오배자의 항산화 활성성분 및 자유라디칼 소거효과 (Constituents of Antioxidative Activity and Free Radical Scavenging Effect from Galla Rhois (Rhus javanica Linne))

  • This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.

Antioxidant Effect of Citri Reticulatae Pericarpium Extract on Oxidative Stress-Mediated Cytotoxicity In Cultures

  • In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

건조 및 추출 방법을 달리한 떡쑥(Gnaphalium affine D. DON)의 항산화 효과에 대한 연구 (Effect of Drying and Extraction Methods on Antioxidant Activity of Gnaphalium affine D. DON)

  • 본 연구는 떡쑥을 동결건조, 음지건조, 열풍건조 후 상온교반과 초음파를 각각 적용하며 물, 80% 메탄올, 80% 에탄올로 추출한 떡쑥 추출물의 항산화 효과를 각 건조 방법 및 추출 조건에 따라 비교하였다. 총 페놀 물질 함량에서는 동결건조 상온교반 80% 에탄올 추출물(13.87 mg QE/g)이 가장 많은 양을 나타냈다. 총 플라보노이드 함량에서는 음지건조 상온교반 80% 메탄올 추출물(12.78 mg QE/g)이 가장 높은 총 플라보노이드 함량을 나타내었다. DPPH radical 소거능에 있어서는 건조 방법 및 교반 조건에 따라서 DPPH radical 소거능에 유의적인 차이가 나타나지 않았으나 80% 에탄올과 80% 메탄올 추출물이 물 추출물보다 유의적으로 높은 소거능을 나타냈다. 환원력은 음지건조 초음파 80% 메탄올 추출물($225.5g{\mu}g$ TERP/g)에서 가장 높게 나타났으며 동결건조 초음파 물 추출물($22.2{\mu}g$ TERP/g)에서 가장 낮게 나타났다. 떡쑥의 환원력에서는 용매에 따라 80% 메탄올, 80% 에탄올, 증류수 순으로 유의적인 차이를 나타내었다. 떡쑥 추출물의 항산화 성분과 항산화능과의 상관관계에 있어서는 DPPH radical 소거능과 환원력에 유의적인 상관관계를 나타냈다. 결과적으로 떡쑥의 DPPH radical 소거 활성과 환원력은 플라보노이드를 포함한 페놀물질로부터 기인하며 떡쑥은 물보다 80% 에탄올과 80% 메탄올이 항산화 물질 추출에 더 효율적인 것으로 나타났다. 80% 에탄올 추출물의 모든 항산화 효과는 건조 및 교반 조건에 유의적인 차이를 보이지 않았으나 물 추출물의 DPPH radical 소거 활성과 80% 메탄올 추출물의 환원력은 열풍건조와 음지건조 처리 시 동결건조보다 높게 나타났다.

염생식물로부터 Peroxynitrite와 DPPH 라디칼 소거 활성 검색 (Screening of Peroxynitrite and DPPH Raoical Scavenging Activities from Salt Marsh Plants)

  • 본 연구에서는 항산화 활성을 갖는 유용한 선도 물질을 찾기 위한 일환으로 20종의 염생식물의 추출물에 대해 peroxynitrite와 DPPH 라디칼 소거 활성을 측정하였다. 그 결과 해당화 (Rosa rugosa), 개망초 (Erigeron annus), 냇씀바귀(Ixeris tamagawaensis), 띠 (Imperata cylindrica), 범행초(Tetragonia tetragonoides), 사철쑥 (Artemisia capillaris)의 MeOH 추출물에서 우수한 DPPH 라디칼 소거 효과가 확인되었다. 또한, 해당화(Rosa rugosa), 개망초 (Erigeron annus), 칠면초 (Suaeda japonica), 냇씀바귀 (Ixeris tamagawaensis), 띠 (Imperata cylindrica), 갯방풍 (Glehnia littoralis), 함초(Salicornia herbacea), 해국 (Aster spathulifolius), 범행초 (Tetragonia tetragonoides)의 MeOH 추출물과 CH$_2$Cl$_2$ 추출물에서 각각 peroxynitrite의 소거효과가 관찰되었다. 그 중에서도 해당화 MeOH 추출물의 DPPH 래디칼 소거효과는 100 $\mu\textrm{g}$/$m\ell$ (f.c.) 농도에서 87.51%로 나타났고, peroxynitrite 소거효과는 5 $\mu\textrm{g}$/$m\ell$ 농도에서 표품의 ONOO를 첨가했을 때 97.03%와 SIN-1을 첨가했을 때 98.48%로서 가장 강력한 항산화활성을 가지는 것으로 나타났다. 해당화의 peroxynitrite의 효과는 대조군으로 사용된 penicillamine보다도 우수하며, L-ascorbic acid와 비슷한 효과였다.

장미과 식물 줄기의 항산화 효과와 총 페놀류 함량에 관한 연구 (Study on the Antioxidant Effect and Total Phenolics Content in Rosaceae Plant Stem)

  • The antioxidant activities and total phenolics of four Rosaceae species Pourthiaea villosa (Thunb.) Decne, Sorbus commixta Hedlund, Sorbaria sorbifolia var. stellipila Maxim and Pyrus pyrifolia (Burm.f.) Nakai were determined. Phenolic content (polyphenol and flavonoid), radical scavenging activities [2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylnezthiazoline-6-sulfoic acid) (ABTS)] and ferrous ion chelating effect were evaluated. Total polyphenol and flavonoid contents were highest in Pourthiaea villosa (Thunb.) Decne and lowest in Pyrus pyrifolia (Burm.f.) Nakai. Phenolic contents of Pourthiaea villosa (Thunb.) Decne was $331.45{\pm}7.78$ and $90.4{\pm}3.5mg{\cdot}g^{-1}$. DPPH and ABTS radical scavenging activities were found to be lowest in Sorbus commixta Hedlund whereas Sorbaria sorbifolia var. stellipila Maxim and Pourthiaea villosa (Thunb.) Decne showed relatively good DPPH and ABTS radical scavenging activities. Ferrous ion chelating effect was highest in Pyrus pyrifolia (Burm.f.) Nakai ($1.05{\pm}0.04mg{\cdot}ml^{-1}$) and lowest in Sorbus commixta Hundlund ($4.22{\pm}0.71mg{\cdot}ml^{-1}$).

도꼬마리 추출물의 산화적 스트레스에 대한 융합연구 (Convergence study of oxidative stress from fraction of Xanthium strumarium L.)

  • 도꼬마리 (X. strumarium )는 국화과에 속하며, '창이자'라는 한약명으로 사용되는 다년생 식물이다. 열매부위와 지상부위의 추출물을 이용하여 융합 식물활성작용을 관찰하였다. 산화적 스트레스에 대항하는 실험법으로는 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH), ABTS assay를 사용하였다. 도꼬마리의 추출물은 단일분리가 아닌 ethanol, petroleum ether, 그리고 ethyl acetate로 추출하였다. 산화적 스트레스연구에 사용되는 radical scavenging 능력을 확인하기 위하여 DPPH radical and ABTS를 투여하였다. 본 연구에서 도꼬마리는 천연유래 항산화 효과를 보였다.

Antioxidant compounds from the stem bark of Sorbus commixta

  • The MeOH extract of Sorbus commixta (Rosaceae) exhibited strong DPPH radical scavenging activity, and through activity-guided fractionation two antioxidant compounds were isolated and identified as $catechin-7-O-{\beta}-D-xylopyranoside$ (1) and $catechin-7-O-\;{\beta}-D-apiofuranoside$ (2) by physicochemical and spectrometric methods. To evaluate the antioxidant effect of these compounds, some in vitro tests, such as the DPPH radical scavenging activity test, the superoxide radical scavenging activity test and the lipid peroxidation inhibitory activity test, were performed. Compounds 1 and 2 showed stronger activities than both a-tocopherol and butylated hydroxy anisole (BHA) in each assay.

마이크로네시아에 서식하는 해조류 Pylaiella littoralis 추출물의 항산화 효과 (Antioxidant Effect of Tropical Seaweed Pylaiella littoralis Extracts Collected from Chuuk Lagoon in Federated States of Micronesia)

  • Pylaiella littoralis was collected in the Chuuk lagoon of the Federated States of Micronesia (FSM). The FSM has a variety of coral reef ecosystems, which provide essential materials, such as minerals, vitamins, essential amino acids, for marine organisms. In this study, the antioxidant activities of ethanol and enzymatic extracts of P. littoralis were evaluated by measuring their scavenging activities on DPPH free radical, Alkyl radical, hydroxyl radical and cell viability. The enzymatic extracts were hydrolyzed to prepare water soluble extracts by using five carbohydrate degrading enzymes (AMG, Celluclast, Termamyl, Ultraflo, and Viscozyme) and five proteases (Alcalase, Flavourzyme, Kojizyme, Neutrase, and Protamex). As a result, the enzymatic extracts prepared by Flavourzyme, Ultraflo, and Kojizyme exhibited the greatest effects in DPPH free radical, alkyl radical scavenging activity and cell viability. Also, these enzymatic extracts had a higher antioxidant effect then commercial antioxidants in DPPH free radical and Alkyl radical scavenging activity. This study suggests that P. littoralis might be a useful source of natural antioxidants for the development of dietary supplements.

오공(蜈蚣)의 항산화효과에 관한 연구 (Antioxidative Effects of Scolopendra subspinipes)

  • Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.

Assai 열매 메탄올 추출물의 항산화 효과 (Antioxidant Effect of Assai Palm Methanolic Extract)

  • Assai 열매추출물의 자유 라디칼 소거능을 측정하기 위하여 DPPH를 이용한 자유 라디칼 소거능 실험을 수행하여 항산화 효과를 측정한 결과, assai열매추출물의 농도가 높을수록 DPPH활성이 뛰어남을 알 수 있었고, ROS를 이용하여 항산화 효과를 확인하였다. 배양된 대식세포에 assai열매추출물을 농도별로 첨가한 결과, 농도가 높을수록 과산화수소에 의해 유도된 산화적 자극이 감소하였다. 또한 세포 생존에 미치는 영향을 알고자 assai열매추출물을 농도별로 첨가하여 24시간후 세포의 형태변화를 MTT assay로 실시한 결과, 산화적 자극에 의해 발생한 세포 손상이 assai 열매추출물의 농도가 높아질수록 감소하는 것이 확인되었다. 따라서 assai열매추출물은 천연 항산화제로서의 가능성을 보였다.
