Cytotoxicity of Daucus Carota L. on Various Cancer Cells (인체 암세포에 대한 당근 추출 성분의 세포독성효과)

  • We investigated the cytotoxic effects of roots and seeds Daucus carota L. on HepG2, HeLa, MCF7, SW626, C6 and NB41A3 cell lines by the MTT assay. Among extracts, the ethylacetate partition layer (DCMEA) of root of Daucus carota L. showed the strongest cytotoxic effects on HepG2, HeLa, C6 and NB41A3 cell lines. On the other hand, methanol(DCM), n-ethylacetate(DCMEA) and n-butanol(DCMB) extract of the seeds of Daucus carota L. also showed significant cytotoxic activities for all six cell lines. $\beta$-carotene, a well-known main component of Daucus carota L. was also tested for its cytotoxic effect. However, in all six cell lines, $\beta$-carotene falied to show significant cytotoxicity. Therefore, the anticancer effect of DCMEA of root fo Daucus carota L. and DCM, DCMH, DCMEA and DCMB extracts of seeds may be caused by components other than $\beta$-carotene. Futher studies are under way to isolate the compounds responsible for the significant cytotoxic activity.

  • Chemoprevention is one of the major strategies for cancer control. It is well established that dietary factors play an important role in modulating the development of certain types of human cancer. The experiment was conducted to determine quinone reductase(QR) activity induction of Daucus carota L. on HepG2 cells. Among various partition layers of roots of Daucus carota L., the ethyl acetate partition layer(DCMEA) and the n-hexane partition layer(DCMH) tested to be most effective which resulted 2.1 and 1.6 respectively compared to the control value of 1.0. In the case of seeds of Daucus carota L. n-butanol partition layer (DCMB) on HepG2 cells at a dose of 200 $\mu\textrm{g}$/$m\ell$ showed the highest induction activity of QR which was 3.0. These results suggest that potentially useful cancer chemoprevention chemicals could be isolated from DCMEA and DCMH of the roots and DCMB of the seeds of Daucus carota L.

  • Daucus species vegetated in the island Dae-heug-san-do off the Korean Peninsula has been recorded as Daucus littoralis Sibth. et Sm. var. koreana Nakai, endemic to Korea in several flora. However, the name was revealed as a naked name, nomen nudum, because the valid publication and type specimens were found anywhere. Comparative observation on morphological characters between the Korean taxon of Daucus and D. littoralis reveals that the Korean vegetated in the island Dae-heug-san-do is not the D. littoralis, but the D. carota L. subsp. carota. The Korean name of the latter is designated newly as San-dang-geun. The new korean name, Seon-dang-geun, of the unrecorded taxon, D. carota subsp. maritimus (Lam.) Batt., is also given.

  • The anticarcinogenic effects of various food components on human cancer cells have received much attention in recent years. The precise effect and mechanisms of anticarcinogens in food materials on cancer cells have rarely been investigated. This study was carried out to determine the effects of Daucus carota L. leaf (DCL) extracts on cytotoxic and chemopreventive effect on human cancer cells. The experiment was conducted to determine cytotoxicity of Daucus carota L. leaf extracts on HepG2, Hela and MCF-7 cells by MTT assay. Among various partition layers of Daucus carota L. leaf, the ethylacetate partition layer (DCLMEA) at 500 $\mu\textrm{g}$/mL was shown to be most effective on MCF-7 cell lines. The four partition layers which are DCLM, DCLMH, DCLMB and DCLMH were less effecitve in inducing cytotoxicity than DCLMEA was. We also determined the induction of intracellular quinone reductase (QR) activity by adding DCL extracts on HepG2 cells. Among various partition layers of DCL extracts, DCLMH and DCLM were tested to be most effective with results such as 4.9 and 4.73 with a control value of 1.0.

  • The activity of Daucus carota L. root $\beta$-glucan synthetase II was observed to increase in the presence of polyamines such as putrescine, spermidine and spermine in vitro, whereas the activity of Daucus carota L. root $\beta$-glucan synthetase I was not affected by the polyamine. The activity $\beta$-glucan synthetase II from Daucus carota L. root protoplasts cultured on medium containing 10-6 M polyamines such as putrescine, spermidine and spermine was observed to be higher than that of the control. Daucus carota L. root protoplasts were observed to have the activities of arginine and ornithine decarboxylases and it was noted that they could produce polyamines, which might have an effect on $\beta$-glucan synthetase II activity.

  • The structure of shoot apex in zygotic and somatic embryos of Daucus carota L. cv. Hongshim was observed by using SEM and longitudinal sections. Shoot apex of zygotic embryo was of an inverted boat shape, and these of two, three and four cotyledon somatic embryos were of an inverted boat shape, a pyramid shape and a convex diamond shape, respectively. In zygotic embryo shoot apex is consisted of small cells which are arranged in layers (tunica) and show corpus in central region. In somatic embryos shoot apices are consisted of somewhat large cells which are arranged in irregularly or slight regularly.

  • The effects of sizes and densities of cells cultured, conditioned medium, and media pH on the somatic embryogenesis of carrot (Daucus carota L.) were examined. A large number of globular embryoids was formed after 4 days in cell culture, and later globular embryoids developed into heart and torpedo shape. High cell density resulted in higher number and better growth of embryos, especially on conditioned medium than Murashige-Skoog medium. The fresh weight and number of embryoids formed increased with the decrease in cell size. The significant reduction in fresh weight and number of embryoids was obtained when culturing cells with diameter of over 90 ${\mu}{\textrm}{m}$. Dry weight and number of embryoids were markedly reduced with medium pH of 4 or 7, but promoted with pH 6.0.

  • The purpose of this study was to investigate whether the extracts of Daucus carota L. could lead to a beneficial outcomes on serum lipid and antioxidative enzyme activity in ovariectomized rats. Sprague-Dawley rats were randomly assigned to one control and three diet groups; carrot seed, carrot root and estrogen after ovariectomy operation. Experimental diets were fed for 8 weeks. The GOT activity was decreased in the carrot extracts treated group than in the control group. The carrot seed extracts treated group showed the lowest SOD and catalase activities compare to other groups. Serum total cholesterol and LDL cholesterol decreased in the carrot seed extracts group than in the control group. From these results, it suggest that carrot seed extracts positively influenced on serum lipid and antioxidative enzyme activity.

Quality Characteristics of Care Food (Jelly) Prepared with Wild Carrot (Daucus carota L.) Juice (야생당근(Daucus carota L.) 주스를 첨가한 개호식품(젤리) 제조 및 품질 특성)

  • This study evaluated the quality characteristics of jelly prepared with different levels (0%, 5%, 10%, 15%, 20%, and 25%) of wild carrot (WC, Daucus carota L.) juice as a care food for the elderly. The lightness, redness, yellowness, and delta (${\Delta}$) values of the jelly (Control, WCJ5, WCJ10, WCJ15, WCJ20, and WCJ25) decreased with increasing amounts of wild carrot juice added. The mechanical properties, such as hardness, springiness, chewiness, and gumminess, of the jelly were decreased with increasing amounts of wild carrot juice added. The total polyphenol and total flavonoid contents of the jelly increased with increasing amounts of wild carrot juice added. The DPPH radical scavenging activity ($IC_{50}$) also increased with increasing amounts of wild carrot juice added. The ${\alpha}$-glucosidase inhibitory effects of wild carrot (WC) and WCJ25 were 71% and 54.4%, respectively, compared to the positive control (acarbose). The lipase inhibitory effects of WC and WCJ25 were 44.2% and 14.4%, respectively, compared to the positive control group (orlistat). On the other hand, the sensory evaluation score was the best at WCJ20, which contained 20% wild carrot juice. In conclusion, WCJ20 or WCJ25 is expected to be a care food for the elderly with respect to texture as well as the antioxidant and enzymatic activity (${\alpha}$-glucosidase inhibitory and lipase inhibitory activities).

Effect of Black Carrot (Daucus carota L.) on the Quality of Pork Hamburger Patties (자색당근 첨가가 돈육 햄버거 패티의 품질에 미치는 영향)

  • The purpose of this study was to investigate the effects of black carrot (Daucus carota L.) on the quality characteristics and antioxidant activity of pork patties. Patties were provided with different amounts (0, 2, 4, 6 and 8%) of black carrot. Notably, the cooking loss rate of the group added with 8% black carrot was lowest (p<0.001). As the content of black carrot increased, the L value decreased and the a, b value increased. The texture analysis results showed that the hardness and chewiness to decrease as the amount of black carrot increased (p<0.05). Evaluation of consumer acceptability revealed that the 4% black carrot groups had a higher score in respect to overall preference and taste to the other groups (p<0.05). For antioxidant activities, both total phenolic compounds content and DPPH were increased (p<0.001) as the amount of added black carrot was increased. As a result, it can be concluded that adding the black carrot to patties in processing can improve antioxidant activities and quality of the patties. Considering various aspects of quality, the suitable amount is 4% of total weight.
