The comet assay (also called the single-cell gel electrophoresis assay) has been widely used for detecting DNA damage and repair in individual cells. Since the conventional methods of evaluating comet assay data using frequency statistics are unsatisfactory we developed a new quantitative measure of DNA damage/repair that is based on all information residing in the dose/time-response curves of a comet experiment. Blood samples were taken from 25 breast cancer patients before undergoing radiotherapy. The comet assay was performed under alkaline conditions using isolated lymphocytes. Tail DNA, tail length, tail moment and tail inertia of the comet were measured for each patient at four doses of $\gamma$-rays (0, 2, 4 and 8 Gy) and at four time points after irradiation (0, 10, 20 and 30 min) using 100 cells each. The resulting three-dimensional dose-time response surface was modeled by multiple regression, and the second derivative, termed 2D, on dose and time was determined. A software module was programmed in SAS/AF to compute 2D values. We applied the new method successfully to data obtained from cancer patients to be assessed for their radiation sensitivity. We computed the 2D values for the four damage measures, i.e., tail moment, tail length, tail DNA and tail inertia, and examined the pairwise correlation coefficients of 2D both on the log scale and the unlogged scale. 2D values based on tail moment and tail DNA showed a high correlation and, therefore, these two damage measures can be used interchangeably as far as DNA repair is concerned. 2D values based on tail inertia have a correlation profile different from the other 2D values which may reflect different facets of DNA damage/repair. Using the dose-time response surface, other statistical models, e.g., the proportional hazards model, become applicable for data analysis. The 2D approach can be applied to all DNA repair measures, Le., tail moment, tail length, tail DNA and tail inertia, and appears to be superior to conventional evaluation methods as it integrates all data of the dose/time-response curves of a comet assay.

In order to study the axial compression performance of sand-lightweight concrete-filled steel tube (SLCFST) stub columns, three circular SLCFST (C-SLCFST) stub column specimens and three SLCFST square (S-SLCFST) stub column specimens were fabricated and static monotonic axial compression performance testing was carried out, using the volume ratio between river sand and ceramic sand in sand-lightweight concrete (SLC) as a varying parameter. The stress process and failure mode of the specimens were observed, stress-strain curves were obtained and analysed for the specimens, and the ultimate bearing capacity of SLCFST stub column specimens was calculated based on unified strength theory, limit equilibrium theory and superposition theory. The results show that the outer steel tubes of SLCFST stub columns buckled outward, core SLC was crushed, and the damage to the upper parts of the S-SLCFST stub columns was more serious than for C-SLCFST stub columns. Three stages can be identified in the stress-strain curves of SLCFST stub columns: an elastic stage, an elastic-plastic stage and a plastic stage. It is suggested that AIJ-1997, CECS 159:2004 or AIJ-1997, based on superposition theory, can be used to design the ultimate bearing capacity under axial compression for C-SLCFST and S-SLCFST stub columns; for varying replacement ratios of natural river sand, the calculated stress-strain curves for SLCFST stub columns under axial compression show good fitting to the test measure curves.
This article presents a comparative analysis of seismic behavior in steel-beam reinforced concrete column (RCS) frames versus steel and reinforced concrete frames. The study evaluates the seismic response and collapse behavior of RCS frames of varying heights through nonlinear modeling. RCS, steel, and reinforced concrete special moment frames are considered in three height categories: 5, 10, and 20 stories. Two-dimensional frames are extracted from the three-dimensional structures, and nonlinear static analyses are conducted in the OpenSEES software to evaluate seismic response in post-yield regions. Incremental dynamic analysis is then performed on models, and collapse conditions are compared using fragility curves. Research findings indicate that the seismic intensity index in steel frames is 1.35 times greater than in RCS frames and 1.14 times greater than in reinforced concrete frames. As the number of stories increases, RCS frames exhibit more favorable collapse behavior compared to reinforced concrete frames. RCS frames demonstrate stable behavior and maintain capacity at high displacement levels, with uniform drift curves and lower damage levels compared to steel and reinforced concrete frames. Steel frames show superior strength and ductility, particularly in taller structures. RCS frames outperform reinforced concrete frames, displaying improved collapse behavior and higher capacity. Incremental Dynamic Analysis results confirm satisfactory collapse capacity for RCS frames. Steel frames collapse at higher intensity levels but perform better overall. RCS frames have a higher collapse capacity than reinforced concrete frames. Fragility curves show a lower likelihood of collapse for steel structures, while RCS frames perform better with an increase in the number of stories.

External low-velocity impact loadings onto the composites cause reduction of stiffness and/or strength. The reductions indicate that internal(external) damages were developed within the composites. These damages could be matrix cracking, fiber/matrix debonding, or delamination between layers. In previous studies, damage evaluation have been done by applying secondary mechanical loading such as buckle-driven compressive, or fatigue, or flexural loadings. An evaluation method by applying indentation loadings on the composites was proposed. The load-displacement curves obtained from the indentation testing provided the extent of damages within the composites due to impact loadings.

The durability of handmade paper prepared by potassium carbonate cooking of paper multiberry (Broussonetia Kazinoki Sieb).was investigated. The potassium carbonate paper before and after heat aging had better strength properties, breaking length and folding endurance than those of the soda paper, except of lower brightness .After 30 days of heat aging , these strength properties were still maintained . however, there was almost no difference in the infrared spectra and the crystallinities calculated by X-ray diffraction curves between the potassium carbonate paper and the soda a paper. The durability of the potassium carbonate paper was though to be due to decrease in the pulp damage caused by weak alkaline cooking condition. The results indicated that the paper prepared by the potassium carbonate cooling method could have better permanence than that prepared by the conventional soda cooking method.
This paper presents numerical studies of stiff fiber pullout behaviors of fiber reinforced cementitious composites based on a progressive damage model. The ongoing debonding process is simulated. Interfacial stress distribution for different load levels is analyzed. A parametric study, including bond strength and the homogeneity index on the pullout behaviors is carried out. The numerical results indicate that the bond stress decreases gradually from loaded end to embedded end along fiber-cement interface. The debonding initially starts from loaded end and propagates to embedded end as load increasing. The embedded length and bond strength affect the load-loaded end displacement curves significantly. The numerical results have a general agreement with the experimental investigation.
This paper introduces a procedure for free span and fatigue analysis of offshore pipelines per DNV-RP-F105, 2002. The new method includes the axial force and deflection load in pipelines. The screening criteria were established to calculate the allowable span lengths in the new design codes. The screening criteria allows a certain amount of vortex-induced vibration due to wave and current loading. However, the induced pipe stresses are very small and usually below the limit stresess of typical S-N curves. In contrast, the conventional criteria did not allow any vortex-induced vibration in the free span of pipelines. Thus, the screening criteria yields reduced allowable span lengths. A simplified procedure was established to calculate the fatigue damage due to long-term current distribution. The long-term current statistics was assumed with a 3-parameter Weibull distribution. The fatigue damage was estimated for the span lengths obtained from the screening criteria for various conditions. Sample calculations show the effect of axial force for various boundary conditions. Comparisons with conventional criteria are included.
In this study, the seismic performance of skewed highway bridges has been assessed by using fragility function methodology. Incremental Dynamic Analysis (IDA) has been used to prepare complete information about the different damage states of a 30 degree skewed highway bridge. A three dimensional model of a skewed highway bridge is presented and incremental dynamic analysis has been applied. The details of the full nonlinear procedures have also been presented. Different spectral intensity measures are studied and the effects of the period on the fragility curves are shown in different figures. The efficiency, practicality and proficiency of these different spectral intensity measures are compared. A suite of 20 earthquake ground motions are considered for nonlinear time history analysis. It has been shown that, considering different intensity measures (IM) leads us to overestimate or low estimate the damage probability which has been discussed completely.

Recently structural damage has been frequently observed in reinforced concrete brdiges due to repeated loads such as vehicular traffic an due to continual overloads by heavy duty trucks. Therefore, the purpose of this experimental stduy is to investigate the damage mechanism due to fatigue behavior of high-strength reinforced concrete beams under repeated loads. From the test results, the relation of cycle loading to deflection is on the mid-span , the crack growth and the modes of failure according to cycle number, fatigue life and S-N curve were observed through the fatigue test. Based on the fatigue test results , high-strength reinforced concrete beams failed to 57 ∼66 percent of the static ultimate strength . Fatigue strength aobut two million cycles from S-N curves was certified by 60 percent of static ultimate strength.
For fatigue analsis of offshore structures, existing methods have been reviewed and a computer code has been developed on PC. As methods to estimate the probabillity distribution of the fatigue stress, three methods(the deterministic method, the stochastic method, and the simplified method) are used in this code, to choose the appropriate method according to the situations. This code estimates damage ratios, fatigue lives, and probabilities of fatigue failure considering scatterness of SN-data, based on linear damage rule and SN-curves. Also, allowable stress for the design extreme wave can calculated by the simplified method.
