Characteristics of a new Grifola frondosa Cultivar "Daebak" with stable pinheading and high yield (발이 안정 및 다수성 잎새버섯 신품종 '대박'의 특성)

  • 'Daebak', a new cultivar of Grifola frondosa, was bred by mating two monokaryotic strains isolated from 'F14309' and 'GMGF44062' at the Mushroom Research Institute, Gyonggi-Do ARES in 2017. The optimum temperature for mycelial growth of 'Daebak' was $25^{\circ}C$ on PDA medium. In bottle cultivation, the culture period of 'Daebak' was 57 days, which was 2 days shorter than that of 'Cham' (control). The pinheading rate of 'Daebak' was 98.4%, which was 24.8% higher than that of 'Cham' and its CV (Coefficient of variation) was 0.6%, 5.3% lower than that of 'Cham'. Regarding the growth characteristics of 'Daebak', the diameter and thickness of the pileus were 27.7 mm and 1.73 mm, respectively, and the diameter and height of the fruiting bodies cluster were 132 mm and 87.2 mm, respectively. The pileus was thinner but the fruiting bodies cluster was larger than that of 'Cham'. Fruiting bodies weighed 139 g per 1,100 ml bottle of 'Daebak', which was 28% higher than that for 'Cham', with a CV of 2.5%, which was 6.2% lower than that of 'Cham'. The yield per 10,000 bottles (used for cultivation) of 'Daebak' was 1,376 kg, 70% higher than that of 'Cham', with a CV of 3.0% that was 11.5% lower than that of 'Cham'. With respect to physical characteristics, the strength and brittleness of the fruiting body of 'Daebak' was less than that of 'Cham'. When considering the period available for sale, the shelf life of 'Daebak' was 42 days, which was 6 days longer than that of 'Cham'.

Identification of Domesticated Silkworm Varieties Using a Whole Genome Single Nucleotide Polymorphisms-based Decision Tree (전장유전체 SNP 기반 decision tree를 이용한 누에 품종 판별)

  • Silkworms, which have recently shown promise as functional health foods, show functional differences between varieties; therefore, the need for variety identification is emerging. In this study, we analyzed the whole silkworm genome to identify 10 unique silkworm varieties (Baekhwang, Baekok, Daebaek, Daebak, Daehwang, Goldensilk, Hansaeng, Joohwang, Kumkang, and Kumok) using single nucleotide polymorphisms (SNP) present in the genome as biomarkers. In addition, nine SNPs were selected to discriminate between varieties by selecting SNPs specific to each variety. We subsequently created a decision tree capable of cross-verifying each variety and classifying the varieties through sequential analysis. Restriction fragment length polymorphism (RFLP) was used for SNP867 and SNP9183 to differentiate between the varieties of Daehwang and Goldensilk and between Kumkang and Daebak, respectively. A tetra-primer amplification refractory (T-ARMS) mutation was used to analyze the remaining SNPs. As a result, we could isolate the same group or select an individual variety using the nine unique SNPs from SNP780 to SNP9183. Furthermore, nucleotide sequence analysis for the region confirmed that the alleles were identical. In conclusion, our results show that combining SNP analysis of the whole silkworm genome with the decision tree is of high value as a discriminative marker for classifying silkworm varieties.

Mycelial growth and fruit body cultural characteristics of a new Grifola frondosa variety, 'Bakyeon' (잎새버섯 신품종 '백연' 균사배양 및 자실체 생육 특성)

  • This study was conducted to reduce the phenomenon of the biased cultivation of certain mushroom varieties and to develop a competitive variety of Grifola frondosa. We developed the first Korean white commercial mushroom strain, 'Bakyeon', by crossing monokaryons derived from brown strains. We have collected and tested the characteristics of mushrooms from domestic and international genetic resources since 2018. We bred the unique domestic variety, 'Bakyeon', which has the following characteristics. The optimal temperature for mycelial growth was 25~28℃ and the optimal temperature for fruit body growth was 16~18℃. The new variety was similar to the control variety (Daebak) in terms of the pileus, which formed a pine cone shape, and the number of days of cultivation. The yield was 94.1 g/bottle, which was 23% lower than the 108.5 g/bottle yield of the control variety. When incubating the parent and control varieties, the replacement line was clear. Moreover, polymerase chain reaction analysis of mycelial DNA resulted in different band patterns between the parent and control varieties, confirming the hybrid species.
