The contribution of damage mechanisms to failure of steam turbine casing made of Cr-Mo-V steel was investigated. Creep-fatigue interaction on the HP side corner of turbine casing was revealed as the root cause of the catastrophic failure performed by metallurgical analysis. The steady-state pressure and transient thermal stress were analyzed based on the actual operating condition of the thermal plant. Damage of creep-fatigue interaction to crack initiation was evaluated with multiaxial effects. The contribution ratio of creep and fatigue to the crack initiation was estimated to 3:1. Temporary geometrical correct action with repair weld was executed. For long-term operation, design improvement of casing equipment for creep resistance should be needed.

Welding fume (WF) induces pulmonary disease including pneumoconiosis. To investigate whether reactive oxygen species-induced oxidative DNA damage occurs during welding fume exposure and the upregulation of DNA repair mechanisms is accompanied, SPF SD rats were exposed to welding fumes with the concentrations of 65.6${\pm}$2.9 mg/㎥(low dose) and 116.8${\pm}$3.9 mg/㎥ (high dose) of total suspended particulate for 2 hrs per day in an inhalation chamber for a total of 2hrs, 15 or 30 days.(omitted)
Oxidative damage to DNA can be caused by excited oxygen species, which are produced by radiation or are by-products of aerobic metabolism. Endogenous evels of 8-hydroxy-2'deoxyguanosine (8-OH-dG), an adduct that results from the damage of DNA caused by hydroxyl radical,have been detected in E. coli and S. typhimurium. Treatment of bacterial cells with various concentrations of hydrogen peroxide caused a moderate increase in the 8-OH-dG content. The enzymatic release of 8-OH-dG from asocorbate/Cu(II)-treated DNA was effected by an extract of E. coli cells. These results indicate that 8-OH-dG is formed in vivo inbacterial DNA through endogenous oxidative mechanisms and on treatment with an oxygen radical-producing agent and that it is repairable.
For detecting toxicity of endocrine disruptors (EDs), rapid, sensitive, and simple methods are needed. Therefore, in this study, a new method in which the different toxic effect of EDs can be monitored using 4 different recombinant bacteria was designed and evaluated. It was found that the recombinant bacteria could monitor the toxic effect, not estrogenic effect, due to EDCs through the measurement of bioluminescence and cell growth rate, which were shown to depend upon a form of cellular toxicity, such as DNA damage, protein damage, oxidative damage, and membrane damage. In addition, it was found that the damage done by EDCs can be divided into several groups based upon the toxic mechanisms of the EDCs

In this study, various damage modes in bending unimorph piezoelectric composite actuators with a thin sandwiched PZT plate during bending fracture tests have been evaluated by monitoring acoustic emission (AE) signals in terms of waveform and peak frequency as well as AE parameters. Three kinds of actuator specimens consisting of woven fabric fiber skin layers and a PZT ceramic core layer are loaded with a roller and an AE activity from the specimen is monitored during the entire loading using an AE transducer mounted on the specimen. AE characteristics from a monolithic PZT ceramic with a thickness of $250{\mu}m$ are examined first in order to distinguish different AE signals from various possible damage modes in piezoelectric composite actuators. Post-failure observations and stress analyses in the respective layers of the specimens are conducted to identify particular features in the acoustic emission signal that correspond to specific types of damage modes. As a result, the signal classification based on waveform and peak frequency analyses successfully describes the failure process of the bending piezoelectric composite actuator exhibiting diverse failure mechanisms. Furthermore, it is elucidated that when the PZT ceramic embedded actuators are loaded mechanical bending loads, the failure process of actuator specimens with different lay-up configurations is almost same irrespective of their lay-up configurations.
In this study, we evaluated and compared the protective effects of two major constituents, baicalein and baicalin, against oxidative DNA and cell damages caused by hydroxyl radical. Antioxidant properties were evaluated using DPPH and hydroxyl radicals scavenging assays and $Fe^{2+}$ chelating assay. ${\varphi}X$ 174 RFI plasmid DNA and intracellular DNA migration assay were used to evaluate the protective effect against oxidative DNA damage. Also, MTT and lipid peroxidation assays were used to evaluate their protective effects against oxidative cell damage. Both baicalein and baicalin prevented intracellular DNA and cells from oxidative damage caused by hydroxyl radical via antioxidant activities. Baicalein demonstrated a stronger antioxidant activity in scavenging DPPH radicals and chelating $Fe^{2+}$ while baicalin scavenged hydroxyl radicals more efficiently. The differences in the level of baicalein and baicalin pose a different pathological pathway for each. The antioxidant activity of baicalin was due to its ability to scavenge hydroxyl radical whilst baicalein was a stronger $Fe^{2+}$ chelator. Further investigation to compare the molecular mechanisms of antitumor activities of baicalein and baicalin is vital to anticancer research.
The aim of this study was to evaluate the effect of galangin towards hydrogen peroxide-induced DNA damage. The calf thymus DNA and Chinese Hamster Lung (CHL) cells were used to measure 8-hydroxy-2'-deoxyguanosine(8-OH2'dG) as an indicator of DNA oxidative damage using high performance liquid chromatography with electrochemical detection. Hydrogen peroxide in the presence of Fe(II) ion induced the formation of 8-OH2'dG in both calf thymus DNA and CHL cells. The DNA damage effects were enhanced by increasing the concentration of Fe(II) ion and inhibited by galangin. In the single cell gel electrophoresis (Comet assay), galangin and dl-a-tocopherol showed an inhibitory effect in CHL on hydrogen peroxide induced DNA single strand breaks. Galangin showed more potent activity than dl-$\alpha$-tocopherol under our experimental conditions. These results indicate that galangin can modify the action mechanisms of the oxidative DNA damage and may act as chemopreventive agents against oxidative stress.
In this paper, a simplified planar model is developed for damage estimation of interlocked caisson systems. Firstly, a conceptual dynamic model of the interlocked caisson system is designed on the basis of the characteristics of existing harbor caisson structures. A mass-spring-dashpot model allowing only the sway motion is formulated. To represent the condition of interlocking mechanisms, each caisson unit is connected to adjacent ones via springs and dashpots. Secondly, the accuracy of the planar model's vibration analysis is numerically evaluated on a 3-D FE model of the interlocked caisson system. Finally, the simplified planar model is employed for damage estimation in the interlocked caisson system. For localizing damaged caissons, a damage detection method based on modal strain energy is formulated for the caisson system.

Cells are constantly exposed to endogenous and exogenous stresses that can result in DNA damage. In response, they have evolved complex pathways to maintain genomic integrity. RUNX family transcription factors (RUNX1, RUNX2, and RUNX3 in mammals) are master regulators of development and differentiation, and are frequently dysregulated in cancer. A growing body of research also implicates RUNX proteins as regulators of the DNA damage response, often acting in conjunction with the p53 and Fanconi anemia pathways. In this review, we discuss the functional role and mechanisms involved in RUNX factor mediated response to DNA damage and other cellular stresses. We highlight the impact of these new findings on our understanding of cancer predisposition associated with RUNX factor dysregulation and their implications for designing novel approaches to prevent cancer formation in affected individuals.

This work describes an investigation into the feasibility of using an acoustic emission (AE) technique to evaluate the integrity of a composite actuator with a PZT ceramic under electromechanical cyclic loading. AE characteristics have been analyzed in terms of the behavior of the AE count rate and signal waveform in association with the performance degradation of the composite actuator during the cyclic tests. The results showed that the fatigue cracking of the composite actuator with a PZT ceramic occurred only in the PZT ceramic layer, and that the performance degradation caused by the fatigue damage varied immensely depending on the existence of a protecting composite bottom layer. We confirmed the correlations between the fatigue damage mechanisms and AE signal types for the actuators that exhibited multiple modes of fatigue damage; transgranular micro damage, intergranular fatigue cracking, and breakdown by a short circuiting were related to a burst type signal showing a shortly rising and slowly decaying waveform with a comparably low voltage, a continuous type signal showing a gradual rising and slowly decaying waveform with a very high voltage and a burst and continuous type signal with a high voltage, respectively. Results from the present work showed that the evolution of fatigue damage in the composite actuator with a PZT ceramic can be nondestructively identified via in situ AE monitoring and microscopic observations.
