The Effects of Dangguijakyak-san and Wuelbigachul-tang on Mesangial Cell Proliferation and on ICAM-1 and ${\beta}1-integrin$ Expression (당귀작약산, 월비가출탕이 Mesangial Cell 증식과 ICAM-l 및 ${\beta}1-integrin$ 발현에 미치는 영향)

  • Objectives : This experiment was conducted to investigate the suppressive effects of Dangguijakyak-san and Wuelbigachul-tang on the expression of ICAM-l and ${\beta}1-integrin$, which mediate cell-cell or cell-matrix interaction, and on the proliferation of mesangial cells. Methods : After in vitro culturing of human mesangial cells with the supernatant which was obtained from the monocytes separated from human blood with Con-A, hydrocortisone, Dangguijakyak-san and Wuelbigachul-tang respectively, we evaluated suppressive effects by measuring the mesangial cell surface enzyme immunoassay or flow cytometry. Results : The results are summarized as follows: 1. Dangguijakyak-san and Wuelbigachul-tang induced marked suppressive effects on the mesangial cell proliferation in the 50% and 25% supernatant concentration stimulating experiments, but hydrocortisone had little effect in these experiments. 2. Dangguijakyak-san and Wuelbigachul-tang induced marked suppressive effects on ICAM-l and ${\beta}1-integrin$ expression, but were less effective than hydrocortisone was. Conclusions : Based on these results, Dangguijakyak-san and Wuelbigachul-tang were found to be effective in the suppression of mesangial cell proliferation and in ICAM-1 and ${\beta}1-integrin$ expression. Further in vitro investigations as conducted above, with the in vivo experiments reflected, may prove that Dangguijakyak-san and Wuelbigachul-tang contribute to the prevention of the glomerular disease.

Effects of Dangguijakyak-san Extract on Nitric Oxide Production and Cytokine Gene Expression in RAW 264.7 Macrophages Cell (당귀작약산이 마우스 대식세포주의 NO 생성 및 사이토카인 유전자 발현에 미치는 영향)

  • The present study was conducted to evaluate the immunomodulatory effects of Dangguijakyak-san(당귀작약산). We investigated the effects of cell proliferation in mouse spleen cell and RAW 264.7 macrophages cells. Dangguijakyak-san enhanced mitogenic activity in the dose-response manner in mouse spleen cells and RAW 264.7 macrophages cells. In nitric oxide (NO) synthesis and iNOS mRNA expression by Dangguijakyak-san, Dangguijakyaksan alone had an effect on NO synthesis and iNOS mRNA expression in RAW 264.7 cells. NO production and iNOS mRNA expression which is excessively induced by LPS decreased after treatment of Dangguijakyak-san. The expressions of cytokine gene by Dangguijakyak-san investigated using reverse transcription polymerase chain reaction (RT-PCR). In RT-PCR, IL-1α, IL-1β and IL-6 mRNA expressions induced in Dangguijakyak-san-treated RAW 264.7 cells. These data indicate that 1) Dangguijakyak-san can modulate various immune response and 2) the immunomodulatory effects of Dangguijakyak-san may be, in part, associated with the regulation of NO synthesis, the expressions of these cytokine as well as the mitogenic effect on spleen cells and macrophages cells.

  • Objectives : The aim of this study was to report the effect of Dangguijakyak-san and acupuncture on postviral olfactory disorder. Methods : Dangguijakyak-san extract granule was administered three times a day. Acupuncture was applied to 11 acupoints(EX-HN3, GV20, GV23 and both side of LI04. LR03, LI20, EX-HN8) for 15 minutes once a week. Results : The score of olfactory threshold test and TDI(Threshold-Discrimination-Identification) score increased by 3 points. The subjective sense of smell was improved for the substances that could not previoulsy be smelled. Conclusions : This study shows the possibility of using Dangguijakyak-san and acupuncture to treat postviral olfactory disorder.

  • Purpose: This master‘s thesis to evaluate the grade of life after medication a Dangguijakyak-san(pasabu) and Gyejibongnyeong-hwan(cheongwal) with in postmenopausal women get lowering quality of life caused by vasomotor symptoms with hot flush. Methods: A subject who signing on the clinical trial written consent by self-will is registered this clinical trial after decided suitable by selection and exception standard, after take a medical experiment and checkup according to clinical trial plan. Registered subject should valuated by settled schedule after take the testing medicine 1,2 during thirty-day. In this period, a subject allocated at relatively better suited experimental group by oriental doctor after consideration of general efficacy and nature of a medicine at second visiting. Results: The result of comparison in the remedial value with sixteen patients who ended the experiment is like next list. 1. There's no regarded difference of comparison in general conditions between two patient groups. 2. There's no regarded difference during observation period before take medicine. 3. By period of measurement, there's something regarded differences most of patients of two parts after taking medicine. 4. There's no regarded difference at alteration phase of each standard according to kind of medicine. 5. There's no abnormal views reflected at allergy, Laboratory and Physical Examination during experiment. Conclusion: This experiment evidence a Dangguijakyak-san and Gyejibongnyeong-hwan can help for improvement generally life quality of postmenopausal women and certify safety of herbs.

A Study on the Regulation of Endometrial and Placental Cell Function by Water Extract of 3 Types of Herbal Medicines and Ethanol Extract on Scutellariae Radix (3종 한약 처방 물 추출물과 황금 에탄올 추출물에 의한 자궁내막과 태반세포 기능조절 연구)

  • Methods: We investigated the ability to induce decidualization of human endometrial stromal cells and invasive ability of human trophoblast cells by water extract of three types herbal medicines (Dangguijakyak-san, Siryung-tang, Antae-eum) and ethanol extract on Scutellariae Radix. Results: In the process of decidualization of endometrial stromal cells, three herbal medicines including Dangguijakyak-san, Siryung-tang, and Scutellariae Radix increased the production of decidual markers such as prolactin (PRL) and insulin-like growth factor-binding protein 1 (IGFBP1). However, Antae-eum increased mRNA levels of PRL and IGFBP and secretion of IGFBP in decidual stromal cells, but not PRL. Four herbal medicines inhibited the invasion of trophoblast cells. Conclusions: Four herbal medicines may play a role in implantation in women with reproductive failures. However, further studies are warranted to elucidate whether these medications are helpful in the maintenance of pregnancy.

The Effect of Dangguijakyak-san on Wound Healing (당귀작약산의 창상 회복에 대한 효과)

  • Objectives We evaluated the wound healing effects of Dangguijakyak-san (DJ) using C57BL/6 mice that were generated open wound. Methods The study was conducted with seven C57BL/6 mice assigned to each group, divided into the normal group, control group, vitamin E group, DJ low-dose group, DJ high-dose group. We measured total polyphenol, flavonoid contents, the size of the wound, liver function, pro-inflammatory cytokine activity in serum, inflammation-related proteins, adhesion molecules and chemokine proteins, collagen-related proteins in skin tissue and histopathological changes by H&E and Masson's staining. Results DJ treatment significantly reduced the area of the wound compared to the control group. Also, inflammatory cytokines were reduced and the expression of anti-inflammatory-related factors (interleukin-4 [IL-4] and IL-10) was significantly increased in the DJ treatment group. We identified that DJ treatment inhibits both pathways of inflammation, the mitogen-activated protein kinases and nuclear factor-κB pathway. Moreover, the protein expressions of Sirt1 (sirtuin 1), MCP-1 (monocyte chemoattractant protein 1), ICAM-1 (intercellular adhesion molecule 1), and VCAM-1 (vascular cell adhesion molecule 1) were decreased by DJ administration. Also, the expression of α-smooth muscle actin and collagen type I alpha 1, collagen-related proteins, that help skin recovery was significantly increased in the DJ treatment group. Histopathologically, a relatively thin epithelial layer could be observed in the DJ administration group, as well as an increase in fibroblasts and collagen fibers. Conclusions These data suggest that DJ treatment is effective in wound healing, suppressing inflammatory proteins, increasing skin repair factors and improving histopathological changes caused by wounds.

The Immuno-Regulatory Effects of Onbi-tang and Dangguijakyak-san in Minimal Change Nephrotic Syndrome (미세변화 현증후군에서 온비탕과 당귀작약산이 면역조절기능에 미치는 영향)

  • The purpose of this research is to investigate the synergistic effect of herb medicines with hydrocortisone and the regulation effect on the immune system of Onbitang and Dangguijakyaksan at the supernatant of PHA-P stimulated PBMC in the patients with minimal change nephrotic syndrome(MCNS). From the measurement of the concentration rate of IL-4, sCD23 and IL-13, the experiment yielded the following results : The Onbitang group showed a greater tendency to suppress IL-4 and IL-13 levels in MCNS group with no statistical significance. It showed very strong suppression in soluble CD23 compared with control group in MCNS group. The Dangguijakyaksan group, though not statistically significant, was inclined to suppress IL-4 level in MCNS group. It shows stronger suppression in sCD23 and IL-13 levels than these of control group in MCNS group. As for the synergistic effect, the group of hydrocortisone with herb medicines(Onbitang or Danguijakyaksan) produced more suppressive effect to IL-13 level in MCNS group than that of hydrocortisone-only group. They also tended to suppress sCD23 and IL-4 levels, though no statistical significance can be given. As to the suppressive effect of 1L-13 level, the group of Onbitang with hydrocortisone showed an increase of 22.6%, while the group of Dangguijakyaksan with hydrocortisone showed 14.7%. So Onbitang is more effective than Dangguijakyaksan. From the above results, a combinative treatment(herb medicines with hydrocortisone) can be an alternative method to substitute for steroid therapy. It can be a more effective therapy than steroid-only therapy because it is expected to reduce side effects and it shows more special effect to suppress IL-13 level. Based on the present results, further investigation concerning the serum IgE elevation is needed.
