는 점이다. 이러한 연구의 의의에도 표본의 대표성 확보, 보안수준 변화요인을 확인하기 위한 시계열 연구, 연구자 및 데이터의 삼각화 등은 연구의 한계이다.
스마트그리드 기술은 기존의 전력망 관리와 제어를 위해 ICT (Information and Communications Technologies)를 이용하여 전력 공급자와 소비자 간에 양방향으로 실시간 전력 정보의 교환을 통하여 에너지 효율을 극대화시키는 것을 목적으로 하고 있다. 본 논문에서는 IEEE 802.11s STDMA (Spatial Time Division Multiple Access) 기반의 다중 채널 스마트그리드의 NAN (Neighborhood Area Network) 네트워크에서 수리적 모델링에 기반한 계층 교차적 설계 기법을 이용하는 "JRS-MS" (Joint Routing and Scheduling for Multi-channel SmartGrid) 알고리즘을 제안한다. 제안 알고리즘은 다중 채널 스마트그리드 NAN 네트워크의 각 데이터 링크에서 데이터 전송량을 적절히 조절하고 동시에 플로우들 간에 간섭이 적은 고속 경로의 탐색을 수행한다. 이를 통하여 각 플로우들의 네트워크 이용률을 높여 전송률을 향상시킨다. 제안 알고리즘과 기존 제안 알고리즘인 JRS-SG (Jointly Routing and Scheduling for SmartGrid) 알고리즘 과의 비교 성능 분석을 통하여 JRS-MS 알고리즘이 다중 홉 NAN 무선 메쉬 네트워크를 경유하는 플로우들의 수가 늘어날 때 주어진 대역폭 자원을 최대로 활용하여 전송 성능을 향상 시킬 수 있음을 보였다.
바이스태틱 역합성 개구 레이다(Inverse Synthetic Aperture Radar: ISAR) 영상은 표적에 대한 2차원 산란분포를 나타내고, 이는 바이스태틱 표적식별에 이용될 수 있다. 하지만 바이스태틱 ISAR 영상은 바이스태틱 기하구조에 따라 그 산란 메커니즘이 다양하게 변화하고, 고유의 왜곡 때문에 표적의 정확한 거리-도플러 정보를 나타낼 수 없다. 따라서 이를 이용한 표적식별에서 효율적인 훈련 DB 구축은 핵심사항이 된다. 최근 모노스태틱 표적식별에서 효율적인 성능을 보였던 비행 시나리오 기반 훈련 데이터베이스(database: DB) 구축 기법을 바이스태틱 고해상도 거리측면도(High Resolution Range Profile: HRRP) 표적식별에 적용하여 바이스태틱 기하구조 내 효율적인 훈련 DB를 구축하는 연구가 수행되었고, 상기 연구는 레이다와 표적 사이의 거리가 충분히 먼 경우, 적은 양의 훈련 DB로도 높은 표적식별 성능을 획득할 수 있음을 보여주었다. 따라서 본 논문에서는 비행 시나리오 기반 훈련 DB 구축 기법을 바이스태틱 ISAR 영상 표적식별에 적용한 후, 그 성능과 효율성을 분석한다.
