The Influence of Abnormally Low Temperatures on Growth and Yield of Hot Pepper(Capsicum Annum L.) (이상저온 조건이 고추의 생육 및 수량에 미치는 영향)

  • Growth and physiological disorders caused by abnormally low temperatures were evaluated in pepper, an important field crop in Korea. In addition, the effects of chemical treatment using glutamine was verified on minimizing the damages by low temperature. The growth of pepper plants in stem length and diameter was suppressed as the temperature decreased from $25^{\circ}C$, and the suppression level was the highest for plants grown for 90 days at $20^{\circ}C$. However, root growth was not affected by the different temperatures. The number of leaf and leaf area decreased at the temperatures below $25^{\circ}C$, an optimum temperature for growth. Fresh weight and dry weight decreased for plants grown at $20^{\circ}C$. Pepper fruit yield also decreased by 11% at $20^{\circ}C$ in comparison to $25^{\circ}C$. Falling blossom rate was different depending on the growth temperature, and the rate was 27.2% at $25^{\circ}C$, 35.2% at $22.5^{\circ}C$, and 41.0% at $20^{\circ}C$, indicating that falling blossom rate increased as temperature decreased. Different growth temperatures did not affected on the level of symptom of calcium deficiency and Phytopathora blight. Falling blossom was severe at abnormally low temperature of $20^{\circ}C$, but the treatment of glutamine reduced falling blossom rate and increased the yield by 7.0% as compared to control. The optimum concentration of glutamine treatment was 10 mg/L for yields.

  • This studies by treatment of 25 individual adults of O. laevigatus and Banker plant with different number of eggs of O. laevigatus were conducted to develop new biological control of the western flower thrips (WFT) in chrysanthemum house. In this work, a treatment of 25 individual adults of O. laevigatus, Banker plant with different number of eggs of O. laevigatus and mixed treatment were applied. Initial WFT density per plant were 5 individuals. WFT density was measured every 7days after treatment for 7 weeks. Biological control of WFT by 25 individual adults of O. laevigatus and Banker plant with 25 eggs of O. laevigatus were applied 1 time and resulted in the high control value of thrips 91.8%. The population of WFT was kept under the economic threshold level (1 individual per plant). For the last three years we developed new banker plant system. This system is being developed to improve biological control of WFT on chrysanthemums.

Effective In Vitro Propagation from Pedicel Culture of Hippeastrum hybridum Hort. 'Dazzler' (아마릴리스 (Hippeastrum hybridum Hort. 'Dazzler') 소화경 배양에 의한 효율적 기내번식)

  • This study was conducted to establish the system of effective in vitro propagation by various explant sources culture of Bippeastrum hybridum Hort, 'Dazzler'. We tested the effects of optimal explant source, plant growth regulators on bulblet formation and plant regeneration. Callus was readily produced on the different tissues excised from floral buds whereas, bulbs and shoots were formed only on pedicel explants as compared with anthers, styles and ovaries. Pedicel is the best optimal explant for in vitro propagation. Two distinct pathways, organogenesis through callus and direct bulblet formation, could be recognized in pedicel culture. Up to the $80-100\%$ of bulblet formation and shoot organogenesis from the pedicel in fifteen days before anthesis were effectively induced by MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA. Plantlet regeneration was successfully achieved from pedicel-derived callus, via shoot bud induction or direct bulblet formation. The bulblets with blooming flower were produced within 2 years.

  • To increase the efficiency of the callus induction in anther culture of Schizandra chinensis, the effects of culture stage, low temperature pretreatment, growth regulators, sucrose and gelling agents were tested on Murashige and Skoog's medium. And the effects of ABA and $AgNO_3$ on organogenesis were investigated. The most suitable stage for anther culture was at the middle to late uninucleate stage, of which the flower size was $6.2{\pm}0.6{\times}4.0{\pm}0.4mm(Length{\times}width)$, and the frequency of callus induction was 13.3%. The effect of low-temperature pretreatment on callus induction was highest as 12.5% in the trearment for 8 days at $4^{\circ}C$. The combination of 1.0 mg/L 2, 4-D and 0.25 mg/L kinetin for callus induction was most effective as 8.3% among various media. The frequency of callus induction was excellent as 10.8% in 4% sucrose. Effect of gelling agents on callus induction was highest as 9.0% on 0.6% Gelrite medium. The prevention of callus browning was effective on the media supplemented with ABA and $AgNO_3$ and rooting was promoted on medium supplemented with 5 mg/L ABA. But shoot was not developed.

Nematicidal Efficacy of Herbal Extracts against Meloidogyne hapla (당근뿌리혹선충에 대한 식물추출물의 살선충 효과)

  • The nematicidal and egg haching inhibitory effects of extracts from 30 herbal plants (total 32 samples) against Meloidogyne hapla J2 juveniles and eggs was tested using the dipping method. At 1,000 ppm, extracts of Daphne genkwa flower buds, Eugenia caryophyllata flowers, Quisqualis indica fruits, and Zingiber officinale rhizomes produced > 80% mortality in J2 juveniles. At 125 ppm, extracts of D. genkwa and Q. indica produced 91 and 99% mortality, respectively. The toxicity of 5 selected plant extracts to M. hapla differed depending on the solvent used (i.e. hexane, methanol, hot water, or cold water). Hot water extracts of Z. officinale and Q. indica produced nematicidal efficacies of 99 and 99%, compared to 36 and 98%, respectively, with cold water extraction. Q. indica extract was highly active against M. hapla regardless of extraction method. The inhibitory effects of Areca catechu, D. genkwa, Desmodium caudatum, Pharbitis nil, Q. indica, and Z. officinale extracts on egg hatching of M. hapla was evaluated. At 1,000 ppm, D. genkwa, P. nil, and Q. indica extracts significantly reduced hatching at 7, 14 and 21 days after treatment. Numbers of juveniles in soil treated with the methanol extract D. genkwa (1,000 ppm) were significantly lower than in untreated soil in trials in pots and in a ginseng (Phanax ginseng) field. These results indicate that Q. indica extracts could be used as an environmental friendly control agent of M. hapla.

Gray Mold of Nephrolepis Caused by Botrytis cinerea (Botrytis cinerea에 의한 네프로레피스 잿빛곰팡이병)

  • In February of 2000-2001, the gray mold disease occurred on nephrolepis (Nephrolepis sp.) grown in a flower nursery farm in Suwen, Korea. Typical symptoms were water-soaked brown or blackish lesions on terminal leaf blades. Severely infected leaves were entirely blighted with grayish fungal mycelia formed on the surface. Conidia of the fungus in mass were hyaline or gray, 1-celled, mostly ellipsoid or ovoid and $13.5{\sim}16.9{\times}6.8{\sim}9.2{\mu}m$ in size. Conidiophores were formed on PDA with $8.7{\sim}11.1{\mu}m$ in width. The sclerotia were readily formed within 2 or 3 days on PDA. In addition, the Biolog database gave the causal fungus a high similarity to Botrytis cinerea (78%) with a match probability of 100%. Pathogenicity of the causal organism was proved according to Koch's postulate. The causal organism was identified as Eotrytis cinerea based on its mycological characteristics and utilization of carbon sources with Biolog system as supporting data. This is the first report of gray mold of nephrolepis caused by Botrytis cinerea in Korea.

A New Early Maturing, High Yielding and High Oleic Acid Rapeseed (Brassica napus L.) of F1 Hybrid "Boram" (조숙, 다수성, 올레인산 고함유 유채 신품종 "보람")

  • "Boram" is a new $F_1$ hybrid of rapeseed (Brassica napus L.) with early maturing, high yield and high oleic acid. This cultivar was developed from the cross between Mokpo-CGMS (male steile line) and 8634-B-1-3-1-8 (restore line) for the production of edible oil and biodiesel in 2006. Yield trials were conducted from 2007 to 2008 and regional adaptation trials were examined at four locations in 2008. "Boram" has green and parted leaf, yellowish flower, and black seed coat. The ripening period of "Boram" is similar to "Sunmang", but 3 days faster than "Tammiyuchae". The average seed yield of the regional adaptation trials was 424 kg/10a, which was 11 and 48% higher than "Sunmang" and "Tammiyuchae" variety. Total oil content of "Boram" was 44.2%. In fatty acid composition, "Boram" has no erucic acid. However, oleic acid content is 68.5%, which is 3.7 and 5.4% higher than "Tammiyuchae" and "Sunmang". And total glucosinolate content was 2.27mg/g. Therefore this variety is recommended as a leading variety at southwestern areas including Jeonnam, Jeonbuk and Kyeongnam provinces and Jeju island of Korea.

Stem Elongation and Photosynthesis Response of Cut Chrysanthemum under Different LEDs Light Intensity (단일처리 후 LEDs 광도에 따른 절화국의 줄기 신장과 광합성반응)

  • The control of stem length is most important external quality aspect in cut chrysanthemum. The present work in conducted in growth chamber and aims at investigating the effect of light intensity on the photosynthesis capacity and stem elongation in cut chrysanthemum. To evaluate the effect, different level of assimilation lighting (LEDs) was given to canopy level 60, 100, 140, and $180mol{\cdot}m^{-2}{\cdot}s^{-1}$, individually, under short-day conditions, and the light treatments were initiated from 7days after planting. There was a positive linear-regression relationship between the light intensity and the net assimilation rate. On the other hand, there was not significant difference in flower buds induction. The growth of stem length, leaf area, and dry weight was increased by increasing the light intensity, whereas the plants grown under 140 and $180mol{\cdot}m^{-2}{\cdot}s^{-1}$ was not differences in those of growth. It indicates that the increased net assimilation rate is not continually coincided with the maximized growth in cut chrysanthemum. It might be considered that the optimal light intensity for stem elongation of cut chrysanthemum is $140mol{\cdot}m^{-2}{\cdot}s^{-1}$ under short-day condition.

Effect of Gamma Ray Irradiation on Seed Germination, Growth and Variant Induction in Hosta plantaginea and Farfugium japonicum (감마선 조사가 옥잠화와 털머위 종자의 발아, 생육 및 변이 유발에 미치는 영향)

  • This study was conducted to select new variants with different characteristics in plant height, leaf color, and petal color of plantain lily (Hosta plantaginea) and Japanese farfugium (Farfugium japonicum). After 10-30Gy ${\gamma}-ray$ irradiation to seeds of both species, seed germination, variant induction, growth of variants, and survival ratios were estimated. Days required for seed germination were lengthened and % seed germination decreased as the dosage of ${\gamma}-ray$ increased over 20Gy. Lethal dose ($LD_{50}$) for plantain lily and Japanese farfugium were 30Gy and 25Gy, respectively. Plantain lily and Japanese farfugium treated by more than 20Gy ${\gamma}-ray$ showed dwarf characteristics in growth, and variations in leaf and petal colors. Total 166 variants in plantain lily were induced by ${\gamma}-ray$ irradiation in a $M_1$ generation, among which 12 promising variants were selected in a $M_2$ generation. Total 57 variants of Japanese farfugium were induced by ${\gamma}-ray$ irradiation in a $M_1$ generation, among which nine promising variants were selected in a $M_2$ generation.

Hybridity Verification of Progenies Obtained from Ovule Culture by Using RAPD Markers in Reciprocal Crosses of Alstroemeria (알스트로메리아 배주배양을 통하여 획득한 정역교배 자손의 혼종성 분석)

  • In this study, we performed ovule culture after reciprocal crosses of two Alstroemeria accessions and investigated genetic contribution of parents by using RAPD markers. The best method was half-ovule culture on MS medium supplemented with $60g{\cdot}L^{-1}$ sucrose and $2.2g{\cdot}L^{-1}$ gelrite at 14 days after pollination. Embryos began to germinate after 6 weeks of culture. The complete plantlets were formed after 4 months of culture. In eight progenies and two parental cultivars, 59 polymorphic bands were obtained out of 89 total bands by RAPD analysis using 7 primers. Eight $F_1$ progenies from the crosses between two accessions using reciprocal crosses showed 1:1 contribution of maternal and paternal parents. It is confirmed that $F_1$ progenies were obtained from parental accessions by using RAPD markers. We conclude this cross combination showed pre-fertilization barriers with incompatibility between stigma or style, and pollen because progeny number was different in each cross combination. Thereby, it warrants overcoming pre-fertilization barrier together with post-fertilization barrier in order to broaden the heterozygosity within progeny populations in Alstroemeria breeding program.
