Coumarins and a Pyrimidine from Angelica gigas Roots

  • Five coumarins and a pyrimidine were isolated from the roots of Angelica gigas. Their structures were elucidated as bergapten (1), decursinol angelate (2), decursin (3), nodakenetin (4), uracil (5) and nodakenin (6) by spectral analysis. Among them, bergapten (1) and uracil (5) were isolated for the first time from this plant part.

Quantitative Analysis of Coumarins from Angelica gigas Using $^1H$-NMR

  • $^1H$-Nuclear magnetic resonance (NMR) spectrometry was applied to the quantitative analysis of coumarins in the roots of Angelica gigas without any chromatographic purification. The experiment was performed by the analysis of each singlet germinal methyl, which was well separated in the range of 1.0-2.0 ppm in the $^1H$-NMR spectrum. The quantity of the compounds was calculated by the ratio of the intensity of each compound to the known amount of internal standard (dimethyl terephthalate). These results were compared with the conventional gas chromatography (GC) method. The contents of decursin and decursinol angelate in A. gigas were determined $1.98{\pm}0.07$, $1.13{\pm}0.08%$ in quantitative $^1H$-NMR method and $2.06{\pm}0.24$, $1.17{\pm}0.24%$ in GC method, respectively. The advantages of quantitative $^1H$-NMR analysis are that can be analyzed to identify and quantify, and no reference compounds required for calibration curves. Besides, it allows rapid and simple quantification for coumarins with an analysis time for only 10 min without any preprocessing.

Anti-inflammatory Activities of Coumarins Isolated from Angelica gigas Nakai on LPS-stimulated RAW 264.7 Cells

  • Five kinds of coumarin compounds were successfully purified from Angelica gigas Nakai by using recyclingpreparative HPLC and identified as decursin (1), decursinol angelate (2), 7-demethylsuberosine (3), marmesin (4), and decursinol (5) by NMR analyses. None of the purified compounds in ethanol showed DPPH radical scavenging activity, while the A. gigas extract (AGNEX) displayed a significant level of activity. Interestingly, compounds 3 in phosphate buffered saline (PBS) showed good $ABTS^+$ radical scavenging activity ($IC_{50}=8.1{\mu}g$/mL) as did compounds 4 and 5. The anti-inflammatory activities of the purified compounds were evaluated and compared using the NO concentration assay and western blot analysis on LPS-stimulated RAW 264.7 cells. NO production was significantly suppressed by all the compounds in a dose-dependent manner among which compounds 1, 2, and 3 showed very good activities with $IC_{50}$ values of 7.4, 6.5, and $7.6{\mu}g$/mL, respectively. Treatment with compounds 1-5 effectively suppressed the expression levels of iNOS, IL-1$\beta$, and COX-2, which are responsible for promoting the inflammatory process. Thus, the ethanol extract and coumarin compounds of A. gigas Nakai hold promise for use as potential anti-inflammatory agents.

압출성형 공정에 따른 참당귀 추출물의 이화화적 특성 (Physiochemical Characteristics of Extruded Angelica gigas Nakai Depending on the Extrusion Processing Parameter)

  • This study was designed to maximize extract yields of functional components in Angelica gigas Nakai by extrusion process. In addition, the optimizing condition of the extrusion process was established to increase the extraction of the functional components in A. gigas. The total polyphenol was increased by 40% compared to a control. And the optimizing condition was that the screw speed was 62.76 rpm, the amount of A. gigas powder was 34.79 kg/h, and the amount of water was 4.44 kg/h. The result of antioxidant activities of A. gigas extrusion, the inhibition of oxidization had the effect of 10.29 ~ 14.59% compared to a control. The content of decursin and decursinol angelate, which were index components, was 6.37%; it was increased by 16.64% compared to a control (5.31%) and showed a significantly difference (p < 0.05). And the optimizing condition was the screw speed 93.71 rpm, the amount of A. gigas powder was 28.67 kg/h, and the amount of water was 9.9 kg/h.

친환경 유기질 비료 시용이 참당귀의 생육과 수량에 미치는 영향 (Effects of Eco-Friendly Organic Fertilizer on Growth and Yield of Angelica gigas Nakai)

  • This experiment was carried out to investigate the effect of several organic compost on the growth and root yield of Angelica gigas Nakai with organic cultivation. After fertilizing the soil with organic fertilizer, the dry weight of liming fertilizer showed a slow change, while microorganism fertilizer decreased about 29% until 20 day after fertilizing. At 110 days after fertilizing, microorganism fertilizer decreased more than liming fertilizer. Liming fertilizer decomposed slowly, while microorganism fertilizer decomposed early on rapidly but gently after 20 days in decomposed rate of organic fertilizer. Dried root yields per 10a of A. gigas were not significance between 277.6kg in conventional fertilizer and 277.7kg, 280.5kg in N 1.5, N 2.0 times of microorganism fertilizer. Decursin and decursinol angelate contents in A. gigas were 9.08 ~ 9.07% from N 1.0 and N 1.5 times in liming fertilizer, and 7.94 ~ 8.12% from N 1.5 times and N 2.0 times in microorganism fertilizer, compared to 7.31% of conventional treatment.

참당귀 추출분말이 Monosodium Iodoacetate로 유발된 흰쥐의 골관절염에 대한 효과 (Effect of Angelica gigas extract powder on progress of osteoarthritis induced by monosodium iodoacetate in rats)

  • 참당귀 추출물의 골관절염에 대한 억제 또는 치료효과를 알고자 monosodium iodoacetate로 유발한 흰쥐의 골관절염 모델을 사용하여 30마리 6주령 SD흰쥐를 정상군(normal), 비처리군(untreated) 및 참당귀 처리군(treated)으로 각각 10마리씩 나누어 실험하였다. 실험기간 동안 실험 개시일부터 4일 간격으로 체중 변화를 측정하였다. 3주간 참당귀 추출물 처리 후 관절연골 내의 glycosaminoglycan (GAG)의 함량 및 proteoglycan (PG) 함량을 측정 비교하였다. 그 결과, 참당귀 추출물은 MIA로 증가한 GAG를 완화시켰으며, PG의 함량을 회복시켰다. 또한, 활액에서의 TNF-a함량은 미약하게 감소하였다. 한편, 경골 관절연골을 분리하여 Safranin-O 염색으로 관절연골 상태를 확인한 결과, MIA 처리로 유발된 관절연골 소실을 억제하였다. 이 참당귀 추출물을 HPLC로 분석한 결과, 주요 성분인 decursin 및 decursinol angelate의 총 함량은 $10.5{\pm}0.2%$이었다.

Antibacterial compounds against fish pathogenic bacteria from a combined extract of Angelica gigas and Artemisia iwayomogi and their quantitative analyses

  • In the search for antibiotic alternatives from safe and effective medicinal plants against fish pathogenic bacteria, we found that a combined extract (CE) of 1:1 (w/w) ratio of Angelica gigas Nakai roots and aerial parts of Artemisia iwayomogi Kitamura showed antibacterial activity against the fish pathogenic bacteria. By antibacterial activity-guided fractionations and isolations, five compounds were isolated and identified as decursinol angelate (1), decursin (2), xanthotoxin (3), demethylsuberosin (4), and 2,4-dihydroxy-6-methoxyacetophenone (5) through spectroscopic analyses, such as nuclear magnetic resonance (NMR) and mass spectrometry (MS). Among the compounds, 1 and 2 showed the highest antibacterial activities against Streptococcus iniae and Vibrio anguillarum, showing minimum inhibitory concentrations (MICs) of 62.5-250 ㎍/mL. Compounds 3, 4, and 5 were also found to be active, with MICs of 31.25-1,000 ㎍/mL for those strains. Furthermore, active compounds, 1 and 2 in CE were simultaneously quantified using high-performance liquid chromatography-tandem MS (HPLC-MS/MS). The average contents of 1 and 2 in CE was 3.68% and 6.14%, respectively. The established method showed reliable linearity (r2 > 0.99), good precision, accuracy, and specificity with intra- and inter-day variations of < 2 % and recoveries of 90.13%-108.57%. These results may be helpful for establishing the chemical profile of CE for its commercialization as an antibiotic alternative in aquaculture.

Analysis of the Components of Guibitang and Fermented Guibi-tang and their Ability to Inhibit Angiotensin-converting Enzyme

  • Guibi-tang is a traditional medicine used for the treatment of colds. We investigated the levels of several compounds in Guibi-tang before and after fermentation with Lactobacillus and tested their ability to inhibit angiotensin-converting enzyme. Six known compounds (decursin, decursinol angelate, nodakenin, liquiritin, formononetin, and 6-gingerol) and 2 unidentified compounds were detected in Guibi-tang (GB) and fermented Guibi-tang (FGB) by an established HPLC-DAD method. The levels of the 6 known compounds were decreased after fermentation. FGB showed more potent inhibition of angiotensin-converting enzyme activity than GB. In conclusion, fermentation with Lactobacillus affects the content of several compounds in GB and improves its angiotensin-converting enzyme inhibitory activity.

Decusinol angelate inhibits UVB-induced MMP-l induction via Mitogen-activated Protein Kinase Pathway in human skin fibroblasts

  • UV-B irradiation increases the synthesis of matrixmetalloproteinase-1 (MMP-1) that degrades skin collagen in human skin. In this work, we investigated the photoprotective effect of decursinol angelate (DEA) extracted Irom Angelica gigas on human skin libroblasts. DEA inhibited UVB-induced MMP-1 induction, which was conlirmed by western blot and ELISA. We examined upstream signal transduction pathway and the action mechanism of DEA on UVB induction of MMp in human skin fibroblasts. (omitted)
