가축분뇨수의 무단방류가 샛강오염에 미치는 영향 (Impact of Livestock-production Wastewater on Water Pollution)

  • Environmental impact assessment survey reflecting farmers` opinion on the residence and production space in rural settlement area by ORD showed that more than 86% of respondents thought their reservoirs and waterways (small rivers) were getting seriously contaminated primarily by garbage and livestock manure. A typical rural settlement unit was taken to assess the impact of improper management of livestock manure in the farms on the water quality of small river flowing down along the villages where swine and dairy farms were situated in Daejook 2, 3-ri, Seolseong-myun, Icheon-gun. Nitrogen compounds such as NO$_3$-N, NO$_2$-N, NH$_3$-N, and phosphorus compound H$_x$PO$_4$, DO, BOD$_5$, COD, and microbial density were analyzed to evaluate water quality at five test sites designated along the water stream. Tests showed. for example, BOD$_5$ at site 4 was average 9.2mg/l which was about 3~8 times higher than that of observation site 2 and 3, at which most livestock houses were situated. This is a clear evidence that the nutrients of livestock manure illegally discharged to small river can lead to an eutrophication of the river at downstream. A soil absorption system with aeration could be one of alternatives to treat the contaminated wastewater by livestock manure. The place at downstream, inbetween observation site 1 and 2, could be the best construction site for the treatment facility from the standpoint of the overall treatment efficiency, An enclosed composting system can also be regarded as a good alternative for treatment of the sludge which is the by-product of the soil absorption system operation.

Survey of American food trends and the growing obesity epidemic

  • The rapid rise in the incidence of obesity has emerged as one of the most pressing global public health issues in recent years. The underlying etiological causes of obesity, whether behavioral, environmental, genetic, or a combination of several of them, have not been completely elucidated. The obesity epidemic has been attributed to the ready availability, abundance, and overconsumption of high-energy content food. We determined here by Pearson's correlation the relationship between food type consumption and rising obesity using the loss-adjusted food availability data from the United States Department of Agriculture (USDA) Economic Research Services (ERS) as well as the obesity prevalence data from the Behavioral Risk Factor Surveillance System (BRFSS) and the National Health and Nutrition Examination Survey (NHANES) at the Centers for Disease Control and Prevention (CDC). Our analysis showed that total calorie intake and consumption of high fructose com syrup (HFCS) did not correlate with rising obesity trends. Intake of other major food types, including chicken, dairy fats, salad and cooking oils, and cheese also did not correlate with obesity trends. However, our results surprisingly revealed that consumption of com products correlated with rising obesity and was independent of gender and race/ethnicity among population dynamics in the U.S. Therefore, we were able to demonstrate a novel link between the consumption of com products and rising obesity trends that has not been previously attributed to the obesity epidemic. This correlation coincides with the introduction of bioengineered corns into the human food chain, thus raising a new hypothesis that should be tested in molecular and animal models of obesity.

Differential Cytokine Regulatory Effect of Three Lactobacillus Strains Isolated from Fermented Foods

  • Lactic acid bacteria (LAB) isolated from fermented foods have potential as a treatment for immune-related disorders and the use of LAB has been increasing worldwide. In this study, the differential cytokine regulatory effect was examined with three isolates of lactobacilli strains; namely, Lactobacillus plantarum K55-5 isolated from dairy product, and L. sakei K101 and L. plantarum K8 previously isolated from kimchi (a Korean traditional fermented vegetable). Production of cytokines such as IL-10, IL-12, IFN-γ, and TNF-α was significantly increased in L. sakei K101- and L. plantarum K55-5-treated splenocytes as compared with controls. The oral administration of L. sakei K101 and L. plantarum K55-5 increased cytokine production in the immunosuppressed mouse splenocytes and blood. NK cell cytotoxic activity was also increased in L. sakei K101- and L. plantarum K55-5-fed mice. On the other hand, L. plantarum K8 did not affect cytokine induction in all the experiments performed in this study. The cytokine-inducing effect of L. plantarum K55-5 was significantly increased by lysates of heat-killed bacteria as compared with live, heat-killed, or supernatant of cell lysates. TNF-α production by lipoteichoic acids (LTAs) isolated from the three isolates of lactobacilli was compared, and it was found that K55-5 LTA had a highest cytokine-inducing ability, which was mediated by TLR2-mediated NF-κB and ERK activation. Taken together, our study suggests that L. plantarum K55-5 and L. sakei K101 can be used for the treatment of immunosuppressed disorders.

건조방법에 따른 분류의 리올로지 특성 (Effect of drying method on rheological properties of milk powders)

  • 건조방법 및 원료성분을 달리하여 분유를 제조하고 이들분유의 몇 가지 리올로지 특성을 Instron Universal Testing Machine으로 측정하여 얻어진 결과는 다음과 같다. 드럼건조된 전지분유의 부피밀도는 $0.382\;g/cm^3$으로 탈지 및 조제 분유보다 높았고, 분무 건조된 탈지 분유의 Hausner ratio, 평정각 및 compressibility 는 각각 1.127, $32.5^{\circ}$, 0.029로 탈지 분유는 자유롭게 흐르는 것을 보여주었으며, 드럼건조된 전지분유의 compressibility는 수분 함량이 4.96%에서 3.89%로 감소함에 따라 0.079에서 0.052로 낮아졌다. 전지분유 그래뉼의 compressibility는 0.056으로서 분유의 compressibility보다 낮았다. 분무 건조된 분유는 구형으로서, 동결건조나 드럼건조된 분유보다 더 견고함을 나타냈다.

Effects of Supplemental Mannanoligosaccharides on Growth Performance, Faecal Characteristics and Health in Dairy Calves

  • Twenty Holstein calves were used to investigate the effects of mannanoligosaccharides (MOS) supplementation in the whole milk on growth performance, faecal score, faecal pH, selected faecal bacterial populations and health during the preweaning period. Healthy calves selected by clinical examination were allocated to one of the two groups (control [CG] and experimental [EG]) at 5 days old. Each group consisted of 5 male and 5 female calves. Each calf in EG was supplemented with 7 g/d of a MOS product (Celmanax) from 5 days to 56 days of age. MOS supplement was mixed with the whole milk once in the morning and administered to the calves in EG via nipple bottle, whereas the calves in CG were fed the whole milk without MOS. Calves were weaned at 56 days of age. The final body weight, average daily weight gain (ADG) and average daily feed intake (ADFI) were statistically similar (p>0.05) but were higher by 3.70%, 6.66%, and 10.97%, respectively, in MOS than in control calves. Feed efficiency (ADG/ADFI) was also similar in two calves group. While faecal scores did not differ on day 5, 7, 14, 21, 28, 42, 49, and 56 between groups, EG had a higher faecal score (p = 0.05) than CG on day 35. Faecal concentration of Lactobacillus was lower (p<0.05) in EG compared with CG. No differences (p>0.05) in faecal concentrations of Bifidobacterium, Clostridium perfringens, and Escherichia coli were found between groups. Although there were no significant differences (p>0.05) in the incidence of diarrhoea, treatment days for diarrhoea and the costs associated with diarrhoea treatments between groups, collectively, the observed reductions in treatment days and the cost of diarrhoea treatments accompanying increases in final body weight, ADG and ADFI for EG may indicate potential benefit of MOS in treatment of diarrhoea.

Production of Functional Whey Protein Concentrate by Monitoring the Process of Ultrafilteration

  • This investigation was undertaken in order to elicit the relationship between the extent of ultrafiltration processing of whey and its effect on composition and yield of resultant whey protein concentrate (WPC). Cheddar cheese whey was fractionated through ultrafiltration to an extent of 70, 80, 90, 95, 97.5% and 97.5% volume reduction followed by I stage and II stage diafiltration. After each level of ultrafiltration, the composition of WPC was monitored. Similarly, the initial whey was adjusted to 3.0, 6.2 and 7.0 pH levels and ultrafiltration was carried out to elicit the effect of pH of ultrafiltration on the composition. Further, initial whey was adjusted to different levels of whey protein content ranging from 0.5 to 1.0 per cent and subjected to ultrafiltration to different levels. The various range of retentate obtained were further condensed and spray dried in order to assess the yield of WPC per unit volume of whey used and the quantity of whey required to produce unit weight of product. With the progress of ultrafiltration, there was a progressive increase in protein content and decrease in lactose and ash content. The regression study led to good relationships with $R^2$ values of more than 0.95 between the extents of permeate removed and the resultant changes in composition of each of the constituents. Whey processed at pH 3.0 had significantly a very low ash content and high protein content as compared to processing at 6.2 and 7.0. The yield of WPC per unit volume of whey varied significantly with the initial protein content. Higher initial protein content led to higher yield of all ranges of WPC and the quantity of whey required per unit weight of spray dried WPC significantly reduced. Regression equations establishing the relationship between initial protein content of whey and the yield of various types of WPC have been derived with very high $R^2$ values of 0.99. This study revealed that, the yield and composition of whey can be monitored strictly by controlling the processing parameters and WPC can be produced depending on the food formulation requirement.

김치 유래 젖산균의 Cell-envelope Proteinase 존재 확인 (Identification of the Cell-envelope Proteinase of Lactic Acid Bacteria Isolated from Kimchi.)

  • 김치로부터 분리된 젖산균들의 16S rDNA 부분 염기서열 결정 결과, 6균주 중 2균주는 Leuconostoc mesenteroides로, 나머지 균주는 Lactobacillus속으로 재동정되었다. 이들 김치유래 젖산균들은 유제품 유래 젖산균과 마찬가지로 cell-envelope proteinase (CEP)로 추정되는 세포외 proteinase 활성을 보유하고 있는 것으로 나타났고, 젖산균의 CEP 특이적 primer를 제작하여 PCR을 수행한 결과, Leu. mesenteroides로부터는 예상 크기의 PCR 산물이 증폭되지 않았지만 Lactobacillus속 균주들로부터는 예상되는 1.2 kb 의 DNA 단편이 증폭되었다. Lactobacillus pentosus KFR1821의 genomic DNA로부터 증폭된 PCR 산물의 추정 아미노산서열은 Lactococcus lactis subsp. cremoris의 PrtP와 95%, Labtobacillus paracasei subsp. paracasei이 PrtP와 92%의 높은 상동성을 보였다. 증폭된 PCR 산물을 probe로 하여 Southern hybridization을 수행한 결과, Lb. pentosus KFR1821의 CEP 유전자는 chromosomal DNA에 암호화 되어있는 것으로 확인되었다.

Effects of Germinated Brown Rice Addition on the Flavor and Functionality of Yogurt

  • This study aimed to investigate the functional and physicochemical properties of yogurt, supplemented with germinated brown rice (GBR) containing γ-aminobutyric acid (GABA), during storage. GBR was produced by soaking brown rice at 30℃, and saccharified germinated brown rice (SGBR) was produced by treating brown rice with α- and β-amylase for 1 h, at 80℃ and 60℃, respectively. Yogurt was manufactured using a commercial starter (YC-X11, CHR. Hansen, Denmark) at 37℃ for 12 h. The fatty acids and GABA contents were analyzed using GC and HPLC, respectively. The fatty acids in the cereal samples consisted of oleic, linoleic, and palmitic acid. The portion of oleic acid was the highest, at 35.65% in GBR, and 32.16% in SGBR. During germination, the oleic acid content increased, whereas linolenic and palmitic acid contents from GBR tended to decrease. Although the portion of saturated fatty acids, such as stearic and myristic acid, decreased significantly (p<0.05), that of unsaturated fatty acids, such as oleic and linoleic acid, increased with an increase in supplementation of BR, GBR, or SGBR in the yogurt. The yogurt, supplemented with cereal samples, showed a tendency of an increase in the concentration of GABA with an increase in the supplementation of the cereal samples. However, yogurt supplemented with GBR showed the highest concentration of GABA, regardless of the supplementation of the cereal samples. These results indicated that yogurt supplemented with BR, GBR, or SGBR could be a promising dairy product.

Bypass Fat Production Using Acid Oil, Its Effect on In Vitro Rumen Fermentation and Effect of Its Feeding on In Sacco DM Disappearance in Sheep

  • Attempts were made in the laboratory to produce bypass fat using acid oil by precipitation and fusion methods. The degree of saponification by both of these methods was above 80 percent. Where heating facilities are not available, precipitation method could be used, otherwise, fusion method of bypass fat production is found to be more convenient, especially for commercial scale operations as handling of large volume of solutions is eliminated. Bypass fat thus produced was tested in vitro for rumen fermentation. Incorporation of acid oil in the incubation medium reduced TVFA conc. from 127.06 to 124.09 mM/l SRL and increased ammonia-N levels from 210.50 to 223 mg/l SRL indicating that the microbial activity was affected on incorporation of acid oil in the incubation medium. However, incorporation of bypass fat in the incubation medium did not significantly affect TVFA conc. as well as ammonia-N levels. In another experiment, nine rumen fistulated sheep in three groups of three each were fed bypass fat at two different levels. Dry matter disappearance in 24 h from the nylon bags suspended in the rumen of animals under different groups was found to be $47.74{\pm}1.10$, $47.55{\pm}0.21$ and $50.74{\pm}1.11$ in group I (control), group II (fed bypass fat 50 g/day) and group III (fed bypass fat 100 g/day), respectively. These studies indicated that it is possible to produce bypass fat from acid oils, a by-product of oil refining process, and its feeding did not affect rumen fermentation.

  • 노인 인구 증가는 유럽과 미국에 걸쳐 몇 년간 관찰되어 왔지만, 일본과 한국과 같은 개발도상국에서도 증가 추세가 현저히 나타나고 있다. 개발도상국에서는 2000년, 65세 이상 노인의 인구비율이 1960년에 비해 약 5배 정도 증가했으며, 2050년에는 전체 인구의 40% 정도를 차지할 것으로 예상되고 있다. 이런 급격한 인구 변화는 이들이 특정 권리와 구매력을 지니는 새로운 사회경제적 집단으로 성장하게 하는 계기가 되었다. 노화가 일어나면, 식이요법으로 어느 정도 조절할 수 있는 인체의 변화가 다양하게 일어난다. 그 중 대표적인 것은 글루타치온 합성과 이용의 균형에 변화가 생기는 것이다. 글루타치온은 인체에서 가장 중요한 항산화 물질이고, 식이 내 시스테인 아미노산에 의해 체내 함량이 결정될 수 있다. 시스테인이 풍부한 펩티드 제품이 기능성 식품 및 식품원료로 개발되었다. 소비자 조사 결과, 이 제품은 숙면과 활력을 제공하는 등 장점이 있음이 밝혀졌다. 동물 임상 실험 결과를 통해 특별히 노인 인구를 대상으로 하여 시스테인 펩타이드의 생리 활성과 대사 과정을 소개하고자 한다.
