Studies on the Composition of Fatty Acid and Protein in Pumpkin Seeds (호박씨의 지방산(脂肪酸) 및 단백질(蛋白質)의 조성(組成)에 관(關)한 연구(硏究))

  • Dehulled pumpkin seeds produced in Korea were defatted and their composition of fatty acid was analyzed by gas chromatography. Soluble protein and insoluble protein were isolated from the dehulled and defatted pumpkin seeds. The protein component was identified by disc-electrophoresis and the amino acid composition of the protein was analyzed. (1) The pumpkin seed contained 46.5% of fat and 25.5% of protein respectively. (2) The fatty acid of the pumpkin seed was compassed chiefly of linoleic acid (51.74%), oleic acid (22.2%), palmitic acid (18.1%) and stearic acid (7.84%) (3) The amino acid composition of the soluble and insoluble seed protein contained most of all the amino acids except for S-contained amino acids. (4) It has been identified by disc-electrophoresis that the soluble protein had 13 bands and the insoluble protein had 4 bands.

Effects of Defatted Safflower Seed Powder on Intestinal Physiology and Fecal Short-Chain Fatty Acids in Overiectomized Female Rats Fed High Cholesterol Diets (고콜레스테롤식에 홍화박의 첨가가 난소절제 흰쥐의 장 생리기능과 분변 Short-Chain Fatty Acid 함량에 미치는 효과)

  • Much attention has been given to the role of safflower (Carthamus tinctorius L.) seed in preventing and treating osteroporosis recently. Although numerous studies were done on effects of safflower oil, no attention was given to the role of dietary fiber of safflower seeds. This study was aimed at investigating physiological significance of defatted safflower seed as a source of dietary fiber. Sprague Dawley female rats were assigned one of 5 groups: ovariectomized control (Ovx-control) group and ovariectomized rats 15% (Ovx-ss15) and 30% defatted safflower seed (Ovx-SS30), sham-operated (Sham) group and a normal group. Cholesterol was supplemented to all diets at 0.5% except the normal diet. Ovariectomized rats were pair-fed isocalorically to the Sham group. Ovariectomy caused heavier body weight, but feeding 30% safflower seed brought back to the level of Sham group. Activities of disaccharidases of jejunal mucosa were significantly lowered in Ovx-control group compared to those of Sham, and supplementation of safflower seed tended to increase the activities. Fecal weight of Ovx-SS15 and Ovx-SS30 were almost twice as those of Ovx-control Sham. Propionic acid and butyric acid concentrations per g of feces of Ovx-SS15 and daily excretion of these fatty acids were significantly increased as compared to those of Sham and Ovx-control. In concentration defatted safflower seed supplementation significantly increased fecal bulk and short chain fatty acid production in large intestine of rats.

Anti-oxidant activities of ethanol extract and fractions from defatted Camellia japonica L. seeds (동백 유박 에탄올추출물 및 분획물의 항산화 활성)

  • The aim of this study was to investigate in vitro antioxidant activities of defatted Camellia japonica L. seeds (DCJS). The DCJS were extracted using ethanol and then fractionated with butanol (BuOH), ethyl acetate (EtOAc), chloroform, and hexane. To evaluate antioxidant activity of extract and fractions from DCJS, we investigated free radical scavenging activities such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), hydroxyl radical (OH), and superoxide anion (O2-) radicals. The five extract and fractions of DCJS dose-dependently increased DPPH, ABTS+ and O2- radical scavenging activities. The BuOH fraction of DCJS showed the highest free radical scavenging activities among other extract and fractions. The contents of total polyphenol and flavonoid in BuOH fraction of DCJS were 23.26 mg GAE/g and 32.39 mg QE/g, respectively. The polyphenol and flavonoids contents of BuOH fraction has highest than other extract and fractions. In addition, BuOH and EtOAc fraction of DCJS contained 102.37 and 165.05 ㎍/g of camelliaside B, respectively. Therefore, DCJS has higher antioxidant activity and may be useful as a natural antioxidant material.

Isolation of Lipoxyeenase Isozymes from Soybean Seeds (대두 Lipoxygenase 이성효소의 분리)

  • Soybean lipoxyeenase isozymes were isolated from acetone-defatted soybean seeds(Glycine max [L.] Merr. variety AmSoy) by ammonium sulfate fractionation, eel filtration, and ion exchange chromatoeraphy. The final preparation of lipoxygenase-1 and -2 obtained was 19- and 32-fold purified, respectively, to the crude extract. But a considerable loss of total enzyme activity occurred during purification. On 7% polyacrylamide gel electrophosis at pH 9.0, employing lipoxigenase specific staining technique, lipoxyeenase-1, -2, and -3 showed distinctive Rf values of 0.38, 0.29, and 0.33, respectively.

Amygdalin Contents in Peaches at Different Fruit Development Stages

  • Amygdalin contents of the seeds, endocarps, and mesocarps from three peach cultivars (i.e., Stone Peach, Hikawa Hakuho, and Bakhyang) were measured at three stages of fruit development (stone-hardening, fruit enlargement, and ripening). The peach samples were dried and defatted with a Soxhlet apparatus, reflux extracted with methanol, and analyzed using reverse phase high-performance liquid chromatography. During all fruit development stages, the amygdalin contents in the seeds were higher than those in the endocarps and mesocarps. The amygdalin contents of the Stone Peach were comparatively higher than the Hikawa Hakuho and Bakhyang (P<0.05). Further, the amygdalin contents during ripening were very low or not detected. Overall, the amygdalin contents of the three peach cultivar samples (seed, endocarp, and mesocarp) increased until the fruit enlargement stage and either remained constant or decreased during ripening.

Enzymatic preparation and antioxidant activities of protein hydrolysates from hemp (Cannabis sativa L.) seeds

  • Hemp (Cannabis sativa L.) seeds have recently been attracting attention as a new high-value-added food material owing to their excellent nutritional properties, and research on the development of functional food materials using hemp seeds is actively progressing. This study aimed to evaluate the antioxidant properties of hemp seed protein hydrolysates. Protein hydrolysates were prepared from defatted hemp seed powder (HS) by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay and SDS-PAGE analysis revealed that HS showed a high degree of hydrolysis after treatment with each enzyme except papain. The total polyphenol content of the protein hydrolysates (<3 kDa) and the RC50 values obtained from two different antioxidant tests showed that alcalase hydrolysate (HSA) had a relatively high level of antioxidant capacity. In addition, treatment with HSA (25-100 ㎍/mL) significantly inhibited linoleic acid peroxidation. These results suggest that hemp seed protein hydrolysates are potential sources of natural antioxidants. Future studies will focus on the identification of active peptides from HSA.

Selective Extraction of Phospholipids from Soybeans with Mixture of Supercritical Carbondioxide and Ethanol (초임계 $CO_2$와 에탄올 혼합물에 의한 대두인지질의 선택적 추출)

  • Supercritical carbondioxide is very effective in removing oils from a variety of seed matrices, devoid of any appreciable amount of phospholipid content. However, the limited solubility of phosphalipids in supercritical carbondioxide leaves behind a potentially valuable by-product in spent seed matrix. Any phospholipid extraction process from the spent matrix must maintain the structure and the quality of phospholipids and must be compatible with the end use of the seed protein meal an animal feed or for human consumption. An initial supercritical carbondioxide extraction of soybean flakes was performed at 32 MPa and $80^{\circ}C$ to extract the oils, leaving the phospholipids in the deflatted soybean flakes, A second step was performed on the defatted soybean flake using $X_{eth}$=0.10, Varying the pressure from 175 MPa to 70.6 MPa and temperature from $60^{\circ}C$ to $80^{\circ}C$. For all supercritical carbon dioxide/ethanol mixture extractions, a fraction rich in phospholipids was obtained. The fractions extracted from defatted soybean flakes were dried and them redissolved in chloroform before HPLC-ELSD analysis. Quantitative and qualitative analysis of phospholipids on soybean seeds, defatted soybean flake, and different extracted phospholipid fractions was carried out, to ascertain the effect of extraction pressure and temperature.

Antioxidant Properties and Quantification of Phenolic Compounds from Safflower (Carthamus tinctorius L.) Seeds

  • The antioxidant properties of twelve phenolic compounds, including matairesinol 4'-O-$\beta$-D-glucoside, 8'-hydroxyarctigenin 4'-O-$\beta$-D-glucoside, matairesinol, 8'-hydroxyarctigenin, N-feruloylserotonin 5-O-$\beta$-D-glucoside, N-(p-coumaroyl)-serotonin-5-O-$\beta$-D-glucoside, N-feruloylserotonin, N-(p-coumaroyl)serotonin, luteolin 7-O-$\beta$-D-glucoside, luteolin, acacetin 7-O-$\beta$-glucuronide, and acacetin, isolated from defatted safflower (Carthamus tinctorius L.) seeds were evaluated with regard to the DPPH, superoxide and hydroxyl radicals. Additionally, levels of phenolic compounds were determined by HPLC in two cultivars of safflower seeds. Among them, four serotonin derivatives showed potent DPPH ($IC_{50}=10.83-21.75\;{\mu}M$) and hydroxyl ($IC_{50}=75.93-374.63\;{\mu}M$) radical scavenging activities, and their activities were significantly stronger than that of ${\alpha}-tocopherol$. Four flavonoids ($IC_{50}=170.65-275.83\;{\mu}M$) and four lignans ($IC_{50}=114.22-406.10\;{\mu}M$) exhibited significant superoxide and hydroxyl radical scavenging activities, respectively, whereas these compounds contained less activity toward the DPPH and hydroxyl radicals than serotonin derivatives. The levels of serotonin derivatives, lignans and flavonoids in safflower seeds of two cultivars ranged from 49.30 to 260.40, 3.72 to 158.90, and 11.72 to 214.97 mg% (dry base), respectively. Of the two cultivars, 'Cheongsu' had somewthat higher concentrations of phenolic compounds than 'Uisan'. These results suggest that phenolic compounds in safflower seeds may playa role as protective phytochemical antioxidants against reactive oxygen-mediated pathological diseases.

An Evaluation of the Anti-oxidant Activity of Fermented Defatted Sesame Seeds (참깨탈지박 발효 추출물의 항산화 활성 평가)

  • This study was conducted to investigate the total lignin content and anti-oxidant activity in extracts of defatted sesame seeds (DSS) fermented with 15 strains of Bacillus subtilis. The anti-oxidant activities of DSS were analyzed both before and after fermentation. The total lignan content of the DSS extracts fermented with BCH3678 (1,613.8 mg/l) and BCH3572 (1,599.5 mg/l) were relatively high compared to other strains. Anti-oxidant activity was determined according to phenolic compound and flavonoid content and DPPH radical scavenging rate; the highest total phenolic compound content was provided by the DSS with SRCM103716 at 2,803.3 mg GAE/g which returned total flavonoid content of 1,553.1 mg/g as strong correlation of its anti-oxidant activity. The DSS extract fermented by SRCM103716 at 37℃ for 24 hr showed the highest DPPH scavenging rate at 66.5%. The fermented DSS extracts, regardless of strain, demonstrated higher anti-oxidative activity than the unfermented control, and these results suggest that such extracts could be useful as a potential source of bioactive compounds.

Contents of Polyphenols and Limonoids in Citron (Citrus junos Sieb. ex Tanaka) Seed Extracts and Their Antioxidant Properties

  • Contents of phytochemicals in citron (Citrus junos Sieb. ex Tanaka) seeds and their effects on antioxidant activities were investigated. Methanol extract of defatted citron seeds contained the highest amounts of total polyphenols followed by 70% ethanol and water extracts. Neohesperidin was the most predominant citrus flavanones in these extracts. The highest amounts of limonoids were found in methanol extract, and this extract was the most efficient in scavenging both DPPH and ABTS radicals. All 3 extracts also exhibited good antioxidant activities against attack of linoleate free radicals on $\beta$-carotene. When methanol extract was sequentially fractionated into ether, ethyl acetate, n-butanol, and water fractions, butanol fraction contained the highest amounts of polyphenols otherwise most of limonoids were concentrated in ethyl acetate fraction. A positive relationship between radical scavenging activities and total polyphenol contents in fractions was observed while antioxidant activity on $\beta$-carotene seemed more related with contents of limonoids and other hydrophobic polyphenols.
