파이토케미컬의 일종인 EGCG는 녹차의 카테킨 성분으로, 세포 내 신호 경로 조절을 통하여 항산화, 항암효과를 나타내는 것으로 알려져 있다. 본 연구에서는 hypoxia 상태인 B16F10 피부암 세포에서 HIF-$1{\alpha}$를 포함한 AMPK의 신호경로를 통하여 EGCG의 apoptosis 유도 효과를 규명하였다. AMPK는 hypoxia, 영양분 결핍, 운동, heat shock 등, 세포 내 ATP의 결핍에 의해서 활성화되며 암세포의 증식을 억제하고 apoptosis를 유도한다. 세포에서 중요한 에너지 센서로서 작용하는 AMPK가 hypoxia 상태의 암세포 내에서는 HIF-$1{\alpha}$의 전사 활성을 유도하는데, HIF-$1{\alpha}$는 hypoxia 상태에서 산소 결핍에 반응하는 첫 번째 전사 조절인자로서 암세포의 생존을 위한 세포내 산소공급과 혈관신생형성을 조절한다. Hypoxia 상태가 아닌 B16F10 세포에서와 hypoxia 상태에서의 B16F10 세포에서 EGCG에 의한 apoptosis 효과를 관찰하였다. 실험 결과, hypoxia 상태에서 EGCG는 더 강한 apoptosis를 유도하며, 혈관신생형성을 조절할 수 있는 HIF-$1{\alpha}$의 전사 활성을 억제시킨다. 이러한 관찰을 통해 EGCG가 hypoxia 상태의 피부암 세포에서 암의 성장과 신생혈관형성을 저해하는 것으로 보인다. 이와 같은 연구는 향후 식품에 첨가된 파이토케미컬을 이용하여 암을 예방하는 연구에서 있어서, 도움이 될 것으로 여겨진다.
Glutathione S-transferase (GST) is a multigene family of phase II detoxifying enzymes that metabolize a wide range of exogenous and endogenous electrophilic compounds. GSTM1 and GSTT1 gene polymorphisms may account for inter-individual variability in coping with oxidative stress. We investigated the relationships between the level of lymphocyte DNA and antioxidative parameters and the effect on GST genotypes. GSTM1 and GSTT1 were characterized in 301 young healthy Korean adults and compared with oxidative stress parameters such as the level of lymphocyte DNA, plasma antioxidant vitamins, and erythrocyte antioxidant enzymes in smokers and non smokers. GST genotype, degree of DNA damage in lymphocytes, erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase (GSH-Px), and plasma concentrations of total radical-trapping antioxidant potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene, and cryptoxanthin were analyzed. Lymphocyte DNA damage assessed by the comet assay was higher in smokers than that in non-smokers, but the levels of plasma vitamin C, ${\beta}$-carotene, TRAP, erythrocyte catalase, and GSH-Px were lower than those of non-smokers (p < 0.05). Lymphocyte DNA damage was higher in subjects with the GSTM1- or GSTT1-present genotype than those with the GSTM1-present or GSTT1- genotype. No difference in erythrocyte antioxidant enzyme activities, plasma TRAP, or vitamin levels was observed in subjects with the GSTM1 or GSTT1 genotypes, except ${\beta}$-carotene. Significant negative correlations were observed between lymphocyte DNA damage and plasma levels of TRAP and erythrocyte activities of catalase and GSH-Px after adjusting for smoking pack-years. Negative correlations were observed between plasma vitamin C and lymphocyte DNA damage only in individuals with the GSTM1-present or GSTT1- genotype. The interesting finding was the significant positive correlations between lymphocyte DNA damage and plasma levels of ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. In conclusion, the GSTM1- and GSTT1-present genotypes as well as smoking aggravated antioxidant status through lymphocyte DNA damage. This finding confirms that GST polymorphisms could be important determinants of antioxidant status in young smoking and non-smoking adults. Consequently, the protective effect of supplemental antioxidants on DNA damage in individuals carrying the GSTM1- or GSTT1-present genotypes might show significantly higher values than expected.
Oxidative stress leads to the induction of cellular oxidative damage, which may cause adverse modifications of DNA, proteins, and lipids. The production of reactive species during oxidative stress contributes to the pathogenesis of many diseases. Antioxidant defenses can neutralize reactive oxygen species and protect against oxidative damage. The aim of this study was to assess the antioxidant status and the degree of DNA damage in Korean young adults using glutathione s-transferase (GST) polymorphisms. The GSTM1 and GSTT1 genotypes were characterized in 245 healthy young adults by smoking status, and their oxidative DNA damage in lymphocytes and antioxidant status were assessed by GST genotype. General characteristics were investigated by simple questionnaire. From the blood of the subjects, GST genotypes; degree of DNA damage in lymphocytes; the erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase; plasma concentrations of total peroxyl radical-trapping potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene and cryptoxanthin, as well as plasma lipid profiles, conjugated diene (CD), GOT, and GPT were analyzed. Of the 245 subjects studied, 23.2% were GSTM1 wild genotypes and 33.4% were GSTT1 wild genotype. No difference in erythrocyte activities of superoxide dismutase, catalase, or glutathione peroxidase, and the plasma TRAP level, CD, GOT, and GPT levels were observed between smokers and non-smokers categorized by GSTM1 or GSTT1 genotype. Plasma levels of ${\alpha}$- and ${\gamma}$-tocopherol increased significantly in smokers with the GSTT1 wild genotype (p < 0.05); however, plasma level of ${\alpha}$-carotene decreased significantly in non-smokers with the GSTM1 wild genotype (p < 0.05). DNA damage assessed by the Comet assay was significantly higher in non-smokers with the GSTM1 genotype; whereas DNA damage was significantly lower in non-smokers with the GSTT1 genotype. Total cholesterol and LDL cholesterol levels were significantly higher in non-smokers with the GSTT1 genotype than those with the GSTT1 wild genotype (p < 0.05). In conclusion, the GSTM1 genotype or the GSTT1 wild genotype in non-smokers aggravated their antioxidant status through DNA damage of lymphocytes; however, the GSTT1 wild type in non-smokers had normal plasma total cholesterol and LDL-cholesterol levels. This finding confirms that GST polymorphisms could be an important determinant of antioxidant status and plasma lipid profiles in non-smoking young adults. Further study is necessary to clarify the antioxidant status and/or lipid profiles of smokers with the GST polymorphism and to conduct a study with significantly more subjects.
In 1995, the Republic of Korea started to establish Techno Park (TP) in order to develop the regional industry while promoting the balanced development of the land. By 2008, 18TPs were established nationwide and have become cradles for developing local industries. And recently evolved forms of TP such as Daedeok Techno Valley and Pangyo Techno Valley emerged. In addition, 19 Centers for Creative Economy and Innovation (CCEI) were established nationwide and Tech-Incubator Program for start-ups (TIPS) was introduced to support and mentor start-ups. TPs in Korea become bureaucratic in course of time, and the new trial of innovation of TP is needed. In Korea, professional TIPS-accelerators mentoring and investing start-ups have a history of only five years. But they support and mentor start-ups efficiently, and have obtained good results. In this paper, we propose that TP attract TIPS-accelerators actively and collaborate with each other to support and mentor start-ups and SMEs effectively.
본 연구는 그동안의 공동체 미디어 실천연구들이 전통적 방식의 지상파(전파) 방송에 국한되어 온 것에 대한 문제의식에서 출발하였다. 기존연구들에서 소홀하게 다루어진 온라인 기반 위에서 실천되고 있는 지역 공동체 미디어의 특성과 콘텐츠 제작 및 유통의 특성을 규명하고자 대덕밸리라디오의 사례를 분석하여 다음과 같은 연구결과와 시사점을 얻을 수 있었다. 첫째, 온라인 공동체 미디어 활동은 지역 공동체의 가치를 담아내는 시민미디어(civic/citizen media) 활동이다. 둘째, 미디어 콘텐츠 제작과 유통전략에 따라 퍼블릭 액세스(Public Access)를 통하지 않고도 공동체의 지역성을 실현하는 효과적인 매체 활용을 할 수 있다. 셋째, 온라인 미디어의 콘텐츠는 공동체를 연결하며 개인이 연결점(node)이 되어 온라인과 오프라인에서 상호작용한다. 따라서 본 연구를 통해 온라인 기반 공동체 미디어 실천은 시민들의 미디어 활동의 지속 가능성을 높일 수 있는 틈새(니치) 미디어로서 기능할 수 있다는 것을 밝힐 수 있었다.
This study investigated the effects of magnetized water supplementation on blood glucose, DNA damage, antioxidant status, and lipid profiles in streptozotocin (STZ)-induced diabetic rats. There were three groups of 4-week-old male Sprague-Dawley rats used in the study: control group (normal control group without diabetes); diabetes group (STZ-induced diabetes control); and magnetized water group (magnetized water supplemented after the induction of diabetes using STZ). Before initiating the study, diabetes was confirmed by measuring fasting blood glucose (FBS > 200 dl), and the magnetized water group received magnetized water for 8 weeks instead of general water. After 8 weeks, rats were sacrificed to measure the fasting blood glucose, insulin concentration, glycated hemoglobin level, degree of DNA damage, antioxidant status, and lipid profiles. From the fourth week of magnetized water supplementation, blood glucose was decreased in the magnetized water group compared to the diabetes group, and such effect continued to the 8th week. The glycated hemoglobin content in the blood was increased in the diabetes group compared to the control group, but decreased significantly in the magnetized water group. However, decreased plasma insulin level due to induced diabetes was not increased by magnetized water supplementation. Increased blood and liver DNA damages in diabetes rats did significantly decrease after the administration of magnetized water. In addition, antioxidant enzyme activities and plasma lipid profiles were not different among the three groups. In conclusion, the supplementation of magnetized water not only decreased the blood glucose and glycated hemoglobin levels but also reduced blood and liver DNA damages in STZ-induced diabetic rats. From the above results, it is suggested that the long-term intake of the magnetized water over 8 weeks may be beneficial in both prevention and treatment of complications in diabetic patients.
High consumption of fruits and vegetables has been suggested to provide some protection to smokers who are exposed to an increased risk of numerous cancers and other degenerative diseases. Carrot is the most important source of dietary ${\beta}$-carotene. Therefore, the objective of this study was to investigate whether carrot juice supplementation to smokers can protect against lymphocyte DNA damage and to compare the effect of supplementationof capsules containing purified ${\beta}$-carotene or a placebo (simple lactose). The study was conducted in a randomized and placebo-controlled design. After a depletion period of 14 days, 48 smokers were supplemented with either carrot juice (n = 18), purified ${\beta}$-carotene (n = 16) or placebo (n = 14). Each group was supplemented for 8 weeks with approximately 20.49 mg of ${\beta}$-carotene/day and 1.2 mg of vitamin C/day, as carrot juice (300 ml/day) or purified ${\beta}$-carotene (20.49 mg of ${\beta}$-carotene, 1 capsule/day). Lymphocyte DNA damage was determined using the COMET assay under alkaline conditions and damage was quantified by measuring tail moment (TM), tail length (TL), and% DNA in the tail. Lymphocyte DNA damage was significantly decreased in the carrot juice group in all three measurements. The group that received purified ${\beta}$-carotene also showed a significant decrease in lymphocyte DNA damage in all three measurements. However, no significant changes in DNA damage was observed for the placebo group except TM (P = 0.016). Erythrocyte antioxidant enzyme was not significantly changed after supplementation. Similarly plasma lipid profiles were not different after carrot juice, ${\beta}$-carotene and placebo supplementation. These results suggest that while the placebo group failed to show any protective effect, carrot juice containing beta-carotene or purified ${\beta}$-carotene itself had great antioxidative potential in preventing damage to lymphocyte DNA in smokers.
셀레늄은 필수미네랄로 항산화작용 등의 체내 주요 작용을 수행한다고 알려져 있다. 만성질환 등의 예방과 치료 등의 바람직한 효능을 기대하여 섭취가 늘고 있어서 주목을 받고 있다. 셀레늄은 암의 억제나 혈중포도당조절 및 지방억제 등의 목적으로도 이용되고 있다. 이러한 다양한 연구에도 불구하고 지방세포를 이용한 지방세포분화나 지방세포사멸 촉진에 미치는 영향은 아직 연구되어 있지 않다. 셀레늄의 지방세포조절을 통한 분자기전에 대한 연구는 늘어가고 있는 비만이나 비만관련대사질환의 조절에 학문적 근거를 제공할 수 있다. 본 연구에서는 셀레늄이 AMP-activated protein kinase (AMPK)를 중심으로 지방 미분화세포인 3T3-L1 지방세포의 분화억제 및 분화된 세포의 사멸작용의 기전을 밝히고자 하였다. AMPK는 세포 내 에너지상태의 센서로 작용하는데 실제적으로 운동의 지방대사 촉진기능이 이 단백질에 기인한다는 사실이 밝혀지면서 각광을 받고 있다. 또한 시상하부에서 AMPK는 호르몬이나 영양소 등의 생리적인 인자들을 통합하고 식욕이나 신체에너지 대사를 총괄하는 물질로 대두되고 있다. 셀레늄을 지방세포의 분화과정에 처리하였을 때에 현저한 지방세포로의 분화가 억제되었고 이미 다 분화된 성숙된 지방세포에 처리하였을 때는 세포사멸(apoptosis)을 유도하는 것을 발견하였으며 이와 동시에 AMPK의 활성화 촉진 및 AMPK의 지질인 acetyl-CoA carboxylase의 $serine^{79}$를 인산화하는 것으로 나타났다. 또한 이러한 셀레늄의 지방세포분화 억제 능력은 AMPK의 활성화 합성물질인 AICAR나 포도의 폴리페놀인 레스베라트롤의 능력과 같은 정도의 억제능을 보여주었다. 또한 AMPK의 항 지방축적작용에서의 필수성을 알아보기 위하여 지방세포에 AMPK 활성화 물질인 AICAR와 억제물질인 Compound C를 사용하여 AMPK활성화를 측정하여 본 결과 지방세포분화와 지방축적의 억제과정에 있어서 AMPK가 필수적임을 밝혀내었다.
본 연구는 기업이 창업 후 죽음의 계곡을 극복하여 지속가능한 성장에 영향을 미치는 요인을 기술, 시장, 입지, 클러스터, INC모델 관점에서 접근하였으며, 대덕클러스터 내 기술기반 기업을 대상으로 성공·실패기업에 대한 사례연구를 진행하여 선행연구의 주요 요인과의 적정성 및 적합성을 탐색하였다. 본 연구 결과는 클러스터 내 기업의 성장을 위해서는 혁신 주체 간 입지 접근성 기반 협업이 중요하며, 기업의 제품·서비스가 시장에서 혁신 가치를 창출하기 위해서는 혁신적인 아이디어(Idea), 시장의 요구(Needs), 요구를 충족할 수 있는 역량(Capability) 등 균형적 성장이 필요함을 제시한다. 대덕클러스터 내 기업 대상 성공·실패 사례분석으로 지속 가능한 기업 성장을 위한 요인을 제시했다는 점에서 연구의 가치가 있다. 그리고 클러스터 내 기업육성을 위해 기업 간 협업 기반 정책적 지원 방안을 제시했다는 점에서 연구의 성과가 있다.
최근 당뇨환자에게서 산화적 손상이 증가됨이 보고되면서 항산화식품의 섭취를 통한 DNA 손상 개선노력이 다각도로 시도되고 있다. 차가버섯을 비롯한 여러 가지 버섯류에 항산화활성 및 혈당강하 효과가 있음이 알려지고 있으므로 본 연구에서는 streptozotocin (STZ) 유발 당뇨쥐를 사용하여 차가버섯에 항산화 효과 및 DNA 손상 억제효과가 있는지, 그리고 이런 항산화효과가 당뇨쥐의 혈당을 감소시키는지를 알아보았다. Sprague-dawley종 수컷 흰쥐(4주령, $110{\pm}6\;g$) 32마리를 대조군, 당뇨군, 저농도 차가버섯투여군 (0.5 g/kg BW) 및 고동도 차가버섯투여군 (5 g/kg BW) 등 4군으로 나누어 동물실험을 진행하였으며, 꼬리 정맥에 STZ을 주사하여 당뇨를 유발시킨 후 4주간 사육하였다. 4주 후에 혈액과 간을 채취하여 혈장 glucose, 혈장 총항산화능 (total radical-trapping antioxidant potential, TRAP) 수준 및 ${\alpha}$-tocopherol 수준, 적혈구 catalase, superoxide dismutase (SOD), 및 glutathione peroxidase (GSH-Px)의 활성도, 그리고 임파구 및 간조직의 DNA 손상 정도를 comet assay로 측정하였다. 실험 결과, 대조군에 비해 당뇨군에서 혈당이 상승하고 (p < 0.05) 혈장 TRAP 수준은 감소하였으며 (p < 0.05) 임파구의 DNA 손상은 증가하였고 (p < 0.05) 나머지 지표들은 차이가 나타나지 않았다. 당뇨군에 비해 저농도 및 고농도 차가버섯 투여군들에서 혈당이 감소하는 경향이 있었고 혈장 TRAP 수준과 ${\alpha}$-tocopherol 수준이 증가하는 경향을 보였으나 모두 유의적인 차이는 아니었으며, 당뇨군에 비해 차가버섯 투여군에서 tail length로 본 임파구 DNA 손상 정도가 유의적으로 감소하였다 (p < 0.05). 나머지 지표들은 당뇨군과 차가버섯 투여군들 간에 차이를 보이지 않았다. 본 연구결과, 차가버섯 열수추출물을 고농도 (5 g/kg BW)로 4주 동안 당뇨유발 흰쥐에게 투여하였을 때 혈당강하 효과는 유의적으로 나타나지 않았으나 당뇨로 인해 유발 된 DNA 손상정도가 정상수준까지 뚜렷하게 감소함을 관찰하였으며 이를 통해 부분적으로 차가버섯의 항산화 효과를 관찰할 수 있었다. 이와 같은 결과는 앞으로 차가버섯을 이용하여 당뇨환자의 DNA손상을 개선시키기 위한 건강기능식품 개발 연구에 활용될 수 있을 것으로 생각된다.

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
