청갓과 청갓김치의 용매별 추출물의 항산화성 (Antioxidant Activity of Various Fractions Extracted from Mustard Leaf (Brassica juncea) and Their Kimchi)

  • 동결건조한 청갓과 담금당일 청갓김치, 15$^{\circ}C$에서 5일간 발효시킨 청갓 김치로부터 극성이 다른 용매들을 사용하여 각각의 용매별 추출물을 얻고 methanol(MeOH), hexane(C$_{6}$ $H_{14}$), dichlorometane(C $H_2$Cl$_2$), ethyl acetate(EtOAc), butanol(BuOH), water($H_2O$)의 각각의 용매분획별(DPPH) 1,1-diphenyl-2-picrylhydrazyl assay에 의한 항산화성을 측정하였고 항산화효과를 상호 비교하였다. 청갓과 담금당일 청갓 김치에서 EtOAc와 n-BuOH에서 얻은 분획층이 다른 분획층보다 항상화효과가 높게 나타났으며 15$^{\circ}C$ 5일간 발효시킨 청갓 김치에서는 C $H_2$Cl$_2$와 EtOAc분획층이 다른 분획층에 비하여 radical scavenger효과가 있음을 확인할 수 있었다. 결과의 차이는 다양한 재료에 의한 발효공정에 기인된다고 판단된다.

  • A phytochemical investigation on the methanolic extract of Cosmos caudatus has led to the isolation of quercetin $3-O-{\beta}-D-arabinofuranoside$ (1), quercetin $3-O-{\beta}-D-rhamnoside$ (2), quercetin $3-O-{\alpha]-D-glucoside$ (3) and quercetin (4). These compounds were shown to be the antioxidative constituents of the plant when evaluated using the ferric thiocyanate (FTC) and thiobarbituric acid (TBA), and radical scavengers based on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays.

해간전(解肝煎)의 항산화(抗酸化) 활성(活性) 및 간세포(肝細胞)의 산화적(酸化的) 손상(損傷)에 대한 보호효과(保護效果) (Antioxidative and Protective Effects of Haeganjeon Extract on Oxidative Damage of Hepatocytes)

  • Objectives: Haeganjeon(HGJ) has been used for the treatment of liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant and protective effects of HGJ extract on oxidative damage of hepatocytes by tert-butyl hydroperoxide(t-BHP). Methods: In the linoleic acid water-alcohol system, the levels of lipid peroxide(LPO) were determined by TBA method. The scavenging effect of HGJ on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical was determined according to the method of Hatano. In the Fenton system(ferrous ion reaction with hydrogen peroxide), the levels of hydroxyl radical induced LPO in rat liver homogenate were determined according to the method of TBA. Inhibitory effect of HGJ on superoxide generation was measured by xanthine-xanthine oxidase system. In order to evaluate antioxidative activity of HGJ in the liver cell, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without HGJ. After 18hr, cells placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydroperoxide(t-BHP) for 2hrs. Viable cells were detected by MTT assay. Conclusions: In the linoleic acid autoxidation system, HGJ extract significantly inhibited the time course of the lipid peroxidation. These effects were similar to those of BHA HGJ extracts showed about 70% scavenging effect on DPPH radical. And HGJ extract inhibited the lipid peroxide formation in rat liver homogenate induced by hydroxyl radical derived from Fenton system. In addition, HGJ extract protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. These result indicated that HGJ extract might playa protective role against oxidative hepatic cell injury by means of free radical scavenger.

추출조건이 해동피 열수 추출물의 항산화 효과에 미치는 영향 (Antioxidant Activity of Water-soluble Extracts from Kalopanacis cortex)

  • 해동피 항산화 물질을 추출하기 위한 열수 추출조건을 냉장처리 여부, 추출온도 및 추출시간을 달리하여 검토하였다. 이러한 조건을 달리하여 추출된 추출물의 총 폴리페놀 화합물의 함량, DPPH radical 소거능 및 HRSA를 측정하였다. 그 결과 총 폴리페놀 함량은 냉장처리 후 추출온도 $95^{\circ}C$, 추출 15시간에서 $612{\mu}g/mL$로써 가장 높았다. DPPH radical 소거능은 냉장처리 후 추출온도 $95^{\circ}C$, 추출 15시간에서 가장 높은 78.8%이었고 대조구인 ascorbic acid의 70.2%보다 높게 나타났다. HRSA는 냉장처리 후 추출온도 $95^{\circ}C$, 추출 15시간에서 ascorbic acid 56.6%보다 높은 69.0%로 가장 높게 나타났다. 따라서 이상의 열수추출에 의한 해동피의 항산화성 활성 및 그 최적 조건을 확립한 것으로 향후 해동피의 기능성식품으로서의 이용개발을 위한 중요한 자료로 이용될 수 있음을 보여 주고 있다.

  • Phellinus linteus has been used as anti-tumor and immuno stimulating agents in folk remedies. From precipitate of MeOH ex. by activited guided fractionation. 5.8-epidioxy ergosta-6.22-dien-3ol. palmitic acid. linoleic acid, and methyl linolate. 3.4-dihydroxybenzoic acid methylester and 4-(3'4'-Dihydroxyphenyl)-3-butene-2one were isolated. DPPH method was used to examine of antioxidative activity of the isolated compounds. As the result, 3.4-dihydroxybenzoic acid methylester, and phenolic compound. 4-(3'4'-Dihydroxyphenyl) -3-butene-2one were found to be a scavenger of 1,1-diphenyl-2-picrylhydrzyl radical. (omitted)

피막이풀의 DPPH 라디칼과 hydroxyl radicals (OH) 항산화 활성 및 리폭시게나아제 저해 효과 (Inhibitory Effect of DPPH Radical Scavenging Activity and Hydroxyl Radicals (OH) Activity of Hydrocotyle sibthorpioides Lamarck)

  • 피막이(Hydrocotyle sibthorpioides Lam.)는 산형과(Araliaceae)의 여러해살이 초본이다. 생약명은 천호유, 예초, 계장초, 변지금으로 한방에서 사용하고 있다. 이 식물의 잎과 줄기로부터 열수 추출물을 이용하여 항산화, 환원력, 리폭시게나아제(lipoxygenase) 저해활성을 조사하였다. 하이드록시 라디칼(-OH)은 거대분자, 탄수화물, 핵산, 지질, 단백질에 유해한 작용을 나타내는데 피막이의 잎 추출물은 8.0 mg/ml 농도일 때 78.6%의 저해활성을 나타내었다. 1, 1- diphenyl 2-picrylhyorazyl (DPPH)은 인체 내 항산화에 잘 알려진 라디칼로 피막이풀의 하이드록시 라디칼 활성 저해는 8.0 mg/ml농도일 때 86.0%였다. 다중불포화지방산 대사 효소에 관여하는 Lipoxygenases (LOXs)의 활성 저해는 8.0 mg/ml농도일 때 55.5%였다. 50%저해를 나타내는 농도(IC50)를 산출할 때 OH 활성, DPPH 활성, LOX 저해에 대해 각각 5.23 mg/ml, 6.44 mg/ml, 3.71 mg/ml이였다. 피막이 잎과 줄기 추출물은 강한 환원력을 나타내었다. 이런 피막이의 항산화 및 환원력 활성, LOX의 높은 저해율, 강환 환원력은 약리학적 소재로 좋은 후보가 될 수 있을 것이다.

산벚나무 가지 추출물 및 용매 분획물의 Nitric Oxide 생성 억제 효과와 여드름 원인균에 대한 항균활성 (Antibacterial and Nitric Oxide Production Inhibitory Activities of Prunus sargentii Branches Extract and Its Fractions against Pathogens of Acne)

  • Background: In this study, we investigated the antibacterial and nitric oxide (NO) production inhibitory activities of 75% ethanol extract of Prunus sargentii branches and its fractions against acne pathogens. Methods and Results: The antibacterial activity against acne causing pathogens was determined using the disc diffusion assay. The ethyl acetate fraction showed higher activities against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. In the DPPH radical and NO scavenging assays, the butanol fraction showed strong DPPH radical and NO scavenging abilities. These activities were related to the total polyphenol and flavonoid contents of butanol fraction. On the other hand, the chloroform and ethyl acetate fractions exhibited the highest NO production inhibitory activity in Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells compared to those exhibited by other fractions. Conclusions: The extract and its ethyl acetate fraction from the branches of P. sargentii exhibited antibacterial activity and could be used as functional materials in antimicrobial related fields. Moreover, the chloroform and ethyl acetate fractions are potential antiinflammatory agents and butanol fraction acts as an effective radical scavenger.

Anti-inflammatory and Radical Scavenging Effects of Spirodela polyrrhiza

  • Anti-inflammatory and anti-oxidant effect of Spirodela polyrrhiza (Lemnaceae), a widely used traditional medicinal plant were investigated. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. 85% MeOH extracts of S. polyrrhiza (0.01, 0.1, 1 mg/mL) suppressed nitric oxide production in interferone-$\gamma$ (IFN-$\gamma$) and lipopoloysaccharide(LPS)-stimulated macrophages. It also attenuated the expression of inflammatory enzymes like inducible NOS (iNOS) and cyclooxygenase-2(COX-2) as assessed by immunoblotting with specific antibodies. Moreover, the values obtained with DPPH radical, superoxide anion and NO radical scavenging assay showed that S. polyrrhiza has potent antioxidant properties as a natural ROS scavenger. The results of the present study suggest the potential use of S. polyrrhiza in the treatment of ROS-mediated chronic inflammatory diseases such as atherosclerosis and rheumatoid arthritis.

후코이단의 분자량이 화장품기능성에 미치는 영향 (The Effect of Fcoidan Molecula Weight on Cosmetic Function)

  • 기능성화장품 소재로 사용하기 위한 후코이단의 특성 및 후코이단 분자량의 영향에 대해 연구하였다. 후코이단은 미역 포자엽에서 추출하였고 접촉글로우방전 전기분해(CGDE)방법에 의해 후코이단 분자량(35~160 kDa)을 조절하였다. 후코이단의 tyrosinase inhibition, 보습력, elastase 활성저해 효과와 항산화력(DPPH assay, radical scavenging)을 측정하여 화장품으로서 가능성을 확인하였다. 후코이단의 보습력은 최고의 보습제 중의 하나인 히아루론산의 보습력보다 높았고, 분자량이 감소함에 따라 보습력이 약간 증가하였다. 주름개선 효과실험인 elastase 활성저해 효과 실험결과 표준 비교물질인 adenosine 보다 높은 elastase 활성저해 효과를 보였다. 최고의 tyrosinase 저해효과, elastase 활성저해 효과, 항산화력(DPPH assay, radical scavenging)을 갖는 후코이단의 분자량은 100 kDa이었다.

Antioxidant Activities of the Ethanol Extract of Hamcho (Salicornia herbacea L.) Cake Prepared by Enzymatic Treatment

  • The antioxidant activities of water ($H_2O$) and ethanol (EtOH) extracts from hamcho (Salicornia herbacea L.) juice and cake prepared by enzymatic treatments were evaluated by in vitro assays against DPPH, superoxide, and hydroxyl radicals. Among the $H_2O$ and EtOH extracts from five different carbohydrases treated, the EtOH extract from viscozyme-treated hamcho cake had higher yield and phenolic content, and exhibited the strongest radical scavenging activity against DPPH ($IC_{50}=186.91\;{\mu}g/mL$), superoxide ($IC_{50}=87.54\;{\mu}g/mL$), and hydroxyl radicals ($IC_{50}=367.07\;{\mu}g/mL$). Antioxidant assay-guided fractionation and purification of the EtOH extract led to isolation and identification of five phenolic compounds, procatechuic, ferulic and caffeic acids, quercetin, and isorhamnetin. Most of these phenolic compounds exhibited considerable DPPH, superoxide, and hydroxyl radical scavenging activities, and in particular, caffeic and ferulic acids had stronger superoxide and hydroxyl radical scavenging activities than the well-known antioxidant radical scavenger, (+)-catechin (p<0.05). Quercetin and isorhamnetin were the primary compounds responsible for the strong antioxidant activity in the EtOH extract of the viscozyme-treated hamcho cake. Meanwhile, these five phenolic compounds were detected in the EtOH extract of the viscozyme-treated hamcho cake at the following levels (dry base of hamcho); procatechuic acid (1.54 mg%), caffeic acid (6.87 mg%), ferulic acid (8.45 mg%), quercetin (12.63 mg%), and isorhamnetin (6.65 mg%). However, three of these phenolic compounds (procatechuic, caffeic acid, and ferulic acids) were detectable in the $H_2O$ extract of viscozyme-treated hamcho juice. These results suggest that the EtOH extract of viscozyme-treated hamcho cake may be a potential source of natural antioxidants.
