한우에서 전장의 유전체 정보를 활용한 연관불평형 및 유효집단크기 추정에 관한 연구 (Estimation of Linkage Disequilibrium and Effective Population Size using Whole Genome Single Nucleotide Polymorphisms in Hanwoo)

  • 본 연구는 한우 유전체 전장에 존재하는 고밀도 단일염기다형을 DNA chip을 이용하여 각각의 유전자형을 구명하고, 동일염색체 내에 존재하는 각 표지인자쌍의 연관불평형을 성 염색체를 제외한 모든 상염색체에서 추정하여 물리적 거리별 연관불평형의 정도를 확인하고 이러한 결과를 이용하여 한우 집단의 유효집단 크기를 추정하기 위하여 실시하였다. 한우개량사업소에서 2005년부터 2008년까지 후대검정에 공시된 후보종모우 및 후대 검정우 288두에 대해 혈액을 채취하고 Bovine SNP 50 DNA Chip을 이용하여 유전자형을 분석하였으며, 총 51,582 표지인자 중 결측률이 10% 이상인 표지인자 1개 및 다형성이 없는 표지인자 10,730개에 대해 사전제거를 실시하고 남은 40,851개의 SNP표지인자를 본 분석에 활용하였다. 연구 결과, 성 염색체를 제외한 상 염색체의 총 SNP표지인자의 길이는 2,541.6 Mb였으며, 염색체별 평균 SNP표지인자간 거리는 0.55에서 0.74로 분포하였으며, EM알고리즘을 이용하여 염색체별 연관불평형을 추정해 보았을 때, 기존의 보고된 연구와 유사하게 표지인자간 거리가 짧을수록 높게 나타나는 지수형태의 그래프를 나타냈으며, SNP표지인자간 거리에 따른 $r^2$를 보면, 0 Mb에서 0.1 Mb일 때 0.136, 0.1-0.2 Mb에서 0.06로 나타났다. Luo (1998)의 연구결과를 한우에 적용시켰을 때, 전체분산의 5%이상 설명하는 양적형질좌위 발굴을 위해서 약 2,000두의 표현형 자료가 필요할 것으로 사료되었다. 또한 한우의 세대별 유효집단 크기에 대해 추정해 본 결과, 현재 한우의 유효집단크기는 84두로 추정되었고, 지금으로부터 약 50세대 이전의 유효집단 크기는 1,150두로 추정되었다. 가축에서 인공수정이 도입(1960년대)된 이 후 개량의 가속화로 인해 한우의 유효집단 크기가 급격히 감소한 것으로 사료되었다.

(U-Th)/He 연령측정법의 원리와 응용 ((U-Th)/He Dating: Principles and Applications)

  • (U-Th)/He 연령측정법은 자연상에 존재하는 $^{238}U$, $^{235}U$$^{232}Th$이 붕괴할 때 알파 입자($^4He$ 원자)가 형성되는 현상을 이용한 연령측정법이다. 모 동위원소인 $^{238}U$, $^{235}U$$^{232}Th$과 붕괴산물인 $^4He$의 양을 측정하여 (U-Th)/He 연령을 구할 수 있다. 이렇게 형성된 $^4He$ 원자는 대부분의 지질학적 시료내에서 비교적 저온에서도 빠르게 확산되는데, 이는 $^4He$가 불활성 기체이고 다른 원소에 비해 작기 때문이다. 따라서 방사성 붕괴에 의한 $^4He$의 형성(ingrowth)과 확산에 의한 $^4He$의 방출(diffusive loss)이 지질학적 시간동안 어떻게 진행 되었느냐에 따라 현재 남아있는 $^4He$의 양이 결정된다. 예를 들어, 인회석내에서의 He 확산은 다른 광물에 비해 빨라서, 비교적 저온인 $80^{\circ}C$에서도 He이 빠르게 인회석 밖으로 방출되는 것으로 알려져 있다. 자연상의 온도조건이 약 $40^{\circ}C$ 이하로 떨어졌을 때 비로소 인회석내 He 확산이 충분히 느려져서 대부분의 He이 인회석내에 보존된다. 따라서 (U-Th)/He 연령은 시료가 $80-40^{\circ}C$를 통과한 시기를 지시한다. 자연상에서 이러한 온도범위에 해당하는 깊이를 "부분 보존대"(partial retention zone)의라 한다. 전통적으로 흔히 쓰이는 폐쇄온도(closure temperature)는 보통의 입자크기와 냉각속도에서, 인회석 경우 약 $60-70^{\circ}C$, 저어콘 및 티탄석의 경우 약 $200^{\circ}C$로 알려져 있다. 특히 인회석의 He 폐쇄온도는 다른 열역사 측정법에 비해 다소 낮기 때문에 비교적 최근의 열역사 또는 천부에서의 지각융기 현상을 기술하는데 매우 유용하게 쓰일 수 있다.

키토산, 곡물아미노산, 목초액의 엽면살포가 포도(Campbell Early)의 품질 및 저장성에 미치는 영향 (Effects of Chitosan, Grain Amino Acid and Wood Vinegar Foliar Spray on the Quality and Storability of Grapes(Campbell Early))

  • 포도 생육 중 유기농자재의 엽면살포가 포도의 품질과 저장성에 미치는 영향을 농약 처리와 비교하였다 포도의 과방중과 열과 발생률은 처리간에 유의적인 차이를 보이지 않았으나 키토산, 목초액, 곡물아미노산 처리에 의하여 과방중 300g이상과의 비율이 농약처리나 무처리에 비하여 높았다. 유기농자재를 처리한 포도의 당도와 환원당은 각각 $15.0\;^{\circ}Brix$, 13.0-14.5%로 농약처리 $14.5\;^{\circ}Brix$, 13.1%에 비하여 높았으나 유의적인 차이는 보이지 않았으며 총산함량은 키토산(0.81%)과, 목초액(0.78%) 처리가 농약(0.72%)처리에 비하여 높은 것으로 나타났다. 무기성분은 칼륨 함량이 가장 높았고 유기농자재 처리(190.0-197.2mg%)에 의하여 증가하였으며 중금속은 철 함량이 가장 높았으며 키토산 처리에 의하여 철과 아연 함량이 증가하였다. 유기농자재 처리 포도를 8주간 MA 저장했을 때 수분감모율은 0.28-0.35%로 낮은 수준이었으며, 환원당 함량은 저장 전에 비하여 감소하였으나 총산 함량은 증가하였으며 경도는 저장 4-6주까지 감소하다 이후 약간 증가하는 경향이었다. 탈립률은 곡물아미노산과 농약 처리가 각각 10.9%, 11.8%로 가장 낮았으며, 부패율은 키토산과 곡물아미노산 처리가 각각2.2 3.7%로 농약 처리 4.8%에 비하여 낮았고, 상품성은 농약 처리가 가장 양호한 것으로 나타났다. 이상의 결과 포도의 유기농재배에 있어서 병해충 방제를 목적으로 키토산, 곡물아미노산, 목초액을 엽면살포 했을 때 과실의 성분 등 품질은 농약 처리와 비교했을 때 차이가 없었으나 포도의 과방중에 의한 등급은 향상되는 것으로 나타났다. 도로터널에서 수소 연료차 수소탱크 폭발시 폭발압력에 대한 기초적 연구 (A basic study on explosion pressure of hydrogen tank for hydrogen fueled vehicles in road tunnels)

  • 수소연료는 환경오염문제를 해소하고 에너지 불균형 및 비용을 절감할 수 있다는 점에서는 화석연료를 대체하는 에너지원으로 부각되고 있다. 수소는 친환경적이나 폭발성이 강하기 때문에 수소연료차의 화재, 폭발 사고에 대한 우려가 매우 높은 실정이다. 연구결과에서 수소사고는 일반적인 화재의 경우 비교적 안전하나, 폭발이 발생하면 매우 위험한 것으로 인식되고 있다. 특히, 터널과 같은 반밀폐공간에서는 위험도가 보다 증가할 것으로 예측되기에 이에 대한 예측방법 및 대책을 마련하기 위한 연구가 수행되고 있다. 이에 본 연구에서는 터널에서 수소폭발시 안전성을 평가하기 위해서 등가 TNT모델의 적용성과 수치해석 방법에 대한 검토를 수행하였다. 6개의 등가 TNT모델과 Weyandt의 실험결과의 폭발압력을 비교·검토하여 모델의 적용성을 평가한 결과, Henrych식이 13.6%의 편차로 가장 근접하는 것으로 나타났다. 수치해석을 이용하여 수소탱크 용량(52, 72, 156 L)과 터널 단면적(40.5, 54, 72, 95 m2)이 폭발압력에 미치는 영향에 대한 검토한 결과, 터널에서 폭발 압력파는 초기에는 대기중에서와 마찬가지로 반구형 형태로 전파되나 벽체에 도달하면 반사파가 형성되며, 일정 거리 이상에서는 평면파로 변형되어 아주 완만한 감쇄율로 전파하는 것으로 나타났다. 등가 TNT모델인 Henrych식은 폭발압력이 급격하게 감소하는 구간에서는 수치해석 결과와 잘 일치하나 폭발압력파가 변형된 이후에는 큰 폭으로 과소평가하는 것으로 나타났다. 수소탱크용량이 동일한 경우에는 터널 단면적이 증가할수록 폭발압력이 감소하며, 단면적이 동일한 경우에는 수소탱크 용량이 52 L에서 156 L로 증가하면 폭발압력은 약 2.5배 정도 증가하는 것으로 나타났다. 인체에 영향을 미치는 한계거리에 대한 평가결과, 수소탱크용량이 52 L인 경우 사망에 이르는 한계거리는 약 3 m, 중상에 이르는 거리는 단면적별로 차이가 있으나 28.5~35.8 m로 나타났다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The frame and leaflet of the polymer valve were made from polyurethane, To evaluate the hemodynamic performance of the polymer valve a comparative study of flow dynamics past a polymer valve and a St. Jude Medical prosthetic valve under physiological pulsatile flow conditions in vitro was made. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. We use combination of MLP and fuzzy filter for pattern recognition to estimate the direction of a muscle and Hogan's method for the force estimation. EMG signals are acquired by using a amputation simulator and 2 dimensional joystick motion.
